Protein-protein interactions regulate key cellular functions and cell signaling pathways in the body; biological systems can be disrupted by protein interactions or lack of interactions that cause cell dysfunction and can lead to disease or illness. However, studying these interactions is complex and difficult to quantify. In this project, we examine the interaction of two putative cancer biomarkers, Bax and PDCDS, using the tetracycline repressor-based mammalian two-hybrid system (trM2H). Compassion fatigue is a term used to describe the unique stressors affecting people in care-giving professions (Joinson in 1992). For nurses, the impact of compassion fatigue may result in stress-related symptoms, job dissatisfaction, decreased productivity, patient satisfaction, safety issues, and job turnover. Atmospheric inversions have become an important tool in quantifying carbon dioxide (CO2) sinks and sources at a variety of spatiotemporal scales, but associated large uncertainties restrain the inversion research community from reaching agreement on many important subjects. We enhanced an atmospheric inversion of the CO2 flux for North America by introducing spatially explicit information on forest stand age for US and Canada as an additional constraint, since forest carbon dynamics are closely related to time since disturbance. More than 40 million Americans suffer from the pains of arthritis. Arthritis is characterized by joint inflammation and cartilage deterioration (3). For many years growth differentiation factor 5 (Gdf5) has been associated with cartilage formation and maintenance (4, 1). GDF5 has been traditionally regarded as a secreted growth factor; however, a new variant of Gdf5 was recently discovered in the nucleus. Nuclear Gdf5 (nGdf5) may present a new mechanism by which Gdf5 regulates cartilage development and preservation.

Metabolomics is an emerging field that entails the detailed characterization of the ensemble of metabolites produced by living organisms; subfields include drug metabolism and natural environmental toxin production. The first part of the dissertation pursued metabolism of glyceollins, i.e., isoflavones produced by soybeans, that are potential cancer therapy agents. In vivo glyceollin metabolites produced in rats were investigated by on-line Liquid Chromatography-Electrospray Ionization Tandem Mass Spectrometry. An odd-electron fragment ion at m/z 148, formed in violation of the even-electron rule, and diagnostic of the glyceollin backbone, was discovered. In the U.S., about 7,000 high school students drop out each school day, representing a loss of talent and ability. Concurrently, there are a decreasing number of enrolled students taking science-related courses at the high school and college levels. Adults, who return to obtain their General Educational Development (GED) certification, are an untapped resource that could be steered toward STEM (Science, Technology, Engineering and Mathematics) careers. Remote sensing provides a variety of methods for classifying forest communities and can be a valuable tool for the impact assessment of invasive species. The emerald ash borer (Agrilus planipennis) infestation of ash trees (Fraxinus) in the United States has resulted in the mortality of large stands of ash throughout the Northeast. This study assessed the suitability of multi-temporal Worldview-2 multispectral satellite imagery for classifying a mixed deciduous forest in Upstate New York. Background Human rights approaches to manage HIV and efforts to decriminalize HIV exposure/transmission globally offer hope to persons living with HIV (PLWH). However, among vulnerable populations of PLWH, substantial human rights and structural challenges (disadvantage and injustice that results from everyday practices of a well-intentioned liberal society) must be addressed. Recent studies of hoplonenemertean planuliform larvae have clarified their development and provided insight into larval evolution within the phylum. However, an assessment of viviparous development using modern techniques is lacking. To help facilitate a comprehensive comparative evaluation of developmental diversity within hoplonemerteans, we have conducted a confocal laser scanning microscopy investigation of the development in Prosorhochmus americanus, one of the few viviparous hoplonemertean species. Phalloidin staining provides evidence of a modified transitory larval epidermis, and reveals that the foregut, midgut, proboscis, central nervous system, and body wall musculature form early in development, consistent with observations for planktonic and encapsulated hoplonemertean …

Ubiquitination by HECT E3 enzymes regulates myriad processes, including tumor suppression, transcription, protein trafficking, and degradation. HECT E3s use a two-step mechanism to ligate ubiquitin to target proteins. The first step is guided by interactions between the catalytic HECT domain and the E2∼ubiquitin intermediate, which promote formation of a transient, thioester-bonded HECT∼ubiquitin intermediate. Here we report that the second step of ligation is mediated by a distinct catalytic architecture established by both the HECT E3 and its covalently linked ubiquitin. Coupling S100a4 To Rhotekin Alters Rho Signaling Output In Breast Cancer Cells, Min Chen, Anne R. Bresnick, Kathleen L. O'Connor

Rho signaling is increasingly recognized to contribute to invasion and metastasis. In this study, we discovered that metastasis-associated protein S100A4 interacts with the Rho-binding domain (RBD) of Rhotekin, thus connecting S100A4 to the Rho pathway. Glutathione S-transferase pull-down and immunoprecipitation assays demonstrated that S100A4 specifically and directly binds to Rhotekin RBD, but not the other Rho effector RBDs. S100A4 binding to Rhotekin is calcium-dependent and uses residues distinct from those bound by active Rho. Interestingly, we found that S100A4 and Rhotekin can form a complex with active RhoA. Geochemical Tracers Linking Submarine Groundwater Discharge To Hypoxia Formation In Long Bay, South Carolina, Usa, Sarah Lynn Chappell

Long Bay, South Carolina has experienced hypoxic conditions (<2mg/L dissolved oxygen) in the nearshore environment, not fully attributed to traditional formation mechanisms. Past research suggested physical, biological, and/or anthropogenic influences on low dissolved oxygen levels. This project aimed to determine the contribution of submarine groundwater discharge (SGD) to hypoxia formation. We measured activities of 222Rnxs, 224Raxs, 223Ra, 228Ra, and 226Ra (3.8d, 3.6d, 11.5d, 5.8yr, 1600yr half-lives, respectively) in nearshore bottom waters from April 2012 through April 2013. Radium activities observed during a hypoxic event on 16-Aug-2012 were up to an order of magnitude higher than those seen during oxic conditions and the highest ever observed in the open ocean, to our knowledge. We determined that a hypoxic water mass, comprised heavily of offshore anoxic SGD, migrated inshore due to physical conditions constraining mixing over a two week time period. High groundwater content suggests that observed hypoxic conditions in nearshore Long Bay may be independent of biological influence once offshore SGD occurs.

This study examined the influence of socio-demographic factors on access and use of ICT by staff of academic libraries in Oyo state. These universities are: Ladoke Akintola University, Lead City University and Bishop Ajayi Crowther.

Data was collected using a self structured questionnaire using convenience sampling technique. The empirical evidence was based on descriptive and statistical analysis using SPSS. The findings revealed that ease of use, perceived benefits and accessibility were highly correlated with the demographic factors (age, level of education and income), however, none of the demographic variables predicted favorably on challenges of ICT usage.

Objectives: Analyses the Indian publications output in glaucoma research during 2002-11 on several parameters including contribution & citation impact of top most productive countries, India’s overall contribution, its growth pattern and citation impact, the share of international collaboration in India’s overall research output, contribution of leading countries and identification of leading foreign collaborating partners, Indian contribution & impact of different types of glaucoma, glaucoma research by sub-fields and glaucoma research output by different population age groups, productivity and impact of leading Indian institutions and authors and pattern of communication of Indian output in most productive journals.

Mining activity in Butte, Montana has taken place, or continues to take place, within the urban residence of Butte itself. This has led to urban areas with high concentrations of toxic metals such as arsenic, lead, copper, zinc, mercury and cadmium. Advances in protein study and gene sequencing has opened the possibility of finding molecular biomarkers whose presence, absence or morphological changes could indicate disease processes in populations exposed to environmental toxins.
