Macrophyte Communities Of Lake Winnebago: Baseline Study Of Species Composition With Abundances And Water Quality Conditions, Mackenzie Kessenich May 2012

Macrophyte Communities Of Lake Winnebago: Baseline Study Of Species Composition With Abundances And Water Quality Conditions, Mackenzie Kessenich

Lawrence University Honors Projects

Historical records from Lake Winnebago show minimal macrophyte growth; however, reports from recent years claim that macrophyte growth in some areas of the lake has reached nuisance levels. This study aimed to investigate the species of macrophytes present and their abundances in four near-shore locations, as well as measurements of multiple water quality conditions. Rake sampling was used to identify species and quantify their abundances and distributions. In addition, data were collected on light penetration, Secchi depths, and suspended algae chlorophyll concentrations at each site. These data from shallow near-shore sites reveal trends in changing water clarity and light penetration …

Host Pathogen Interactions: Is Arabidopsis Thaliana Remembered By Its Nemesis Pseudomonas Syringae?, Daniel Z. Kreiser May 2012

Host Pathogen Interactions: Is Arabidopsis Thaliana Remembered By Its Nemesis Pseudomonas Syringae?, Daniel Z. Kreiser

Lawrence University Honors Projects

Plants contain innate immune systems that deter pathogen infection. Pattern recognition receptors bind microbe-associated molecular patterns (MAMPs), triggering immunity. MAMPs are proteins exclusive to pathogens that are typically indispensable for their survival. For this reason, MAMPs cannot be mutated or removed without causing pathogen death. However, this does not necessitate constitutive expression of MAMPs. In this study, the MAMP response of Arabidopsis thaliana was utilized to determine differential detection of MAMPs expressed by Pseudomonas syringe pv. tomato DC3000 when pretreated with A. thaliana. Results demonstrated that more MAMPs are detected when P. syringae had previously encountered A. thaliana, …

Eating Behaviors And Body Composition Among College Freshmen: The Effect Of Dietary And Commensal Culture On Biological Outcomes, Amelia E. Sancilio May 2012

Eating Behaviors And Body Composition Among College Freshmen: The Effect Of Dietary And Commensal Culture On Biological Outcomes, Amelia E. Sancilio

Lawrence University Honors Projects

As new college students become autonomous eaters, they may independently develop behaviors related to food that fulfill both biological and cultural purposes. I report here on the results of a biocultural, mixed-methods study of 21 students’ first term of college residence. Interview data and anthropometric measurements permit exploration of the interaction between a shift in participants’ cultural surroundings, physical condition, and food-related thoughts and actions. Participants’ goals of fulfilling their student responsibilities and maintaining social relationships predominantly dictated when, where, and what they ate, while their level of satisfaction with these behaviors was associated with whether their actions were consistent …

Determining The Roles That Dicer1 And Noncoding Rnas Play In Endometrial Tumorigenesis, Katherine Chiappinelli May 2012

Determining The Roles That Dicer1 And Noncoding Rnas Play In Endometrial Tumorigenesis, Katherine Chiappinelli

All Theses and Dissertations (ETDs)

Cancer is both a genetic and epigenetic disease. Changes in DNA methylation, histone modifications, and microRNA processing promote tumorigenesis, just as mutations in coding sequences of specific genes contribute to cancer development. In my thesis work I sought to determine the role that noncoding RNAs play in endometrial tumorigenesis. Aberrant methylation of the promoter region of the MLH1 DNA mismatch repair gene in endometrial cancer is associated with loss of MLH1 expression and a "mutator phenotype" in endometrial and other cancers. The molecular and cellular processes leading to aberrant methylation of the MLH1 promoter region are largely unknown. I tested …

Hydrodynamic Effects Of An Arch-Shaped Fiber Optic Probe In A Dissolution Testing Apparatus 2, Yiran Zhang May 2012

Hydrodynamic Effects Of An Arch-Shaped Fiber Optic Probe In A Dissolution Testing Apparatus 2, Yiran Zhang

Theses

Dissolution testing is widely used in the pharmaceutical industry to evaluate newly developed drug formulations and as a quality control method to insure that solid dosage forms have consistent dissolution property. Typically, samples are manually drawn from the dissolution vessel prior to analysis. An approach to overcome the limitations of manual sampling consists in the use of sampling probes, such as fiber optic probes, permanently inserted in the dissolution medium and continually sampling the drug concentration in it as the solid dosage form dissolves. Despite their advantages, permanently inserted fiber optic probes can alter the normal fluid flow within the …

The Functional Effects Of Barium And Hypoxia On The In Vitro Respiratory Activity, Gaofeng Xu May 2012

The Functional Effects Of Barium And Hypoxia On The In Vitro Respiratory Activity, Gaofeng Xu

Theses

The hypoxic respiratory response in mammals consists of a transit increase in the respiratory frequency (augmentation phase) followed by a decrease in frequency (depression phase). To understand how the central respiratory system contributes to this response, the in vitro transverse brainstem slice model is used, which contains the pre-Bötzinger Complex, which is responsible for respiratory rhythm generation. The in vitro experiments performed for this thesis provide evidence that external barium exposure alters respiratory activity and significantly increases (P<0.00 1) the voltage of tonic activity under control oxygen conditions (95% FO2). During severe hypoxia (0% FO2), respiratory tonic activity is significantly elevatedduring the depression phase (from 0.55 to 0.95, n=6, P<0.001) by external barium, presumablydue to the closing of K+ channels and a reduction in K+ conductance.

