Multidomain enzymes often rely on large conformational motions to function. However, the conformational setpoints, rates of domain motions and relationships between these parameters and catalytic activity are not well understood. To address this, we determined and compared the conformational setpoints and the rates of conformational switching between closed unreactive and open reactive states in four mammalian diflavin NADPH oxidoreductases that catalyze important biological electron transfer reactions: cytochrome P450 reductase, methionine synthase reductase and endothelial and neuronal nitric oxide synthase. Sentencing credit laws provide opportunities for inmates to gain a reduction in their prison sentence, and such laws have at least four intended goals: 1) reducing prison populations; 2) promoting prosocial behavior during imprisonment by offering inmates incentive for good behavior and/or deterring them from engaging in antisocial behavior; 3) reducing recidivism by providing offenders incentive for good behavior and participation in rehabilitative programming; and, 4) lowering correctional costs (Lawrence & Lyons, 2011; Weisburd & Chayet, 1989). The purpose of this study was to determine whether there is a difference in Laricobius nigrinus Fender fecundity between field-collected and laboratory-reared beetles. Laricobius nigrinus used for this study were separated into four cohorts based on origin: Cohort 1 – Wild-caught from Seattle, WA – native population; Cohort 2 – Wild-caught from Banner Elk, NC – naturalized population; Cohort 3 – Laboratory-reared F1generation from Seattle, WA parents; and Cohort 4 – Laboratory-reared F1 generation from Banner Elk, NC parents. Each year, there are an estimated 3.8 million sports-related concussions that occur in the United States alone, the majority occurring during football collisions. These staggering numbers occur even with the use of mandated protective helmets, which are designed to decrease potential brain injuries. The outer shell of a football helmet acts as a shield for vulnerable areas of the cranium by providing the initial impact force dispersion to allow a more distributed load to be transferred to the inner foam padding. Students with special needs often miss out on classroom curricula for specialized instruction. While these services are valued for educational benefits, this instruction method often has negative impacts on social-emotional development and targets students for their differing needs.

Integrated collaborative teaching models include collaborative teaching among general and special educators in an inclusive environment. In this descriptive study, the author examined integrated collaborative teaching as a delivery model to increase responsiveness to the needs of all learners through academic and social inclusion.

The growth of the Hispanic population in Nebraska was accompanied by a significant increase in Hispanic entrepreneurship at the beginning of the 2000s. From 2002 to 2007, the 3,065 Hispanic-owned businesses constituted a small but dynamic and resilient segment of business growth in the state. Did participation in self-employment decrease for Hispanics after the economic crisis of 2008? What are the characteristics of Hispanic-owned businesses and Hispanics business-owners in Nebraska? Do some of these characteristics reveal social or economic barriers that could prevent these businesses from succeeding?

On December 4, 2014, the U.S. Census Bureau released five‐year American Community Survey (ACS) estimates for the combined years of 2008 through 2012, making available social, economic, housing and demographic statistics for Nebraska’s Legislative Districts. Also available are estimates for every community and county in Nebraska and the nation.

The charts and tables in this report are based on the Legislative District boundaries for the 103rd Legislature that were approved by the Legislature in 2011 and were based on data from the 2010 Census. In recent decades, the promotion of policies that are environmentally friendly has become an important goal in U.S. foreign policy. One way that the United States has influence over protecting the environment is through the policies attached to projects funded by the multilateral development banks (MDBs). This gives the U.S. the ability to indirectly fund projects in developing countries. Using data provided by the United States Treasury, I examined U.S. voting decisions on projects from 2004 to 2011. The protective mechanisms of microglia cells help to maintain central nervous system (CNS) homeostasis and function.1 Microglia are innate immune cells that constantly survey their surrounding CNS microenvironment for pathogens, damaged cells, and inflammatory molecules. Sugars from our diet, including glucose and fructose, combine with endogenous proteins non-enzymatically and form advanced glycation-end products (AGEs).2 AGEs are shown to produce reactive oxygen species, leading to inflammation and cellular damage that may be mediated by microglia.3 Fructose consumption has become increasingly prevalent within the American diet, as it is a lower cost sweetener.4 Microglia become activated and phagocytic in the presence of …

Objective: As a result of the heightened reactivity fructose demonstrates compared to glucose and our current knowledge of glucose advanced glycation end-products, the aim of this research was to further elucidate the proinflammatory pathways involved in the response to fru-AGE exposure, including the effects of fru-AGEs on cell-surface receptor expression. We hypothesized that once microglia were activated in response to fru-AGE exposure, there would be an increase in the expression of RAGE and TLR4 to facilitate the proinflammatory cascade.

Over-the-counter (OTC) medications are used by various populations as an adjunct to help improve various aspects of life, such as sleep cycles, disease prevention, and mood. The purpose of this study is to compile all of the available data from human trials of over-the-counter medications used in patients with Autism Spectrum Disorders (ASD). OTCs have been used in the treatment of ASD to minimize social impairments, suppress repetitive behaviors, and enhance quality of sleep to improve daytime behaviors. Peer tutoring services for a pathophysiology and therapeutics course have historically been made available to pharmacy students, but they were greatly underutilized. The role of peer tutors had been exclusively to hold office hours during which students could ask questions or review material. Peer tutoring services were updated and expanded based on the needs of the students. The objectives of this project were to develop, implement, and assess the impact of peer-led organized weekly review sessions in a pathophysiology and therapeutics course.

HMG-CoA reductase inhibitors, also known as statins, are a ubiquitous class of medication used for lowering cholesterol. In-vitro and animal studies have suggested that statins can activate osteoblast differentiation and have anabolic effects on bones; however, observational and experimental studies in humans have shown conflicting results.1-5 The exact mechanism of statins on bone growth is unknown; however, there are several hypotheses. The “Lipid Hypothesis” (Figure 1) suggests that lipid oxidation leads to activation of PPARγ, and production of isoprostanes including isoPGF2α and isoPGEα. Chlorpromazine is a phenothiazine antipsychotic which is often used in hospice and palliative care to treat hiccups, delirium, and nausea. With the discontinuation of the commercial oral solution concentrate, there is a need to prepare this product by extemporaneous compounding. This study was initiated to identify an easy-to-prepare formulation for the compounding pharmacist. A stability study was also conducted to select the proper storage conditions and establish the beyond-use date. Chlorpromazine HCl powder and the Ora-Sweet® syrup vehicle were used to prepare the 100 mg/mL solution. Once the feasibility was established, a batch of the solution was prepared and packaged …

DNA can be damaged by several kinds of endogenous and exogenous reactive nitrogen species. Under nitosative stress, uracil (U), hypoxanthine (I), xanthine (X) and oxanine (O) are four major deaminated DNA bases derived from cytosine (C), adenine (A) and guanine (G) respectively. To repair this type of DNA damage, several different repair pathways are involved.

My dissertation work mainly focused on the uracil-DNA glycosylase (UDG) superfamily, which includes several groups of enzymes that recognize the damaged DNA bases and initiate the base excision repair (BER) pathway, one of the most important repair pathways to deal with deaminated DNA bases.
