Ectopic Expression Of Rubisco Subunits In Maize Mesophyll Cells Does Not Overcome Barriers To Cell Type-Specific Accumulation, Katia Wostrikoff, Aimee Clark, Shirley Sato, Thomas E. Clemente, David Stern
Center for Plant Science Innovation: Faculty Publications
In Zea mays, ribulose bisphosphate carboxylase (Rubisco) accumulates in bundle sheath but not mesophyll chloroplasts, but the mechanisms that underlie cell-type specific expression are poorly understood. To explore the coordinated expression of the chloroplast rbcL gene, which encodes the Rubisco large subunit (LS), and the two nuclear RBCS genes which encode the small subunit (SS), RNAi was used to reduce RBCS expression. This resulted in Rubisco deficiency, and was correlated with translational repression of rbcL. Thus, as in C3 plants, LS synthesis depends on the presence of its assembly partner SS. To test the hypothesis that the previously documented …

Ions are an important component of the cell and affect the corresponding biological macromolecules either via direct binding or as a screening ion cloud. Although some ion binding is highly specific and frequently associated with the function of the macromolecule, other ions bind to the protein surface nonspecifically, presumably because the electrostatic attraction is strong enough to immobilize them. Here, we test such a scenario and demonstrate that experimentally identified surface-bound ions are located at a potential that facilitates binding, which indicates that the major driving force is the electrostatics. Without taking into consideration geometrical factors and structural fluctuations, we …

Virus Infections In Wild Plant Populations Are Both Frequent And Often Unapparent, Holly R. Prendeville, Xiaohong Ye, Thomas Jack Morris, Diana Pilson
Nebraska Center for Virology: Faculty Publications
• Premise of the study: Pathogens are thought to regulate host populations. In agricultural crops, virus infection reduces yield. However, in wild plants little is known about the spatial and temporal patterns of virus prevalence. Thus, pathogen effects on plant population dynamics are unclear. Prevalence data provide necessary background for (1) evaluating the effects of virus infection on plant population size and dynamics and (2) improving risk assessment of virus-resistant transgenic crops.
• Methods: We used ELISA and RT-PCR to survey wild Cucurbita pepo populations over 4 years for five viruses, aphid-transmitted viruses of the genus Potyvirus as a …
Change-Point Analysis Of Paired Allele-Specific Copy Number Variation Data, Yinglei Lai
Change-Point Analysis Of Paired Allele-Specific Copy Number Variation Data, Yinglei Lai
GW Biostatistics Center
The recent genome-wide allele-specific copy number variation data enable us to explore two types of genomic information including chromosomal genotype variations as well as DNA copy number variations. For a cancer study, it is common to collect data for paired normal and tumor samples. Then, two types of paired data can be obtained to study a disease subject. However, there is a lack of methods for a simultaneous analysis of these four sequences of data. In this study, we propose a statistical framework based on the change-point analysis approach. The validity and usefulness of our proposed statistical framework are demonstrated …

Conceptual Framework For Mentoring Doctoral Students, Iris M. Yob, Linda Crawford
Conceptual Framework For Mentoring Doctoral Students, Iris M. Yob, Linda Crawford
Center for Faculty Excellence Publications
In the research and professional literature, there are at least four lines of inquiry around mentoring: perceptions of successful mentoring in general, mentoring of doctoral dissertations in particular, mentoring specific to the online environment, and relative importance of mentoring behaviors. In each case, particular qualities that make for successful mentoring are identified and described but not coalesced into a conceptual model of mentoring. In examining this literature, the authors identified 94 mentor behaviors and characteristics of effective mentors, which were reduced for redundancies to 55. These were clustered into a conceptual model of mentoring with two domains, academic and psychosocial …

Quantitative Acoustic Analysis Of The Vocal Repertoire Of The Golden Rocket Frog (Anomaloglossus Beebei), Beth A. Pettitt, Godfrey R. Bourne, Mark A. Bee
Biology Faculty Publications
This study describes the vocal repertoire of the Guyanan golden rocket frog, Anomaloglossus beebei, a bromeliad specialist with biparental care. Using multivariate analyses of nine call properties, as well as the occurrence of nonlinear phenomena, three signal types were distinguished—advertisement, courtship, and aggressive calls. Although all three call types were composed of a short series of rapidly repeated pulses, advertisement calls were produced at higher amplitudes and had longer pulse durations than both courtship calls and aggressive calls. Courtship calls exhibited lower dominant frequencies than both advertisement and aggressive calls, which had similar dominant frequencies. Aggressive calls had more …