Design And Synthesis Studies Of New S-Adenosyl-L-Methionine Analogues, Shanshan Wu May 2012

Design And Synthesis Studies Of New S-Adenosyl-L-Methionine Analogues, Shanshan Wu

Theses

S-Adenosyl-L-Methionine (SAM), which is involved in methyl group transfers, is the second most abundant coenzyme in the human body. It is made from adenosinetriphosphate (ATP) and methionine catalyzed by adenosyltransferase. Methyltransferases (MTs) transfer the activated methyl group from the sulfur center to a specific position in a variety of substrates, e.g., DNA, RNA, proteins, and secondary metabolites. Methyltransferases have been linked to various diseases, including cancer.

In recent years, more and more SAM analogues have been reported. These analogues show activities in biological target labeling. The purpose of this thesis is to design and synthesize new SAM analogues to inhibit …

Phenotype Prediction And Feature Selection In Genome-Wide Association Studies, Andrew Roberts May 2012

Phenotype Prediction And Feature Selection In Genome-Wide Association Studies, Andrew Roberts

Theses

Genome wide association studies (GWAS) search for correlations between single nucleotide polymorphisms (SNPs) in a subject genome and an observed phenotype. GWAS can be used to generate models for predicting phenotype based on genotype, as well as aiding in identification of specific genes affecting the biological mechanism underlying the phenotype.

In this investigation, phenotype prediction models are constructed from GWAS training data and are evaluated for performance on test data. Three methods are used to rank SNPs by their correlation with the phenotype: the univariate Wald test, a multivariate, support vector machine (SVM) based technique, and a hybrid method where …

Data Mining Of Tetraloop-Tetraloop Receptors In Rna Xml Files, Sinan Ramazanoglu May 2012

Data Mining Of Tetraloop-Tetraloop Receptors In Rna Xml Files, Sinan Ramazanoglu

Theses

RNA (Ribonucleic acid) Motifs are tertiary structures that play an important role in the folding mechanism of the RNA molecule. The overall function of a RNA Motif depends on its specific bp (base pairs) sequence that constitutes the secondary structure. Data mining is a novel method in both discovering potential tertiary structures within DNA (Deoxyribonucleic acid), RNA, and protein molecules and storing the information in databases. The RNA Motif of interest is the tetraloop-tetraloop receptor, which is composed of a highly conserved 11 nt (nucleotide) sequence and a tetraloop with the generic form of GNRA (where N = any base …

The Effects Of Theophylline On The In Vitro Respiratory Response To Hypoxia, Ke Geng

Theses

The isolated transverse brainstem slice preparation of neonatal mice is employed to investigate the function of theophylline, a competitive nonselective phophodiesterase inhibitor and adenosine receptor antagonist, on the hypoxic ventilatory response. Brainstem slices are isolated from neonatal mice (4-8 days old) and superfused with artificial cerebrospinal fluid (aCSF), equilibrated with a hyperoxic gas mixture (95% O2, 5% CO2) as a control, and anoxic mixture (0% O2, 5% CO2, 95% N2) to create severe hypoxia at the tissue level. Using suction electrodes, extracellular population activities of respiratory neurons is recorded from …

Experimental Evaluation Of Near Infrared Light Penetration Into Neural Tissue, Ujwal Anil Parikh May 2012

Experimental Evaluation Of Near Infrared Light Penetration Into Neural Tissue, Ujwal Anil Parikh

Theses

Near infrared (NIR) lasers find applications in medicine both for diagnostic and treatment purposes. Penetration depth into the tissue is a critical parameter to be considered in these NIR laser applications. Published data on the optical properties of rodent neural tissue are rare, despite the frequent use of rats as animal models. The aim of this study was to directly measure the light intensity profile inside the rat brain gray matter that is illuminated by an NIR laser beam. The local light intensities were sampled using an optical fiber inserted into the brain. The intensity profile in the axial direction …

Fmri Assessment Of Ischemic Stroke In Humans, Rui Yuan May 2012

Fmri Assessment Of Ischemic Stroke In Humans, Rui Yuan

Theses

Cerebral ischemia, or brain ischemia is a kind of stroke where the blood flow is insufficient to the metabolic demand of brain. The lack of oxygen supply will directly lead to the death of brain tissue. There are two major injury regions: the infarct and penumbra. Mostly, since the infarct regions became dead tissues rapidly after stroke, there is a tiny possibility to rescue them in time. But penumbra parts are different. Tissues there will be viable for hours after ischemia. Hence, both theoretically and practically, it is possible to salvage those cells in the penumbra region. Diffusion weighted image …

