Transcriptional Regulation Of Neurogenic Atrophy-Induced Gene Expression By Muscle Ring Finger-1 And Myogenic Regulatory Factors, Theodore Olson Jan 2014

Transcriptional Regulation Of Neurogenic Atrophy-Induced Gene Expression By Muscle Ring Finger-1 And Myogenic Regulatory Factors, Theodore Olson

UNF Graduate Theses and Dissertations

Skeletal muscle wasting is a consequence of numerous physiological conditions, including denervation, corticosteroid treatment, immobilization, and aging. The E3 ubiquitin ligases, MuRF1 and MAFbx, are induced under nearly all atrophy conditions and are believed to play a key role in protein degradation in atrophying muscle. However, the preliminary data described in this study provides new evidence that MuRF1 may also act as a transcriptional modulator of atrophy-induced gene activity, including the regulation of MAFbx and MuRF1 expression. To characterize the transcriptional regulation of MuRF1 and MAFbx, reporter gene constructs containing fragments of the proximal promoter regions of these genes were …

Promoting A Shared Vision: Identifying Intersections Between Food And Fiber Systems Literacy Benchmarks And Illinois State Board Of Education Standards, Daniel P. Ingold Mr.

Research Papers

Understanding of agricultural systems has been an accepted and necessary aspect of a child’s education for centuries. Conventional agricultural education has proven effective in creating well-trained agricultural professionals and scholars, but has had the unintentional effect of limiting access to agricultural concepts to the non-agricultural student. This effect has potentially negative cultural consequences considering the importance agricultural issues. In response, agricultural educators have carried out an initiative to promote agricultural literacy in the classroom through an integration of agricultural concepts into core curriculum. A Guide to Food and Fiber Systems Literacy Benchmarks (Leising, 1998) is an example of programs that …

Developing Rating Curves In A Small Iowa Watershed To Evaluate Nutrient Loading In Streams, Jennifer Mary Shepeck

Dissertations and Theses @ UNI

Dry Run Creek, a tributary to the Cedar River in northeast Iowa, is a watershed under high pressure from the surrounding region. With its headwaters located in farm fields and urban drainage ditches, it is subject to flash flooding and erosion and is on the DNR’s list of impaired waters. Dry Run Creek is similar to other small watersheds across eastern Iowa. As tributaries to the larger rivers that eventually flow into the Mississippi River and then to the Gulf of Mexico, they are significant contributors to the nutrient loading causing the hypoxia in the Gulf. The purpose of this …

The Effects Of Sampling Design On Abundance And Distribution Of Bottlenose Dolphins In The St. Johns River, Florida, Samantha Ryanne Nekolny Jan 2014

The Effects Of Sampling Design On Abundance And Distribution Of Bottlenose Dolphins In The St. Johns River, Florida, Samantha Ryanne Nekolny

UNF Graduate Theses and Dissertations

The National Oceanic and Atmospheric Administration (NOAA) is tasked with upholding the regulations prescribed in the Marine Mammal Protection Act of 1972. One of the ways this is accomplished is by producing regular stock assessment reports. For these reports, knowledge about a species abundance and distribution is a vital component, allowing informed management decisions to be made that may potentially reduce the natural and anthropogenic impacts on the organisms. In 2010, an Unusual Mortality Event (UME) occurred in the lower St. Johns River, resulting in an unusually large number of bottlenose dolphin strandings. As the data regarding this population was …

The Performance Of Simple Artificial Floating Wetland Communities And Their Effects On Aquatic Nutrient Levels And Algal Abundance, Bradley L. Sleeth Jan 2014

The Performance Of Simple Artificial Floating Wetland Communities And Their Effects On Aquatic Nutrient Levels And Algal Abundance, Bradley L. Sleeth

UNF Graduate Theses and Dissertations

Harmful algal blooms are exponential increases in autotrophic microorganisms that proliferate in such a way that the surrounding environment, the local economy and the health of regional populations are negatively affected. Among the causes of these blooms are anthropogenic inputs of excess nitrogen and phosphorus into the environment through overfertilization. Floating treatment wetlands (FTW) have emerged as a novel method of reducing the negative impacts of these nutrient inputs by using artificial rafts to float normally emergent wetland plants on the surface of water bodies to assimilate excess nutrients. Because their use is so new, only limited research has been …

