Thirteen mono-N-acyl derivatives of 2,6-diaminopimelic acid (DAP)—new potential inhibitors of the dapE-encoded N-succinyl-l,l-diaminopimelic acid desuccinylase (DapE; EC 3.5.1.18)—were analyzed and characterized by infrared (IR) and nuclear magnetic resonance (NMR) spectroscopies and two capillary electromigration methods: capillary zone electrophoresis (CZE) and micellar electrokinetic chromatography (MEKC). Structural features of DAP derivatives were characterized by IR and NMR spectroscopies, whereas CZE and MEKC were applied to evaluate their purity and to investigate their electromigration properties. Effective electrophoretic mobilities of these compounds were determined by CZE in acidic and alkaline background electrolytes (BGEs) and by MEKC in acidic and alkaline BGEs …

Characterizing The Fitness Landscapes Of Gut Symbionts In Defined Community And Diet Contexts, Meng Wu Dec 2014

Characterizing The Fitness Landscapes Of Gut Symbionts In Defined Community And Diet Contexts, Meng Wu

Arts & Sciences Graduate Student Theses and Dissertations

A species' niche is the description of all the environmental conditions required to permit a population of that species to persist, including the effects of the population on those conditions. This definition includes the species' resource requirements, as well as stress tolerances and interactions with other species acting as competitors, predators, parasites, and mutualists. The human gut microbiota serves as a microbial `metabolic organ' tasked in part with the biotransformation of many components of our diet. Relatively little is known about the factors that allow members of the human gut microbiota to persist in a habitat that experiences marked changes …

Hyperfiltration subjects podocytes to increased tensile stress and fluid flow shear stress (FFSS). We showed a 1.5- to 2.0-fold increase in FFSS in uninephrectomized animals and altered podocyte actin cytoskeleton and increased synthesis of prostaglandin E2 (PGE2) following in vitro application of FFSS. We hypothesized that increased FFSS mediates cellular changes through specific receptors of PGE2. Presently, we studied the effect of FFSS on cultured podocytes and decapsulated isolated glomeruli in vitro, and on solitary kidney in uninephrectomized sv129 mice. In cultured podocytes, FFSS resulted in increased gene and protein expression of cyclooxygenase (COX)-2 but not COX-1, prostanoid receptor EP2 …

Essays On Battle Clusters In Internal Armed Conflicts And Insurgencies: Concept, Causes And Consequences, Chifeng Liu

Theses and Dissertations

With the purpose to re-conceptualize the intensity of internal armed conflicts, I argue that the characteristics such as location, scale and duration of “major battlefields,” i.e. the spatio-temporal clusters of combat events are important in the evaluation of impact of modern civil wars and insurgencies. I start with elaborating and constructing a new concept of conflict magnitude with battle clusters using up-to-date geo-referenced data and spatial statistic methods. In the second article, I first explain the location and duration of major battlefields. The findings indicate that an area with multiple non-state armed groups (NSAGs) is more likely to become main …

Birds are the most species-rich class of tetrapod vertebrates and have wide relevance across many research fields. We explored bird macroevolution using full genomes from 48 avian species representing all major extant clades. The avian genome is principally characterized by its constrained size, which predominantly arose because of lineage-specific erosion of repetitive elements, large segmental deletions, and gene loss. Avian genomes furthermore show a remarkably high degree of evolutionary stasis at the levels of nucleotide sequence, gene synteny, and chromosomal structure. Despite this pattern of conservation, we detected many non-neutral evolutionary changes in protein-coding genes and noncoding regions. These analyses …

Implementation Of A Helicobacter Pylori Test And Treat Program In Rural Nepal Facilitated By A Nurse Practitioner Doctor Of Nursing Practice Comprehensive Project Report, Shelley Bloom

Doctor of Nursing Practice (DNP) Projects

There are 30 million people in Nepal, 85% live in rural areas and have access to 15% of health care resources. This deficit in health care resources is supplemented by services provided by non-governmental organizations who organize short-term medical missions. At a rural Nepalese health post 45% of the patients complained of abdominal discomfort. As the prevalence of helicobacter pylori is 50-90% in Southeast Asia, the patients believed that they had helicobacter pylori.

