Characterization Of A Reductively-Activated Elimination Pathway Relevant To The Biological Chemistry Of The Kinamycins And Lomaiviticins, Seann P. Mulcahy, Christina M. Woo, Weidong Ding, George A. Ellestad, Seth B. Herzon

The lomaiviticins (1 and 2) and kinamycins (3–5) are bacterial metabolites with potent antimicrobial and antiproliferative activities. Herein we establish that 1–5 are capable of generating electrophilic acylfulvene intermediates (6) under mildly reducing conditions. These acylfulvenes 6 are formed by a multistep process comprising two-electron reduction and loss of dinitrogen to form an ortho-quinone methide, followed by elimination. Based on these studies, the structure of the product formed from 1 in DNA-cleavage assays is proposed (26). We also show that the bis(hydroxynaphthoquinone) substructures of the lomaiviticins activate the metabolites toward reduction. Finally, based on COMPARE and time-dependent cell response profiling …

The large Southeast Asian genus Desmoxytes is slightly rediagnosed. A number of troglomorphic, most likely troglobitic, species occur in southern China. A key is provided to all 10 Desmoxytes spp. currently known from China, including three new presumed troglobites: Desmoxytes eupterygota sp. n. from Hunan Province, as well as Desmoxytes spinissima sp. n. and Desmoxytes lui sp. n. from Guangxi Province. “Desmoxytes” philippina Nguyen Duc & Sierwald, 2010, from the Philippines, is formally removed from Desmoxytes, but not assigned to another genus. It probably belongs in a new genus in the subfamily Australiosomatinae, tribe Antichiropodini, close to the Bornean Euphyodesmus …

Though the term "model minority" was only coined in the 1960s to apply to Asian-Americans, in the white imagination, a "model" way to be a minority has always existed. This model has gone through various iterations from the "happy" blacks of the antebellum years to supposedly over-achieving Asian-Americans of the 1960s. I argue that we are witnessing the emergence of a new model minority that includes not only Asian-Americans but other high-achieving minorities as well. This model minority is characterized by economic success, formal education, American values, and conservative racial politics or a complete silence about race. Because an inability …

Understanding and specification of the higher altitudes of the atmosphere with global coverage over all local times is hampered by the challenges of obtaining direct measurements in the upper atmosphere. Methods to measure the properties of the atmosphere above the stratopause is an active area of scientific research. In this thesis, we revisit the use of infrasound as a passive remote sensing technique for the upper atmosphere. Signals from the Tungurahua volcano in Ecuador are used to investigate the behavior of the upper atmosphere. The primary objective of this study was to evaluate the psychometric properties of the Psychological Maltreatment (PM) and Neglect subscales of the Computer Assisted Maltreatment Inventory (CAMI; DiLillo et al., 2010). The CAMI is a retrospective self-report measure that assesses multiple forms of child maltreatment (i.e., sexual, physical, psychological, neglect, exposure to interparental violence) retrospectively from adults. The CAMI’s PM and Neglect subscales were administered to a geo-graphically diverse sample of 400 college students and a sample of 412 newlyweds. Fire blight caused by the bacterium Erwinia amylovora results in millions of dollar in losses worldwide. It is the most important disease problem for apple and pear growers in Utah. Currently the only effective management strategy is application of the antibiotic streptomycin. In 2006, resistant isolates were detected in an apple orchard in Utah County. To determine the distribution of resistant isolates, samples collected in 2006, 2007, 2010 and 2011 from orchards across Utah were tested for resistance to streptomycin. Isolates were screened at 0, 100 and 1000ppm of streptomycin. Bacteria were spread on LB agar. A hole was punched …

Dubbed the “cradle of the Amazon,” and covering more than 2.4 million acres in the Amazon River Basin, Ecuador’s Yasuní National Forest is perhaps the most biodiverse place on Earth. Almost 200 species of mammal, 560 fish species, 300 reptile and amphibian species and close to 600 different types of birds can be found within the parks parameters. In just 2.5 acres of the park can be found as many tree species as in the U.S. and Canada combined. In addition to its ecological diversity, Yasuní supports several tribes of indigenous people, which remain voluntarily isolated in the forest. In …

