Proximity-Labeling Of Near Neighbors Of Lamin A And Lamin A-Δ50 (Progerin)., Mohammad Sabri

In an attempt to isolate and identify proteins that differentially interact with or locate near lamin A and progerin, we used a previously described method named BioID (proximity-dependent biotin identification). This method is based on fusion of a promiscuous E. coli biotin-protein ligase (BL) to a targeting protein (in this study, lamin A-GFP and progerin-GFP). The biotin ligase biotinylates amino residues in proteins that are near-neighbors of the fusion protein. To create the fusion proteins, BL was sub-cloned from a pcDNA3.1 MCS-BirA(R118G)-HA plasmid donated by Kyle Roux from University of South Dakota. The BL fragment was ligated into a pNEBR-X1-lamin …

This study analyzes the association between walkability and housing value in neighborhoods of Lincoln, Nebraska in order to observe the impact of walkability as one of prominent smart growth principles in creating economic value. The study estimates walkability through walkability components in each neighborhood of Lincoln based on the Lawrence Frank walkability index model. For the housing value assessment, a hedonic regression model was created to estimate median housing value in Lincoln based on the census block group dataset. Methicillin-resistant Staphylococcus aureus (MRSA) is a problem nationally and affects many people from all walks of life. Community associated MRSA is often ignored and not viewed as significant, thus there is a lack of education on the topic. One area that is at a particular risk for the transmission of Community Associated MRSA is physical activity centers. There have been previous hints that the transiting planet WASP-3b is accompanied by a second planet in a nearby orbit, based on small deviations from strict periodicity of the observed transits. Here we present 17 precise radial velocity (RV) measurements and 32 transit light curves that were acquired between 2009 and 2011. These data were used to refine the parameters of the host star and transiting planet. This has resulted in reduced uncertainties for the radii and masses of the star and planet. The RV data and the transit times show no evidence for an additional planet in the system. …

OBJECTIVE: To examine the incidence and reverse of obesity among young low-income children and variations across population subgroups.

METHODS: We included 1.2 million participants in federally funded child health and nutrition programs who were 0 to 23 months old in 2008 and were followed up 24 to 35 months later in 2010–2011. Weight and height were measured. Obesity at baseline was defined as gender-specific weight-for-length >/=95th percentile on the 2000 Centers for Disease Control and Prevention growth charts. Obesity at follow-up was defined as gender-specific BMI-for-age >/=95th percentile. We used a multivariable log-binomial model to estimate relative risk of …

Assessment of drug susceptibility has become an integral part of influenza virus surveillance. In this study, we describe the drug resistance profile of influenza A(H3N2) virus, A/Mississippi/05/2011, collected from a patient treated with oseltamivir and detected via surveillance. An MDCK cell-grown isolate of this virus exhibited highly reduced inhibition by the neuraminidase (NA) inhibitors (NAIs) oseltamivir (8,005-fold), zanamivir (813-fold), peramivir (116-fold), and laninamivir (257-fold) in the NA inhibition assay. Sequence analysis of its NA gene revealed a known oseltamivir-resistance marker, the glutamic acid-to-valine substitution at position 119 (E119V), and an additional change, threonine to isoleucine at position 148 (T148I). Adenocarcinoma of pancreas is recognized for its poor prognosis, as it progresses asymptomatically and is rarely diagnosed at early stage. According to American cancer society pancreatic cancer, it has lowest survival rate in all cancer types, with less than 6% five year survival rate. Surgical resection is only possible for 20% of diagnosed cases and current chemotherapy has only 15% response rate. Even the most favored drug, Gemcitabine, increases survival time only by a few months, depending on the stage at diagnosis. Some bioactive food components or nutraceuticals have shown chemopreventive and chemotherapeutic effects without added side effects. Garcinol is …

This study uses practitioner research to examine secondary mathematics teachers’ learning of literacy integration practices in the context of a district-wide literacy professional development series. The author, a secondary mathematics curriculum and instruction facilitator in a large, Midwestern suburban district, engaged in a two-year partnership with seventeen Mathematics Teacher Facilitators (MTFs) who taught literacy practices to their colleagues via a train-the-trainer model. This study provides an explicit rendering of professional development practices and ongoing, job-embedded learning vignettes of six MTF’s experiences in (a) teaching literacy practices to their colleagues and (b) how they learned and enacted these practices in their …

The Impact Of Caring For Children With Mental Retardation On Families As Perceived By Mothers In Karachi, Pakistan., Arusa Lakhani, Irma Gavino, Aisha Yousafzai

OBJECTIVE:

To assess how families perceive the positive and negative impacts of caring for a child with mental retardation.

METHODS:

The quantitative descriptive study was conducted from January to August 2007 and comprised 54 families attending a private day-care centre for children with special needs in Karachi, Pakistan. The Kansas Inventory of Parental Perceptions was used to assess mothers' perceptions on the impact of caring for a child with mental retardation. Positive contributions, social comparisons with others, understanding of disability and perception of control were assessed. SPSS 16 was used for statistical analysis.

The present study investigated the induction and possible role of activating transcription factor 3 (ATF3) in the corpus luteum. Postpubertal cattle were treated at midcycle with prostaglandin F2α(PGF) for 0–4 hours. Luteal tissue was processed for immunohistochemistry, in situ hybridization, and isolation of protein and RNA. Ovaries were also collected from midluteal phase and first-trimester pregnant cows. Luteal cells were prepared and sorted by centrifugal elutriation to obtain purified small (SLCs) and large luteal cells (LLCs). Real-time PCR and in situ hybridization showed that ATF3 mRNA increased within 1 hour of PGF treatment in vivo. Western blot and immunohistochemistry demonstrated …

Therefore, appellate cases continue to emerge that interpret and apply the former rules, providing insight and raising questions about how the new rules have changed the face of evidence in Georgia. Polycarbonate (PC) is a tough, transparent engineering thermoplastic. Its impact strength and ability undergo large plastic deformations without shatter make PC an ideal protective material for impact-resilient eyewear, aircraft windows and transparent armor. A good understanding of the response of this material to large deformations at high strain rates is critical for its utilization in these applications. To this end, a striker-less Kolsky bar device is employed in this work for the needed material characterization.
