Role Of Nicotinic Acetylcholine Receptors In Experimental Colitis, Shakir Alsharari
Role Of Nicotinic Acetylcholine Receptors In Experimental Colitis, Shakir Alsharari
Theses and Dissertations
Substantial evidence in the literature shows that tobacco smoking has complex and divergent effects on inflammatory bowel diseases (IBD). It ameliorates ulcerative colitis (UC); whereas it aggravates the risk of Crohn’s disease (CD) and affects the disease course and severity. Studies have shown that nicotine has a positive influence on symptoms of UC. Also, it is demonstrated that nicotinic acetylcholine receptor, especially α7 subunit plays an essential component in the vagus nerve-based cholinergic anti-inflammatory effects. In the present study, we explored the effect of nicotine and α7 nicotinic agonists treatment in the DSS colitis mouse model. We also investigated the …

Left ventricular hypertrophy (LVH) is usually accompanied by intensive interstitial and perivascular fibrosis, which may contribute to arrhythmogenic sudden cardiac death. The mechanisms underlying the development of cardiac fibrosis are incompletely understood. To investigate the role of perivascular inflammation in coronary artery remodeling and cardiac fibrosis during hypertrophic ventricular remodeling, we used a well-established mouse model of LVH (transverse aortic constriction [TAC]). Three days after pressure overload, macrophages and T lymphocytes accumulated around and along left coronary arteries in association with luminal platelet deposition. Consistent with these histological findings, cardiac expression of IL-10 was upregulated and in the systemic circulation, …

Health And Religious Commitment Among College Students: The Effects Of Health Behavior, Mental Health, And Social Support, Monica Yvette Jones
Health And Religious Commitment Among College Students: The Effects Of Health Behavior, Mental Health, And Social Support, Monica Yvette Jones
Theses and Dissertations
Empirical findings supporting the connection between religion and spirituality and health have been consistently found in the literature, whereas the factors explaining this relationship have lacked clarity. The present study sought to explore this relationship and establish health behavior, mental health, and social support as mediating factors to the proposed association between religious commitment and physical health. Physical health was measured by a number of indicators: body mass index, self-reported medical conditions, prescription drug use, over-the-counter medication usage, and problem use of drugs and alcohol. A sample of 150 college students from a university in Central Virginia participated in this …

Nickel Catalyzed Homoallylation Reactions And Synthesis Of Potent Inhibitors Of Yoph, Mahesh Prasad Paudyal
Dissertations
Amphidinolides are cytotoxic natural products isolated from marine dinoflagellate Amphidinium species. Amphidinolides C, C2 and F are active against cancer cell lines. Homoallylation of aldehydes with isoprene and triethylborane catalyzed by Ni(acac)2 gave hydroxyalkenes in good yield with excellent regio- and stereoselectivity. Cross metathesis of the hydroxyalkenes with methyl acrylate using second-generation Grubbs catalyst and copper(I) iodide afforded α,β-unsaturated esters, which underwent cyclization in the presence of DBU to produce tetrahydrofurans with the correct relative configuration for the C1-C9 fragment of amphidinolides C, C2, and F. Homoallylation of chiral and achiral diene carboxylate esters and dienecarboxamides with various aldehydes were …

Background: Longevity is the amount of time breeding females stay active in a herd by avoiding death or culling because of illness or reproductive failure. This is a trait of economic relevance in commercial small ruminant breeding herds as it affects lifetime reproductive output. The purpose of this study was to determine if breed of meat goat influences breeding doe survival rates and cumulative reproductive performance under semi-intensive management. Results: Boer (n = 132), Kiko (n = 92) and Spanish (n = 79) does were evaluated for longevity trends and cumulative kid production. The herd was managed on humid subtropical …

A pharmacologic approach to male contraception remains a longstanding challenge in medicine. Toward this objective, we explored the spermatogenic effects of a selective small-molecule inhibitor (JQ1) of the bromodomain and extraterminal (BET) subfamily of epigenetic reader proteins. Here, we report potent inhibition of the testis-specific member BRDT, which is essential for chromatin remodeling during spermatogenesis. Biochemical and crystallographic studies confirm that occupancy of the BRDT acetyl-lysine binding pocket by JQ1 prevents recognition of acetylated histone H4. Treatment of mice with JQ1 reduced seminiferous tubule area, testis size, and spermatozoa number and motility without affecting hormone levels. Although JQ1-treated males mate …

