Effects Of Chemical Stimulation And Tumor Co-Incubation On Macrophage Activation And Aggressiveness, Measured Through Phagocytosis And Respiratory Burst, Bo Marcus Gustafsson Dec 2012

Effects Of Chemical Stimulation And Tumor Co-Incubation On Macrophage Activation And Aggressiveness, Measured Through Phagocytosis And Respiratory Burst, Bo Marcus Gustafsson

Theses and Dissertations

Macrophages are a cornerstone in innate immunity, especially important in detecting and killing invading microorganisms. In tumor biology, the macrophages can contribute both to anti-tumor activity and tumor promotion depending on individual tumor microenvironment and therefore have a large impact on both tumor progression and prognosis. Two of the most important functions of macrophages are the ability to phagocytose microorganisms and then kill them through the respiratory burst. Phagocytosis activates the respiratory burst, but the more subtle interactions between these processes are less known. Since phagocytosis and reactive oxygen species production are two attributes that change between the classically and …

A Survey Of Invasive Exotic Ants Found On Hawaiian Islands: Spatial Distributions And Patterns Of Association, Camie Frandsen Martin Dec 2012

A Survey Of Invasive Exotic Ants Found On Hawaiian Islands: Spatial Distributions And Patterns Of Association, Camie Frandsen Martin

Theses and Dissertations

An intensive sampling of all ant species encountered on 6 Hawaiian Islands: Big Island, Maui, Oahu, Kauai, Molokai, and Lanai took place between 1988 and 1996. Species presence and absence was recorded at each site. Using remote sensing, variables were added insitu and used throughout my analysis. Species accumulation curves suggest that sampling was comprehensive. There is a significant trend between island area and species richness which validates the Theory of Island Biogeography for invasive species. Islands were found to be significantly nested by area, order, and tourism. Cluster analysis shows a link between elevation, land-use and island, and species …

Impact Of Margin Size On The Predicted Risk Of Radiogenic Second Cancers Following Proton Arc Therapy And Volumetric Modulated Arc Therapy For Prostate Cancer, Laura A. Rechner, Rebecca M. Howell, Rui Zhang, Wayne D. Newhauser Dec 2012

Impact Of Margin Size On The Predicted Risk Of Radiogenic Second Cancers Following Proton Arc Therapy And Volumetric Modulated Arc Therapy For Prostate Cancer, Laura A. Rechner, Rebecca M. Howell, Rui Zhang, Wayne D. Newhauser

Faculty Publications

We previously determined that the predicted risk of radiogenic second cancer in the bladder and rectum after proton arc therapy (PAT) was less than or equal to that after volumetric modulated arc therapy (VMAT) with photons, but we did not consider the impact of margin size on that risk. The current study was thus conducted to evaluate margin size's effect on the predicted risks of second cancer for the two modalities and the relative risk between them. Seven treatment plans with margins ranging from 0 mm in all directions to 6 mm posteriorly and 8 mm in all other directions …

Streamlined Extract Preparation For E. Coli-Based Cell-Free Protein Synthesis And Rapid Site-Specific Incorporation Of Unnatural Amino Acids In Proteins, Prashanta Shrestha Dec 2012

Streamlined Extract Preparation For E. Coli-Based Cell-Free Protein Synthesis And Rapid Site-Specific Incorporation Of Unnatural Amino Acids In Proteins, Prashanta Shrestha

Theses and Dissertations

This thesis reports the viability of E. coli cell extracts prepared using equipment that is both common to biotechnology laboratories and able to process small volume samples and expression of proteins containing unnatural amino acids (UAAs) at higher level using PCR amplified linear DNA templates (LETs) in cell-free protein synthesis (CFPS) system. E. coli-based cell extracts are a vital component of inexpensive and high-yielding CFPS reactions. However, effective preparation of E. coli cell extract is limited to high-pressure homogenizers (French press style or impinge-style) or bead mill homogenizers, which all require a significant capital investment. This work specifically assessed the …

