Older patients with acute myeloid leukemia (AML) and myelodysplastic syndrome have often been excluded from myeloablative-conditioning regimens containing busulfan because of non-disease-related morbidity and mortality. We hypothesized that busulfan clearance (BuCL) in older patients (> 60 years) would be reduced compared to that in younger patients, potentially explaining observed differences in busulfan tolerability. AML patients in three CALGB hematopoietic cell transplantation studies were treated with a conditioning regimen using IV busulfan, dosed at 0.8 mg/kg. Plasma busulfan concentrations were determined by LC-MS and analyzed by non-compartmental methods. BuCL was normalized to actual (ABW), ideal (IBW), or corrected (CBW) body weight …

Objective. To compare the efficacy and safety of intravenous (IV) abatacept, a selective T cell costimulation modulator, versus placebo for the treatment of active class III or IV lupus nephritis, when used on a background of mycophenolate mofetil and glucocorticoids. Methods. This was a 12-month, randomized, phase II/III, multicenter, international, double-blind study. A total of 298 patients were treated in 1 of 3 IV treatment arms: placebo, abatacept at the standard weight-tiered dose (approximating 10 mg/kg), or abatacept at 30 mg/kg for 3 months, followed by the standard weight-tiered dose (abatacept 30/10). The primary end point, time to confirmed complete …

Purpose Molecular risk stratification of acute myeloid leukemia (AML) is largely based on genetic markers. However, epigenetic changes, including DNA methylation, deregulate gene expression and may also have prognostic impact. We evaluated the clinical relevance of integrating DNA methylation and genetic information in AML. Methods Next-generation sequencing analysis of methylated DNA identified differentially methylated regions (DMRs) associated with prognostic mutations in older ( 60 years) cytogenetically normal (CN) patients with AML (n = 134). Genes with promoter DMRs and expression levels significantly associated with outcome were used to compute a prognostic gene expression weighted summary score that was tested and …

The objective of this study was to study genetic factors that influence quantitative anticyclic citrullinated peptide (anti-CCP) antibody levels in RA patients. We carried out a genome-wide association study (GWAS) meta-analysis using 1975 anti-CCP + RA patients from three large cohorts, the Brigham Rheumatoid Arthritis Sequential Study (BRASS), North American Rheumatoid Arthritis Consortium (NARAC) and the Epidemiological Investigation of RA (EIRA). We also carried out a genome-wide complex trait analysis (GCTA) to estimate the heritability of anti-CCP levels. GWAS-meta-analysis showed that anti-CCP levels were most strongly associated with the human leukocyte antigen (HLA) region with a P-value of 2 x …

Purpose Lenalidomide is an immunomodulatory agent with therapeutic activity in chronic lymphocytic leukemia (CLL). In preclinical models, lenalidomide acted synergistically with rituximab. The CLL Research Consortium initiated a phase II study to evaluate this combination in treatmentnaive patients. Patients and Methods Lenalidomide was initiated at 2.5 mg/day and was escalated based on treatment tolerability to a maximum of 10 mg/day, for 21 days/cycle, for a maximum of seven cycles. Rituximab was administered at the end of cycle 1 and was continued for seven cycles. Patients received allopurinol and aspirin for prophylaxis. Results Sixty-nine patients enrolled onto one of two age-specific …

A Five-Step Process For Interactive Parent–Adolescent Communication About Hiv Prevention: Advice From Parents Living With Hiv/Aids, Laura L. Edwards, Janet S. Reis

University Author Recognition Bibliography: 2014

The authors investigated how parents living with HIV/AIDS communicate about HIV prevention with their 10- to 18-year-old children. Semistructured interviews with 76 mothers and fathers were analyzed for (a) their experiences discussing HIV prevention with adolescents and (b) advice for other parents about how to best broach HIV-related topics. Interactive conversations were regarded as particularly effective. A five-step process for interactive communication emerged as a result of these discussions. Parents emphasized that adolescents should have a “voice” and a “choice” in HIV-related talks. Health care professionals can facilitate adolescent sexual health by encouraging parents to actively involve their children in …

Winter Wheat Quality Responses To Water, Environment, And Nitrogen Fertilization, Hyejin Park, David E. Clay, R. G. Hall, Tulsi P. Kharel, Sharon A. Clay, Sanghun Lee

Agronomy, Horticulture and Plant Science Faculty Publications

Decreasing carbon (C) footprints by reducing nitrogen (N) and water inputs has been speculated to have negative impacts on wheat grain yield and flour processing quality. The objective of this study was to determine the impact of N and water stress on winter wheat grain yield, protein composition, and dough quality. Wheat fertilized at two N rates (unfertilized and recommended) was grown under water-stressed and well-watered environments. Nitrogen and water stress were measured using the 13C isotopic approach. Research showed that (1) N fertilizer and the water-management environment produced similar impacts on wheat quality and yield loss due to N …

Identification Of Differential Agronomic Traits In Early Stage Teosinte, Flint, Dent, And Sugar (Sweet) Corn Varieties In Competition With Weeds, S.A. Hansen, S. A. Clay, David Horvath, S. Flint-Garcia Jan 2014

Identification Of Differential Agronomic Traits In Early Stage Teosinte, Flint, Dent, And Sugar (Sweet) Corn Varieties In Competition With Weeds, S.A. Hansen, S. A. Clay, David Horvath, S. Flint-Garcia

Agronomy, Horticulture and Plant Science Faculty Publications

Weed competition reduces corn yield. Today’s corn monoculture relies heavily on herbicide inputs to maintain yield. However, teosinte, corn’s ancestor, was successfully grown in mixed production systems. Harnessing genes and traits that allow teosinte to be a better competitor but which may have been lost during corn domestication could help producers reduce herbicide inputs and maintain yield. The first step to finding those genes is to identify varieties of corn and lines of teosinte that have a higher tolerance of weeds or greater weed suppressive ability. Five introductions of teosinte and 14 varieties of corn (including dent, heritage, and sweet …