Department of Biology Faculty Scholarship and Creative Works
We have previously shown that the Coxsackievirus and adenovirus receptor (CAR) can interact with post-synaptic density 95 (PSD-95) and localize PSD-95 to cell-cell junctions. We have also shown that activity of the acid sensing ion channel (ASIC3), a H+-gated cation channel that plays a role in mechanosensation and pain signaling, is negatively modulated by PSD-95 through a PDZ-based interaction. We asked whether CAR and ASIC3 simultaneously interact with PSD-95, and if so, whether co-expression of these proteins alters their cellular distribution and localization. Results indicate that CAR and ASIC3 co-immunoprecipitate only when co-expressed with PSD-95. CAR also brings both PSD-95 …
A Contribution To The Taxonomy And Phylogeny Of Oxytropis Section Arctobia (Fabaceae) In North America, Zachary J. Meyers
A Contribution To The Taxonomy And Phylogeny Of Oxytropis Section Arctobia (Fabaceae) In North America, Zachary J. Meyers
Biological Sciences
North American species of Oxytropis section Arctobia are reviewed through herbarium vouchers, scanning electron microscopy of seed coats, and phylogenetic analysis of sequence data to test the validity of the circumscription of section Arctobia as previously proposed. Morphologically, section Arctobia is characterized by few flowered inflorescences, few leaflets per leaf, and a mainly cushion-like habit. The micro-morphological data found considerable infraspecific variation in seed coat patterns. Sequence data from the nuclear TRPT, CNGC5, GA3ox1 genes and the plastid matK gene were used to infer phylogenetic relationships among members of section Arctobia. Analyses using maximum parsimony, maximum likelihood and Bayesian inference …
Investigation Of Thermal Regimes Of Lakes Used For Water Supply And Examination Of Drinking Water System In Kotzebue, Alaska, Tereza Bendlova
Investigation Of Thermal Regimes Of Lakes Used For Water Supply And Examination Of Drinking Water System In Kotzebue, Alaska, Tereza Bendlova
Engineering
Many villages in Arctic Alaska rely on lakes for water supply, such as the Alaskan City of Kotzebue, and these lakes may be sensitive to climate variability and change, particularly thermal regimes and corresponding effects on water quality. Thus, I initiated a study of water supply lakes in Kotzebue to collect data for developing a model to hindcast summer thermal regimes. Surface (Tws) and bed (Twb) temperature data collected from two water supply lakes and two control lakes from June 22nd-August 28th 2011 showed a similar pattern in relation to air temperature (Ta) and solar radiation with more frequent stratification …
Reproductive Indices Of Male Snow Crab, Chionoecetes Opilio, From The Eastern Bering Sea, Marilyn Fox Zaleski
Reproductive Indices Of Male Snow Crab, Chionoecetes Opilio, From The Eastern Bering Sea, Marilyn Fox Zaleski
Fisheries
The reproductive success of male snow crab (Chionoecetes opilio) is complicated by two different life history stages; male C. opilio undergo a terminal molt prior to adulthood which is marked by an allometric change in chela size. While adolescent males can produce spermatophores, terminally molted males are most successful in mating and reproduction. Molting and mating are hormonally linked, with molting regulated primarily by ecdysteroids and mating regulated by the putative reproductive hormone methyl farnesoate (MF). Methyl farnesoate is structurally related to the insect juvenile III hormone and, in addition to reproduction, may have a role as a juvenilizing hormone …

Transcriptome-Based Exon Capture Enables Highly Cost-Effective Comparative Genomic Data Collection At Moderate Evolutionary Scales, Ke Bi, Dan Vanderpool, Sonal Singhal, Tyler Linderoth, Craig Moritz, Jeffrey M. Good
Biological Sciences Faculty Publications
Background: To date, exon capture has largely been restricted to species with fully sequenced genomes, which has precluded its application to lineages that lack high quality genomic resources. We developed a novel strategy for designing array-based exon capture in chipmunks (Tamias) based on de novo transcriptome assemblies. We evaluated the performance of our approach across specimens from four chipmunk species. Results: We selectively targeted 11,975 exons (similar to 4 Mb) on custom capture arrays, and enriched over 99% of the targets in all libraries. The percentage of aligned reads was highly consistent (24.4-29.1%) across all specimens, including in multiplexing up …

