Choppi: A Web Tool For The Analysis Of Immunogenicity Risk From Host Cell Proteins In Cho-Based Protein Production, Chris Bailey-Kellogg, Andres H. Gutiérrez, Leonard Moise, Frances Terry, William D. Martin, Anne S. De Groot

Institute for Immunology and Informatics Faculty Publications

Despite high quality standards and continual process improvements in manufacturing, host cell protein (HCP) process impurities remain a substantial risk for biological products. Even at low levels, residual HCPs can induce a detrimental immune response compromising the safety and efficacy of a biologic. Consequently, advanced-stage clinical trials have been cancelled due to the identification of antibodies against HCPs. To enable earlier and rapid assessment of the risks in Chinese Hamster Ovary (CHO)-based protein production of residual CHO protein impurities (CHOPs), we have developed a web tool called CHOPPI, for CHO Protein Predicted Immunogenicity. CHOPPI integrates information regarding the possible presence …

Effect Of Temperature On Feeding Period Of Larval Blacklegged Ticks (Acari: Ixodidae) On Eastern Fence Lizards, Eric L. Rulison, Roger A. Lebrun, Howard S. Ginsberg Jan 2014

Effect Of Temperature On Feeding Period Of Larval Blacklegged Ticks (Acari: Ixodidae) On Eastern Fence Lizards, Eric L. Rulison, Roger A. Lebrun, Howard S. Ginsberg

Plant Sciences and Entomology Faculty Publications

Ambient temperature can influence tick development time, and can potentially affect tick interactions with pathogens and with vertebrate hosts. We studied the effect of ambient temperature on duration of attachment of larval blacklegged ticks, Ixodes scapularis Say, to eastern fence lizards, Sceloporus undulatus (Bosc & Daudin). Feeding periods of larvae that attached to lizards under preferred temperature conditions for the lizards (WARM treatment: temperatures averaged 36.6°C at the top of the cage and 25.8°C at the bottom, allowing behavioral thermoregulation) were shorter than for larvae on lizards held under cool conditions (COOL treatment temperatures averaged 28.4°C at top of cage …

Relative Humidity And Activity Patterns Of Ixodes Scapularis (Acari: Ixodidae), K. A. Berger, Howard S. Ginsberg, L. Gonzalez, Thomas N. Mather

Plant Sciences and Entomology Faculty Publications

Laboratory studies have shown clear relationships between relative humidity (RH) and the activity and survival of Ixodes scapularis Say (blacklegged tick). However, field studies have produced conflicting results. We examined this relationship using weekly tick count totals and hourly RH observations at three field sites, stratified by latitude, within the state of Rhode Island. Records of nymphal tick abundance were compared with several RH-related variables (e.g., RH at time of sampling and mean weekly daytime RH). In total, 825 nymphs were sampled in 2009, a year of greater precipitation, with a weighted average leaf litter RH recorded at time of …

Unique Cytokine Signature In The Plasma Of Patients With Fibromyalgia, Jamie Sturgill, Elizabeth Mcgee, Victoria Menzies Jan 2014

Unique Cytokine Signature In The Plasma Of Patients With Fibromyalgia, Jamie Sturgill, Elizabeth Mcgee, Victoria Menzies

School of Nursing Publications

Fibromyalgia (FMS) is a chronic pain syndrome with a complex but poorly understood pathogenesis affecting approximately 10 million adults in the United States. The lack of a clear etiology of FMS has limited the effective diagnosis and treatment of this debilitating condition. The objective of this secondary data analysis was to examine plasma cytokine levels in women with FMS using the Bio-Plex Human Cytokine 17-plex Assay. Post hoc analysis of plasma cytokine levels was performed to evaluate patterns that were not specified a priori. Upon examination, patients with FMS exhibited a marked reduction in cytokines such as IL-4, IL-5, and …

Guided Imagery For Stress And Symptom Management In Pregnant African American Women, Nancy Jallo, R. Jeanne Ruiz, R. K. Elswick Jr., Elise French

School of Nursing Publications

The purpose of this study was to evaluate the efficacy of a guided imagery (GI) intervention for stress reduction in pregnant African American women beginning early in the second trimester. This prospective longitudinal study of 72 women used a randomized controlled experimental design with two groups conducted over 12 weeks. The intervention was a CD with 4 professionally recorded tracts designed and sequenced to influence study variables. Participants in both GI and usual care (UC) completed measures and donated 5 cc of blood at baseline, 8 weeks and 12 weeks. Participants also completed a daily stress scale. A mixed-effects linear …

