Tethered IL-15 to augment the therapeutic potential of T cells expressing chimeric antigen receptor: Maintaining memory potential, persistence, and antitumor activity
Adoptive immunotherapy can retarget T cells to CD19, a tumor-associated antigen (TAA) expressed on B-cell malignancies, by the expression of a chimeric antigen receptor (CAR). Infusion of CAR-modified T cells for the treatment B-cell malignancies has demonstrated promise in preclinical and clinical trials. These data highlight the ability of infused CD19-specific T cells to be synchronously activated by large burdens of CD19+ leukemia and lymphoma. This can lead to dramatic antitumor effects, but also exposes the recipient to …

Car-Modified T Cells Capable Of Distinguishing Normal Cells From Malignant Cells, Hillary G. Caruso
Dissertations and Theses (Open Access)
T cells can be redirected to target tumor-associated antigen (TAA) by genetic modification to express a chimeric antigen receptor (CAR), which fuses the specificity derived from an antibody to T-cell activation domains to result in lysis of TAA-expressing cells. Due to the potential for on-target, off-tissue toxicity, CAR+ T-cell therapy is currently limited to unique or lineage-restricted TAAs. Glioblastoma, a grade IV brain malignancy, overexpresses epidermal growth factor receptor (EGFR) in 40-50% of patients. EGFR also has widespread normal tissue expression. To target EGFR on glioblastoma while reducing the potential for normal tissue toxicity, EGFR-specific CAR generated from cetuximab, …

This dissertation consists of three projects in the area of group testing. The method of group testing, through the use of pooling, has proven to be an efficient method of reducing the time and cost associated with screening for a binary characteristic of interest, such as infection status. The salient feature of group testing that provides for these gains in efficiency is that testing is performed on pooled specimens, rather than testing specimens one-by-one. In Chapter 1, we present a general introduction of group testing. Typically, the statistical literature surrounding group testing has investigated the implementation of pooled testing for …

Human papillomaviruses (HPVs) have been reported to infect epithelial trophoblastic cells of the placenta, induce cell death and even cause placental malfunction associated with spontaneous preterm delivery. To date, no study has been conducted to determine the role of HIV on HPV genotype distribution and pathogenesis in the placental compartment. This is despite the evidence that the human immunodeficiency virus (HIV) can decrease the cellular immune response and increase the incidence of malignant cancers in HPV patients. Therefore, in this study, we analyzed 200 genomic DNA (gDNA) samples extracted from paraffin embedded placental tissues of HIV positive and HIV negative …

Chemical Engineering Undergraduate Honors Theses
The United Nations estimates that water-borne illness accounts for 80% of deaths in developing nations and that nearly 1 in 6 do not have access to clean water (1). However, despite the ability to produce water in the developed world from complex streams such as wastewater or seawater, challenges remain in providing this basic resource in the developing world. Too often the approach has been to apply a high-tech solution, which requires significant technical expertise to run and significant financial resources to repair. Furthermore, as every developing nation faces different challenges, a new solution must be provided for each situation. …

Water System For Developing Countries / Disaster Relief Made With Local Materials, Dong Joon Lee
Chemical Engineering Undergraduate Honors Theses
As the equipment builder of the team, I was responsible for building the treadle pump, the sand filter tank, the hand pump, and generating the instructions for each equipment. Each team member was responsible for certain parts of the water purification system, but everyone worked cohesively on all parts of the design when assistance was necessary. The original design for the treadle pump was a two-man powered pump which discharged water at 15 gallons per minute. Since our system only required less than half of the flow rate, and required another person to operate the bicycle, the pump was redesigned …

At the beginning of this project, I elected to focus mainly on the electrolysis portion. I completed a lot of research on electrolysis in general, and then specifically on brine electrolysis, which is transforming a salt water solution into a sodium hypochlorite solution, or bleach. I researched all of the different types of electrode materials and purchased several electrodes in order to perform experiments. I researched several methods of electrolysis in use today. I performed several calculations about the electrolysis reaction, including the electric potential required to drive the reaction (approximately 1.0 volt), as well as what concentration of salt …
Prison Health Care After The Affordable Care Act: Envisioning An End To The Policy Of Neglect, Evelyn Malave
Prison Health Care After The Affordable Care Act: Envisioning An End To The Policy Of Neglect, Evelyn Malave
Hofstra Law Faculty Scholarship
Inadequate prison health care has created a health crisis for reentering prisoners and their communities--a crisis that is exacerbated by barriers to employment and other collateral consequences of release. This Note will first examine how current Eighth Amendment doctrine has failed to sufficiently regulate prison health care so as to have any significant effect on the crisis. Next, it will argue that the Affordable Care Act (ACA) alters the Eighth Amendment analysis by triggering a change in the "evolving standards of decency" that guide the doctrine. Specifically, this Note will argue that, after the passage of the ACA, releasing sick, …

