A unique avian-origin A/H7N9 influenza virus has so far caused 134 cases with 44 deaths. Probing the host factors contributing to disease severity, we found that lower levels of plasma inflammatory cytokines on hospital admission correlated with faster recovery in 18 patients with A/H7N9 influenza virus, whereas high concentrations of (in particular) IL-6, IL-8, and macrophage inflammatory protein-1β were predictive of a less favorable or fatal outcome. Analysis of bronchoalveolar lavage samples showed up to 1,000-fold greater cytokine/chemokine levels relative to plasma. Furthermore, patients with the rs12252-C/C IFN-induced transmembrane protein-3 (IFITM3) genotype had more rapid disease progression and were less …

International Journal of Exercise Science: Conference Proceedings

Introduction: Catabolism of muscle and loss of function are complications that can occur during the course of HIV infection, and are commonly seen in a majority of adolescents with vertically transmitted Human Immunodeficiency Virus-type 1 (HIV-1). The levels of CD4+ and CD8+ lymphocytes, reference markers for the treatment of vertically transmitted HIV-1, also decline as the disease progresses. Alterations on these reference markers may be associated with neuromuscular force parameters in sedentary adolescents that have potential as prognostic indicators for treatment administration. Objective: To investigate the relationship between maximal isometric muscular contraction force and levels of CD4+ and CD8+ lymphocytes …

Infections And Immune Disturbances: Clinical-Based Outcomes, Stephane Bermon Jan 2014

Infections And Immune Disturbances: Clinical-Based Outcomes, Stephane Bermon

International Journal of Exercise Science: Conference Proceedings

Whatever their level or type of sport practice, athletes are exposed to infectious diseases or immune-related problems. This susceptibility is partly explained by exercise-induced immuno-suppression. Moreover, many athletes are exposed to either pathogens or physico-chemical influences while practising their sports. Indeed, the respiratory tract, the digestive tract as well as the skin and mucosa can be seriously challenged in some aerobic outdoor or indoors sports as well as in some contact sports. At the upper respiratory tract level, and especially after exercise-associated hyperventilation, symptoms may develop, but it is sometimes difficult for the medical staff to confirm any infectious origin. …

Change Point Testing In Logistic Regression Models With Interaction Term, Youyi Fong, Chongzhi Di, Sallie Permar Jan 2014

Change Point Testing In Logistic Regression Models With Interaction Term, Youyi Fong, Chongzhi Di, Sallie Permar

UW Biostatistics Working Paper Series

The threshold effect takes place in situations where the relationship between an outcome variable and a predictor variable changes as the predictor value crosses a certain threshold/change point. Threshold effects are often plausible in a complex biological system, especially in defining immune responses that are protective against infections such as HIV-1, which motivates the current work. We study two hypothesis testing problems in change point models. We first compare three different approaches to obtaining a p-value for the maximum of scores test in a logistic regression model with change point variable as a main effect. Next, we study the testing …

Department of Biological Sciences Publications

Lactococci isolated from non-dairy sources have been found to possess enhanced metabolic activity when compared to dairy strains. These capabilities may be harnessed through the use of these strains as starter or adjunct cultures to produce more diverse flavor profiles in cheese and other dairy products. To understand the interactions between these organisms and the phages that infect them, a number of phages were isolated against lactococcal strains of non-dairy origin. One such phage, ΦL47, was isolated from a sewage sample using the grass isolate L. lactis ssp. cremoris DPC6860 as a host. Visualization of phage virions by transmission electron …

Antiretroviral therapy (ART) can reduce HIV levels in plasma to undetectable levels, but rather little is known about the effects of ART outside of the peripheral blood regarding persistent virus production in tissue reservoirs. Understanding the dynamics of ART-induced reductions in viral RNA (vRNA) levels throughout the body is important for the development of strategies to eradicate infectious HIV from patients. Essential to a successful eradication therapy is a component capable of killing persisting HIV infected cells during ART. Therefore, we determined the in vivo efficacy of a targeted cytotoxic therapy to kill infected cells that persist despite long-term ART. …

Ara H 6 Complements Ara H 2 As An Important Marker For Ige Reactivity To Peanut, Audrey E. Koid, Martin D. Chapman, Robert G. Hamilton, Ronald Van Ree, Serge A. Versteeg, Stephen C. Dreskin, Stef J. Koppelman, Sabina Wünschmann

Department of Food Science and Technology: Faculty Publications

The similarities of two major peanut allergens, Ara h 2 and Ara h 6, in molecular size, amino acid sequence, and structure have made it difficult to obtain natural Ara h 6 free of Ara h 2. The objectives of this study were to purify natural Ara h 6 that is essentially free of Ara h 2 and to compare its IgE reactivity and potency in histamine release assays to Ara h 2. SDS-PAGE of the highly purified allergen (<0.01% Ara h 2) revealed a single 14.5kD band and the identity of Ara h 6 was confirmed by LC-MS/MS. Ara h 6 showed a higher seroprevalence in chimeric-IgE ELISA (n=54), but a weaker biological activity in basophil histamine release assays than Ara h 2. Purified Ara h 6 will be useful for diagnostic IgE antibody assays, as well as molecular and cellular studies to investigate the immunological mechanisms of peanut allergy.

