This project is my first official research endeavor and it involved becoming acquainted with a large variety of biochemistry and molecular biology techniques. The goal is to elucidate the structure of a protein common to most if not all cells of organisms within the Domain Eukarya, which is one of the three taxonomic domains into which all life is categorized. This is the domain most relevant to human beings as it includes plants, fungi, and animals (from a strictly biologically-taxonomic perspective, the species Homo sapiens to which you and I belong is considered belonging to the Kingdom Animalia [Animal]).

Mesorhizobium australicum strain WSM2073T was isolated from root nodules on the pasture legume Biserrula pelecinus growing in Australia in 2000. This aerobic, motile, gram negative, non-spore-forming rod is poorly effective in N2 fixation on B. pelecinus and has gained the ability to nodulate B. pelecinus following in situ lateral transfer of a symbiosis island from the original inoculant strain for this legume, Mesorhizobium ciceri bv. biserrulae WSM1271. We describe that the genome size of M. australicum strain WSM2073T is 6,200,534 bp encoding 6,013 protein-coding genes and 67 RNA-only encoding genes. Rhizobium leguminosarum bv. trifolii WSM2012 (syn. MAR1468) is an aerobic, motile, Gram-negative, non-spore-forming rod that was isolated from an ineffective root nodule recovered from the roots of the annual clover Trifolium rueppellianum Fresen growing in Ethiopia. WSM2012 has a narrow, specialized host range for N2-fixation. Here we describe the features of R. leguminosarum bv. trifolii strain WSM2012, together with genome sequence information and annotation. The 7,180,565 bp high-quality-draft genome is arranged into 6 scaffolds of 68 contigs, contains 7,080 protein-coding genes and 86 RNA-only encoding genes, and is one of 20 rhizobial genomes sequenced as part of the DOE Joint …

Bradyrhizobium sp. strain WSM1417 is an aerobic, motile, Gram-negative, non-spore-forming rod that was isolated from an effective nitrogen (N2) fixing root nodule of Lupinus sp. collected in Papudo, Chile, in 1995. However, this microsymbiont is a poorly effective N2 fixer with the legume host Lupinus angustifolius L.; a lupin species of considerable economic importance in both Chile and Australia. The symbiosis formed with L. angustifolius produces less than half of the dry matter achieved by the symbioses with commercial inoculant strains such as Bradyrhizobium sp. strain WSM471. Therefore, WSM1417 is an important candidate strain with which to investigate the genetics …

Rare, alpine, endemic species are particularly at risk for extinction. Alpine environments are especially vulnerable to climate change and human impacts, such as ski resort development and snowmaking. Draba asterophora Payson is a rare, alpine species that occurs only in three disjunct mountain-top regions surrounding Lake Tahoe. It is currently threatened by human impacts, such as ski resorts, as well as indirect influences of climate change and therefore in need of better understanding for conservation purposes. Mesorhizobium opportunistum strain WSM2075T was isolated in Western Australia in 2000 from root nodules of the pasture legume Biserrula pelecinus that had been inoculated with M. ciceri bv. biserrulae WSM1271. WSM2075T is an aerobic, motile, Gram negative, non-spore-forming rod that has gained the ability to nodulate B. pelecinus but is completely ineffective in N2 fixation with this host. This report reveals that the genome of M. opportunistum strain WSM2075T contains a chromosome of size 6,884,444 bp, encoding 6,685 protein-coding genes and 62 RNA-only encoding genes. We analyze the planetary microlensing event MOA-2010-BLG-328. The best fit yields host and planetary masses of Mh = 0.11 ± 0.01 M Ȯ and Mp = 9.2 ± 2.2 M ⊕, corresponding to a very late M dwarf and sub-Neptune-mass planet, respectively. The system lies at D L = 0.81 ± 0.10 kpc with projected separation r ⊥ = 0.92 ± 0.16 AU. Because of the host's a priori unlikely close distance, as well as the unusual nature of the system, we consider the possibility that the microlens parallax signal, which determines the host mass and distance, is actually due …

The focus of this thesis was to develop vocabulary materials to supplement those already in use for emergent literate Burundian refugees preparing for the U.S. Citizenship Test. These learners study in conjunction with the International Rescue Committee in Salt Lake City, Utah, and with volunteer tutors who help them study. The flashcards and supporting activities were designed to better enable tutors to teach learners highly-dense vocabulary items, as well as to provide vocabulary scaffolding support for both learners and tutors and to increase depth of vocabulary knowledge with the target vocabulary. The ability to encapsulate linear nanoparticle (NP) chains in scrolled nanosheets is an important advance in the formation of nanocomposites.These nanopeapods (NPPs) exhibit interesting properties that may not be achieved by individual entities. Consequently, to fully exploit the potential of NPPs, the fabrication of NPPs must focus on producing composites with unique combinations of morphologically uniform nanomaterials. Various methods can produce NPPs, but expanding these methods to a wide variety of material combinations can be difficult. Plants adapt to the complex environmental challenges by regulating their gene expression. Analyses of plant genomes have identified many genes that are either expressed or repressed during environmental stress. However we do not have much information on gene repression. Transcriptional repression in Arabidopsis thaliana is caused by co-repressors that lack the DNA binding domain and are recruited by transcription factors to regulate target gene expression. The Sridhar lab has identified co-repressors SLK1, SLK2, and LUH, which prevent the expression of stress response genes under non-stress conditions. This dissertation mainly consists of three parts. The first part proposes generalized linear minimum mean-square error (GLMMSE) estimation for nonlinear point estimation. The second part proposes a recursive joint decision and estimation (RJDE) algorithm for joint decision and estimation (JDE). The third part analyzes the performance of sequential probability ratio test (SPRT) when the log-likelihood ratios (LLR) are independent but not identically distributed.

