Cooperative Tumor Suppression By Arf And P53, Jason Thomas Forys Nov 2013

Cooperative Tumor Suppression By Arf And P53, Jason Thomas Forys

All Theses and Dissertations (ETDs)

Cancer is a complex genetic disease characterized by the inactivation of tumor suppressor genes and enhanced activity of oncogenes leading to deregulated cellular proliferation. Two tumor suppressor genes, p53 and Arf, play important roles in protecting cells against numerous biological stresses. In response to oncogenic signals, increased ARF expression leads to the activation of p53, which in turn leads to the cessation of cell division or induction of an apoptotic response. Interestingly, p53 coordinates repression of Arf transcription, setting up a negative feedback loop with currently unknown physiological significance. Cells that lack p53 express elevated levels of ARF, but it …

Trabecular Evidence For A Human-Like Gait In Australopithecus Africanus, Meir M. Barak, Daniel E. Lieberman, David Raichlen, Herman Pontzer, Anna G. Warrener, Jean-Jacques Hublin

Publications and Research

Although the earliest known hominins were apparently upright bipeds, there has been mixed evidence whether particular species of hominins including those in the genus Australopithecus walked with relatively extended hips, knees and ankles like modern humans, or with more flexed lower limb joints like apes when bipedal. Here we demonstrate in chimpanzees and humans a highly predictable and sensitive relationship between the orientation of the ankle joint during loading and the principal orientation of trabecular bone struts in the distal tibia that function to withstand compressive forces within the joint. Analyses of the orientation of these struts using microCT scans …

Taxis Toward Hydrogen Gas By Methanococcus Maripaludis, Kristen A. Brileya, James M. Connolly, Carey Downey, Robin Gerlach, Matthew W. Fields

Biology Faculty Publications and Presentations

Knowledge of taxis (directed swimming) in the Archaea is currently expanding through identification of novel receptors, effectors, and proteins involved in signal transduction to the flagellar motor. Although the ability for biological cells to sense and swim toward hydrogen gas has been hypothesized for many years, this capacity has yet to be observed and demonstrated. Here we show that the average swimming velocity increases in the direction of a source of hydrogen gas for the methanogen, Methanococcus maripaludis using a capillary assay with anoxic gas-phase control and time-lapse microscopy. The results indicate that a methanogen couples motility to hydrogen concentration …

Optical Imaging Of Breast Cancers, Yan Xu Nov 2013

Optical Imaging Of Breast Cancers, Yan Xu

Doctoral Dissertations

Breast cancer is the most common cancer and the second major cause of cancer death among women. Screening tries to detect the disease in the early stage, and can save more lives. Diffuse optical tomography (DOT) in the near-infrared (NIR) spectrum is a promising noninvasive approach for functional diagnostic breast imaging. Our group has explored the use of ultrasound (US)-guided DOT to improve lesion localization and light quantification accuracy. In this dissertation, several special cases of breast cancer imaging were studied with theUSguided DOT technique.

Firstly, the heterogeneous absorption distributions of advanced cancers were characterized. A series of simulations and …

National Park Service Cave Ecology Inventory And Monitoring Framework, Gretchen M. Baker, Steven J. Taylor, Shawn Thomas, Rick Olson, Kathy Lavoie, Marie Denn, Steven Thomas, Hazel Barton, Kurt Helf, Rene Ohms, Joel Despain, Jim Kennedy, David Larson

National Cave and Karst Management Symposium 2013

A team developed the Cave Ecology Inventory and Monitoring Framework for National Park Service (NPS) units. It contains information for NPS cave managers across the United States to determine how to inventory and monitor cave ecology. Due to the wide geographical scope of NPS caves and their many different types, the document does not prescribe exact protocols. Instead, it provides guidance for what types of inventory and monitoring are possible, a framework for deciding how to prioritize inventory and monitoring activities, and references to specific protocols that are already in place at NPS cave parks.

Keywords: cave ecology, cave microbiology, …

Geologic Management Of Cave And Karst Resources On National Forest System Lands, Johanna L. Kovarik

National Cave and Karst Management Symposium 2013

The United States Department of Agriculture Forest Service (Forest Service) manages caves, karst systems, and associated resources on over 780,000 square kilometres of public land. The management of these resources is mandated by the Federal Cave Resources Protection Act (FCRPA) of 1988 and other federal acts, and is guided by the Code of Federal Regulations (CFR) and the Forest Service Manual (FSM). The FCRPA and its implementing regulations at 36 CFR Part 290 – Cave Resources Management provide the framework for management guidance in the FSM. Known caves and karst areas occur in over 100 National Forests and Grasslands, and …

Conceptualizing Communication Capital For A Changing Environment, Leo Wayne Jeffres, Guowei Jian, Sukki Yoon Nov 2013

Conceptualizing Communication Capital For A Changing Environment, Leo Wayne Jeffres, Guowei Jian, Sukki Yoon

Communication Faculty Publications

With rapidly evolving technologies, boundaries between traditional modes of communication have blurred, creating an environment that scholars still describe from viewpoints as researchers in interpersonal, organizational or mass communication. This manuscript looks at the social capital literature and argues for conceptualizing “communication capital” to help understand the impact of communication phenomena in a changing environment. The literature has treated interpersonal communication variables as components of social capital and mass communication variables as factors affecting social capital, but scholars long ago recognized their reinforcing nature, leading us to develop a concept of communication capital merging symbolic activity across domains in its …