In resource-poor communities, there is a gap between best practices and the care delivered. To decrease the gap in one rural Nepalese health post, a Nurse Practitioner …

Biofilm Microenvironment Analysis Systems For Medicine And Agriculture, Jinzi Deng Dec 2014

Biofilm Microenvironment Analysis Systems For Medicine And Agriculture, Jinzi Deng

Doctoral Dissertations

Biofilms are aggregated bacteria embedded in extracellular polymeric substances (EPS). Biofilms are the dominant growth form of bacteria in most natural environments, and are also important in many industrial and in vivo settings. The function of a biofilm system is inherently different from the function of the same cells dispersed in liquid culture. Existing methods to study biofilm either do not maintain the essential biofilm architecture or do not capture the dynamic nature of their responses. This research describes the development of a set of biofilm microenvironment analysis systems that maintain the essential micro-scale structure of biofilms. These systems are …

How Disgust Affects Romantic Attraction: How Our Moods Affect Our Judgments Of Attractiveness, Nicholas Phelan Dec 2014

How Disgust Affects Romantic Attraction: How Our Moods Affect Our Judgments Of Attractiveness, Nicholas Phelan

Theses

Facial attractiveness is an important part of our initial judgments of people and in our ability to determine their compatibility as a mating partner. Just as mating is evolutionarily important, the emotion disgust is one of the most evolutionarily beneficial emotions as it helps keep us away from poisonous and possibly infectious substances or foods. We instinctively avoid people with facial scars and other facial deformities because these are seen as a sign of illness or generally poor well-being. Facial attraction is one component of how we choose a mate. The current experiment examined whether or not being disgusted created …

Background

Bladder cancer is a significant health problem in rural areas of Africa and the Middle East where Schistosoma haematobium is prevalent, supporting an association between malignant transformation and infection by this blood fluke. Nevertheless, the molecular mechanisms linking these events are poorly understood. Bladder cancers in infected populations are generally diagnosed at a late stage since there is a lack of non-invasive diagnostic tools, hence enforcing the need for early carcinogenesis markers.

Methodology/Principal Findings

Forty-three formalin-fixed paraffin-embedded bladder biopsies of S. haematobium-infected patients, consisting of bladder tumours, tumour adjacent mucosa and pre-malignant/malignant urothelial lesions, were screened for bladder …

Resistance to antibiotics is on the rise at an alarming pace. The problem is exacerbated by the lack of new class of therapeutic agents and targets. Modes of resistance include alteration of drug target, modification of ligand, and reduction of the intracellular concentration of drug. Dihydrofolate reductase (DHFR) is a validated drug target for the treatment of infectious diseases and cancer. The binding of ligands to DHFR inhibits the downstream folate pathway that is vital for purine and thymidylate synthesis. Trimethoprim (TMP), a clinically used antifolate exhibits resistance in both gram-positive and gram-negative bacteria. In this thesis, methods have been …

White-Nose Syndrome Initiates A Cascade Of Physiologic Disturbances In The Hibernating Bat Host, Michelle L. Verant, Carol U. Meteyer, John R. Speakman

KIP Articles

The physiological effects of white-nose syndrome (WNS) in hibernating bats and ultimate causes of mortality from infection with Pseudogymnoascus (formerly Geomyces) destructans are not fully understood. Increased frequency of arousal from torpor described among hibernating bats with late-stage WNS is thought to accelerate depletion of fat reserves, but the physiological mechanisms that lead to these alterations in hibernation behavior have not been elucidated. We used the doubly labeled water (DLW) method and clinical chemistry to evaluate energy use, body composition changes, and blood chemistry perturbations in hibernating little brown bats (Myotis lucifugus) experimentally infected with P. destructans to better understand …
