This research studies the expressive and receptive skills of four Deaf children with the aim of investigating how different linguistic environment, such as the amount of input and practice, will affect their sign language acquisition. The study was conducted through observations of the subjects in the formal learning activity sessions and their regular activities in three different settings: home, church and shopping mall. Henckelia sect. Loxocarpus is a small taxon consisting of c. 26 taxa distributed in West Malesia. The present study is the first comprehensive taxonomic revision for all species of Loxocarpus. This study has four objectives. The first one is to provide a review of macro-morphological diversity and variation within Loxocarpus. Second, to carry out phylogenetic analyses to determine whether Loxocarpus is distinct from or is a section within Henckelia based on DNA sequences and morphological data. Erycibe is a big taxon consisting about 75 taxa distributed from western India and Sri Lanka across tropical and subtropical Asia to the northern tip of Queensland, Australia. The present study is focusing on the morphology and taxonomy of Erycibe in Peninsular Malaysia. The results show that Peninsular Malaysia consists of nineteen taxa. At present, seventeen taxa have scientific names with an additional two new taxa known as Erycibe. sp. A and E. sp. B. Four taxa are endemic to Peninsular Malaysia. Yogurt is generally recognized as a healthy and multifunctional food. It is a coagulated milk product obtained from the lactic acid fermentation by the action of Lactobacillus bulgaricus and Streptococcus thermophilus. In this present studies, dragon fruit Hylocereus polyrhizus and Hylocereus undatus extract was added in milk and their effects during yogurt fermentation were investigated. The changes in pH and Titratable Acidity (TA) were measured during fermentation and after 14 days of storage at 4°C. Acinetobacter baumannii is an opportunistic pathogen with increasing relevance in nosocomial infections. They cause a wide range of clinical complications, such as pneumonia, septicemia, urinary tract infection, wound infection, and meningitis, particularly in immunocompromised patients. Development And Validation Of Chromatography And Spectrometric Methods For Characterization And Impurity Profiling Of Seized Heroin For Forensic Intelligence Purposes / Chan Kar Weng, Kar Weng Chan

Heroin profiling has been undertaken to derive forensic intelligence from the seized samples and make predictions as per the criminal activities. Six major tasks were performed to collect the relevant data from 311 heroin case samples submitted in 2010. Ten visual and physical characteristics were first collected from the case samples in Task 1. Subsequently in Task 2, five types of plastic films were successfully distinguished by attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR) from the plastic packages used to contain the drug substances. This technique is rapid, robust and able to show repeatable (RSD < 2%) and reproducible (RSD < 3%) quantitative results when data normalization is used to minimize the variations arising from the poor optical contact and other uncontrolled factors. In Task 3, all the samples were analyzed by gas chromatography-mass spectrometry (GC-MS) and gas chromatography-flame ionization detection (GC-FID) to determine the levels of eight major compounds. Prior to analysis, the methods were also optimized and validated using chemical standards. The GC-FID method proved to be rapid (< 12 min), simple, precise (RSD < 5%), accurate (recoveries 99 – 102%) and robust for high throughput analysis. On the other hand, the GC-MS method is also found suitable for the profiling purpose. Furthermore, 8 links of simulated samples were analyzed following the procedures adopted by the case samples. This simulated dataset has successfully achieved eleven Normalizationselected + Standardization (Nselected + S) quotients for origin level sample classification where the clustering of case samples using principal component analysis (PCA) and hierarchical cluster analysis (HCA) operating with Ward-Manhattan is concerned. With the opium-based alkaloids, a novel tetrahedron model has also been developed to simplify the chemometric procedure involved in sample classification. Based on the purity level determined from the iii previous task, the samples were analyzed using a sample weight equivalent to 15 mg heroin base or a 650 mg constant sample weight in Task 4 for impurity profiling. This was done after 12 impurity peaks were identified and a GC-FID method was optimized and validated. The method was also demonstrated to be rapid (< 35 min), precise (RSD < 10%) and stable. Both of the sample weight approaches were statistically validated using samples of known relationships. Normalization + Standardization (N + S) was found to be the best pretreatment method and it performed particularly well in HCA operating with the Ward-Squared Euclidean and Ward-Squared Pearson. This technique was then applied to the case samples to evaluate the sample-to-sample relationships at the manufacturing level. Task 5 was carried out by quantifying trace elements. A method using inductively coupled plasma-mass spectrometry (ICP-MS) was validated using chemical standards and it proved stable for analysis. Highly variable ICP-MS data were often obtained but the intra-batch variation can be minimized via the Normalization + Fourth root (N + 4R) pretreatment method. This was verified by six batches of related samples using Ward-Squared Euclidean through HCA. Furthermore, various sources of metal contamination were also evaluated. The investigation showed that trace metals could only be used for street level sample classification. All the physical and chemical data were finally updated on a database in Task 6.

