Detection of population density is a form of communication for certain phytobacteria that usually involves the association of small diffusible chemical signals, particularly N-acyl homoserine lactones. It is pondered whether or not this molecule and the phytotoxin, coronatine, play a significant role in the directional motility of the wild type (Pst DC3000) and mutant strains (Pst DC3118, Pst DB29, and Pst psy/IR-) of Pseudomonas syringae pv. tomato on the adaxial surface of Arabidopsis thaliana leaves. It's Not Too Difficult: A Plea To Resurrect The Impossibility Defense, Ken Levy

The impossibility defense has baffled legal scholars, courts, and attorneys. This defense is especially difficult for two main reasons. First, it is not obvious which kinds of attempts it exculpates, which kinds of attempts it does not exculpate, and why it works only for the former and not the latter. Second, courts that have not resolved the first set of difficulties have only compounded the confusion by misstating and misapplying the impossibility defense. In particular, they have mistaken factual impossibility for legal impossibility and have interpreted certain factual impossibility situations as hybrid-impossibility situations. The expansion of Philippine aquaculture is essential to addressing poverty and food insecurity in the midst of a seemingly irreversible decline in national capture fisheries. Concomitant with its rise, however, are compounding environmental problems caused by climate change, lackluster governance and irresponsible farming practices. It is equally important to ensure aquaculture sustainability so that fish demand from a rapidly growing population is continually met and that fish farmers’ socioeconomic well-being is secured. As such, programs have been developed that aim to enjoin the government and private sectors with fishing communities to realize this goal.

Hantaviruses, family Bunyaviridae, are present throughout the globe in a variety of mouse, rat, mole, vole, shrew, or bat species. Hantaviruses persist for the lifetime of the animal reservoir, while causing no signs or symptoms of disease. Only the rodent-borne hantaviruses cause disease in humans. In contrast, a “spillover” infection of a hantavirus into a nonreservoir rodent species results in an asymptomatic acute infection. We and others in the field are interested in understanding the biology of these virus-host interactions and mechanisms that underlie these three very different outcomes. The creative thesis that I have written is a memoir. It is not complete in its detail or ambition but instead focuses on themes and topics of self-transformation, reinvention and the unintentional consequences of several nervous breakdowns and experiments with psychedelic substances. This thesis is just a beginning for a longer more drawn out memoir that will explore the themes above but also discuss family, one’s sense of self worth in the world and how patterns between fathers and sons repeat themselves. I hope to eventually publish this work in a commercial setting.

Complete Congenital Stationary Night Blindness (cCSNB) is a rare hereditary retinal disorder characterized by abnormal night vision. cCSNB is caused by postsynaptic defects in On bipolar cells (BCs) and is identified by the presence of an electroretinogram (ERG) with a normal a-wave, corresponding to photoreceptor function, and the absence of a b-wave, corresponding to a failure of On BC function. Through the study of genetic mutations in mouse that result in no b-wave ERG phenotypes, several proteins have been identified that play crucial roles in On BC signal transmission. Legionella pneumophila is a facultative intracellular pathogen that infects a wide array of protozoan hosts and human alveolar macrophages. L. pneumophila is dependent on a functional Dot/Icm type IVB secretion system that translocates bacterial effector proteins into the host cell cytosol. L. pneumophila genomes encode more than 250 effector proteins, many of which inhibit host cellular processes to form a favorable niche termed the Legionella-containing vacuole (LCV). The eukaryotic-like Dot/Icm translocated effector AnkB contains two eukaryotic-like ankyrin protein-protein interacting domains, one eukaryotic-like F- box domain and an eukaryotic C-terminal CaaX motif. Polychlorinated biphenyls (PCBs) are persistent environmental toxicants, present in 100% of US adults and dose-dependently associated with nonalcoholic fatty liver disease (NAFLD) in epidemiologic studies. PCBs are predicted to interact with receptors previously implicated in xenobiotic/energy metabolism and NAFLD. These receptors include the aryl hydrocarbon receptor (AhR), pregnane xenobiotic receptor (PXR), constitutive androstane receptor (CAR), peroxisome proliferator-activated receptors (PPARs), liver-X-receptor and farnesoid-X-receptor. This study evaluated the hepatic effects of the PCB mixture, Aroclor 1260, whose composition mimics human bioaccumulation patterns, in a mouse model of diet-induced obesity. The work presented in this dissertation outlines the role of stem cells in the remodeling of adipose tissue under conditions of nutrient excess. Obesity-induced variations in adipose tissue stem cell distribution were uncovered by analysis of the stromal vascular fraction isolated from mice fed a high fat diet for several weeks. Bone marrow cell transplantation was used to determine the derivation of progenitor cells found in adipose tissue; and supplementation of depleted progenitor cell populations achieved via direct cell transplantation, helped to determine the contribution of these progenitor cells to the remodeling process. Skeletal muscle constitutes a highly plastic and malleable tissue that rapidly and profoundly adapts to various environmental and physiological stimuli. This response takes place by means of regulated signaling transduction which elicits changes in gene expression, biochemical, and metabolic properties. Adult muscle is composed by alignment of multinucleated, post mitotic muscle fibers formed during embryonic and neonatal development. Residing in the niche of mature muscle fibers is a small population of undifferentiated muscle progenitors termed satellite cells. The type IIa sodium-phosphate cotransporter (Npt2a) is expressed in the apical membrane of the renal proximal tubule and is responsible for the reabsorption of the majority of the filtered load of phosphate. Parathyroid hormone (PTH) is secreted by the parathyroid gland in response to a decrease in serum calcium or an increase in serum phosphorus, and acutely induces phosphaturia through the rapid stimulation of endocytosis of Npt2a and its subsequent lysosomal degradation. Chronic PTH stimulation leads to sustained phosphaturia, but the mechanisms for the chronic regulation of Npt2a by PTH remain unclear. We hypothesize that PTH decreases Npt2a mRNA levels …

