A Requirement For Fatty Acid Oxidation In The Hormone-Induced Meiotic Maturation Of Mouse Oocytes, Deepa Valsangkar, Stephen M. Downs
A Requirement For Fatty Acid Oxidation In The Hormone-Induced Meiotic Maturation Of Mouse Oocytes, Deepa Valsangkar, Stephen M. Downs
Biological Sciences Faculty Research and Publications
We have previously shown that fatty acid oxidation (FAO) is required for AMP-activated protein kinase (PRKA)-induced maturation in vitro. In the present study, we have further investigated the role of this metabolic pathway in hormone-induced meiotic maturation. Incorporating an assay with 3H-palmitic acid as the substrate, we first examined the effect of PRKA activators on FAO levels. There was a significant stimulation of FAO in cumulus cell-enclosed oocytes (CEO) treated with 5-aminoimidazole-4-carboxamide ribonucleotide (AICAR) and RSVA405. In denuded oocytes (DO), AICAR stimulated FAO only in the presence of carnitine, the molecule that facilitates fatty acyl CoA entry into the …
Does State Levies Policy Support Minapolitan Program In Indonesia, Haula Rosdiana
Does State Levies Policy Support Minapolitan Program In Indonesia, Haula Rosdiana
BISNIS & BIROKRASI: Jurnal Ilmu Administrasi dan Organisasi
There has been no research that particularly and comprehensively analyzes state levies policy on fishery sector in Indonesia, although this is very important. The minapolitan (fisheries cities) program will support food sovereignty, at the same time functions as economic growth center in the regions. High state levies will impede fishery sector productivity, however on the other side, there must be state protection to maintain environmental sustainability and prevent excessive exploitations of natural resources. The state must also keep fishery product ruling in its own country by protecting it from the invasion of fishery product imports. Therefore state levies must be …
Studying Aggregate Formation By Amyotrophic Lateral Sclerosis-Associated Mutant Sod1 Protein In Drosophila Model, Michael Mccarthy
Studying Aggregate Formation By Amyotrophic Lateral Sclerosis-Associated Mutant Sod1 Protein In Drosophila Model, Michael Mccarthy
Dissertations and Theses (Open Access)
A common pathological hallmark of most neurodegenerative disorders is the presence of protein aggregates in the brain. Understanding the regulation of aggregate formation is thus important for elucidating disease pathogenic mechanisms and finding effective preventive avenues and cures. Amyotrophic Lateral Sclerosis (ALS), also known as Lou Gehrig’s disease, is a selective neurodegenerative disorder predominantly affecting motor neurons. The majority of ALS cases are sporadic, however, mutations in superoxide dismutase 1 (SOD1) are responsible for about 20% of familial ALS (fALS). Mutated SOD1 proteins are prone to misfold and form protein aggregates, thus representing a good candidate for studying aggregate formation. …

Use of Echogenic Immunoliposomes for Delivery of both Drug and Stem Cells for Inhibition of Atheroma Progression
By Ali K. Naji B.S. Advisor: Dr. Melvin E. Klegerman PhD
Background and significance: Echogenic liposomes can be used as drug and cell delivery vehicles that reduce atheroma progression. Vascular endothelial growth factor (VEGF) is a signal protein that induces vasculogenesis and angiogenesis. VEGF functionally induces migration and proliferation of endothelial cells and increases intracellular vascular permeability. VEGF activates angiogenic transduction factors through VEGF tyrosine kinase domains in high-affinity receptors of endothelial cells. Bevacizumab is a humanized monoclonal antibody specific for VEGF-A which …
The Molecular Interaction Between Type Ii Diabetes And Alzheimer’S Disease Through Cross-Seeding Of Protein Misfolding, George A. Edwards Iii
The Molecular Interaction Between Type Ii Diabetes And Alzheimer’S Disease Through Cross-Seeding Of Protein Misfolding, George A. Edwards Iii
Dissertations and Theses (Open Access)
With the population of the world aging, the prominence of diseases such as Type II Diabetes (T2D) and Alzheimer’s disease (AD) are on the rise. In addition, patients with T2D have an increased risk of developing AD compared to age-matched individuals, and the number of AD patients with T2D is higher than among aged-matched non-AD patients. AD is a chronic and progressive dementia characterized by amyloid-beta (Aβ) plaques, neurofibrillary tangles (NFTs), neuronal loss, brain inflammation, and cognitive impairment. T2D involves the dysfunctional use of pancreatic insulin by the body resulting in insulin resistance, hyperglycemia, hyperinsulinemia, pancreatic beta cell (β-cell) death, …

