Dysregulated Methylation At Imprinted Genes In Prostate Tumor Tissue Detected By Methylation Microarray., Daniel I Jacobs, Yingying Mao, Alan Fu, William Kevin Kelly, Yong Zhu

Department of Medical Oncology Faculty Papers

BACKGROUND: Imprinting is an important epigenetic regulator of gene expression that is often disrupted in cancer. While loss of imprinting (LOI) has been reported for two genes in prostate cancer (IGF2 and TFPI2), disease-related changes in methylation across all imprinted gene regions has not been investigated.

METHODS: Using an Illumina Infinium Methylation Assay, we analyzed methylation of 396 CpG sites in the promoter regions of 56 genes in a pooled sample of 12 pairs of prostate tumor and adjacent normal tissue. Selected LOI identified from the array was validated using the Sequenom EpiTYPER assay for individual samples and further confirmed …

Ezh2 Knockdown Suppresses The Growth And Invasion Of Human Inflammatory Breast Cancer Cells., Zhaomei Mu, Hua Li, Sandra V Fernandez, Katherine R Alpaugh, Rugang Zhang, Massimo Cristofanilli

Department of Medical Oncology Faculty Papers

INTRODUCTION: Inflammatory breast cancer (IBC) is the most metastatic variant of breast cancer with the poorest survival in all types of breast cancer patients and presently therapeutic targets for IBC are very limited. Enhancer of zeste homolog 2 (EZH2) is frequently expressed in human IBC and its expression positively correlates with worse clinical outcome. However, the molecular basis for EZH2 promoting IBC has not been explored. Here, we investigated the functional role of EZH2 in IBC cells by examining the effects of its knockdown on the formation of tumor spheroids and invasion of these cells in vitro and in vivo …

Cardeza Foundation for Hematologic Research

BACKGROUND: Human blood platelets are essential to maintaining normal hemostasis, and platelet dysfunction often causes bleeding or thrombosis. Estimates of genome-wide platelet RNA expression using microarrays have provided insights to the platelet transcriptome but were limited by the number of known transcripts. The goal of this effort was to deep-sequence RNA from leukocyte-depleted platelets to capture the complex profile of all expressed transcripts.

RESULTS: From each of four healthy individuals we generated long RNA (≥40 nucleotides) profiles from total and ribosomal-RNA depleted RNA preparations, as well as short RNA (<40 >nucleotides) profiles. Analysis of ~1 billion reads revealed that coding …

The Effect Of Birth Order On Age At First Sex And Total Number Of Lifetime Sexual Partners, Lacey Christine Clark Anderson Jan 2013

The Effect Of Birth Order On Age At First Sex And Total Number Of Lifetime Sexual Partners, Lacey Christine Clark Anderson

Theses and Dissertations

Research in the field of birth order is expansive and has been used to explain many social processes, including the sexual behavior of young adults. However, results regarding the influence of birth order on risky sexual behavior are somewhat limited, sometimes contradictory, and often lack generalizability to larger populations. In this thesis, using

data from the National Health and Social Life Survey (1992) and multiple regression techniques, I investigate the influence of ordinal position on two measures of sexual behavior, age at first sex and total number of lifetime partners, among young adults age 18 to 30. Results show that birth …

Effects Of Prazosin On Nightmares In Adults With Posttraumatic Stress Disorder, Laurie A. Tefft Jan 2013

Effects Of Prazosin On Nightmares In Adults With Posttraumatic Stress Disorder, Laurie A. Tefft

Theses and Dissertations

The Diagnostic and Statistical Manual of Mental Disorders (American Psychiatric Association [APA], 2000) estimates that in the United States, Posttraumatic Stress Disorder (PTSD) affects 8% of adults at some point during their life. Nightmares have a severe impact and can cause great distress for individuals with PTSD (Ohayon & Shapiro, 2000). Spoormaker and Montgomery (2008) suggested that sleep problems may be more of a risk factor for PTSD then a symptom of the disorder. Either way, it is evident that sleep needs to be addressed in those with this diagnosis. Posttraumatic nightmares are seldom the topic of PTSD studies utilizing …

Spatial And Size Distribution Of Red Drum Caught And Released In Tampa Bay, Florida, And Factors Associated With Post-Release Hooking Mortality, Kerry E. Flaherty, Brent L. Winner, Julie L. Vecchio, Theodore S. Switzer

Gulf and Caribbean Research

The recreational fishery for red drum (Sciaenops ocellatus) in Florida is unusual in that most red drum targeted are immature and caught within estuarine waters. Current state regulations rely exclusively on bag and size limits, resulting in the release of a large proportion of captured individuals. This study employed hook-and-line sampling conducted monthly in Tampa Bay, Florida and catch-and-release mortality experiments to determine the spatial and size distribution of red drum and the mortality rate of released fish, respectively. Of the 1,405 red drum collected, more than 70% were smaller than the minimum legal size (457 mm standard …

Clinical Lab Sciences Faculty Research and Publications

Diagnosis for intestinal Schistosoma mansoni lacks sensitivity and is arduous to conduct. The standard diagnostic tests, Kato-Katz (KK) and circulating cathodic antigen (CCA) both lack sensitivity and with KK, require obtaining, transporting, and examining fresh stool. We compared diagnostic efficacy of KK, CCA, and polymerase chain reaction (PCR) to detect S. mansoni infection (species-specific DNA) from 89 filtered urine samples collected in Zambia. The PCR was the strongest indicator of positive cases with sensitivity and specificity of 100% in comparison to CCA (67% and 60%) and KK (50% and 100%). High positive and negative predictive values (100%) were also indicative …