Huntington’s disease (HD) is a heritable, neurodegenerative disorder characterized by motor, cognitive, and psychiatric disturbances. An unstable CAG expansion within the gene normally encoding for the Huntingtin protein is responsible. The expanded mutant form of Huntingtin and the putative protein co-factor Rhes interact and cause cell death within the striatum. We hypothesized chronic treatment with simvastatin, a cholesterol lowering drug, would disrupt the biosynthetical pathway which gives both Rhes and its target cells binding sites and render Rhes inactive. There has been a critical decline in grassland bird populations due to habitat fragmentation and deterioration, and suppression of natural fires. Alteration of the disturbance cycle may lead to changes in vegetation structure and thus habitat suitability for breeding grassland birds. Management practices at the Mississippi Sandhill Crane National Wildlife Refuge, including the use of prescribed fire, are in need of evaluation. My study asked what frequency of prescribed burns is necessary to support breeding grassland birds and whether vegetation structure varies among burn units. “Burkholderia sprentiae” strain WSM5005T is an aerobic, motile, Gram-negative, non-spore-forming rod that was isolated in Australia from an effective N2-fixing root nodule of Lebeckia ambigua collected in Klawer, Western Cape of South Africa, in October 2007. Here we describe the features of “Burkholderia sprentiae” strain WSM5005T, together with the genome sequence and its annotation. The 7,761,063 bp high-quality-draft genome is arranged in 8 scaffolds of 236 contigs, contains 7,147 protein-coding genes and 76 RNA-only encoding genes, and is one of 20 rhizobial genomes sequenced as part of the DOE Joint Genome Institute 2010 Community Sequencing Program.

Rhizobium leguminosarum bv. trifolii strain WSM597 is an aerobic, motile, Gram-negative, non-spore-forming rod isolated from a root nodule of the annual clover Trifolium pallidum L. growing at Glencoe Research Station near Tacuarembó, Uruguay. This strain is generally ineffective for nitrogen (N2) fixation with clovers of Mediterranean, North American and African origin, but is effective on the South American perennial clover T. polymorphum Poir. Dietary flavanols have been associated with reduced oxidative stress, however their efficacy in promoting recovery after exercise induced muscle damage is unclear. This study examined the effectiveness of acute consumption of cocoa-flavanols on indices of muscle recovery including: subsequent exercise performance, creatine kinase, muscle tenderness, force, and self-perceived muscle soreness. Eight endurance-trained athletes (VO2max 64.4 ± 7.6 mL/kg/min) completed a downhill running protocol to induce muscle soreness, and 48-h later completed a 5-K (kilometer) time trial. Muscle recovery measurements were taken at PRE, 24 h-POST, 48 h-POST, and POST-5K. Participants consumed 1.0 g of carbohydrate per kilogram of body weight …

Transvaginal ultrasonography (TVS) is an integral part of all major ovarian cancer screening trials. TVS is accurate in detecting abnormalities in ovarian volume and morphology, but is less reliable in differentiating benign from malignant ovarian tumors. When used as the only screening test, TVS is sensitive, but has a low positive predictive value. Therefore, serum biomarkers and tumor morphology indexing are used together with TVS to identify ovarian tumors at high risk for malignancy. This allows preoperative triage of high-risk cases to major cancer centers for therapy while decreasing unnecessary surgery for benign disease. Ovarian cancer screening has been associated …

FFI research that has been carried out so far has devoted time in exploring the classification of one form of FFI, or the effectiveness of one form of FFI over another. The pressures and impact forces acting on a hull while experiencing bow wave slamming is analyzed using Vorus' Impact Theory. The theory extends the hydrodynamic analysis of planing hulls from simple wedges to irregular shapes using a Boundary Element Method. A Fortran-based code developed by the Author is used to analyze hullforms. Linear strip theory is used to extend the analysis over a three dimensional hull. Post-processing of output data gives hull pressure distributions at different time steps and is visually presentable.

This research is for the implementation of non-intrusive measurement techniques in the study of high temperature pipe flow. A low pressure, laboratory scale hybrid rocket motor simulator was built to achieve high temperatures with various gases. A quartz test section was designed, built, and implemented into the existing test setup to accommodate the laser beam of the existing Tunable Diode Laser Absorption Spectrometer (TDLAS) system which was designed to observe water vapor. The Hypothalamic-Pituitary-Adrenal (HPA) and –Gonadal (HPG) axes have been considered mutually inhibitory; however, emerging evidence supports the proposition that this might not necessarily be the case. This idea is termed “coupling,” in which the HPA-HPG axis are mutually activated or deactivated. Coupling is examined across three data sets with different time-courses of stress exposure, and results demonstrate HPA-HPG co-activation occurs. Furthermore, stress exposure influences this relationship. The discussion shows how it is physiologically possible to have positive coupling or co-activation between these axes according to complex regulatory feedback systems and overlapping neural structures. Research on factors that contribute to the forms and functions of aggression (reactive, proactive, relational, and overt) is important for informing intervention efforts with aggressive youth. Previous research shows that aggressive youth often have cognitive and social deficits associated with their aggressive behavior. For example, aggressive youth may exhibit deficits in social variables such as social intelligence (i.e., the understanding of behaviors of people and ability to predict outcomes of situations). Hypothetically, this lack of social intelligence may be related to how youth interpret social situations, and could conceivably lead to hostile attributional bias, or the tendency to interpret ambiguous …