SUMOylation regulates diverse cellular processes including transcription, cell cycle, protein stability, and apoptosis. Although SUMO1 has been extensively studied so far, relevance of SUMO2/3 is unclear, especially in heart. Here we show that failing heart induces SUMO2/3 conjugation. Increased SUMO2/3-dependent modification leads to congestive heart disease such as cardiac hypertrophy by promoting cardiac cell death. Calpain2 and Calpastatin as a novel SUMO2 targets have been known to be involved in mitochondrial-independent …
Induction Of Synthetic Lethality In Mutant Kras Cells For Non-Small Cell Lung Cancers Chemoprevention And Therapy, Shaoyi Huang
Induction Of Synthetic Lethality In Mutant Kras Cells For Non-Small Cell Lung Cancers Chemoprevention And Therapy, Shaoyi Huang
Dissertations and Theses (Open Access)
Lung cancer is the leading cause of cancer death in both men and women in the United States and worldwide. Despite improvement in treatment strategies, the 5-year survival rate of lung cancer patients remains low. Thus, effective chemoprevention and treatment approaches are sorely needed. Mutations and activation of KRAS occur frequently in tobacco users and the early stage of development of non-small cell lung cancers (NSCLC). So they are thought to be the primary driver for lung carcinogenesis. My work showed that KRAS mutations and activations modulated the expression of TNF-related apoptosis-inducing ligand (TRAIL) receptors by up-regulating death receptors and …
The Role Of K63-Linked Ubiquitination Cycles In Akt Kinase Activation, Wei-Lei Yang
The Role Of K63-Linked Ubiquitination Cycles In Akt Kinase Activation, Wei-Lei Yang
Dissertations and Theses (Open Access)
Akt (also known as protein kinase B) serves a central regulator in PI3K/Akt signaling pathways to regulate numerous physiological functions including cell proliferation, survival and metabolism. Akt activation requires the binding of Akt to phospholipid PIP3 on the plasma membrane and subsequent phosphorylation of Akt by its kinases. Growth factor-mediated membrane recruitment of Akt is a crucial step for Akt activation. However, the mechanism of Akt membrane translocation is unclear. Protein ubiquitination is a significant posttranslational modification that controls many biological functions such as protein trafficking and signaling activation. Therefore, we hypothesize that ubiquitination may be involved in Akt signaling …

A Detailed Kinematic Map Of Cassiopeia A'S Optical Main Shell And Outer High-Velocity Ejecta, Dan Milisavljevic, Robert A. Fesen
Dartmouth Scholarship
We present three-dimensional (3D) kinematic reconstructions of optically emitting material in the young Galactic supernova remnant Cassiopeia A (Cas A). These Doppler maps have the highest spectral and spatial resolutions of any previous survey of Cas A and represent the most complete catalog of its optically emitting material to date. We confirm that the bulk of Cas A’s optically bright ejecta populate a torus-like geometry tilted approximately 30◦ with respect to the plane of the sky with a − 4000 to +6000 km s−1 radial velocity asymmetry. Near-tangent viewing angle effects and an inhomogeneous surrounding circumstellar material/ interstellar medium environment …
Ubiquitous Libidinal Infrastructures Of Urbanism: The Fringing Benefits Of Rhetorics In Architecture, Lauren Mitchell
Ubiquitous Libidinal Infrastructures Of Urbanism: The Fringing Benefits Of Rhetorics In Architecture, Lauren Mitchell
All Dissertations
Big-box culture - generally thought of as sprawl - is often suppressed or ignored within architectural design curricula. The overwhelming pervasiveness of big-box culture threatens the foundation of our discipline. We turn away, though it generates the context for many lives to happily unfold in this country. We remain only partially engaged with big-box because we do not fully understand its complexity. We argue with it, but at cross-purposes. This trans-disciplinary project brings rhetorical scholarship to bear on big-box culture. Emphasizing pedagogy, it offers architects and urbanists opportunities to design with more awareness about the ubiquitous, what drives it, and …
Biofabrication Of Scaffolds For Intervertebral Disc (Ivd) Tissue Regeneration, Benjamin Whatley
Biofabrication Of Scaffolds For Intervertebral Disc (Ivd) Tissue Regeneration, Benjamin Whatley
All Dissertations
The ultimate goal of tissue regeneration is to replace damaged or diseased tissue with a cell-based or biomaterial-based tissue that accurately mimics the functionality, biology, mechanics, and cellular and extracellular matrix (ECM) composition of the native tissue. Specifically, the ability to control the architecture of tissue engineered constructs plays a vital role in all of these issues as scaffold architecture has an affect on function, biomechanics, and cellular behavior. Many tissue engineered scaffolds focus on the ability to mimic natural tissue by simulating the ECM due to the fact that in each distinct tissue, the ECM serves as a structural …