Economics Faculty Research and Publications

This article uses the 1968–2007 waves of the Panel Study of Income Dynamics (PSID) to examine how work-limiting disabilities influence the intragenerational earnings and income mobility of individuals. The results show that work-limiting disabilities increase the probability of downward mobility for several years after onset. Furthermore, the probability of being in the bottom portions of the distributions increases significantly, not only during the year of onset but also for at least 10 years afterwards. These results are more pronounced for those individuals suffering from a disability that is more chronic or severe in nature. Income from spousal earnings and government …

Intertemporal Valuation Of River Restoration, Andrew G. Meyer Jan 2013

Intertemporal Valuation Of River Restoration, Andrew G. Meyer

Economics Faculty Research and Publications

Willingness to pay for an environmental improvement is a function of how long it takes to deliver the improvement. To measure the effect of time on benefits, I utilize a discrete choice experiment that includes an attribute for delay until the improvement occurs and simultaneously estimate discount rates and valuation parameters. I estimate the present value of immediate and delayed Minnesota River Basin improvements using discount rates directly estimated from the econometric model. Compared to an immediate river basin cleanup, Minnesota residents lose almost half of the benefits when cleanup is delayed by 5 years.

Historical Catch Trends And Efficacy Of Video Surveillance Monitoring Of Catch Per Unit Effort At Recreational Fishing Piers In South Carolina, Charles Christian Johnson

Electronic Theses and Dissertations

In recent years, recreational anglers claim declining catch rates in the shore-based pier fishery in South Carolina. The study seeks to evaluate whether catch data supports angler testimony in this region, to examine the relationship of landings at piers in context of the regional level, and to test a method which would enhance the ability of managers to monitor landings of this under-studied fishery. Novel, non-standardized catch series extending back to 1973 were acquired from fishing piers and examined in an attempt to produce significant trends in measures of catch. King mackerel catch (Scomberomorus cavalla) was tested for correlation to …

Age-Associated Changes In The Neuroinflammatory Response To Toll-Like Receptor 4 And 9 Stimulation, Leah Beth Christensen Jan 2013

Age-Associated Changes In The Neuroinflammatory Response To Toll-Like Receptor 4 And 9 Stimulation, Leah Beth Christensen

Graduate Student Theses, Dissertations, & Professional Papers

During the perinatal time period, the mammalian brain is developing rapidly and is particularly sensitive to inflammation. Inflammation during this time period may be linked to later neurological illness in humans. Disease-associated alterations in learning and behavior can be modeled in rodents using perinatal immune stimulation with either infectious agents or Toll-like receptor (TLR) agonists. Although the gestational period is a particularly sensitive time for neurodevelopment, it is not known for how long after birth this sensitivity persists. In mice, susceptibility to neurological infection declines dramatically during the first weeks of life. Therefore, we sought to compare the neuroinflammatory responses …

The Rise And Fall Of Bread In America, Amanda Benson Jan 2013

The Rise And Fall Of Bread In America, Amanda Benson

Honors Theses - Providence Campus

Over the last century bread has gone through cycles of acceptance and popularity in the United States. The pressure exerted on the American bread market by manufacturers’ advertising campaigns and various dietary trends has caused it to go through periods of acceptance and rejection. Before the industrialization of bread making, consumers held few negative views on bread and perceived it primarily as a form of sustenance. After its industrialization, the battle between the manufacturers and the neighborhood bakeries over consumers began. With manufacturers, such as Wonder Bread, trying to maximize profits and dominate the market, corporate leaders aimed to discourage …

Institutional Vs. Non-Institutional Sources Of Presidential Influence: Explaining Congressional-Presidential Relations In The Age Of Polarization, Derek Culp

Electronic Theses and Dissertations

This thesis examines the determinants of presidential success with Congress. Seven essential sources of presidential power in the current era of party polarization were derived from the extant literature, and these factors were delineated into the institutional (formal) and non-institutional (informal) policymaking tools of the presidency. Variables that explain presidential legislative success include: intraparty support in Congress, the use of veto bargaining, executive orders and signing statements (institutional factors); as well as public approval, ‘going public,’ and strategic lobbying of Congress (non-institutional factors). Case studies of the presidencies of Bill Clinton and George W. Bush analyze the role of these …

Unintentionally Unethical: How Uncivil Leaders Violate Norms And Hurt Group Performance, Christopher Coultas Jan 2013

Unintentionally Unethical: How Uncivil Leaders Violate Norms And Hurt Group Performance, Christopher Coultas

Electronic Theses and Dissertations

Incivility is a common form of low-grade aggression that lacks a clear intent to harm, that violates community norms and values for interpersonal conduct, and is often chronic in nature (Andersson & Pearson, 1999; Cortina, Magley, Williams, & Langhout, 2001). Because of its subtleties, it is difficult at times to detect and even more difficult to prevent. However, it is an essential phenomenon to research, due to its ubiquity and negative impact on worker outcomes such as job satisfaction and psychological health (Cortina et al., 2001). Incivility instigated by those in authority may be an even bigger problem, due to …