Osteoblast And Odontoblast Specific Regulatory Actions Of Runx2 For The Development Of Mineralized Tissues, Mitra Darice Adhami Jan 2014

Osteoblast And Odontoblast Specific Regulatory Actions Of Runx2 For The Development Of Mineralized Tissues, Mitra Darice Adhami

All ETDs from UAB

The Runx2 transcription factor is necessary for commitment and differentiation of mesenchymal cells into the chondroblasts, osteoblasts and odontoblasts. Runx2 induces the expression of several key genes involved in mineralization of both the bone and dentin matrices. Global Runx2-null mice are completely void of mineralized tissue and do not express osteoblast markers, indicating that Runx2 is required for commitment to the osteoblast lineage. Tooth development in Runx2-null mice is arrested in the bell stage, prior to the differentiation of odontoblasts. Thus, Runx2 is essential for development of osteoblast and odontoblast lineages. However, the functional requirements of Runx2 after commitment to …

Fatigue Testing Of Straight And Angled Abutments In A High Torque Dental Implant System, Rana M. Atieh Jan 2014

Fatigue Testing Of Straight And Angled Abutments In A High Torque Dental Implant System, Rana M. Atieh

All ETDs from UAB

Root form dental implants are a common solution used today to replace missing teeth. However current designs still suffer from a number of limitations. Angled abutments are used for incisors, where maximum human bite forces are normally < 200N, while straight abutments are used for molars subjected to higher forces > 200 N. The aim of this study was to evaluate the mechanical performance of a novel dental implant system by fatigue testing straight and 15-degree angled abutments. It was hypothesized that the novel thread design will dissipate the load evenly throughout the implant allowing the system to withstand normal chewing forces. Implants with 4.2mm diameter were tested according to ISO 14801 in specimen …

Questing The Beast: From Malory To Milton, Malorie A. Sponseller Jan 2014

Questing The Beast: From Malory To Milton, Malorie A. Sponseller

College of Graduate Studies: Theses & Dissertations

The Questing Beast is a Medieval creature that has received little scholarly attention. Because of her labile nature, she is difficult to identify and therefore challenging to study. When previously analyzed, she has been considered only in her Medieval context. By comparing the Questing Beast from Perlesvaus, the Post-Vulgate Cycle, and the Prose Tristan, four identifying characteristics can be found: she is symbolic, she is multi-formed, she is a mother that gives birth, and she produces a barking noise most often made by her unborn young. Of these four signs, the last is the most prevalent and identifiable. …

Modulation Of Immunity Via Antibodies Displayed On In Situ Forming Peptide Co-Assemblies, Yi Wen Jan 2014

Modulation Of Immunity Via Antibodies Displayed On In Situ Forming Peptide Co-Assemblies, Yi Wen

Electronic Theses and Dissertations

Many diseases are associated with disruption of immune homeostasis. Monoclonal antibodies (mAb) targeting soluble antigens or immune cell surface molecules have been developed to modulate immunity. However, systemic administration can lead to severe immune related side effects. This dissertation investigated a versatile platform to display antibodies locally in diseased tissues to minimize unwanted systemic exposure. This system is a stimuli-responsive and injectable formulation utilizing in situ forming peptide co-assemblies and two linker proteins. The self-assembling peptides EAK16-II and EAKIIH6 co-assemble into fibrous structures with accessible His-tags when exposed to salts. Anti-His-tag antibodies (αH6-IgG) bind to these His-tags and provide fragment …

A New Framework For Assessing Clinical Data Transparency Initiatives, Erika Lietzan Jan 2014

A New Framework For Assessing Clinical Data Transparency Initiatives, Erika Lietzan