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
Rift Valley fever (RVF) is a severe disease affecting both humans and a number of agriculturally important livestock species. The causative agent, RVF virus (RVFV), is primarily transmitted through mosquito bites, with transmission also occurring by exposure to infectious aerosols and direct contact with infected body fluids such as blood. Presently, there are no licensed vaccines or medicines to prevent or treat severe RVFV infection in humans. Favipiravir (T-705) is a novel compound licensed for the treatment of influenza in Japan and presently in Phase III clinical trials in the US, which has demonstrated favorable activity against an attenuated strain …

Evaluation Of Pro-Inflammatory Biomarkers As Potential Early Indicators Of Acute Respiratory Distress Syndrome (Ards), Makda Solomon Gebre
Undergraduate Honors Capstone Projects
Infections with the 2009 pandemic influenza A(HlNl) virus often leads to acute respiratory distress syndrome (ARDS) in high-risk patients. In these patients, ARDS has a mortality rate of 40-50 % [1]. Patients may die before any intervention is possible since there are few if any early biomarkers that are indicative of ARDS and can be used for diagnosis. In a search for biomarkers that signal early disease progression in mice, multiple acute phase proteins involved with inflammatory responses to infectious stimuli were proposed. Three biomarkers were evaluated for this project. These included C-reactive protein (CRP), Serum Amyloid A (SAA) and …

The emergence of antibiotic-resistant bacterial strains underscores the importance of identifying new drug targets and developing new antimicrobial compounds. Lysine and meso-diaminopimelic acid are essential for protein production and bacterial peptidoglycan cell wall remodeling and are synthesized in bacteria by enzymes encoded within dap operon. Thereforedap enzymes may serve as excellent targets for developing a new class of antimicrobial agents. The dapE-encoded N-succinyl-L,L-diaminopimelic acid desuccinylase (DapE) convertsN-succinyl-L,L-diaminopimelic acid to L,L-diaminopimelic acid and succinate. The enzyme is composed of catalytic and dimerization domains, and belongs to the M20 peptidase family. To understand the specific role …

Few, if any, tribulations in the field of public health have managed to transcend the epidemiological transition from acute to chronic disease as malnutrition has. Officially, malnutrition is a category of diseases that includes micronutrient deficiency, under-nutrition (calories/protein), as well as excessive nutrition. The World Food Program estimates that 842 million people worldwide do not have access to enough food, approximately 146 million of which are children. The most serious types of hunger are known as Severe Acute Malnutrition (SAM) and Moderate Acute Malnutrition (MAM), with 20 million and 35 million respective cases each year occurring in children less than …

Investigations Of Novel Mechanisms Of Action For Anti-Bacterial And Anti-Cancer Agent Development, Jenson Verghese
Theses and Dissertations
The development of drugs and therapeutic agents for combating infections and human malignancies continues to be a forefront area in both academic and industrial research. This is driven by the rapid emergence of multi-drug resistant bacterial strains and accumulating mutations in cancer targets that is quickly rendering our current arsenal of drugs ineffective for these therapies. Unless new drugs with novel mechanisms of action are identified and developed at a faster pace, we face a losing battle in managing these diseases. The first part of this work concerns with the natural product Simocyclinone D8 (SD8). Simocyclinone D8 is an angucyclinone …

Dengue is one of the most rapidly spreading mosquito-borne viral diseases in the world and inflicts significant health, economic and social burdens on populations. In this dissertation, I studied different aspects of modeling of dengue and vector-borne diseases in general. Among various dengue models that have appeared in literature, some explicitly model the mosquito population, while others model them implicitly. In spite of extensive use of both modeling approaches, little guidance exists for which type of model should be preferred. I developed a Bayesian approach that uses a Markov chain Monte Carlo (MCMC) method to fit disease models to epidemiological …

