Geography and the Environment Poster Presentations

Enhancing Thermal Tolerance By Eliminating The Pejus Range: A Comparative Study With Three Decapod Crustaceans, Jennifer A. Jost, Stephanie M. Podolski, Markus Frederich

Marine Sciences Faculty Publications

Marine invertebrates in the intertidal and subtidal zones are often exposed to highly variable environmental conditions, especially rapid changes in temperature. The ability to survive at different temperatures has previously been described using an extended version of Shelford’s law of tolerance, with optimum, pejus (Latin: ‘turning worse’), and pessimum ranges, and the respective thresholds, critical (Tc) and pejus (Tp) temperatures, that mark the transition from one range into the next. The width of the pejus range, in which the scope for activity gradually declines, varies among species. We tested the hypothesis that the width of the …

Evidence For A Grooming Claw In A North American Adapiform Primate: Implications For Anthropoid Origins, Stephanie Maiolino, Doug M. Boyer, Jonathan I. Bloch, Christopher C. Gilbert, Joseph Groenke

Publications and Research

Among fossil primates, the Eocene adapiforms have been suggested as the closest relatives of living anthropoids (monkeys, apes, and humans). Central to this argument is the form of the second pedal digit. Extant strepsirrhines and tarsiers possess a grooming claw on this digit, while most anthropoids have a nail. While controversial, the possible presence of a nail in certain European adapiforms has been considered evidence for anthropoid affinities. Skeletons preserved well enough to test this idea have been lacking for North American adapiforms. Here, we document and quantitatively analyze, for the first time, a dentally associated skeleton of Notharctus tenebrosus …

Biology And Pathogenesis Of Acanthamoeba., Ruqaiyyah Siddiqui, Naveed Ahmed Khan Jan 2012

Biology And Pathogenesis Of Acanthamoeba., Ruqaiyyah Siddiqui, Naveed Ahmed Khan

Department of Biological & Biomedical Sciences

Acanthamoeba is a free-living protist pathogen, capable of causing a blinding keratitis and fatal granulomatous encephalitis. The factors that contribute to Acanthamoeba infections include parasite biology, genetic diversity, environmental spread and host susceptibility, and are highlighted together with potential therapeutic and preventative measures. The use of Acanthamoeba in the study of cellular differentiation mechanisms, motility and phagocytosis, bacterial pathogenesis and evolutionary processes makes it an attractive model organism. There is a significant emphasis on Acanthamoeba as a Trojan horse of other microbes including viral, bacterial, protists and yeast pathogens.

Aggregative Feeding Behavior In Sea Urchins Leads To Destructive Grazing In A Nova Scotian Kelp Bed, Colette Feehan, R. E. Scheibling, J. S. Lauzon-Guay Jan 2012

Aggregative Feeding Behavior In Sea Urchins Leads To Destructive Grazing In A Nova Scotian Kelp Bed, Colette Feehan, R. E. Scheibling, J. S. Lauzon-Guay

Department of Biology Faculty Scholarship and Creative Works

Grazing aggregations of the sea urchin Strongylocentrotus droebachiensis drive the transition between alternative ecosystem states in Nova Scotia, from productive kelp beds to less productive barrens. This transition can be initiated by the formation of gaps within a kelp bed, containing dense aggregations of sea urchins. We examined the importance of local density of sea urchins and pre-existing gaps in a kelp canopy in mediating the formation of destructive grazing aggregations of sea urchins in a kelp bed. We transplanted 14 000 adult sea urchins from a barrens on the Atlantic coast of Nova Scotia into ̃4.5 m 2 plots …

Protein Docking Prediction Using Predicted Protein-Protein Interface, Bin Li, Daisuke Kihara

Faculty Publications

Background: Many important cellular processes are carried out by protein complexes. To provide physical pictures of interacting proteins, many computational protein-protein prediction methods have been developed in the past. However, it is still difficult to identify the correct docking complex structure within top ranks among alternative conformations. Results: We present a novel protein docking algorithm that utilizes imperfect protein-protein binding interface prediction for guiding protein docking. Since the accuracy of protein binding site prediction varies depending on cases, the challenge is to develop a method which does not deteriorate but improves docking results by using a binding site prediction which may …