Adaptation Of An Evidence-Based Intervention To Promote Colorectal Cancer Screening: A Quasi-Experimental Study, Shin-Ping Tu, Alan Chun, Yutaka Yasui, Alan Kuniyuki, Mei-Po Yip, Vicky Taylor, Roshan Bastani

Internal Medicine Publications

Background To accelerate the translation of research findings into practice for underserved populations, we investigated the adaptation of an evidence-based intervention (EBI), designed to increase colorectal cancer (CRC) screening in one limited English-proficient (LEP) population (Chinese), for another LEP group (Vietnamese) with overlapping cultural and health beliefs.

Methods Guided by Diffusion of Innovations Theory, we adapted the EBI to achieve greater reach. Core elements of the adapted intervention included: small media (a DVD and pamphlet) translated into Vietnamese from Chinese; medical assistants distributing the small media instead of a health educator; and presentations on CRC screening to the medical assistants. …

Successful curative treatment of severe pulmonary arterial hypertension with luminal obliteration will require a thorough understanding of the mechanism underlying the development and progression of pulmonary vascular lesions. But the cells that obliterate the pulmonary arterial lumen in severe pulmonary arterial hypertension are incompletely characterized. The goal of our study was to evaluate whether inhibition of CXC chemokine receptor 4 will prevent the accumulation of c-kit+ cells and severe pulmonary arterial hypertension. We detected c-kit+ cells expressing endothelial (von Willebrand Factor) or smooth muscle cell/myofibroblast (α-smooth muscle actin) markers in pulmonary arterial lesions of SU5416/chronic hypoxia rats. We found increased …

CFTR is a dynamically regulated anion channel. Intracellular WNK1-SPAK activation causes CFTR to change permeability and conductance characteristics from a chloride-preferring to bicarbonate-preferring channel through unknown mechanisms. Two severe CFTR mutations (CFTRsev) cause complete loss of CFTR function and result in cystic fibrosis (CF), a severe genetic disorder affecting sweat glands, nasal sinuses, lungs, pancreas, liver, intestines, and male reproductive system. We hypothesize that those CFTR mutations that disrupt the WNK1-SPAK activation mechanisms cause a selective, bicarbonate defect in channel function (CFTRBD) affecting organs that utilize CFTR for bicarbonate secretion (e.g. the pancreas, nasal sinus, vas …

Oral Supplementation With Non-Absorbable Antibiotics Or Curcumin Attenuates Western Diet-Induced Atherosclerosis And Glucose Intolerance In Ldlr−/− Mice – Role Of Intestinal Permeability And Macrophage Activation, Siddhartha S. Ghosh, Jinghua Bie, Jing Wang, Shobha Ghosh

Internal Medicine Publications

Association between circulating lipopolysaccharide (LPS) and metabolic diseases (such as Type 2 Diabetes and atherosclerosis) has shifted the focus from Western diet-induced changes in gut microbiota per se to release of gut bacteria-derived products into circulation as the possible mechanism for the chronic inflammatory state underlying the development of these diseases. Under physiological conditions, an intact intestinal barrier prevents this release of LPS underscoring the importance of examining and modulating the direct effects of Western diet on intestinal barrier function. In the present study we evaluated two strategies, namely selective gut decontamination and supplementation with oral curcumin, to modulate Western-diet …

Impact Of Teacher Attitude And Urbanicity Through The Implementation Of Positive Behavior Support Programming, Theodore Caleris Jan 2014

Impact Of Teacher Attitude And Urbanicity Through The Implementation Of Positive Behavior Support Programming, Theodore Caleris

One of the major questions that both urban and non-urban public school districts are faced with is the relationship between the predictors of school climate, student behavior, staff perceptions and the influence these variables have on school climate. School districts are under public scrutiny to produce positive achievement results for all students. Much research suggests that schools that are considered urban are generally considered unsafe, unproductive and unable to produce students that are competitive in a 21st century global economy. In the major urban centers, student populations are decreasing, flooding the first-ring school districts. First-ring school districts are the individual …