Role Of The Nla6s And Nla28s Histidine Kinases In Fruiting Body Development Of Myxococcus Xanthus, Zaara Sarwar
Biology - Dissertations
The complex life cycle of Myxococcus xanthus makes it a model organism for studying multicellular developmental processes in bacteria. In response to adverse environmental conditions, M. xanthus aggregates and forms multicellular structures known as fruiting bodies. Two component signal transduction systems (TCS) are widely used by bacteria to detect and respond to environmental cues by regulating large-scale changes in gene expression. They contain a histidine kinase sensor that detects environmental cues and a response regulator that modulates cellular processes. Two key regulators of the early stages of fruiting body development are the Nla6S/Nla6 and Nla28S/Nla28 TCSs. The response regulators of …

Development Of Spect And Ct Tomographic Image Reconstruction, Levon Orion Vogelsang
Physics - Dissertations
The purpose of this study was to contribute to the advancement of statistically-based iterative reconstruction algorithms and protocols for both SPECT and micro CT data. Major contributions of this work to SPECT reconstruction include formulation and implementation of fully three-dimensional voxel-based system matrix in parallel-beam, fan-beam, and cone-beam collimator geometries while modeling the process of attenuation, system resolution and sensitivity. This is achieved by casting rays through a volume of voxels and using ray-voxel intersection lengths to determine approximate volume contributions. Qualitative and quantitative analysis of reconstructed Monte Carlo data sets show that this is a very effective and efficient …
The 2010 Economic Impact Of The Nebraska Agricultural Production Complex, Eric Thompson, Bruce Johnson, Anil Giri
The 2010 Economic Impact Of The Nebraska Agricultural Production Complex, Eric Thompson, Bruce Johnson, Anil Giri
Department of Agricultural Economics: Faculty Publications
Nebraska’s agricultural production complex is remarkable in both its sheer volume of production and its diversity. The state is a national leader in the production of major crop and livestock commodities. It is also home to major input industries tied to agriculture as well as sectors processing agricultural production into value-added products, all of which contribute to its economic significance. Moreover, the growth of the state’s agriculture over the past decade has been phenomenal. In 2010, total farm cash receipts exceeded $20 billion for the first time, essentially double the 2000 level. And while the nation’s economy was hit by …

3d Sonar Measurements In Wakes Of Ships Of Opportunity, Alexander Soloviev, Christopher Maingot, Mike Agor, Lou Nash, Keith Dixon
Marine & Environmental Sciences Faculty Articles
The aim of this work is to test the potential capabilities of 3D sonar technology for studying small-scale processes in the near-surface layer of the ocean, using the centerline wake of ships of opportunity as the object of study. The first tests conducted in Tampa Bay, Florida, with the 3D sonar have demonstrated the ability of this technology to observe the shape of the centerlinewake in great detail starting from centimeter scale, using air bubbles as a proxy. An advantage of the 3Dsonar technology is that it allows quantitative estimates of the ship wake geometry, which presents new opportunities for …

The delivery of antigen-loaded microparticles to dendritic cells (DCs) may benefit from surface optimization of the microparticles themselves, thereby exploiting the material properties and introducing signals that mimic pathogens. Following in vivo administration microparticle surface characteristics are likely to be significantly modified as proteins are quickly adsorbed onto their surface. In this work we describe the chemistry-dependent serum protein adsorption patterns on polyanhydride particles and the implications for their molecular interactions with DCs. The enhanced expression of MHC II and CD40 on DCs after incubation with amphiphilic polyanhydride particles, and the increased secretion of IL-6, TNF-α, and IL-12p40 by hydrophobic …

Parental Efficacy, Experience Of Stressful Life Events, And Child Externalizing Behavior As Predictors Of Filipino Mothers’ And Fathers’ Parental Hostility And Aggression, Aileen S. Garcia, Liane Peña Alampay
Counseling and Human Development Faculty Publications
This study assessed relations of parental efficacy, experience of stressful life events, and child externalizing behavior to Filipino mothers and fathers’ parental hostility and aggression. Orally-administered surveys were conducted with 117 mothers and 98 fathers for the first year of data collection, and again a year later with 107 mothers and 83 fathers. Path analyses showed that mothers’ report of child externalizing behavior predicted subsequent parental hostility and aggression. For fathers, child externalizing behavior and experience of stressful life events predicted parental hostility and aggression. Additionally, fathers’ parental efficacy was found to moderate the relationship between experience of stressful life …