Faculty Publications

Biopharmaceutical companies submit vast amounts of clinical data and analysis to support approval of their medicines, expecting the information to be kept confidential, as has been the practice of regulators around the world for decades. Over the last ten years, however, pressure has been mounting for regulators or industry to release this information. Legal scholars have generally taken the view that no relevant doctrines or bodies of law preclude the release of this material and that public policy considerations compel its release. This article argues that the scholarship to date has overlooked key considerations: the special issues presented by operation …

Paper 1 and Paper 2 include the first systematic studies of the properties, structures of zinc aluminophosphate (ZAP) glasses and phase relationships in the ZnO-Al₂O₃-P₂O₅ system, respectively. Adding alumina to a …

The Effect Of Composition On The Viscosity, Crystallization And Dissolution Of Simple Borate Glasses And Compositional Design Of Borate Based Bioactive Glasses, Kathryn Lynn Goetschius Jan 2014

The Effect Of Composition On The Viscosity, Crystallization And Dissolution Of Simple Borate Glasses And Compositional Design Of Borate Based Bioactive Glasses, Kathryn Lynn Goetschius

Doctoral Dissertations

"Borate glasses have recently been developed for a variety of medical applications, but much less is known about their structures and properties than more common silicate glasses. Melt properties and crystallization tendency for compositions in the Na2O-CaO-B2O3 system were characterized using differential thermal analysis and viscosity measurements. Characteristic viscosity (isokom) temperatures varied with the ratio between the modifier content (Na2O+CaO) and B2O3, particularly at lower temperatures, consistent with the changes in the relative concentrations of tetrahedral borons in the glass structure.

Similar glasses were used to study dissolution processes …

Francisella Tularensis Lvs Invasion Of Primary Murine Macrophages And Mtor Signaling, Michael Warren Edwards Jan 2014

Francisella Tularensis Lvs Invasion Of Primary Murine Macrophages And Mtor Signaling, Michael Warren Edwards

All ETDs from UAB

Francisella tularensis is an intracellular pathogen and the etiologic agent of tularemia. Because virulent strains cause morbidity and lethality in human, the attenuated live vaccine strain (LVS) has been used to study F. tularensis pathogenesis in murine models, since the disease in mice resembles human tularemia. Despite growing knowledge about host responses to Francisella infection, there is scarce information on the cell signaling events involved in Francisella host cell entry and in early immune re-sponses. Therefore, the purpose of this dissertation was to assess the involvement of the mammalian target of rapamycin (mTOR) in these processes. First, we evaluated the …

The Role Of Wnt5a In Tumor Suppression And Differentiation, Stephanie Easter Jan 2014

The Role Of Wnt5a In Tumor Suppression And Differentiation, Stephanie Easter

All ETDs from UAB

TGF-beta and the Wnt signaling pathways play key roles in regulating ductal growth and branching morphogenesis in the mammary gland. Disruption to this signaling can cause deregulation of the normal mammary stem cell population and foster an environment favorable for tumorigenesis and recurrence. Recent studies show that TGF-beta up-regulates expression of the non-canonical Wnt, Wnt5a, in the mammary gland and that Wnt5a antagonizes canonical Wnt/beta-catenin signaling in mammary epithelial cells. Canonical Wnt/beta-catenin signaling has been strongly implicated in expanding the mammary stem/progenitor cell pool. Here, we test the hypothesis that Wnt5a suppresses the expansion of the mammary stem and progenitor …

Following the PRECEDE half of the PRECEDE-PROCEED planning model, the author examined the health issue of pedophilia within the State of Alabama. There are three constructs by which a health issue may be examined using PRECEDE: Predisposing, Reinforcing, and Enabling. The extant literature on pedophilia and child sexual abuse (CSA) was reviewed organized by these constructs. PRECEDE also consists of five assessment phases: social assessment; epidemiological assessment; behavioral assessment; educational, environmental, and ecological assessments; and administrative …

Mitochondrial Genetics And Cellular Metabolism Regulate Tumorigenicity And Metastatic Potential, Kyle Paul Feeley Jan 2014

Mitochondrial Genetics And Cellular Metabolism Regulate Tumorigenicity And Metastatic Potential, Kyle Paul Feeley

