Differential Protein Expression Of Caco-2 Cells Treated With Selenium Nanoparticles Compared With Sodium Selenite And Selenomethionine, Linglin Fu, Xuxia Yan, Xinming Ruan, Junda Lin, Yanbo Wang

The study was designed to determine the differential protein expression of Caco-2 cells treated with different forms of selenium including sodium selenite, selenomethionine (Se-Met), and selenium nanoparticles (nano-Se). Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and mass spectrometry (MS) were used to identify the differentially expressed proteins. The results indicated that seven protein spots, ubiquitin-conjugating enzyme E2 (E2), glutathione synthetases (GS), triosephosphate isomerase (TSP), T-complex protein 1 subunit zeta (TCPZ), lamin-B1, heterogeneous nuclear ribonucleoprotein F (hnRNP F), and superoxide dismutase [Cu-Zn] (Cu, Zn-SOD) were significantly different among all the groups. Study Abroad And Moral Development, Andrew B. Johnson, Danielle R. Hathcock

Why should students study abroad? The standard answer universities give cites three types of benefit: academic, cultural, and professional. We argue that this answer sells the value of study abroad short. Just as important as any of these benefits is the value study abroad has in promoting moral development. Drawing on key ideas of the seminal developmental psychologists Jean Piaget and Lawrence Kohlberg, we make the case that study abroad can facilitate moral development, whether one understands morality in utilitarian, Kantian, or sentimentalist terms. It does so by helping students take the perspective of those who are culturally different, inducing …

The genus Fusarium has a wide host range and causes many different forms of plant disease. These include seed rot and seedling blight diseases of cultivated plants. The Fusarium-caused diseases of wild plants are less well-known. In this study we examined Fusarium sp. n-caused disease development on non-dormant seeds of the important rangeland weed Bromus tectorum as part of broader studies of the phenomenon of stand failure or ‘die-off’ in this annual grass. We previously isolated an undescribed species in the Fusarium tricinctum species complex from die-off soils and showed that it is pathogenic on seeds. It can cause high …

An advanced mesoscale spring-mass model is used to mimic fabric surface motion. The fabric surface is represented by a high-quality triangular surface mesh. Both the tensile stiffness and the angular stiffness of each spring are determined by the material's Young's modulus and Poisson ratio, as well as the geometrical characteristics of the surface mesh. The spring-mass system is a nonlinear Ordinary Differential Equation (ODE) system solved by fourth order Runge-Kutta method. Remodeling of bone is an important indicator of bone health, and disorders in bone remodeling may lead to bone diseases such as osteoporosis. This dissertation analyzes nationalisms in the work of Canadian magazine illustrators in Toronto and New York, 1880 to 1960. Using a continentalist approach--rather than the nationalist lens often employed by historians of Canadian art--I show the existence of an integrated, joint North American visual culture. Drawing from primary sources and biography, I document the social, political, corporate, and communication networks that illustrators traded in. I focus on two common visual tropes of the day--that of the pretty girl and that of wilderness imagery. The production of lipids and proteins for growth, as well as nucleotides for the replication of the genome, are key requirements that loom as importantly as the need for the production of energy in the transformed metabolic state of cancer cells. The remodeling of the TCA cycle (extensive anaplerosis and cataplerosis) reflects this need for biosynthetic macromolecules. Cells require purines for critical biological processes that can be created by the de novo biosynthetic pathway or maintained by the salvage pathways. The de novo pathway in humans requires 6 enzymes, some of which are multifunctional. Adenylosuccinate lyase (ASL) catalyzes step 8 of the de novo pathway converting susuccinylaminoimidazolecarboxamide ribonucleotide (SAICAR) to aminoimidazolecarboxamide ribotide (AICAR). It also catalyzes the second step of the purine nucleotide cycle by converting SAMP to AMP. A short isoform of ASL (sASL) is ubiquitously expressed in all cell types at a lower concentration. Orbitofrontal cortex (OFC) is situated at the ventral floor of the prefrontal cortex region. OFC and some of its related structure (ventral striatum, ventral tegmental area, basolateral amygdala) are considered to be part of the valuation system that is essential to the decision-making process. The enoyl-acyl-carrier-protein reductase (ENR) catalyzes the last reaction in the elongation cycle in the fatty acid biosynthesis type II (FAS-II) pathway. To date, there are four known ENR isoenzymes: FabI, FabK, FabL, and FabV. We have rigorously characterized the FabV ENR from Burkholderia mallei (BmFabV) and have shown that this enzyme catalyzes substrate reduction via an ordered bi-bi mechanism, in which NADH binds first to the enzyme followed by the enoyl substrate [Lu, H. (2010) Biochemistry 49, 1281-1289]. However, this pathogen contains both FabI and FabV ENRs, and mechanistic insights into ENR substrate recognition are lacking in pathogens that solely …

