This method is validated by referring to the APHA 3500 B- Cr Colorimetric Method. The current colorimetric method of determination of hexavalent chromium in wastewater is carried out by using UV-VIS Spectrocopy. A study on the development, effects and control of biofouling on replanted mangrove seedlings (40-50cm height) was carried out, from April 2008 to July 2010, at Kampung Sungai Haji Dorani and Kampung Sungai Limau, Selangor, Malaysia. The dominant biofoulers were barnacles (Amphibalanus amphitrite), which were the main causal organism of mangrove seedling damage or death. The high density (>100 no./ 10 cm section of plant stem) and cover (>30 %) of barnacles appeared to obstruct development of new leaves, shoots and roots, as well as imposing weight loads as high as six times the weight of the seedlings. …

Bactrocera carambolae and Bactrocera papayae fruit flies, also known as the carambola and papaya fruit fly respectively, are significant pest fruit flies in Malaysia. They are capable of causing losses in the agricultural industry by infesting host fruits and making them unable to be sold. These two species are grouped in the Bactrocera dorsalis species complex, and are oftentimes difficult to distinguish from one another due to similar and intermediate morphological characteristics. ST332 is a 332 bp gene marker used in a patented Salmonella kit for detection of foodborne pathogen, Salmonella Typhi. As the gene is specific in DNA based detection by using polymerase chain reaction (PCR), its specificity in immunoassay is studied. For this purpose, an expression system of the gene is constructed and the expressed recombinant ST332 protein is tested for reactivity by hybridizing it towards typhoid patients sera through dot blotting. A nationwide investigation was carried out to (1) determine the distribution of Culex quinquefasciatus and other species of mosquitoes in stagnant water in residential areas, (2) investigate the genetic diversity of Cx. quinquefasciatus, (3) quantify the insecticide susceptibility status of Cx. quinquefasciatus, (4) characterize the biochemical mechanisms of insecticide resistance in Cx. quinquefasciatus, (5) characterize the molecular mechanisms of insecticide resistance in Cx. quinquefasciatus from 13 states and one federal territory in Malaysia. Streptococcus salivarius is known to produce different bacteriocin-like inhibitory substances (BLIS). In this study six S. salivarius strains were isolated from different Malaysian subjects. The isolates were identified using morphological and biochemical characterization and 16S rDNA gene sequencing. Levansucrase enzyme was characterized from sucrose-enriched culture of strain S. salivarius YU10. BLIS production was investigated using simultaneous and deferred antagonism tests that showed YU10 and NU10 as the best BLIS-producing strains among the Malaysian isolates in this study. Strain HJEFF was the only S. salivarius isolate which produced a more limited quantity of BLIS in liquid medium. However, all other strains …

Quorum sensing (QS) is a communication tool of bacteria that is involved in the regulation of bacterial virulence determinants, therefore making it an interesting target to attenuate pathogens. In this study, the anti-QS activities of four Malaysia‟s edible, local plants, namely, Melicopelunu-ankenda, Piper betle, Gnetumgnemon and Piper nigrum were explored by testing the hexane, chloroform and methanol extracts of the plants on Chromobacteriumviolaceum CV026, Escherichia coli [pSB401], E. coli [pSB1075],Pseudomonas aeruginosa PA01 and P. aeruginosaPA01lecA::lux. Quorum sensing (QS) is a phenomenon which refers to bacterial cell-to-cell communication in a population manner. QS regulates diverse activities in bacteria. Bacteria that possess QS activity communicate through secreted signal molecules or autoinducer. The Gram-negative bacteria and archaeon use acyl homoserine lactones (AHLs) as a QS signal to achieve regulation through the luxI-luxR homolog system, wheareas Gram-positive bacteria use small peptides as autoinducers. One of the biggest problems caused by soft drink poly(ethylene terephthalate) PET bottles is the waste problem resulting in environmental pollution. To economically reduce this waste, this study presents a method to recycle the PET bottle by glycolysis and convert it into a polyester polyol that can be used to make polyurethane foam. Common soft-drink PET bottles were glycolysed by glycerol in the presence of zinc acetate as a catalyst. Dependence on glycolysis conversion on reaction temperature and the Arrhenius plot, activation energy obtained of 36.5 kJ/mol. The present study investigated the effects of Allium sativum or Cinnamomum verum water extract on fermentation of milk and subsequent changes in yogurt bacteria growth and fermentation products on the organoleptic, nutritional and functional values of yogurt. Three types of milk (cow, camel and goat) were incubated (41°C) with starter culture in the presence of A. sativum or C. verum water extract until pH of yogurt was reduced to 4.5. The health of our environment can be measured by monitoring moths. Moths are especially useful as biological indicators given that they are sensitive to environmental changes, widespread and can be found in various habitats. Therefore, by monitoring their ranges and numbers, we can obtain essential clues about our changing environment such as climate change, air pollution and the effects of new farming practices. The antifeedant effects studies of Alpinia galanga rhizome and Chrysanthemum indicum on Coptotermes gestroi, Coptotermus curvignathus and Macrotermus carbonarius were conducted in the laboratory. Dual choice bioassays were conducted using methanolic extract and essential oil of A. galanga. Two paper discs (4.0 cm diameter) were placed in Petri dishes (9 cm diameter). One disc treated with A. galanga or C. indicum extract and another disc was treated with solvent hexane or methanol as control. Ten termites were placed in the treated petri dish. Percentage of antifeedant was determined by calculating the difference in the paper consumption of treated and control. …

The phytochemical and bioactivity studies on Mitrella kentii species from Annonaceae family have been performed. The extraction of phytochemicals from dried and ground plants were performed using conventional method extractions; Soxhlet extractor and Cold-extraction. The isolation of pure compounds from crude extracts were carried out by using various chromatographic methods such as column chromatography (CC), thin layer chromatography (TLC) and preparative thin layer chromatography (PTLC). MicroRNAs (miRNAs) are endogenous 18-25nt RNAs that regulate genes at the protein translation level. They can act as tumor suppressors or oncogene and are often dysregulated in various cancers. ANXA7 is a tumor suppressor gene that encodes for a Ca2+-dependent membrane-binding protein and its expression was found to be reduced or lost in advanced androgen-independent prostate cancers. This research aims to identify miRNAs that are regulated by the ANXA7 tumor suppressor gene and the putative pathways involved in prostate cancer. Morphologies and morphometries as diagnostic characters are shown statistically to be able to differentiate between species and intraspecific variants. Intraspecific variants or morphovariants exist in nature but the variations they possessed are not enough to consider them as species (differentiation index values for morphovariants is 50% less than the values for species). These variations are due to genetic difference resulting from cross-fertilisations between different individuals of the same species. Allamanda cathartica L. is a potential medicinal plant which starts to achieve public awareness due to its nutritional value that able to treat various types of diseases, especially in the treatment of jaundice, malaria and cancer. Tissue culture studies were undertaken to examine the effects of different plant growth regulators (PGRs) on the callus induction from leaf and stem explants of this species.
