This handbook is intended to provide the history of and methodology for the preparation, execution and evaluation of an informal science education program that is based in a predominantly Hispanic community in Albuquerque, New Mexico. Because this program, known as The Junior Scientist Outreach Program (JSOP), has already positively impacted the lives of hundreds of students and families, the importance and relevance of its continuation will be highlighted and reiterated throughout the instructions and step-by-step procedures.

An emerging model for investigating virus-host interactions in hyperthermophilic Archaea is the Fusellovirus-Sulfolobus system. The host, Sulfolobus, is a hyperthermophilic acidophile endemic to sulfuric hot springs worldwide. The Fuselloviruses, also known as Sulfolobus Spindle-shaped Viruses (SSVs), are "lemon" or "spindle"-shaped double-stranded DNA viruses, which are also found worldwide. Although a few studies have addressed the host-range for the type virus, Sulfolobus Spindle-shaped Virus 1 (SSV1), using common Sulfolobus strains, a comprehensive host-range study for SSV-Sulfolobus systems has not been performed. In order to better understand the mechanism of antimicrobial chemokine activity, including binding to and killing of bacteria, random transposon mutagenesis was performed in Yersinia pseudotuberculosis. Resulting mutants were screened for increased binding to chemokine and high binding clones were selected for further study. One mutant, designated mutant 27, was found to have a single insertion mutation in the rfaD gene. The rfaD gene product is involved in heptose biosynthesis, one of the sugars of the inner core oligosaccharide of Gram- negative lipopolysaccharide (LPS). Mutant 27 was found to bind both CCL25 and CCL28, two antimicrobial chemokines, more efficiently than …

Nutrient deprivation based on the loss of essential amino acids by catabolic enzymes in the microenvironment is a critical means to control in ammatory responses and immune tolerance. Here we report the novel nding that Tph-1 (tryptophan hydroxylase-1), a synthase which catalyses the conversion of tryptophan to serotonin and exhausts tryptophan, is a potent regulator of immunity. In models of skin allograft tolerance, tumor growth, and experimental autoimmune encephalomyelitis, Tph-1 de ciency breaks allograft tolerance, induces tumor remission, and intensi es neuroin ammation, respectively. All of these effects of Tph-1 de ciency are independent of its downstream product serotonin. Because …

Chronic Ketamine Administration Modulates Midbrain Dopamine System In Mice, Sijie Tan, Wai Ping Lam, Maria S. M. Wai, Wan-Hua Amy Yu, David T. Yew

Ketamine is an anesthetic and a popular abusive drug. As an anesthetic, effects of ketamine on glutamate and GABA transmission have been well documented but little is known about its long-term effects on the dopamine system. In the present study, the effects of ketamine on dopamine were studied in vitro and in vivo. In pheochromocytoma (PC 12) cells and NGF differentiated-PC 12 cells, ketamine decreased the cell viability while increasing dopamine (DA) concentrations in a dose-related manner. However, ketamine did not affect the expression of genes involved in DA synthesis. In the long-term (3 months) ketamine treated mice, significant increases …

Mueller, Tippins, and Bryan’s contrast of the current limitations of science education with the potential virtues of citizen science provides an important theoretical perspective about the future of democratized science and K–12 education. However, the authors fail to adequately address the existing barriers and constraints to moving community-based science into the classroom. We contend that for these science partnerships to be successful, teachers, researchers, and other program designers must reexamine questions about traditional science education and citizen-science programs and attend to certain dimensions, including: framing these projects around the nature of science, creating a dialog with experts and allowing access …

Mueller, Tippins, and Bryan (2012) presented a new conceptualization of citizen science that is meant to facilitate emerging trends in the democratization of science and science education to produce civically engaged students. I review some relevant trends in the field of citizen science, for clarity here referred to as public participation in scientific research (PPSR), and present overlooked styles and outcomes of PPSR. Education efforts should seize the opportunity to emphasize the key and distinct roles students can play in both the science and the values elements that inform decision-making processes.

Flow cytometry analysis of propidium iodide (PI) stained nuclei isolated from leaf tissues was used to estimate the genome size of American ginseng (Panax quinquefolius L.) and evaluate inhibitor y effects of its secondary compounds on PI intercalation. American ginseng nuclear DNA content was estimated to be 10.05 ± 0.04 pg/2C, and therefore its haploid (1C) genome size is 4914 Mbp. There was no significant inhibition of PI fluorescence of reference standard nuclei co-processed with American ginseng. This indicates that secondary compounds of American ginseng do not interfere with PI intercalation. By comparison, the genome size of American ginseng is …

Just as Nebraska Athletics has long been regarded as a pioneer in the field of collegiate strength and conditioning programs, was among the first to emphasize the link between health and nutrition and athletic performance and stood in the vanguard in life skills training for student-athletes, the University of Nebraska–Lincoln (UNL) now is taking the lead by integrating athletics and research and using cutting-edge imaging technology to better elucidate the biological underpinnings of behavior and performance. No other university in the nation is conducting research in this way.

Background: Imatinib mesylate is currently the drug of choice to treat chronic myeloid leukemia. However, patient resistance and cytotoxicity make secondary lines of treatment, such as omacetaxine mepesuccinate, a necessity. Given that drug cytotoxicity represents a major problem during treatment, it is essential to understand the biological pathways affected to better predict poor drug response and prioritize a treatment regime.
Methods: We conducted cell viability and gene expression assays to determine heritability and gene expression changes associated with imatinib and omacetaxine treatment of 55 non-cancerous lymphoblastoid cell lines, derived from 17 pedigrees. In total, 48,803 transcripts derived from Illumina …

Many beneficiaries believe that Medicare provides substantial coverage for nursing home care. The coverage actually available is of short duration, incomplete, and riddled with limitations. This article discusses the various requirements to receive Medicare benefits for skilled nursing facility admissions, and it also discusses the appeals process for denied benefit claims.

While Medicare appears to afford generous coverage for hospital care, statutory limitations and changes in medical practice often make access to coverage problematic. The difficulties that Medicare beneficiaries face when receiving coverage for inpatient hospital care are discussed in this article.

One of the most troublesome issues with end-of-life decision making is the withdrawal of tube feeding, which became more problematic following the Wisconsin decision in the Edna M.F. case. This article examines case law on this issue, reviews some of the medical literature on the effects of withdrawing a feeding tube, reviews case law developments in other states, and articulates what would be a workable approach to this issue.