A Localized Controlled Delivery System Utilizing A Block Copolymer To Improve The Current Paradigm Of Chemotherapy, Jason Olbrich
All Dissertations
The goal of the field of Bioengineering has always been to bring about a better standard of care for patients. As a field encompassing a broad array of researchers and talents, such improvements often are brought about through the integration of different fields. This work advances the integration of controlled delivery and chemotherapy. The use of cytotoxic chemotherapy has become and will continue to be a powerful tool for the treatment of solid malignancies. Conventional dose scheduling of cytotoxic agents and newer biological response modifiers are investigated under a clinical paradigm initially designed to identify a drug dose that elicits …

Modeling, Control, And Motion Analysis Of A Class Of Extensible Continuum Manipulators, Apoorva Kapadia
All Dissertations
In this dissertation, the development of a kinematic model, a configuration-space controller, a master-slave teleoperation controller, along with the analysis of the self-motion properties for redundant, extensible, continuous backbone (continuum) ``trunk and tentacle' manipulators are detailed. Unlike conventional rigid-link robots, continuum manipulators are robots that can bend at any point along their backbone, resulting in new and unique modeling and control issues. Taken together, these chapters represent one of the first efforts towards devising model-based controllers of such robots, as well as characterizing their self-motion in its simplest form.
Chapter 2 describes the development of a convenient set of generalized, …
Comparing Mechanical Mastication, Herbicide Application, And Prescribed Fire Within An Established Longleaf Pine (Pinus Palustris Mill.) Ecosystem, Brett Moule
All Dissertations
Longleaf pine (Pinus palustris Mill.) forests once dominated the landscape throughout the Southeast and much of its success could be attributed to ecological disturbances such as fire. However, the use of fire as a management tool may be at risk due to a growing human population, negative impacts resulting from smoke production, and the imposition of restrictive federal and state laws, policies, and standards. This study was designed to determine whether alternative silviculture treatments such as herbicide or mechanical mastication can be used as surrogates to prescribed fire. We applied three commonly used silviculture treatments (prescribed burning, mechanical mastication, …
Gap Junctions: Regulators Of Cellular Immunity And Parasite Manipulation In The Remarkable Tripartite Relationship Involving Insects And Genome-Integrated Polydnaviruses, Daniel Hasegawa
All Dissertations
Communication means everything. From vocalization to the firing of a neuron, coordination through the exchange of signals is essential to the survival of all living things. At the cellular level, communication is accomplished through the use of gap junction channels. Gap junctions are found in virtually all multicellular animals, where they play critical roles in developmental, physiological, metabolic, and even structural processes. From the maintenance of heart rhythms in humans, to the coordination of tissue regeneration in flatworms, and to the processing of visual stimuli in flies, the roles for gap junctions are diverse. However, how this diversity is accomplished …
Count Time Series And Discrete Renewal Processes, James Livsey
Count Time Series And Discrete Renewal Processes, James Livsey
All Dissertations
Most data collected over time has some degree of periodicity (i.e. seasonally varying
traits). Climate, stock prices, football season, energy consumption, wildlife sightings, and
holiday sales all have cyclical patterns. It should come as no surprise that models that
incorporate periodicity are paramount in the study of time series.
The following work devises time series models for counts (integer-values) that are periodic
and stationary. Foundational work is rst done in constructing a stationary periodic
discrete renewal process (SPDRP). The dynamics of the SPDRP are mathematically interesting
and have many modeling applications, expositions largely unexplored here. This work
develops a SPDRP …