Keap1-Knockdown Decreases Fasting-Induced Fatty Liver Via Altered Lipid Metabolism And Decreased Fatty Acid Mobilization From Adipose Tissue, Jialin Xu, Ajay C. Donepudi, Jamie E. Moscovitz, Angela Slitt

Biomedical and Pharmaceutical Sciences Faculty Publications

Aims: The purpose of this study was to determine whether Nrf2 activation, via Keap1-knockdown (Keap1-KD), regulates lipid metabolism and mobilization induced by food deprivation (e.g. fasting).

Methods and Results: Male C57BL/6 (WT) and Keap1-KD mice were either fed ad libitum or food deprived for 24 hours. After fasting, WT mice exhibited a marked increase in hepatic lipid accumulation, but Keap1-KD mice had an attenuated increase of lipid accumulation, along with reduced expression of lipogenic genes (acetyl-coA carboxylase, stearoyl-CoA desaturase-1, and fatty acid synthase) and reduced expression of genes related to fatty acid transport, such as fatty acid translocase/CD36 …

Managing A Globally Unique Nexus Of Acid Mine Drainage, Karst, And World Heritage Site, Peter J. Mills, Pj (Phil) Hobbs

National Cave and Karst Management Symposium 2013

The Cradle of Humankind World Heritage Site (COH WHS), South Africa, is located downstream of the actively decanting West Rand Goldfield (a.k.a. the Western Basin). It is therefore the only UNESCO-protected karst landscape in the world that is under threat from acid mine drainage (AMD). The perceived threat has generated wide and considerable concern for the preservation of the fossil sites. This perception has been fuelled by a poor understanding of the surface and groundwater resources of the WHS which, in turn, has precipitated often alarmist reporting in the media, some even hinting at the possible delisting of the site …

Incorporating Cave And Karst Management Into The Forest Plan Revision Process Of Arizona Forests, Ray Keeler, Richard Bohman Nov 2013

Incorporating Cave And Karst Management Into The Forest Plan Revision Process Of Arizona Forests, Ray Keeler, Richard Bohman

National Cave and Karst Management Symposium 2013

Arizona National Forest land managers have a multitude of tasks and priorities; historically, caves and karst management has not been amongst the priorities receiving any significant allocation of resources. When caves and karst management is not included in the Forest Plan, even when large and significant cave and karst areas exist, active cave and karst management often falls below the waterline of available manpower and resources. Additionally, there is entropy associated with normal changes in personnel assignments and new staff coming onboard. When combined with a general lack of written policies and guidelines, these personnel transitions lead to unnecessary degradation …

Postglacial Faunal Records From Cave Deposits In Jasper National Park, Alberta, Canada, Christopher N. Jass, Greg Horne, Dave Critchley

National Cave and Karst Management Symposium 2013

In 2009, we initiated an on-going, long term research project that focuses on exploration of fossil-bearing cave deposits in Jasper National Park. Specifically, we were interested in understanding patterns of mammalian re-colonization of mountainous regions following late Pleistocene deglaciation. Our work has focused on the identification of fossil-bearing cave deposits, excavation and sampling of those deposits, and radiocarbon dating of recovered remains. Examined sites have at least an age of 9600 years BP. Research at four cave sites, located in relatively close proximity to one another, is contributing to an improved understanding of the late Quaternary record of Jasper National …

Role Of Faah-Like Anandamide Transporter In Anandamide Inactivation, Kwannok Leung, Matthew W. Elmes, Sherrye T. Glaser, Dale G. Deutsch, Martin Kaczocha

Publications and Research

The endocannabinoid system modulates numerous physiological processes including nociception and reproduction. Anandamide (AEA) is an endocannabinoid that is inactivated by cellular uptake followed by intracellular hydrolysis by fatty acid amide hydrolase (FAAH). Recently, FAAH-like anandamide transporter (FLAT), a truncated and catalytically-inactive variant of FAAH, was proposed to function as an intracellular AEA carrier and mediate its delivery to FAAH for hydrolysis. Pharmacological inhibition of FLAT potentiated AEA signaling and produced antinociceptive effects. Given that endocannabinoids produce analgesia through central and peripheral mechanisms, the goal of the current work was to examine the expression of FLAT in the central and peripheral …

Cooperative Cave Management In The Era Of Wns, Scott House

National Cave and Karst Management Symposium 2013

With declining budgets, less money than ever is available for meaningful cave management, including baseline inventories and monitoring. The spread of White Nose Syndrome (WNS) has made it even more difficult to effectively manage cave resources. In the region of the Ozark Plateau, Cave Research Foundation is continuing to provide these services, and more, by working within guidelines of various agencies and private landowners. These include Ozark National Scenic Riverways and Buffalo National River (NPS) Mark Twain National Forest (USFS) the Missouri Department of Conservation, Missouri Department of Natural Resources – Division of State Parks, U.S. Fish and Wildlife Service, …
