A serology-based tiered approach has, to date, provided the most effective means of laboratory confirmation of clinically suspected cases of Lyme disease, but it lacks sensitivity in the early stages of disease and is often dependent on subjectively scored immunoblots. We recently demonstrated the use of immuno-PCR (iPCR) for detecting Borrelia burgdorferi antibodies in patient serum samples that were positive for Lyme disease. To better understand the performance of the Lyme disease iPCR assay, the repeatability and variability of the background of the assay across samples from a healthy population (n = 36) were analyzed. Steller sea lion (Eumetopias jubatus) populations have undergone precipitous declines through their western Alaskan range over the last four decades with the leading hypothesis to explain this decline centering around changing prey quality, quantity, or availability for this species (i.e., nutritional stress hypothesis). Under chronic conditions of reduced food intake sea lions would conserve energy by limiting energy expenditures through lowering of metabolic rate known as metabolic depression. Mediators of monocyte migration, complement receptor-3 (CR3), and chemokine ligand-4 (CCL4) were measured in response to recovery modalities following resistance exercise. Thirty resistance-trained men (23.1 +/- 2.9 y; 175.2 +/- 7.1 cm; 82.1 +/- 8.4 kg) were given neuromuscular electric stimulation (NMES), cold water immersion (CWI), or control (CON) treatments immediately following resistance exercise. Blood samples were obtained preexercise (PRE), immediately (IP), 30 minutes (30 P), 24 hours (24 H), and 48 hours (48 H) after exercise for measurement of circulating CCL4 and CR3 expression on CD14+ monocytes, by assay and flow cytometry. Limnology has been greatly influenced by The lake as a Microcosm (Forbes, 1887), which described a holistic focus on the internal machinations of singular, island-like aquatic ecosystems. I consider three persistent influences of The lake as a Microcosm: as an organizing paradigm for the teaching of limnology relative to its practice; the idea that inland waters are like islands, and the replicability of types of inland waters. Based on inspection of recent peer-reviewed literature and 32 limnology texts, we teach limnology according to Forbes but do not practice it in that holistic context. Instead, we practice limnology as aquatic ecology. …

The effective delivery of therapeutics to disease sites significantly contributes to drug efficacy, toxicity and clearance. Here we demonstrate that clinically approved iron oxide nanoparticles (Ferumoxytol) can be utilized to carry one or multiple drugs. These so called 'nanophores' retain their cargo within their polymeric coating through weak electrostatic interactions and release it in slightly acidic conditions (pH 6.8 and below). The loading of drugs increases the nanophores' transverse T2 and longitudinal T1 nuclear magnetic resonance (NMR) proton relaxation times, which is proportional to amount of carried cargo. The purpose of this research study was to explore the impact of virtual character design on user emotional experience and user behavior in a simulated environment. With simulation training increasing in popularity as a tool for teaching social skills, it is essential that social interactions in virtual environments provide authentic opportunities for practice (Swartout et al., 2006). Establishing a chronology of variation in isotopic values can reveal frailty associated with biological and social age, as well as highlight individuals who vary from typical patterns. Although general dietary characteristics and infant feeding practices were previously unknown for subadults excavated from the cemetery at Alytus, Lithuania (14th-18th centuries), previous research concludes that Alytus' subadults experienced high rates of physiological, metabolic, non-specific stress, in addition to specific diseases like tuberculosis. Staphylococcus aureus (SA), an opportunistic pathogen colonizing the anterior nares in approximately 30% of the human population, causes severe hospital-associated and community-acquired infections. SA nasal carriage plays a critical role in the pathogenesis of staphylococcal infections and SA eradication from the nares has proven to be effective in reducing endogenous infections. Pinnipeds exhibit a wide range of lactation strategies that vary from just a few days to nearly three years in duration. Phocids have a relatively short, intense nursing period culminating with weaning after just a few days or weeks, while dependent otariids generally take several months of consuming a combined milk and solid food diet before being completely independent. This study identified what Twitter followers were discussing in reference to the top five megachurches in size. The methods utilized within this study involved an examination of all tweets hashtagging the top five megachurches over a sixteen day timeframe. Background N-terminal peptide of procollagen type III (P3NP) and C-terminal agrin fragment (CAF) are circulating biomarkers that are related to lean body mass in older adults. P3NP is a circulating marker reflective of muscular structural remodeling while CAF is a circulating marker of neuromuscular remodeling. As resistance exercise is an established intervention that can effectively improve muscle quality, we sought to evaluate circulating biomarker changes corresponding to a resistance exercise intervention in older adults. Maternal cigarette smoking has adverse effects on pregnancy outcomes. The granulocyte-macrophage colony-stimulating factor (GM-CSF) is an essential cytokine for a normal pregnancy. We investigated the impact of cigarette smoke extract (CSE) on GM-CSF expression in human cytotrophoblast cells and suggested a cellular mechanism underlying the CSE-induced GM-CSF expression. An immortalized normal human trophoblast cell line (B6Tert-1) was treated with CSE. The viability and proliferation of the CSE-treated B6Tert-1 cells were evaluated, and the expression of GM-CSF in these cells was quantified at the mRNA and the protein levels by means of reverse-transcription and quantitative polymerase chain reaction (RT-qPCR); and enzyme-linked …

Even though this is a well-explored topic, many challenges remain when confronted with data from real world situations. This study investigates the representation of gender in technology-related video advertisements. This thesis quantitatively and qualitatively examined 54 of the most recent commercials by the top nine Fortune 500 technology companies. A total of 407 characters were coded and quantitatively analyzed while the videos themselves were qualitatively assessed with particular attention given to the videos' themes and the interactions between the characters and the technology products and services. Results of the analyses showed that there were more male, Caucasian characters than any other character type based on gender and race/ethnicity. Taking The Strict Account Of Techne Seriously: An Interpretive Direction In Plato's Republic, Kenneth Knies

I argue that the strict account of techne agreed to by Socrates and Thrasymachus in Republic I provides a useful framework for addressing a central question of the dialogue as a whole: how philosophy might belong to the polis. This view depends upon three positions: 1) that Plato invites us to interpret the relationship between techne and polis outside the terms of the city-soul analogy, 2) that the strict account contributes to a compelling description of vocational work, and 3) that this description determines what Socrates means by a true polis, and thus frames the problem of philosophy’s political inclusion.

Currently, we do not understand how much biodiversity may be maintained in any environment, especially not in the structured environments of biofilms, which constitute the dominant mode of microbial life. Although maintenance is associated with the complex spatial structure of biofilm architecture and resulting ecological opportunities, how both the abiotic and biotic environment defines available niches remains poorly understood. Freshwater wetland ecosystems are a valuable resource, but current policies fail to prevent their continuing destruction. Policy-makers increasingly use decentralized wetland-buffer programs to address such policy failings, but scant research has evaluated whether these programs are both ecologically and socially effective. In the following dissertation, I address this gap using two complementary projects.
For the first project, I assessed whether forested buffers are an effective tool for maintaining viable populations of pool-breeding amphibians. Such species spend their early life stages in vernal pools, which are small, highly productive wetlands, but use the surrounding forest during juvenile and adult life-stages. Though …