Although the United States has not yet adopted a national greenhouse gas cap-and-trade program, carbon trading is nonetheless an active industry. Private companies continue to participate in voluntary carbon markets despite the closure of the Chicago Climate Exchange. On the east coast, the Regional Greenhouse Gas Initiative (“RGGI”) forges into its seventh year, while in the west, California, a pioneer of cap- and-trade, launched its own statewide greenhouse gas cap and trade program. While these state efforts to curb greenhouse gases are laudable, the disparate cap-and-trade programs have yielded inconsistent definitions of carbon credits. To promote efficiency in this new …

Here, we determined the potential of photochemotherapy, namely the application of photodynamic compounds followed by exposure to a suitable source of UV-visible radiation against corneal pathogen, Acanthamoeba. Organometallic macromolecule, tin porphyrin [Sn(IV)porphyrin] was synthesized and purity confirmed using nuclear magnetic resonance spectroscopy. The Sn(IV)porphyrin was tested against a keratitis isolate of Acanthamoeba castellanii belonging to the T4 genotype using growth and viability assays. The effects of Sn(IV)porphyrin on A. castellanii binding to and cytopathogenicity of human corneal epithelial cells in vitro were tested. The metalloporphyrin showed potent amoebistatic effects. This investigation was aimed to probe the pharmacological base of medicinal use of Acorus calamus in ischemic heart diseases. Effect on heart parameters was studied in isolated rabbit heart while coronary vasodilator effect was studied in isolated bovine coronary arterial rings, suspended in tissue baths filled with Krebs solution, maintained at 37degreesC, aerated with carbogen and responses were measured on PowerLab data acquisition system. In Langendorrf's perfused rabbit heart, the crude extract of Acorus calamus (Ac.Cr) at 0.01-10 mg/mL partially suppressed force of ventricular contractions (FVC), heart rate (HR) and coronary flow (CF). Thymoquinone (TQ) is a bioactive component found in many medicinal herbs. In this study, we report the smooth and cardiac muscle relaxant activities of this compound. TQ concentration dependently suppressed spontaneously contracting rabbit jejunum while also relaxed high K+-(80 mM) induced contractions in jejunum and guinea-pig ileum, indicating activity at voltage-operated Ca++ channels (VOCC). Further, TQ displaced Ca++ concentration-response curves, obtained in a Ca++-free environment, to the right, showing blockade of VOCC. Similar activity was observed with verapamil, a standard VOCC blocker. TQ also exhibited nonadrenergic relaxation of agonist-induced contractions in guinea-pig trachea. Background: The aqueous-methanolic extract of Amaranthus spinosus (A. spinosus Linn.,) whole plant, was studied for its laxative, spasmolytic and bronchodilator activities to validate some of its medicinal uses.
Methods: The crude extract of A. spinosus was studied in-vivo for bronchodilator and laxative activities and in-vitro using isolated tissue preparations which were mounted in tissue baths assembly containing physiological salt solutions, maintained at 37°C and aerated with carbogen, to assess the spasmolytic effect and to find out the possible underlying mechanisms.

Haitian-American author Edwidge Danticat evokes the Haitian tradition of storytelling in many of her novels and short story collections. A tradition formulated by vodou religion and the amalgamation of African cultures, storytelling acts to entertain, educate and enlighten the people of Haiti. Additionally, her novels are often written in the context of traumatic events in Haitian history. While Danticat's works have been studied with focus on their depiction of storytelling and of trauma, little has been done on the restorative power that storytelling provides. An ethnographic pre-assessment study of the behavioral dynamics of men who have sex with men (MSM) and injecting drug users (IDU) was conducted in 2009 as an integral part of the 2010 Integrated Biological and Behavioural Surveillance Survey (IBBSS). The study took place in Northern and Southern Nigeria and provided rich contextual information on the logistic and scientific factors that would aid successful conduct of the IBBSS and secure buy-in for it among study respondents. A new class of distributions recently developed involves the logit of the beta distribution. Among this class of distributions are, the beta-Normal (Eugene et al. [15]); beta-Gumbel (Nadarajah and Kotz [18]); beta-Exponential (Nadarajah and Kotz [19]); beta-Weibull (Famoye et al. [6]); beta-Rayleigh (Akinsete and Lowe [3]); beta-Laplace (Kozubowshi and Nadarajah [20]); and beta-Pareto (Akinsete et al. [4]), among a few others. Many useful statistical properties arising from these distributions and their applications to real life data have been discussed in literature.