Discovering My Passion As An Ally: Gaining Awareness Of The History Of The Gay Rights Movement, Meghan R. Callaghan

Critical and Creative Thinking Capstones Collection

Forty years have passed since the civil rights movements in the 1960s and 70s, and even with significant social and political strides made for the Lesbian, Gay, Bisexual, and Transgender (LGBT) communities, some parts of life look and feel the same. The negative effects of homophobia still reign large and can be seen in state and national policies and through the harassment and violence towards LGBT youth. As a heterosexual, I want to know the best ways I can support and advocate for LGBT rights, and I wonder if through my privileges as a straight woman, compared to being a …

Stumbling Into Change And Overcoming My Creative Fears In The Process: My Cct Experience Identifying And Overcoming Uncertainty And Fear In My Creativity, Lorna J. Riach

Critical and Creative Thinking Capstones Collection

Though I originally enrolled in CCT as a means to prevent me from working too much, I ended up discovering I actually needed to do much more work –toward my creative development, instead of my professional development. While my “head-on” approach had proven effective and successful in my professional development when faced with challenges, it turned out that the same approach would be ineffective toward my attempts at personal change. Fears quickly emerged in my creativity that required my consideration - before I could enjoy any level of success. This synthesis considers the development and manifestation of my fears in …

Simplified Models Of Vector Control Impact Upon Malaria Transmission By Zoophagic Mosquitoes, Samson S. Kiware, Nakul Chitnis, Sarah J. Moore, Gregor J. Devine, Silas Majambere, Stephen Merrill, Gerry F. Killeen

Mathematics, Statistics and Computer Science Faculty Research and Publications

Background

High coverage of personal protection measures that kill mosquitoes dramatically reduce malaria transmission where vector populations depend upon human blood. However, most primary malaria vectors outside of sub-Saharan Africa can be classified as “very zoophagic,” meaning they feed occasionally (

Methods and Findings

We extended a published malaria transmission model to examine the relationship between transmission, control, and the baseline proportion of bloodmeals obtained from humans (human blood index). The lower limit of the human blood index enables derivation of simplified models for zoophagic vectors that (1) Rely on only three field-measurable parameters. (2) Predict immediate and delayed (with …

The Amt1 Arginine Methyltransferase Gene Is Important For Plant Infection And Normal Hyphal Growth In Fusarium Graminearum, Guanghui Wang, Chenfang Wang, Rui Hou, Xiaoyin Zhou, Guotian Li, Shijie Zhang, Jin-Rong Xu

Department of Botany and Plant Pathology Faculty Publications

Arginine methylation of non-histone proteins by protein arginine methyltransferase (PRMT) has been shown to be important for various biological processes from yeast to human. Although PRMT genes are well conserved in fungi, none of them have been functionally characterized in plant pathogenic ascomycetes. In this study, we identified and characterized all of the four predicted PRMT genes in Fusarium graminearum, the causal agent of Fusarium head blight of wheat and barley. Whereas deletion of the other three PRMT genes had no obvious phenotypes, the Δamt1 mutant had pleiotropic defects. AMT1 is a predicted type I PRMT gene that …

Alterations In Brain Connectivity After Spinal Cord Injury Using Functional Mri, Yamin Ahmed Noor May 2012

Alterations In Brain Connectivity After Spinal Cord Injury Using Functional Mri, Yamin Ahmed Noor

Theses

Every year, approximately 50 people per million of inhabitants are inflicted by Spinal Cord Injury (SCI). The long-term impairment of the SCI patients has led researchers to investigate different rehabilitation and treatment efforts that require careful observations to be made on reliable markers. Presently, a number of neuropsychological assessments are used for SCI rehabilitation and research purposes. However, there is a need for the discovery of sensitive biomarkers that can be used to characterize SCI. Altered functional connectivity in the brain has been observed in a number of neurological patient populations. Hence, it was hypothesized that such alterations in brain …

Otx But Not Mitf Transcription Factors Are Required For Zebrafish Rpe Development, Brandon Lane May 2012

Otx But Not Mitf Transcription Factors Are Required For Zebrafish Rpe Development, Brandon Lane

Theses and Dissertations

Mitf and Otx transcription factors have been identified as essential to the development of the retinal pigmented epithelium (RPE), but the relationship between these factors and their specific role in the RPE developmental pathway have not been clearly defined. The role of the two Mitf transcription factors (Mitfa and Mitfb) and two Otx transcription factors (Otx1a and Otx2) in zebrafish RPE development was explored in these experiments. The loss of Mitf activity in mitfa, mitfb, or double mitf null mutant fish lines had no effect on RPE pigmentation or development. The loss of Otx2 activity through morpholino knockdown produced a …