Host cells respond to bacterial lipid A through Toll-like receptor 4 (TLR4). Activation of TLR4 by lipid A triggers a response that involves two main adaptor proteins, MyD88 and TRIF. MyD88-dependent gene expression is associated with proinflammatory protein production, while TRIF-dependent gene expression is essential for optimal activation of adaptive immunity by antigen-presenting cells. Detoxified, monophosphoryl lipid A agonists (MPLA or synthetic MLA) were previously suggested to elicit TRIF-biased TLR4 signaling; that is, induction of weaker MyD88-associated gene expression but relatively intact TRIF-dependent gene expression when compared to fully active diphosphoryl lipid A (lipid A). Fairy tales and other forms of fantastic literature have fascinated children and adults for generations. The scholarship on these types of works indicate that many find their folkloric origins in oral storytelling, and those roots may be quite ancient. However, some of the earliest examples of recorded literature capture stories of magic and fantasy. Invasive species are one of the leading causes of biodiversity loss and their presence can significantly impact the structure and function of native ecosystems. In this dissertation, the impacts of the invasive honeysuckle shrub, Lonicera maackii, on exotic earthworm populations, leaf litter decay responses, and inorganic and organic nutrient pools in an urban woodland park are assessed. Chapter 1 provides a brief review of honeysuckle's effects on forest ecosystems. Chapter 2 describes honeysuckle’s effects on exotic earthworm populations and the seasonal importance of macroinvertebrates to leaf litter decomposition in urban woodlands. Detrusor-sphincter dyssynergia after chronic spinal cord injury (SCI) leads to over-distention, high bladder pressures and vesico-ureteric reflux into the kidney, resulting in repeated infections and pyelonephritis. Given the importance/limited knowledge of SCI effects on the upper urinary tract, this study assessed expression of two proteins in the kidneys of contused (T9) male rats relative to controls that are indicative of tissue stress/damage (TGFβ, CD11b). In tumor cells, glutamine metabolism provides anaplerotic carbon for the TCA cycle, fatty acid synthesis, and precursors for the production of nucleotides and glutathione. This metabolic alteration is primarily driven by disruptions in oncogenic or tumor suppressor function and results in glutamine dependency for tumor cell survival. Troglitazone, a PPARγ agonist, has been reported to alter glutamine metabolism. This thesis project addresses whether troglitazone treatment could disrupt glutamine metabolism in glutamine-dependent tumor cells. Results obtained with troglitazone treatment include: dose-dependent inhibition of cell proliferation, glutamine uptake, glutaminolytic protein expression, steady-state ATP levels, and glutamine carbon flux into the TCA cycle. …

A relationship between periodontal disease and oral squamous cell carcinoma (OSCC) has been demonstrated in recent studies. Porphyromonas gingivalis’ antigen have been found in oral cancer cells. The embryonic program “epithelial-mesenchymal transition” (EMT) is thought to trigger cancer invasion by allowing tumor cell propagation. The objective of this study was to examine the role of P. gingivalis in the expression of ZEB1, vimentin, and repression of miRNA-200 family as EMT prognostic markers in cancer cell metastasis. The purpose of this case study was to gain a full understanding of the AHGA with a focus on the economic and social motivations of the original farmers to bring tourism to the farm. The ability of science to produce experimental data greatly surpasses our current ability to effectively visualize, conceptualize, and integrate the vast volumes of available data into a unified understanding of how complex biological systems work. This inability is a hindrance to scientific progress, and is particularly daunting when one considers multidimensional and shape-based observations as in the field of regenerative biology. Trade secret law arose to help companies protect confidential information (e.g., the Coca-Cola formula) from competitors seeking to copy their innovative efforts. But companies increasingly use trade secret law to block a wide swath of information from the scrutinizing eyes of consumers, public watchdog groups, and potential improvers. Companies can do this, in part, because trade secret law lacks clear limiting doctrines that consider the social benefits of unauthorized use. For example, trade secret law makes no allowance for the departing employee that uses proprietary information to create a substantially improved product or disclose public health risks.

In recent years, a large number of biomedical studies have demonstrated that the bacteria that contribute to periodontal disease can migrate outside the oral cavity, causing a host of systemic infections. Yet, to date, only one bioarchaeological investigation has addressed this co-occurring disease process in a past population. The results of this thesis confirm the bioarchaeological visibility of the correlation between oral and systemic disease based on data derived from a sample of white and black adults from the Robert J. The objectives of this research were to 1) evaluate the effects of stocking rate on forage nutrient quality 2) quantify the relative proportions of current vs. previous year growth being consumed in early summer upland range pastures and 3) determine how grazing effects forage nutrient quality in subirrigated meadows in the Nebraska Sandhills. Experiment 1 was a two year study conducted on the experimental upland range paddocks at Gudmundsen Sandhills Laboratory. There are multiple breeds of beef cattle available to utilize in breeding systems to maximize production and economics. Calving difficulty (dystocia) is a significant cost to beef production and is more prevalent in first-calf heifers. The objectives of this study were to estimate genetic parameters and breed differences for calving difficulty and birth weight as a first step towards the development of across-breed adjustment factors for calving difficulty.
