Microbial populations have very important roles in wetlands because they are highly involved in nutrient cycling processes essential to health of the ecosystem. This study examines the changes that occur in bacterial and methanogen abundance in the soil in wetlands that have been taken over by the invasive reed Phragmites australis compared to soils that are inhabited by cattail species (Typha). To measure microbial abundance, soil samples were collected from a freshwater marsh, DNA was extracted from the soil, and bacterial DNA was amplified using primers specific to the Eubacterial l6s rRNA and methanogen …

Plant cell walls are the world's most abundant source of renewable biomass. Consisting mainly of carbohydrates, including mannans, plant cell walls are vital to humanity as a source of food, health products, and biofuels. Previous research suggests diverse functions of mannans and implicates several members of the CELLULOSE SYNTHASE-LIKE A (CSLA) gene family as mannan synthases, proteins involved in mannan backbone synthesis. Prior research using Arabidopsis thaliana (A. thaliana) cs/a single mutants showed no obvious phenotypic abnormalities, with the exception of cs/a7, which was embryo lethal; however, ectopic expression of CSLA9 complemented the csla7 mutant phenotype, suggesting csla7 …

Though science fiction is dominated by males, strong female protagonists in this genre have become very popular. Current dystopian young adult literature starring girls incorporates romance, blending a traditionally masculine genre with a typically feminine genre. The heroines in dystopian young adult novels are empowered by their own femininity and are able to bring about societal change, taking power not generally given to young females in patriarchal societies. Romance, an often ridiculed genre, has recently been incorporated into science fiction narratives in order to create more authentic characters who deal with both societal problems and personal relationships. Tyrosine-phosphorylated FAK is required for the hypertrophic response of cardiomyocytes to growth factors and mechanical load, but the role of FAK serine phosphorylation in this process is unknown. Endothelin-1 (ET-1; 1-100nM, 2-30min) and other hypertrophic factors induced a time- and dose-dependent increase in FAK-S910 phosphorylation in neonatal rat ventricular myocytes (NRVM). FAK-S910 phosphorylation required ETAR-dependent activation of PKCδ and Src via parallel Raf-1→MEK1/2→ERK1/2 and MEK5→ERK5 signaling pathways. Using co-immunoprecipitation, TIRF-microscopy and FRAP, ET-1 stimulation of NRVM expressing a nonphosphorylatable, S910A-FAK mutant decreased the interaction of paxillin and vinculin within costameres. Little historical research has been done on the role that radio stations in the northern United States played in spreading information related to the Civil Rights Movement. Owned and operated by Chess Records label owners Leonard and Phil Chess, the Chicago radio station WVON 1450 AM, "The Voice of the Negro," was one of the most popular African American radio stations in the country during the 1960s. How Do Secondary Science Teachers Understand And Implement Technological Design In Their Classrooms?, Kathryn H. Heroux

This qualitative, phenomenological study examined how ten secondary science teachers from a variety of different schools around the country understand and implement technological design in their classrooms. The results of this study indicate that these teachers were drawn to the tenets of technological design because of its inherent challenge to their own pedagogical strategies, and its ability to stimulate and motivate their students. The future of science education with respect to technological design is examined, and compared with the standards that will emerge in the new science standards soon to be finalized and circulated.

The ways in which African American women negotiate the intersections of popular media, dominant discourses of beauty, and identity are rarely explored. This work brings into focus how African American women consume, understand, and make meaning of mediated images and representations of African American women. In order to inform this particular research project, this study engages a constellation of literature and theoretical perspectives and explores historical representations of African American women and beauty messages they contain. CXCR4 is a G protein-coupled receptor (GPCR) that binds to the chemokine, stromal cell-derived factor-1 (SDF-1alpha; a.k.a. CXCL12). The SDF-1alpha/CXCR4 signaling axis plays an essential role during embryogenesis in the development of the heart, brain and vasculature and in the adult mediating immune cell trafficking and stem cell homing to the bone marrow. Dysregulation of SDF-1alpha/CXCR4 signaling is linked to several pathological conditions, including cardiovascular disease, immunological disorders as well as cancer growth and metastasis. With Fluorescence Resonance Energy Transfer (FRET), we are able to detect changes in the structure and affinity of the PLB-SERCA regulatory complex in live cells. Using this approach, we have detected a high level of PLB-SERCA interaction even at Ca2+ concentrations known to fully relieve PLB inhibition of SERCA, suggesting that dissociation is not required for relief of inhibition. We also detect no real-time change in PLB-SERCA binding over the course of a single Ca2+ transient in paced myocytes. The effect of Ca2+ on the PLB-SERCA interaction is best described as a reduced affinity with no change in the structure …