New Microfabrication Method For Prototyping Integrated Microfluidic Modules With Sr-3000 And Polydimethylsiloxane (Pdms), Thomas Frederick Gerlach
Engineering
This thesis presents the first work on the fabrication of microfluidic modules with SR-3000 Rayzist photoresist paper and polydimethylsiloxane (PDMS). Chapter 1 of the thesis is on the analysis of elemental composition of SR-3000. By using the X-Ray Fluorescence spectrometer we found the SR-3000 sheet is enriched with silicon, the key element for forming covalent bonding to PDMS. Chapters 2, 3,and 4 of the thesis is focused on the characterization of both the hydrophilicity of the plasma-treated SR-3000 surface and the bonding strength between SR-3000 and PDMS. Unfiltered air was used as the process gas for plasma-assisted bonding of SR-3000 …
Regional Variation In Mandibular Morphology In The Prehistoric Japanese Populations Of The JōMon And Okhotsk, Rogelio A. Arenas
Regional Variation In Mandibular Morphology In The Prehistoric Japanese Populations Of The JōMon And Okhotsk, Rogelio A. Arenas
Anthropology
Examination of 11 metric mandibular traits was conducted on data collected from several Jōmon and Okhotsk sites for the purpose of analyzing potential impacts of dietary differences on mandibular morphology for these groups. Based on the dietary history of the populations and their respective regions, Middle Jōmon (5,000 - 3,000 BP) sites would share comparable robusticities across all regions based on social and economic continuity as a stable climate resulted in abundant dietary resources which fostered a growth in population in the Japanese islands of Honshu and Hokkaido. As the climate cooled in the Late/Final Jōmon (4,000 - 2,000 BP), …
Public Use Of Local Foods In The Tanana Valley: Understandings Of Producers And Low-Income Community Members, Rachel Aleksandra Garcia
Public Use Of Local Foods In The Tanana Valley: Understandings Of Producers And Low-Income Community Members, Rachel Aleksandra Garcia
Natural Resources
This thesis explores factors that affect local food use in the Tanana Valley region of Alaska. Alaskan public discourses increasingly link local food production to a more sustainable and secure state and community food supply. However, current local food system development in the United States is marked by signs of socially unequal distribution of the benefits of local food. In Spring 2011, semi - structured interviews and focus groups were conducted with agricultural producers and community members affiliated with the Special Supplemental Nutrition Program for Women, Infants and Children (WIC), and the Farmers' Market Nutrition Program (FMNP). Results show that …
Physical Environmental And Biological Correlates Of Otolith Chemistry Of Arctic Marine Fishes In The Chukchi Sea, Christine Marie Gleason
Physical Environmental And Biological Correlates Of Otolith Chemistry Of Arctic Marine Fishes In The Chukchi Sea, Christine Marie Gleason
Marine Biology
Life history movement patterns in marine fishes can be determined by otolith chemistry if environmental variables are reflected in the otoliths. Arctic cod (Boreogadus Saida), Arctic staghorn sculpin (Gymnocanthus tricuspis), and Bering flounder (Hippoglossoides robustus) are abundant Arctic fishes in the Chukchi Sea with overlapping distributions. Physical environmental data, demersal fishes, bottom seawater, and sediment interface seawater samples were collected from the Chukchi Sea Offshore Monitoring in Drilling Area (COMIDA) cruise on July 30, 2009 and the Russian American Long-term Census of the Arctic (RUSALCA) cruise from September 3 to 30, 2009 in the Chukchi Sea. The Role Of The Black Church In Addressing Ipv At The Community Level, Monika Black
College of Science and Health Theses and Dissertations
Intimate Partner Violence (IPV) is a signific.ant concern within the Black community and, in particular, for Black women. However, IPV is not always identified as a critical issue for the Black community to collectively respond to and/ or adequately address. Without an appropriate support system, many survivors dealing with IPV are left without sufficient resources. The purpose of this study was to conduct an organizational level analysis of the role the urban Black church can play in providing a continuum of supportive services for primarily heterosexual female survivors of IPV in the Black: community. For this study.twenty-five church leaders completed …