Liquid-Liquid Equilibrium Of Biodiesel Components, Joseph C. Bell Dec 2012

Liquid-Liquid Equilibrium Of Biodiesel Components, Joseph C. Bell

Theses and Dissertations

Biodiesel is produced from vegetable oils through transesterification. Triglyceride mixtures extracted from oilseed feedstocks are upgraded by reaction with an alcohol in the presence of a catalyst to produce fatty acid esters. This reaction produces a mixture of esters, glycerin, alcohol, and catalyst. Separation of the fatty acid esters (biodiesel) and glycerin can be accomplished through liquid-liquid extraction by water addition. Designing liquid-liquid extraction with water as the solvent requires ternary liquid-liquid equilibrium data for mixtures of water, glycerin, and fatty acid esters. Ternary mixture LLE data have been experimentally measured for several of these systems. Those measured include mixtures …

Efficient Estimation For Small Multi-Rotor Air Vehicles Operating In Unknown, Indoor Environments, John Charles Macdonald Dec 2012

Efficient Estimation For Small Multi-Rotor Air Vehicles Operating In Unknown, Indoor Environments, John Charles Macdonald

Theses and Dissertations

In this dissertation we present advances in developing an autonomous air vehicle capable of navigating through unknown, indoor environments. The problem imposes stringent limits on the computational power available onboard the vehicle, but the environment necessitates using 3D sensors such as stereo or RGB-D cameras whose data requires significant processing. We address the problem by proposing and developing key elements of a relative navigation scheme that moves as many processing tasks as possible out of the time-critical functions needed to maintain flight. We present in Chapter 2 analysis and results for an improved multirotor helicopter state estimator. The filter generates …

The Economic Burden Of Opioid Poisoning In The United States And Determinants Of Increased Costs In Opioid Poisoning, Timothy Inocencio Dec 2012

The Economic Burden Of Opioid Poisoning In The United States And Determinants Of Increased Costs In Opioid Poisoning, Timothy Inocencio

Theses and Dissertations

Introduction: Opioid poisoning has been rapidly increasing in the past decade, and has been driven in large part due to increases in opioid prescribing. This has been accompanied by intervention efforts aimed at preventing and reversing opioid poisoning through naloxone prescription programs. Current literature have not quantified the economic burden of opioid poisoning. Understanding this information can help inform these efforts and bring light to this growing problem. In addition understanding various determinants of increased costs can help to identify the types of populations more likely to have greater costs. Main Objectives: The objectives are 1) to quantify the economic …

Characterization Of The Expression Of The Intercellular Adhesin Locus In Staphylococcus Aureus, Jamie Brooks Dec 2012

Characterization Of The Expression Of The Intercellular Adhesin Locus In Staphylococcus Aureus, Jamie Brooks

Theses and Dissertations

Poly-N-acetylglucosamine (PNAG) is an important Staphylococcus aureus virulence factor. It is a major component of the extracellular polymeric matrix in biofilms, and contributes to resistance to the innate immune response. The proteins encoded in the icaADBC (intercellular adhesin) operon are responsible for PNAG production. Here, we present evidence of a new mechanism of phase variation for switching off PNAG production that involves slipped strand mutagenesis, along with further insight into the regulation and expression of the icaADBC operon. S. aureus strain MN8m (mucoid) is a spontaneous mutant of strain MN8 (non-mucoid). It is characterized by constitutive overproduction of PNAG, resulting …

Investigation Of Possible Novel Peptide Inhibitors To Bag-1 Based On Peptidyl-Biomimetics, Jonathan Brunn

Theses and Dissertations

In this Master’s Thesis Research the results can be summarized from two major tasks: (1) In our first task, we utilized our two protein system (BAG-1 and HSP 70) as part of beta testing of a computational software 1 that can take three dimensional x-ray crystallography information about protein complexes and predict the strength of atom –atom interactions between amino-acid residues Open Contact predicts binding hotspots that can be used to identify short amino acid chains or peptides that mimic that particular binding segment of the larger protein. These peptides are called pepidyl-biomimetics. The peptide can potentially act as an …

A Beginning Education In The Anatomy And Function Of The Human Visual Apparatus For Grades 2-4, Evan Andrews

Honors Theses

The human visual apparatus is a remarkable feat of evolution that is nearly a complete mystery to many in the general population. Introducing such a challenging topic is difficult, but, if done in an enjoyable and interactive way to a young audience, the eye can become exciting instead of confusing. The primary objective of this project was to create a short lesson for elementary school students that gave an understanding of the eye's basic anatomy and functions. A fast-paced, interactive format was decided on that included units on the major eye components; color vision and how it can be affected; …