Usp8 Promotes Smoothened Signaling By Preventing Its Ubiquitination And Changing Its Subcellular Localization, Ruohan Xia, Hongge Jia, Junkai Fan, Yajuan Liu, Jianhang Jia Jan 2012

Usp8 Promotes Smoothened Signaling By Preventing Its Ubiquitination And Changing Its Subcellular Localization, Ruohan Xia, Hongge Jia, Junkai Fan, Yajuan Liu, Jianhang Jia

Molecular and Cellular Biochemistry Faculty Publications

The seven transmembrane protein Smoothened (Smo) is a critical component of the Hedgehog (Hh) signaling pathway and is regulated by phosphorylation, dimerization, and cell-surface accumulation upon Hh stimulation. However, it is not clear how Hh regulates Smo accumulation on the cell surface or how Hh regulates the intracellular trafficking of Smo. In addition, little is known about whether ubiquitination is involved in Smo regulation. In this study, we demonstrate that Smo is multi-monoubiquitinated and that Smo ubiquitination is inhibited by Hh and by phosphorylation. Using an in vivo RNAi screen, we identified ubiquitin-specific protease 8 (USP8) as a deubiquitinase that …

Tumor Suppression By Cell Competition Through Regulation Of The Hippo Pathway, Chiao-Lin Chen, Molly C. Schroeder, Madhuri Kango-Singh, Chunyao Tao, Georg Halder

Biology Faculty Publications

Homeostatic mechanisms can eliminate abnormal cells to prevent diseases such as cancer. However, the underlying mechanisms of this surveillance are poorly understood. Here we investigated how clones of cells mutant for the neoplastic tumor suppressor gene scribble (scrib) are eliminated from Drosophila imaginal discs. When all cells in imaginal discs are mutant for scrib, they hyperactivate the Hippo pathway effector Yorkie (Yki), which drives growth of the discs into large neoplastic masses. Strikingly, when discs also contain normal cells, the scrib− cells do not overproliferate and eventually undergo apoptosis through JNK-dependent mechanisms. However, induction of apoptosis does …

Serotonin 1a Receptor-Mediated Signaling Through Erk And Pkcα Is Essential For Normal Synaptogenesis In Neonatal Mouse Hippocampus, A. Mogha, Sara R. Guariglia, Priya Ranjan Debata, Guang Y. Wen, Probal Banerjee

Publications and Research

Aberrant expression of the presynaptic serotonin 1A receptor (5-HT1A-R) because of a polymorphism in the 5-HT1A-R gene is associated with severe depression in human, whereas its absence up to postnatal day 21 (P21) in the forebrain of mice results in heightened anxiety in adulthood. These observations collectively indicate that the 5-HT1A-R has a crucial role in brain development. To understand the mechanistic underpinnings of this phenomenon, we used organotypic slice cultures of hippocampi from C57BL6 mice (C57) at P15, which coincides with the peak of neonatal synaptogenesis. Stimulation of the hippocampal 5-HT1A-R caused a dramatic increase …

Derivation Of A Novel Efficient Supervised Learning Algorithm From Cortical-Subcortical Loops, Ashok Chandrashekar, Richard Granger Jan 2012

Derivation Of A Novel Efficient Supervised Learning Algorithm From Cortical-Subcortical Loops, Ashok Chandrashekar, Richard Granger

Dartmouth Scholarship

Although brain circuits presumably carry out powerful perceptual algorithms, few instances of derived biological methods have been found to compete favorably against algorithms that have been engineered for specific applications. We forward a novel analysis of a subset of functions of cortical-subcortical loops, which constitute more than 80% of the human brain, thus likely underlying a broad range of cognitive functions. We describe a family of operations performed by the derived method, including a non-standard method for supervised classification, which may underlie some forms of cortically dependent associative learning. The novel supervised classifier is compared against widely used algorithms for …

Identification And Quantification Of Protein Carbonylation By Mass Spectrometry, Qingyuan Liu Jan 2012

Identification And Quantification Of Protein Carbonylation By Mass Spectrometry, Qingyuan Liu

Theses and Dissertations

Accumulated evidence indicates oxidative stress plays important roles in disease and aging. Under oxidative stress, lipid peroxidation (LPO) leads to reactive carbonyl species (RCS) that can modify a wide range of biomolecules including protein, DNA and carbohydrate. In this dissertation, we investigate the modification of two model proteins, human serum albumin (HSA) and aconitase (ACO), by the LPO-relevant a, b-unsaturated aldehydes, acrolein (ACR) and 4-hydroxy-2-nonenal (HNE). The investigation is focused on the characterization and quantification ACR and HNE addition to the model proteins. A correlation between HNE modification and ACO activity is also determined. These results provide insights into the …