Hemodynamic stress and neurohumoral signaling are common causes of cardiac hypertrophy. These extrinsic stress stimuli typically act on GPCR and induce a cascade of signal transduction to re-program terminally differentiated myocytes to grow in length or width. The compensatory hypertrophic response can enhance cardiac output briefly due to increased work load. However, prolonged stress results in maladaptive changes in the heart and gradually deteriorates ventricular function to supply blood throughout the body. Sustained hypertrophic signaling can also progress toward heart failure.

The following dissertation presents findings from a year-long evaluation of informal scientific education programs at the Museum of Science and Industry in Chicago, Illinois. Science identity, rather than scientific knowledge, was the analytic lens through which the programs' effectiveness was assessed. A goal of the Museum generally, and the programs specifically, is to increase public identification with the field of science. Science identity was assessed using a novel survey instrument and three focus groups. Hierarchical linear models found a positive relationship between time enrolled in the Science Minors program and the development of science identity. These analyses also point to …

The purposes of the current study were to 1) examine the associations of racial discrimination to internalizing symptoms, externalizing symptoms and perceived life satisfaction in African American adolescents, and 2) determine Africultural cluster profiles based on indicators of racial socialization, racial identity and culturally relevant coping strategies 3) examine whether cluster profile buffers stress exposed African American adolescents from increased internalizing symptoms, anger and decreased life satisfaction. "Aliens Found in Waiting" is a case study of the Walosas Club chapter of the Women of the Ku Klux Klan in the suburban Chicago community of Oak Park, Illinois. Through the use of a rare manuscript collection this study provides a glimpse into the daily lives of suburban Klanswomen grappling with changing demographics in the village where they lived. Examination of the Walosas Club also provides a new context for the study of suburban history. New suburban historians have traditionally viewed suburbs as battlegrounds for the intersection of race, class and gender. Oak Park adds religion as an agent …

Far less is known about the effects of functional communication-based toileting interventions for students with developmental disabilities in a school setting. Furthermore, the currently available toileting interventions for students with disabilities include some undesirable procedures such as the use of punishment, unnatural clinic/university settings as opposed to more natural school/home settings, and prompt-based as opposed to communicative, self-initiating skills. The current study examined the effectiveness of FCT on the incidence of communicative, self-initiated toileting and of toileting accidents without any punishment components, which were often considered as necessary in traditional toileting procedures. Excessive alcohol consumption is associated with increased fracture risk, delayed bone healing and fracture non-union. Binge alcohol consumption has occurred in 25-40% of all orthopaedic trauma cases. The canonical Wnt pathway, through tight regulation of stabilized beta-catenin, plays an essential and pivotal role in the formation of new bone and cartilage to initiate bone repair. We sought to determine the molecular mechanisms behind alcohol-related fracture complications by testing the hypothesis that binge alcohol exposure deregulates canonical Wnt signaling in the fracture callus, leading to impaired healing.