Amyloid formation, the aggregation of normally soluble proteins or polypeptides into highly ordered beta-sheet structures, is a characteristic feature of many human diseases including Alzheimer's disease, Parkinson's disease and type 2 diabetes. I centered my research on islet amyloid polypeptide (IAPP), a neuroendocrine hormone that forms fibrillar amyloid deposits in the extracellular space of the pancreatic islets of Langerhans in type 2 diabetes. Although whether IAPP amyloid formation is a cause or consequence of the disease is controversial, the aggregation process has been shown to induce beta-cell apoptosis and reduce beta-cell mass, which is a hallmark feature of type 2 …

Cancer remains the second leading cause of death in the United States, accounting for nearly a quarter of all deaths, and is exceeded only by heart disease. Traditional chemotherapy uses highly potent cytotoxic agents to kill rapidly dividing cancer cells, relying on the premise that these fast-growing cancer cells are more likely to be killed by these drugs. Psychiatric disorders are the most common and costly forms of disease and injury worldwide. Despite this, our ability to accurately identify and treat these disorders is limited, due in part to deficient understanding of the underlying mechanisms of dysfunction. The current study aimed to identify genetically-influenced neurobehavioral traits--individual differences evident in both self-report and neural response--that might help improve classification and treatment of mental disorders. The world has been going through one of worst economic recessions in history with severe job market downturns. In October 2009, the U.S. civilian unemployment rate reached a historical 10% and nearly 50% of the unemployed exhausted their 26-week regular Unemployment Insurance (UI) benefits and consequently, UI has been extended to 99 weeks. In this thesis, I examine two novels that feature characters with disabilities and that are often taught in secondary English classes. These texts will be analyzed through a disability studies perspective - a form of analysis that is not often taught in schools. The disability studies perspective involves looking at disability in a critical light and examining the social, cultural, and political ramifications of assumptions that are made about those considered to be " disabled" by the able-bodied majority. The conventional argument in literary and gender studies of nineteenth-century United States culture has been that the home was women's claimed sphere of influence, whereas men were excluded from the home because of their economic, cultural, and societal commitments to the public sphere. Even as Catharine Beecher, Lydia Maria Child, Sarah Josepha Hale, and other adherents to a conservative domestic ideology closely associated the private sphere with women, these same writers identified specific roles and spaces that they expected men to occupy in the home. Tumor suppressor protein p53 responds to cellular stress by activating transcriptional and transcription-independent programs that induce apoptosis, growth arrest or senescence. Given its critical tumor suppressing role, p53 is often mutated in human cancers. Once altered by missense mutations in the DNA-binding domain, mutant p53 (mutp53) protein becomes highly stabilized in tumors, which is critical for mutp53′s oncogenic gain-of function. Current insight into the mechanism of tumor-specific stabilization of mutp53 is fragmentary and largely derived from ectopically constructed cell systems. We find that in human cancer cells endogenous mutp53, despite its ability to interact with MDM2, suffers from a profound …