Aquaporin 4 Is A Ubiquitously Expressed Isoform In The Dogfish (Squalus Acanthias) Shark, Christopher P. Cutler, Bryce Maciver, Gordon Cramb, Mark L. Zeidel

Biology: Faculty Publications

The dogfish ortholog of aquaporin 4 (AQP4) was amplified from cDNA using degenerate PCR followed by cloning and sequencing. The complete coding region was then obtained using 5′ and 3′ RACE techniques. Alignment of the sequence with AQP4 amino acid sequences from other species showed that dogfish AQP4 has high levels (up to 65.3%) of homology with higher vertebrate sequences but lower levels of homology to Agnathan (38.2%) or teleost (57.5%) fish sequences. Northern blotting indicated that the dogfish mRNA was approximately 3.2 kb and was highly expressed in the rectal gland (a shark fluid secretory organ). Semi-quantitative PCR further …

The Complications Of Surrogacy: A New Jersey Court Refuses To Uphold A Surrogacy Arrangement, But Awards Full Custody To The Intended Father, Joanna L. Grossman Jan 2012

The Complications Of Surrogacy: A New Jersey Court Refuses To Uphold A Surrogacy Arrangement, But Awards Full Custody To The Intended Father, Joanna L. Grossman

Hofstra Law Faculty Scholarship

Surrogacy arrangements, in which a set of intended parents contract with a woman to gestate a child for them in exchange for money, have many potential complications. Studies of surrogacy suggest that the vast, vast majority of surrogacy contracts are actually carried out without a hitch. But when things go wrong, they can go drastically wrong, leaving courts and the parties to the agreement to sort out a situation that was never intended to occur.

An ongoing litigation in New Jersey, A.G.R. v. D.R.H., presents just such a case. There, a gay male couple, S.H. and D.R.H., decided to …

Spiritual Economies: Redemptive Violence, The Body And Social Care, Perry W. Benson Jan 2012

Spiritual Economies: Redemptive Violence, The Body And Social Care, Perry W. Benson

Theses and Dissertations

“Spiritual Economies” asserts that the emancipatory drive in international social care to integrate spirituality and religion in culturally sensitive practice, is much needed given the demographic and social changes engendered by globalization. Ethical codes guiding the practice of international social care, furthermore, have mandated that spiritual and religious beliefs are an important part of cultural diversity. An extensive examination and analysis of the discourse on the integration of religion and spirituality in the predominantly secular social services, however, yielded a number of concerns among international social care scholars and practitioners regarding the inclusion of religion due to the potential for …

The Impact Of Resiliency And Biculturalism On Subjective Well Being In A Northern Plains American Indian Tribe, Paula Maureen Carter Jan 2012

The Impact Of Resiliency And Biculturalism On Subjective Well Being In A Northern Plains American Indian Tribe, Paula Maureen Carter

Theses and Dissertations

For generations American Indians struggled with the burden of historical trauma. Despite this trauma, American Indian people have survived and many flourish. With the majority of American Indian research focusing on deficits in this population it is difficult to gain a perspective that positive, strength-based factors may contribute to this cultural survival. The intent of this study is to present evidence for the position that resiliency exists in this population and that the resilience is directly related to acculturation, specifically a bicultural orientation. Further, subjective well-being is impacted by resiliency and biculturalism. Participants from a Northern Plains Ojibwa tribe completed …

Fully Enclosed Microfluidic Paper-Based Analytical Devices, Kevin M. Schilling, Anna L. Lepore, Jason A. Kurian, Andres W. Martinez

Chemistry and Biochemistry

This article introduces fully enclosed microfluidic paper-based analytical devices (microPADs) fabricated by printing toner on the top and bottom of the devices using a laser printer. Enclosing paper-based microfluidic channels protects the channels from contamination, contains and protects reagents stored on the device, contains fluids within the channels so that microPADs can be handled and operated more easily, and reduces evaporation of solutions from the channels. These benefits extend the capabilities of microPADs for applications as low-cost point-of-care diagnostic devices.
