Influence Of Exposure To Benzo[A]Pyrene On Mice Testicular Germ Cells During Spermatogenesis, Hueiwang Anna Jeng, Silvina M. Bocca

The objective of this study was to assess the toxicological effect of exposure to benzo(a)pyrene, B[a]P, on germ cells during spermatogenesis. Mice were exposed to B[a]P at 1, 10, 50, and 100 mg/kg/day for 30 days via oral ingestion. Germ cells, including spermatogonia, spermatocytes, pachytene spermatocytes, and round spermatids, were recovered from testes of mice exposed to B[a]P, while mature spermatozoa were isolated from vas deferens. Reproductive organs were collected and weighed. Apoptotic response of germ cells and mature spermatozoa were qualified using the terminal deoxynucleotidyl transferase mediated deoxy-UTP nick end labeling (TUNEL) assay. B[a]P exposure at

Size And Age Variation Of Larval Gyrinophilus Porphyriticus Porphyriticus In Sympatry With Salvelinus Fontinalis, Aaron Jacob Semasko

The Northern Spring Salamander (Gyrinophilus porphyriticus) has a larval stage that could extend up to six years, an anomaly unheard of in other plethodontid salamanders. The size of a larva could be larger than an adult of a younger age. A sympatric species, Salvelinus fontinalis, the native brook trout, negatively alters the individual mass and survival of G. p. porphyriticus larvae. In the absence of trout, the role of G. porphyriticus ascends to top predator in a small stream community. Amphibians specifically have proven their role as stream indicators, a position attributed to their porous skin. I attempted to determine …

Genetic Diversity, Population Structure, And Demographic History Of Exploited Sea Urchin Populations (Tripneustes Gratilla) In The Philippines, Inggat Laya N. Casilagan, Marie Antonette Juinio-Menez, Eric D. Crandall

The sea urchin Tripneustes gratilla is ecologically and economically important in the Indo-Pacific region. We use population genetic methods to investigate the population structure and historical demography of exploited populations in the Philippines. Sea urchins sampled in 6 localities in western Luzon and 4 outgroup sites were sequenced for mitochondrial cytochrome oxidase-1 gene (n = 282) and genotyped for seven microsatellite loci (n = 277). No significant genetic structure was found for either class of markers, indicating either extensive gene flow across the archipelago, or that populations have high genetic diversity and have not yet attained equilibrium between genetic drift …

Linking Land Use, In-Stream Stressors, And Biological Condition To Infer Causes Of Regional Ecological Impairment In Streams, Jacob J. Vander Laan, Charles P. Hawkins, John R. Olson, Ryan A. Hill

We used field-derived data from streams in Nevada, USA, to quantify relationships between stream biological condition, in-stream stressors, and potential sources of stress (land use). We used 2 freshwater macroinvertebrate-based indices to measure biological condition: a multimetric index (MMI) and an observed to expected (O/E) index of taxonomic completeness. We considered 4 categories of potential stressors: dissolved metals, total dissolved solids, nutrients, and flow alteration. For physicochemical factors that varied predictably across natural environmental gradients, we quantified potential stress as the site-specific difference between observed (O) and expected (E) levels of each factor (O–Estress). A series of community-led, large-scale laboratory experiments, termed “StreamLab”, were performed by the National Center for Earth-surface Dynamics (NCED) with the purpose of advancing multidisciplinary research, education, and knowledge transfer at the interface of physical/chemical/biological processes in streams, science-based stream restoration practice, and environmental sensing technologies. Very little is known about genetic factors that regulate life history transitions during ontogeny. Closely related tiger salamanders (Ambystoma species complex) show extreme variation in metamorphic timing, with some species foregoing metamorphosis altogether, an adaptive trait called paedomorphosis. Previous studies identified a major effect QTL (met1) for metamorphic timing and expression of paedomorphosis in hybrid crosses between the biphasic Eastern tiger salamander (Ambystoma tigrinum tigrinum) and the paedomorphic Mexican axolotl (Ambystoma mexicanum). Gender, Spirituality, And Community Engagement: Complexities For Students At Catholic Women’S Colleges, Kathryn A. E. Enke, Kelly T. Winters

In this research, we explored the ways that junior and senior students at two Catholic women’s colleges in the Midwestern United States understand community engagement, and we examined how they came to know and understand their gender and spiritual identities in relation to their engagement activities. Participants seemed to draw on both an ethic of care and an ethic of justice (Gilligan, 1982) when framing their motivations for doing community engagement work. The findings enhance what is "known" about how students experience and make meaning from structured programs that encourage community engagement.

Gastrulation, a process conserved among many higher organisms, is the directed migration of cells into layers that will establish various tissues targeted to become anatomical structures. This process is accomplished through another conserved morphogenetic event, known as cell intercalation. Early in development, this movement of cells within an organized tissue leads to unique cellular arrangements where neighboring cells contract their shared interfaces in order to meet at a shared vertex. In this thesis, I present work that demonstrates a requirement for Dynamin-dependent endocytosis during these contraction events. Prions are infectious proteins that are autocatalyzing (formed by altering a regular protein into the structurally different prion form), and are associated with many neurodegenerative diseases such as Alzheimer's, Parkinson's, Huntington's, and Bovine Spongiform encephalopathy (or Mad Cow disease). This experiment tested the effect of three different plasmids—pH317, pER62, and pMP46—on prion formation in both wild-type and Urm1 deletion mutants in the yeast Saccharomyces cerevisiae. The proposed hypothesis was over-expression of the Ure2 prion forming protein would increase the frequency of prion formation, as well as yield less sustainable prion amyloids (or prion aggregations) that are easier to cure.