The goal of this project is to develop liquid chromatography tandem mass spectrometry (LC/MS/MS) methods for the analysis of different, novel micropollutants in waste water and natural water. Micropollutants are compounds that can exert adverse environmental and human health effects at low concentrations. Most of these compounds include personal care products and pharmaceuticals, such as antibiotics and controlled substances. Mixed Lineage Leukemia (MLL) is a multidomain protein whose gene is translocated in a subset of AML leukemias. Translocation of the MLL gene is present in approximately five percent of adult acute leukemias and ten percent of pediatric leukemias (Daser, A 2004, Look, A 1997, Huret, J 2001) Patients presenting in the clinic at the time of diagnosis with an MLL fusion have been shown to respond poorly to treatment and have a worse prognosis than matched wild type MLL patients (Rubnitz, J 1994, Rubnitz, J 1999). Capillaries are comprised of two cell types: the endothelial cells (ECs) which line the lumen of the capillary, and the supporting pericytes which are closely apposed to the abluminal surface of the capillary wall (sometimes called mural cells). These pericytes are phenotypic derivations of mesenchymal organ stromal cells (from smooth muscle cells and fibroblasts in blood vessels, for example), are recruited and incorporate into the neocapillaries, and are known to mediate reciprocal interactions with ECs during angiogenesis. In this dissertation, I argue in favor of an alternative approach to the problem of global poverty, one that is distinct in significant ways from current philosophical approaches of utilitarianism, human rights, or an ethic of care. While these approaches have their merits, I will argue that they are all insufficient (in various ways) to deal with the problem of global poverty. Given their shortcomings, a new approach is needed. My alternative ethical approach contains two major facets: It provides a compelling moral motivation to act to end global poverty, and it also suggests specific initiatives that will prove effective …

Combining multiple regression estimates with meta-analysis has continued to be a difficult task. A variety of methods have been proposed and used to combine multiple regression slope estimates with meta-analysis, however, most of these methods have serious methodological and practical limitations. The purpose of this study was to explore the use of robust variance estimation for combining commonly specified multiple regression models and for combining sample-dependent focal slope estimates from diversely specified models. T cells play a central role in the immune response to fight against pathogens and orchestrate other immune cells. An overreactive immune response can lead to autoimmune diseases, therefore the immune system must possess negative regulation mechanisms. In the periphery, naturally arising regulatory T cells (nTregs) negatively regulate immune responses and play an important role in maintaining immune homeostasis. When antigens are present, conventional CD4+ T cells recognize their cognate antigen and proliferate. Thirty to 45% of patients who suffer a myocardial infarction (MI) develop anxiety disorders and/or depression, and the development of mood and anxiety disorders following MI is a major risk factor for cardiovascular morbidity and mortality. Depressed post-MI patients have an increased risk of fatal arrhythmias, which are often precipitated by emotional stressors. However, the central nervous system mechanisms that contribute to post-MI anxiety and arrhythmia susceptibility are unknown. The American horseshoe crab (HSC), Limulus polyphemus, is an economically and ecologically important species in the coastal ecosystem. Horseshoe crabs inhabit the continental shelf and estuaries from Maine to the central Gulf Coast and the Yucatán Peninsula. Although the presence of horseshoe crabs in southern Mississippi is known locally, there are limited data specific to the area and population, particularly regarding spawning and nesting habitat. Surveys of HSC presence, habitat use, and behavior on Mississippi barrier islands were conducted between March and November 2007 to 2009. PURPOSE—This study investigated the efficacy and safety of vorinostat, a deacetylase (HDAC) inhibitor, in the treatment of laser-induced corneal haze following photorefractive keratectomy (PRK) in rabbits in vivo and transforming growth factor beta 1 (TGFβ1) -induced corneal fibrosis in vitro.
METHODS—Corneal haze in rabbits was produced with −9.00 diopters (D) PRK. Fibrosis in cultured human and rabbit corneal fibroblasts was activated with TGFβ1. Vorinostat (25 μm) was topically applied once for 5 minutes on rabbit cornea immediately after PRK for in vivo studies. Background: A series of 2-amino-4-aryl-4H-benzo[h or f]chromene-3-carbonitrile derivatives were synthesized and evaluated for inhibition of Src kinase and cell proliferation in breast carcinoma (BT-20) cell lines.
Methods: The one-pot, three-component reaction of a or beta-naphthol, malonitrile and an aromatic aldehyde in the presence of diammonium hydrogen phosphate was afforded the corresponding 2-amino-4-aryl-4H-benzo[h or f]chromene-3-carbonitrile derivatives, All target compounds were evaluated for inhibition of Src kinase and cell proliferation in breast carcinoma (BT-20) cell lines.