All ETDs from UAB

Current paradigms of carcinogenic risk suggest that genetic, hormonal, and environmental factors combine to influence an individual's predilection for breast cancer and related metastatic tumor formation. The genetic component, in particular, has become the focus of many emergent studies. A renewed focus on cancer metabolism and the Warburg effect has similarly cast a spotlight on the role, if any, of the mitochondrion in directing disease progression. Analysis of the direct contribution of mitochondrial DNA on tumorigenicity is made possible through the use of mitochondrial-nuclear exchange (MNX) mice in which nuclei from normal FVB mice (the background strain of the tg: …

An Evolutionary Study Of Dynamic Cognitive Game Captchas: Automated Attacks And Defenses, Song Gao Jan 2014

An Evolutionary Study Of Dynamic Cognitive Game Captchas: Automated Attacks And Defenses, Song Gao

All ETDs from UAB

CAPTCHAs represent a primary defense mechanism against online attacks and resource abuse. However, existing CAPTCHA solutions (e.g., distorted static text-based CAPTCHAs) suffer from significant security problems. Recent research has demonstrated that many current forms of CAPTCHAs can be solved with a high accuracy using automated techniques. This vulnerability associated with traditional CAPTCHAs thus provides a sound motivation to explore CAPTCHA alternatives. In this dissertation, we study a broad class of game-based CAPTCHAs, called the Dynamic Cognitive Game (DCG) CAPTCHAs, which challenge the user to perform a game-like cognitive task interacting with a series of dynamic objects. Specifically, we focus on …

Mitochondrial Dysfunction In Response To Neurotoxins And The Role Of Mitophagy, Samantha Giordano Jan 2014

Mitochondrial Dysfunction In Response To Neurotoxins And The Role Of Mitophagy, Samantha Giordano

All ETDs from UAB

Parkinson's disease (PD) is the second most common neurodegenerative disorder. Two major factors in both familial and sporadic PD are mitochondrial dysfunction and insufficient autophagy. My thesis research focuses on the interplay between these activities in PD. To investigate the common and differential effects of PD-inducing neurotoxins on mitochondrial bioenergetics and their relationships to cell survival, we used an in vitro culture system, differentiated dopaminergic SH-SY5Y neuroblastoma cells. We found that the neurotoxins rotenone, 1-methyl-4- phenylyridinium (MPP+) and 6-hydroxydopamine (6-OHDA), decreased mitochondrial respiration and induced cell death in these cells. The extent and characteristics of mitochondrial dysfunction in response to …

Regulation Of Skeletal Muscle Plasticity Using Engineered Configurations Of The Transcription Factors Myod And E12, Vladimir Grubisic Jan 2014

Regulation Of Skeletal Muscle Plasticity Using Engineered Configurations Of The Transcription Factors Myod And E12, Vladimir Grubisic

All ETDs from UAB

Skeletal muscle fibers, although terminally differentiated, can change their features, such as contraction speed and endurance, and throughout adult life in response to variety of stimuli, like exercise. This process is called plasticity and is mainly regulated by the type of nerve activity. Over the past several decades, research has shown that the properties of myofibers are also determined during muscle development. Ultimately, the change in phenotype requires a change in gene expression which is, in turn, regulated by transcription factors (TFs). Basic helix-loop-helix (bHLH) TFs regulate myogenesis and the central role belongs to the myogenic differentiation 1 (MyoD), often …

The Role Of Human Ficolin-2 During Invasive Pneumococcal Disease, Allison Marie Brady Jan 2014

The Role Of Human Ficolin-2 During Invasive Pneumococcal Disease, Allison Marie Brady

All ETDs from UAB

Streptococcus pneumoniae (pneumococcus) is a significant human pathogen that typically resides in the nasopharynx of humans. The major virulence factor of pneumococcus is its capsular polysaccharide, of which it can express over 90 unique serotypes. However, there is great disparity in which serotypes colonize or cause disease, though the reason for these differences are not well understood. Serotype 11A is one such serotype that frequently colonizes the nasopharynx of children, but rarely causes disease. Interestingly, serotype 11E, a variant of 11A, varies only in the O-acetylation of a galactose residue in its polysaccharide repeating unit. This difference in structure is …