Brewery spent grain (BSG) which was used as the main organic material in this research is the by-product of brewing industries. Every 100 liter of beer generates 20kg of BSG. A Malaysian brewery (Carlsberg (M) Sdn. Bhd) alone generates 31,000 tonnes of BSG yearly. Almost all the BSG are being dumped into landfills, since there is no proper and separate guideline for industrial organic waste management in Malaysia. BSG materials are rich in nutrients and high in fermentable sugar as well as lignin, cellulose and hemicellulose. Streptomyces spp. have been the most abundant sources of all types of antibiotics. The success of marine natural products has promoted the marine environment as a source of novel chemical diversity for drug discovery. In Malaysia, the marine actinomycetes niche remains virtually unexplored and is a promising resource for the biotechnological applications including drug discovery. Screening for antifungal activity revealed that six out of 44 putative Streptomyces spp. isolated from sponges from Tioman Island showed strong antifungal activities against Candida albicans and Schizosaccharomyces pombe. Panus giganteus, a culinary and medicinal mushroom consumed by selected indigenous communities in Malaysia is currently being considered for large scale cultivation. This study was performed to investigate the medicinal potential of P. giganteus fruiting bodies and wheat grains fermented by P. giganteus including antioxidant, genoprotective and hepatoprotective properties. Ethanol extracts of P. giganteus fruiting bodies, wheat grains fermented by P. giganteus and unfermented wheat grains exhibited moderate antioxidant properties by virtue of DPPH free radical scavenging activity, reducing power, antioxidant capacity and inhibition of lipid peroxidation. This study was conducted to evaluate the effects of topical application of an ethanol extract of Goniothalamus umbrosus on the rate of skin wound healing closure and the histology of healed wound. The animal groups were topically treated with 0.2 ml of each vehicle (gum acacia), Intrasite gel, 100 and 200 mg/ml of ethanol leaf extract, respectively. We investigated the effects of nanosecond pulse electric fields (nsPEFs) on Jurkat and PANC1 cells, which are human carcinoma cell lines, in the presence of Tween 80 (T80) at a concentration of 0.18% and demonstarted an enhanced killing effect. We used two biological assays to determine cell viability after exposing cells to nsPEFs in the presence of T80 and observed a significant increase in the killing effect of nsPEFs. Nanoparticle research is often performed in vitro with little emphasis on the potential role of cell culture medium. In this study, gold nanoparticle interactions with cell culture medium and two cancer cell lines (human T-cell leukemia Jurkat and human pancreatic carcinoma PANC1) were investigated. Gold nanoparticles of 10, 25, 50, and 100 nm in diameter at fixed mass concentration were tested. Size distributions and zeta potentials of gold nanoparticles suspended in deionized (DI) water and Dulbecco's Modified Eagle's Media (DMEM) supplemented with fetal calf serum (FCS) were measured using dynamic light scattering (DLS) technique. In earlier studies, we found that permeabilization of mammalian cells with nsPEF was accompanied by prolonged inhibition of voltage-gated (VG) currents through the plasma membrane. This study explored if the inhibition of VG Na+ current (INa) resulted from (i) reduction of the transmembrane Na+ gradient due to its influx via nsPEF-opened pores, and/or (ii) downregulation of the VG channels by a Ca2+ -dependent mechanism. We describe a new phenomenon of anodotropic pseudopod-like blebbing in U937 cells stimulated by nanosecond pulsed electric field (nsPEF). In contrast to "regular," round-shaped blebs, which are often seen in response to cell damage, pseudopod-like blebs (PLBs) formed as longitudinal membrane protrusions toward anode. PLB length could exceed the cell diameter in 2 min of exposure to 60-ns, 10-kV/cm pulses delivered at 10-20 Hz. Both PLBs and round-shaped nsPEF-induced blebs could be efficiently inhibited by partial isosmotic replacement of bath NaCl for a larger solute (sucrose), thereby pointing to the colloid-osmotic water uptake as the principal driving force for bleb …

A large-scale in vitro 3D tumor model was generated to evaluate gene delivery procedures in vivo. This 3D tumor model consists of a "tissue-like" spheroid that provides a micro-environment supportive of melanoma proliferation, allowing cells to behave similarly to cells in vivo. This functional spheroid measures approximately 1 cm in diameter and can be used to effectively evaluate plasmid transfection when testing various electroporation (EP) electrode applicators. In this study, we identified EP conditions that efficiently transfect green fluorescent protein (GFP) and interleukin 15 (IL-15) plasmids into tumor cells residing in the 3D construct. The identification of individuals from commingled human remains can be a difficult task. Osteometric sorting is often utilized for these purposes since it sorts remains based on size. This research investigates whether asymmetry of the humerus is present, if it is due to an individual’s hand preference, and if this asymmetry can adversely affect the osteometric sorting method. The osteometric sorting formulae for pair-matching was used to classify individuals as asymmetric when elements were considered significantly different in size. Significant asymmetry was seen in multiple measurements and the asymmetry could be due to handedness. This relationship appeared more consistent with …

The IUCN Red List has classified all subspecies of tigers (Panthera tigris) as endangered with prey depletion being widely recognized as one of the primary drivers of tiger declines. Due to substantial energetic requirements, tigers can only survive and reproduce in areas with healthy prey populations. This may be particularly important for Amur tigers (P. t. altaica) in the Russian Far East, living at the northern limits and with the lowest prey densities of any tiger population. Few studies have been able to rigorously investigate annual prey requirements for any tiger population. We deployed Global Positioning System (GPS) collars on …