Codon pair bias, the preferential pairing of certain codons over others, has been observed in all species analyzed ranging from bacteria to mammals. Previous research has shown that artificial coupling of underrepresented codon pairs (codon pair deoptimization) in viral genomes (such as poliovirus and influenza virus) reduces gene function and leads to virus death or attenuation. Although the reason for this phenomenon is unclear, increasing underrepresented codon pairs in an RNA viral coding region consequently increases the frequency of CpG and UpA dinucleotides. The adenovirus L4-22K protein is multifunctional and critical for different aspects of viral infection. Following DNA replication, increasing amounts of the L4-22K protein are observed prior to the appearance of other late viral proteins. We found that L4-22K controls viral gene expression at the post-transcriptional level and regulates the accumulation of the L4-33K protein, another critical viral regulator, at the level of alternative pre-mRNA splicing. This initiates an activation cascade of late viral proteins, while early genes are being turned off. Missing in Metastasis (MIM), also known as MTSS1, is a scaffold protein that is down-regulated in multiple metastatic cancer cell lines compared to non-metastatic counterparts. MIM regulates cytoskeletal dynamics and actin polymerization, and has been implicated in the control of cell motility and invasion. MIM has also been shown to bind to a receptor PTP, PTP-delta, an interaction that may provide a link between tyrosine phosphorylation-dependent signaling and metastasis. The main goal of the research is evaluation of the plasma jet driven magneto-inertial fusion (PJMIF) concept via simulations. To achieve this goal, the development of mathematical models and numerical algorithms for PJMIF has been performed, and large-scale simulation studies have been conducted. Francisella tularensis is a Gram-negative, facultative intracellular pathogen and the causative agent of tularemia. F. tularensis is a Tier 1 agent of bioterrorism that is highly lethal via the pulmonary route of infection. F. tularensis invades host cells, escapes the phagosome, and replicates in the cytosol prior to being released upon host cell death. The molecular mechanisms behind the virulence of F. tularensis are largely unknown. Among the described virulence factors of F. tularensis is a functional type I secretion system (T1SS). The T1SS is important for the secretion of virulence factors from the bacterial cytoplasm to the extracellular environment. …

The amygdala, a subcortical structure in the anterior temporal lobe, plays important roles in integrating sensory information from different brain areas, assigning emotional valence and effecting adaptive behavioral, physiological, and cognitive responses. Sensory inputs from cortical and thalamic sources, carrying highly processed sensory information and low level, more direct sensory information, respectively, converge in the basolateral nucleus of the amygdala (BLA). Here, neuromodulation by acetylcholine (ACh), released from inputs arising from the nucleus basalis of Meynert (NBM) in the basal forebrain, plays a major role in synaptic integration during different emotional arousal states. This dissertation advocates a model of dialogue as an approach to education and to philosophical discourse itself that is capable of acknowledging and accommodating the manifold possibilities for human thought. Contemporary philosophy is permeated by a loss of certainty and the increasingly problematic nature of the concept of truth, and the complication of these concepts exposes significant limitations in the ways we conceptualize thought itself. Working in this vein, Gilles Deleuze identifies what he calls an " image of thought," which characterizes the forms we have traditionally allowed thinking to take. This dissertation examines ways the environment conditions individual incentives to participate in collective action. I hypothesize that as resources become scarce, individuals are more willing to participate in individually-costly collective action, including intragroup cooperation and intergroup hostility. The first and second of three studies establish a unit-of-selection based, game-theoretic foundation for intergroup conflict. The final study explores macro-level implications for the relationship between climate change and violence in Africa. For human beings and other highly social creatures, cooperation is the key adaptation with which we respond to environmental challenges. The rapid development of information technology produces vast amounts of data with numerous attributes. These multi-dimensional datasets offer tremendous opportunities for studying existing behavioral patterns and for predicting future developments. However, the high-dimensional space exceeds human comprehension. More sophisticated visualization techniques than the arsenal of standard plots are needed. First, we introduce an interactive navigation technique to help the analysts explore within the multi-dimensional data spaces. We employ a network-based interface and pair it with a parallel coordinates plot.