Assessing The Optimal Virulence Of Malaria‐Targeting Mosquito Pathogens: A Mathematical Study Of Engineered Metarhizium Anisopliae, Bernhard Konrad, Michael R. Lindstrom, Anja Gumpinger, Jielin Zhu, Daniel Coombs

School of Mathematical & Statistical Sciences Faculty Publications

Background

Metarhizium anisopliae is a naturally occurring fungal pathogen of mosquitoes. Recently, Metarhizium has been engineered to act against malaria by directly killing the disease agent within mosquito vectors and also effectively blocking onward transmission. It has been proposed that efforts should be made to minimize the virulence of the fungal pathogen, in order to slow the development of resistant mosquitoes following an actual deployment.

Results

Two mathematical models were developed and analysed to examine the efficacy of the fungal pathogen. It was found that, in many plausible scenarios, the best effects are achieved with a reduced or minimal pathogen …

N-Substituted 5-Amino-6-Methylpyrazine-2,3-Dicarbonitriles: Microwave-Assisted Synthesis And Biological Properties, Aidan Coffey, Jiahui Guo, Et. Al.

Department of Biological Sciences Publications

In this work a series of 15 N-benzylamine substituted 5-amino-6-methyl-pyrazine-2,3-dicarbonitriles was prepared by the aminodehalogenation reactions using microwave assisted synthesis with experimentally set and proven conditions. This approach for the aminodehalogenation reaction was chosen due to its higher yields and shorter reaction times. The products of this reaction were characterized by IR, NMR and other analytical data. The compounds were evaluated for their antibacterial, antifungal and herbicidal activity. Compounds 3 (R = 3,4-Cl), 9 (R = 2-Cl) and 11 (R = 4-CF3) showed good antimycobacterial activity against Mycobacterium tuberculosis (MIC = 6.25 µg/mL). It was found that the lipophilicity is …

In the outskirts of Medellin, Colombia, impoverished rural schoolchildren have cause for hope. The Colombian Coffee Growers’ Association wants to hire them. Why? Because these children have developed the independent thinking, communication, and work skills that will make them an asset to the industry. They developed these skills in their multigrade primary schools, where children do most of their learning in competence-based groups, while the teacher functions as guide and coach.

In Kenya, a teenage boy is also celebrating. A primary school dropout who once survived outside the law, he now runs his own small business, lives on his own, …

Biological Sciences Faculty Research & Creative Works

Multiple sclerosis (MS), characterized by chronic inflammation, demyelination, and axonal damage, is a complicated neurological disease of the human central nervous system. Recent interest in adipose stromal/stem cell (ASCs) for the treatment of CNS diseases has promoted further investigation in order to identify the most suitable ASCs. To investigate whether MS affects the biologic properties of ASCs and whether autologous ASCs from MS-affected sources could serve as an effective source for stem cell therapy, cells were isolated from subcutaneous inguinal fat pads of mice with established experimental autoimmune encephalomyelitis (EAE), a murine model of MS. ASCs from EAE mice and …

Inhibition Of Methicillin-Resistant Staphylococcus Aureus-Induced Cytokines Mrna Production In Human Bone Marrow Derived Mesenchymal Stem Cells By 1,25-Dihydroxyvitamin D3, Aparna Maiti, William A. Jiranek

Orthopaedic Surgery Publications

Background Methicillin-resistant Staphylococcus aureus (MRSA) is the predominant cause of bone infection. Toll like receptors (TLRs) are an important segments of host response to infection and are expressed by a variety of cells including human mesenchymal stem cells (hMSCs). The active form of Vitamin D, 1,25-dihydroxyvitamin D3 (1,25(OH)2D3) has potent immunoregulatory properties, but the mechanism remains poorly understood. The genomic action of 1,25(OH)2D3 is mediated by vitamin D receptor (VDR), hormone-regulated transcription factor. VDR interacts with co-activators and co-repressors are associated with chromatin histone modifications and transcriptional regulation. The aim of our study is to explore MRSA-induced TLRs-mediated pro-inflammatory cytokines …

The genus Corynebacterium consists of nearly seventy species and is closely related to the genera Mycobacterium, Nocardia, and Rhodococcus. The characteristic traits of Corynebacteria include cells that are shaped like straight rods with clubbed ends, as well as an extra cell wall layer consisting of mycolic acids covalently bound to the peptidoglycan layer which adds an additional layer of protection against antibiotics. Corynebacteria are very well studied and outstandingly important for the large scale biotechnological production of amino acids and nucleotides.[1] Moreover, Corynebacteria produce several pathogens that affect humans and livestock. Toxins produced by C. diphtheriae …