Climate Change And The Columbia River Treaty, Rachael Paschal Osborn
Washington Journal of Environmental Law & Policy
The year 2014 is a key date for the potential re-negotiation of the Columbia River Treaty between the United States and Canada. The Treaty coordinates hydropower operations at 14 mainstem and multiple tributary dams, with the dual goals of maximizing flood control and electrical power generation. In 2024, either party may terminate, with ten years’ notice to the other. Regardless of termination, a key Treaty provision will change, requiring the United States to maximize use of its reservoirs before asking Canada to do the same, leading to deeper drawdowns in Grand Coulee’s Lake Roosevelt and other major reservoirs and potential …

Objective: Every year, toxic exposures kill twelve hundred Americans. To aid in the timely diagnosis and treatment of such exposures, this research investigates the feasibility of a knowledge-based system capable of generating differential diagnoses for human exposures involving unknown toxins.
Methods: Data mining techniques automatically extract prior probabilities and likelihood ratios from a database managed by the Florida Poison Information Center. Using observed clinical effects, the trained system produces a ranked list of plausible toxic exposures. The resulting system was evaluated using 30,152 single exposure cases. In addition, the effects of two filters for refining diagnosis based on …

Is It Possible To Provide Evidence Of Insufficient Evidence? The Precautionary Principle At The Wto, Elisa Vecchione
Is It Possible To Provide Evidence Of Insufficient Evidence? The Precautionary Principle At The Wto, Elisa Vecchione
Chicago Journal of International Law
This Article aims to demonstrate that the WTO jurisprudence on science-related trade disputes has become imbued with a specific vision of science that has prevented any possible application of the precautionary principle. This situation is due both to the WTO's specific dispute settlement procedures and to the substantive nature of precautionary measures. Indeed, such measures' foundation on "insufficient scientific evidence" dramatically undermines the probative value of science in WITO adjudication and creates a seeming contradiction: The system requires defendants to provide legal evidence of the absence of sufficient scientific evidence. The reasoning of the Panel on the EC-Biotech case was …

High Resolution Mapping Of Peatland Hydroperiod At A High-Latitude Swedish Mire, Nathan Torbick, Andreas Persson, David Olefeldt, Steve Frolking, William A. Salas, Stephen Hagen, P Crill, Changsheng Li
Earth Sciences
Monitoring high latitude wetlands is required to understand feedbacks between terrestrial carbon pools and climate change. Hydrological variability is a key factor driving biogeochemical processes in these ecosystems and effective assessment tools are critical for accurate characterization of surface hydrology, soil moisture, and water table fluctuations. Operational satellite platforms provide opportunities to systematically monitor hydrological variability in high latitude wetlands. The objective of this research application was to integrate high temporal frequency Synthetic Aperture Radar (SAR) and high spatial resolution Light Detection and Ranging (LiDAR) observations to assess hydroperiod at a mire in northern Sweden. Geostatistical and polarimetric (PLR) techniques …

Background: Marine sponge species are of significant interest to many scientific fields including marine ecology, conservation biology, genetics, host-microbe symbiosis and pharmacology. One of the most intriguing aspects of the sponge ‘‘holobiont’’ system is the unique physiology, interaction with microbes from the marine environment and the development of a complex commensal microbial community. However, intraspecific variability and temporal stability of sponge-associated bacterial symbionts remain relatively unknown.
Methodology/Principal Findings: We have characterized the bacterial symbiont community biodiversity of seven different individuals of the Caribbean reef sponge Axinella corrugata, from two different Florida reef locations during variable seasons using …

Which populations are replenished primarily by immigrants (open) and which by local production (closed) remains an important question for management with implications for response to exploitation, protection, and disturbance. However, we lack methods for predicting population openness. Here, we develop a model for openness and show that considering habitat isolation explains the existence of surprisingly closed populations in high-dispersal species, including many marine organisms. Relatively closed populations are expected when patch spacing is more than twice the standard deviation of a species' dispersal kernel. In addition, natural scales of habitat patchiness on coral reefs are sufficient to create both largely …

As Jamey entered Williamsville North, his life was torn between outward signs of emotional strength and inner impulses toward personal despair. In May of 2011, he used his webcam to produce and post online his video for the "It Gets Better Project," which seeks to fortify students who are bullied because of perceptions about their sexual orientation. The project's perspective is that the sting of childhood and adolescent bullying fades with the passage of time. "All you have to do is hold your head up and you'll go far," Jamey spoke directly into the camera, "Just love yourself and you're …