Impact Of Physical And Psychosocial Workplace Hazards On Employee Health: An Irish T Ale Of Civil Servant Workers, Kyle R. Stanyar
All Dissertations
Obesity, mental health problems, and absenteeism are both economic and health burdens for employers and employees. Research suggests that physical and psychosocial hazards in the workplace contribute to health risks and health problems among employees. There is a need for researchers to examine how exercise, diet, and age interact with the negative effects of workplace hazards upon health. Hypotheses 1a through 3b predicted that physical and psychosocial workplace hazards would negatively impact body mass index (BMI), general mental health, and sickness absences. Further, hypotheses 4a through 9b predicted that exercise and diet would buffer stress from occupational hazards upon BMI, …

Long-Term Mindfulness Meditation: Anxiety, Depression, Stress And Pain, Is There A Connection For Public Health?, Sara Spowart
USF Tampa Graduate Theses and Dissertations
Long-term mindfulness meditation for anxiety, depression, pain and stress has not been adequately investigated in academic literature. The majority of literature in relation to mindfulness meditation and these ailments concerns Kabat-Zinn's Mindfulness-Based Stress Reduction (MBSR) program. This is a short-term 6-8 week program that requires a licensed professional, is limited in availability, high cost, and combines other elements aside from mindfulness meditation, such as yoga, martial arts, group discussion, poetry and other teaching techniques meant to increase mindfulness concentration. Furthermore, efficacy studies, which have assessed the long-term impact of MBSR are inconclusive. Although these studies on long-term practice demonstrate a …
Assessing The Impacts Of Unrestricted Pesticide Use In Small-Scale Agriculture On Water Quality And Associated Human Health And Ecological Implications In An Indigenous Village In Rural Panam[Aacute], Sarah Louise Watson
USF Tampa Graduate Theses and Dissertations
In 2014, the global pesticide industry's projected worth is $52 billion and by 2020, the developing world will make up one-third of the world's chemical production and consumption. Pesticides can have unintended negative consequences for human health and the environment, especially in the developing world where regulations are loose or nonexistent. One country with unrestricted use of pesticides is Panam[aacute], especially in Santa Rosa de Cucunatí. In this indigenous village, small-scale farmers and ranchers spray paraquat, glyphosate, picloram, and 2,4-D at higher elevations than the spring water source of a gravity-fed water system, the river, and the village. The objective …

Myocarditis Complicating Plasmodium Vivax Malaria., Noreen Nasir, Saima Lalani, Zahra Aziz Samani, Aysha Almas
Department of Medicine
Myocarditis complicating Plasmodium vivax malaria is an extremely rare complication. We report this development in a young girl who was diagnosed to have P. vivax malaria on the basis of peripheral smear. While undergoing antimalarial treatment, she developed respiratory distress requiring invasive mechanical ventilation and inotropic support due to cardiogenic shock secondary to myocarditis. Cardiovascular complications are well recognized with Plasmodium falciparum malaria. Nevertheless, a high index of suspicion should be maintained for the same in Plasmodium vivax infection especially if symptoms of heart failure develop in a young patient.

High Prevalence Of Ranavirus Infection In Permanent Constructed Wetlands In Eastern Kentucky, Usa, Jennifer Strong
University Presentation Showcase Event
Two important drivers of global amphibian declines are land-use change and infectious diseases. Hundreds of permanent wetlands have been constructed on ridge tops for wildlife management in eastern Kentucky within the same landscape as natural, ephemeral wetlands. Constructed wetlands support a different amphibian community than natural wetlands, and some of these species have been associated with ranavirus outbreaks. Our objective was to test for occurrence of ranavirus and amphibian chytrid fungus (Batrachochytrium dendrobatidis) in amphibian populations of natural and constructed wetlands in the Daniel Boone National Forest, Kentucky. We selected Eastern Newts (Notopthalmus viridescens) and Wood Frogs (Lithobates sylvaticus) as …

Background: A simple sensitivity analysis technique was developed to assess the impact of misclassification and verify observed prevalence differences between distinct populations.
Methods: The prevalence of self-reported comorbid diseases in 4,331 women with surgically-diagnosed endometriosis was compared to published clinical and population-based prevalence estimates. Disease prevalence misclassification was assessed by assuming over-reporting in the study sample and under-reporting in the general (comparison) population. Over- and under-reporting by 10%, 25%, 50%, 75%, and 90% was used to create a 5×5 table for each disease. The new prevalences represented by each table cell were compared by p-values, prevalence odds ratios, and 95% …