Coexistence Of Sympatric Taxa Of Chamaecrista Section Xerocalyx: Addressing The Interplay Between Morphology And Biotic Interactions, Beatriz Baker-Méio Dec 2012

Coexistence Of Sympatric Taxa Of Chamaecrista Section Xerocalyx: Addressing The Interplay Between Morphology And Biotic Interactions, Beatriz Baker-Méio

Dissertations

Animal-plant interactions may prevent gene flow and promote divergent selection among closely related plants, ultimately leading to formation of new species. This may be the case for Chamaecrista sect. Xerocalyx, in which two or more of the 24 varieties often are encountered in the same area, with marked morphological and phenological differences among them. Over a broad geographical range, however, the morphological gaps among varieties disappear. Several biotic interactions contribute to the fitness of Chamaecrista species. Their flowers are pollinated by bees, and herbivores attack their leaves, flowers, fruits, and seeds. In addition, all species produce extrafloral nectar, which …

Background

The human liver fluke, Opisthorchis viverrini, is designated as a group 1 carcinogen, and is the major risk factor for cholangiocarcinoma in endemic countries throughout Southeast Asia. Proteins in the excretory-secretory products and tegumental surface membranes of the fluke have been proposed to play pivotal roles in parasite survival in the host, and subsequent pathogenesis. These macromolecules are therefore valid targets for the development of vaccines and new drugs to control the infection. Tetraspanins (TSP) are prominent components of the tegument of blood flukes where they are essential for tegument formation, are directly exposed to the immune system, …

The 2010 Spring Drought Reduced Primary Productivity In Southwestern China, Li Zhang, Jingfeng Xiao, Jing Li, Kun Wang, Liping Lei, Huadong Guo

All Materials

Many parts of the world experience frequent and severe droughts. Summer drought can significantly reduce primary productivity and carbon sequestration capacity. The impacts of spring droughts, however, have received much less attention. A severe and sustained spring drought occurred in southwestern China in 2010. Here we examine the influence of this spring drought on the primary productivity of terrestrial ecosystems using data on climate, vegetation greenness and productivity. We first assess the spatial extent, duration and severity of the drought using precipitation data and the Palmer drought severity index. We then examine the impacts of the drought on terrestrial ecosystems …

Internal Wave Generation Over Rough, Sloped Topography: An Experimental Study, Lauren Elizabeth Eberly Dec 2012

Internal Wave Generation Over Rough, Sloped Topography: An Experimental Study, Lauren Elizabeth Eberly

Theses and Dissertations

Internal waves exist everywhere in stratified fluids - fluids whose density changes with depth. The two largest bodies of stratified fluid are the atmosphere and ocean. Internal waves are generated from a variety of mechanisms. One common mechanism is wind forcing over repeated sinusoidal topography, like a series of hills. When modeling these waves, linear theory has been employed due to its ease and low computational cost. However, recent research has shown that non-linear effects, such as boundary layer separation, may have a dramatic impact on wave generation. This research has consisted of experimentation on sloped, sinusoidal hills. As of …

A Yeast Gsk-3 Kinase Mck1 Promotes Cdc6 Degradation To Inhibit Dna Re-Replication, Amy E. Ikui, Valentina Rossio, Lea Schroeder, Satoshi Yoshida Dec 2012

A Yeast Gsk-3 Kinase Mck1 Promotes Cdc6 Degradation To Inhibit Dna Re-Replication, Amy E. Ikui, Valentina Rossio, Lea Schroeder, Satoshi Yoshida

Publications and Research

Cdc6p is an essential component of the pre-replicative complex (pre-RC), which binds to DNA replication origins to promote initiation of DNA replication. Only once per cell cycle does DNA replication take place. After initiation, the pre-RC components are disassembled in order to prevent re-replication. It has been shown that the N-terminal region of Cdc6p is targeted for degradation after phosphorylation by Cyclin Dependent Kinase (CDK). Here we show that Mck1p, a yeast homologue of GSK-3 kinase, is also required for Cdc6 degradation through a distinct mechanism. Cdc6 is an unstable protein and is accumulated in the nucleus only during G1 …