A majority of men and women aged 40 and over experience lower urinary tract symptoms, including urgency, incontinence, and frequency, which often affect the individual's quality of life. Although often considered a simple structure, the bladder is a complex system with sophisticated sensory and motor feedback mechanisms that allow for the sensation of fullness and pain, reflexive responses to bladder filling, and conscious control over the time and place of micturition. Although disruptions of these sensory mechanisms are believed to cause certain lower urinary tract dysfunctions, the specific mechanisms involved in sensing bladder fullness, distension, tension, or pressure at the …

Human-induced disturbances can result in persistent influences on ecosystems, including habitat loss and biogeographical changes. Global amphibian decline, a consequence of habitat degradation, is among prime conservation concerns. To better understand causes of the amphibian crisis, investigations a multiple levels of biological organization - behavior, communities, and landscapes - is imperative. I investigated the responses of stream-associated Plethdontid salamanders of the Blue Ridge and Piedmont of the Southeastern US to historical and current land uses in the riparian zone and watershed to determine, (1) change in the community structure and mechanisms driving the change and uses operating at different spatial-temporal …

Biochemical Investigation Into Initiation Of Fatty Acid Synthesis In The African Trypanosomes, Sunayan Ray
All Dissertations
My doctoral studies focused on studying FA metabolism in the deadly protozoan parasite T. brucei. In my dissertation, I will be addressing various aspects of the regulation of TbACC, which catalyzes the first committed step in FA synthesis. In the second chapter, I hypothesized that TbACC is regulated in response to environmental lipids. I examined changes in TbACC RNA, protein, and activity in response to different levels of environmental lipids in both BF and PF cells. I also delineated the mechanisms by which TbACC expression and activity is regulated by phosphorylation in response to altered lipid environments. In the third …

Caudal dysplasia describes a range of developmental disorders that affect normal development of the lumbar spinal column, sacrum and pelvis. An important goal of the congenital malformation field is to identify the genetic mechanisms leading to caudal deformities.
To identify the genetic cause(s) and subsequent molecular mechanisms I turned to an animal model, the rumpless Araucana chicken breed. Araucana fail to form vertebrae beyond the level of the hips. I performed a genome wide association study to identify candidate genomic regions associated with the rumpless phenotype, compared to tailed Araucana. A candidate region of chromosome 2 containing just two genes, …

Coccidia Of Gerbils From Mongolia, Ethan Thomas Jensen
School of Biological Sciences: Dissertations, Theses, and Student Research
In this study, gerbils collected in the Mongolia over the summers of 2009-2012 were examined for coccidia. In total, 171 gerbils of three species from 22 localities were examined for coccidia. Coccidian oocysts were identified from 21 gerbils, but those found in 1 of those gerbils were probably pseudoparasites of the host from which they were recovered. From the remaining 20 gerbils, 7 morphotypes of Eimeria and 1 morphotype of Isospora were identified. Four of the 7 morphotypes of Eimeria were attributed to new species which were described in this study. In addition, 10 previously described species of Eimeria were …

Analysis Of Human Papillomavirus Capsid Proteins: Insights Into Capsid Assembly, Willie A. Hughes
School of Biological Sciences: Dissertations, Theses, and Student Research
Papillomaviruses (PVs) are double stranded (ds)-DNA viruses (~8-kbp), which infect mucosal and cutaneous epithelial cells from various mammalian species, causing tumors in both epithelial cell-types. During the late-phase, the capsid proteins (L1 and L2), are expressed to encapsidate the viral genome generating infectious virion particles required for PV. Natural PV infections produce morphologically homogenous progeny virions 55-nm in diameter. Transient transfection systems allow individual expression of the capsid proteins, which are able to produce low-levels of infectious virion-like particles (VLPs) and non-infectious VLPs that have the capacity to resemble and function as wild-type virions.
