This study demonstrated a method for toughening a highly crosslinked anhydride cured DGEBA epoxy using rubbery block copolymer grafted SiO2 nanoparticles. The particles were synthesized by a sequential reversible addition-fragmentation chain transfer (RAFT) polymerization. The inner rubbery block poly(n-hexyl methacrylate) (PHMA) had a glass transition temperature below room temperature. The outer block poly(glycidyl methacrylate) (PGMA) was matrix compatible. A rubbery interlayer thickness of 100% and 200% of the particle core radius was achieved by grafting a 20 kg/mol and a 40 kg/mol PHMA at a graft density of 0.7 chains/nm2 from the SiO2 surface. The 20 kg/mol rubbery …

A Comparative Review Of “How To” Books For Parents Of Adhd Children And “How To” Books For Parents Of Typical Children, Bora Pajo, Paul H. Stuart Dr.

All Faculty and Staff Scholarship

Although an increasing number of children are diagnosed with Attention Deficit Hyperactivity Disorder (ADHD) and take medications to control their behaviors, a well-publicized controversy persists about whether ADHD refers to a common neurobehavioral disorder of childhood or to various medicalized temperamental, educational, and cultural differences and difficulties of children. Moreover, behaviors indicative of ADHD are commonly found among all children—although with different frequencies. This paper aims to identify and compare the type of information provided to parents of ADHD and non-ADHD diagnosed children through readily available self-help books. Searches using popular online bookstores were conducted to rank and select the …

River otters (Lontra canadensis) are highly active, semi-aquatic mammals indigenous to a range of elevations and represent an appropriate model for assessing the physiological responses to diving at altitude. In this study, we performed blood gas analyses and compared blood chemistry of river otters from a high-elevation (2357 m) population at Yellowstone Lake with a sea-level population along the Pacific coast. Comparisons of oxygen dissociation curves (ODC) revealed no significant difference in hemoglobin-oxygen (Hb-O2) binding affinity between the two populations - potentially because of demands for tissue oxygenation. Instead, high-elevation otters had greater Hb concentrations (18.7 …

Orai1 Deficiency Leads To Heart Failure And Skeletal Myopathy In Zebrafish., Mirko Völkers, Nima Dolatabadi, Natalie Gude, Patrick Most, Mark A Sussman, David Hassel

Center for Translational Medicine Faculty Papers

Mutations in the store-operated Ca²⁺ entry pore protein ORAI1 have been reported to cause myopathies in human patients but the mechanism involved is not known. Cardiomyocytes express ORAI1 but its role in heart function is also unknown. Using reverse genetics in zebrafish, we demonstrated that inactivation of the highly conserved zebrafish orthologue of ORAI1 resulted in severe heart failure, reduced ventricular systolic function, bradycardia and skeletal muscle weakness. Electron microscopy of Orai1-deficient myocytes revealed progressive skeletal muscle instability with loss of myofiber integrity and ultrastructural abnormalities of the z-disc in both skeletal and cardiac muscle. Isolated Orai1-deficient cardiomyocytes showed loss …

The genetic diversity within and among parasite populations provides clues to their evolutionary history. Here, we sought to determine whether mitochondrial and microsatellite DNA variation could be used to evaluate the extent of differentiation, gene flow and historical reproductive isolation among the freeze resistant parasites Trichinella nativa and the Trichinella T6 genotype infecting wolverines (Gulo gulo) in Nunavut, Canada. To this end, we genotyped Trichinella isolates derived from the diaphragms of 39 wol- verines from this locale to reference strains of T. nativa and the Trichinella T6 genotype. Results showed that among a subset of 13 isolates examined, individuals resembled …

Superarmed Glycosyl Donors In Chemical Glycosylation, Hemali Deepthika Premathilake

Dissertations

Only recently has the tremendous biological significance and therapeutic potential of carbohydrates and conjugates thereof (glycoproteins, glycolipids, proteoglycans, etc.) begun to emerge. As the appreciation for the biological roles of carbohydrates a growing demand for efficient and scalable methods towards the synthesis of these challenging molecules has become even more imperative. This has led to the development of a variety of expeditious strategies for oligosaccharide assembly. Amongst these strategies, the so-called armed-disarmed strategy introduced by Fraser-Reid is of particular attractiveness as it allows for chemoselective oligosaccharide synthesis. Recently, the conformational changes as well as the strategic placement of common protecting …

What Fescue Toxicosis Is Really Doing Inside Your Animals, Glen E. Aiken Jan 2012

What Fescue Toxicosis Is Really Doing Inside Your Animals, Glen E. Aiken

Forage Symposium at the Kentucky Cattlemen’s Convention

Eighty years have passed since Dr. F.N. Fergus collected seed of tall fescue from that hillside at the Suiter Farm in Menifee County that led to the commercial release of the cultivar ‘Kentucky 31’. Fescue provided an opportunity to replace the briar and weed patches that dominated the rocky hillsides of Kentucky with productive forage. Plantings of tall fescue were numerous in the state during the 1940s and 1950s, and its hardiness and adaptability resulted in the grass spreading over much of the middle and upper southeastern USA, eventually covering a region we now call the “fescue belt”. It did …

War Crimes Prosecution – Lessons Learned From Bosnia And Herzegovina, Iva Vukusic

Conflict, Memory, and Reconciliation: Bridging past, present, and future

Since the war ended fifteen years ago, war crimes prosecution has been the dominant tool of addressing war crimes committed during the four-year-long conflict in Bosnia and Herzegovina (1992-1995). The prosecution has been conducted at the International Criminal Tribunal for the former Yugoslavia (ICTY), the national institutions (Court of BiH, Sarajevo) and the local courts (cantonal and district in Republika Srpska, the Federation of BiH and the Brcko District – all entities established as a result of the Dayton Peace Accords). All these institutions have been criticized by various individuals in different capacities, for different reasons – some of these …

The Role Of Mitochondrial Dysfunction In Neurodegenerative Proteinopathies And Aging., Alejandro Ocampo Jan 2012

The Role Of Mitochondrial Dysfunction In Neurodegenerative Proteinopathies And Aging., Alejandro Ocampo

Open Access Dissertations

Age-related neurodegenerative proteinophaties, including polyglutamine (polyQ) diseases such as Huntington’s disease, are a group of disorders in which a single protein or a set of proteins misfold and aggregate resulting in a progressive and selective loss of anatomically or physiologically related neuronal systems. Despite evidence showing a clear relationship between mitochondrial dysfunction, aging and neurodegenerative proteinophaties, the extent of the mitochondrial respiratory chain deficits, the involvement of mitochondrial dysfunction and the mechanisms responsible for these processes are largely unknown. Using yeast models of cellular aging and polyQ disorders we show that mitochondrial dysfunction is an important contributor to the process …

Serotonin (5-HT) regulates important biological and psychological processes including mood, and may be associated with the development of several psychiatric disorders. An association between psychopathology and genes that regulate 5-HT neurotransmission is a robust area of research. Identification of the genes responsible for the predisposition, development, and pharmacological response of various psychiatric disorders is crucial to the advancement of our understanding of their underlying neurobiology. This review highlights research investigating 5-HT transporter (5-HTTLPR) polymorphism, because studies investigating the impact of the 5-HTTLPR polymorphism have demonstrated significant associations with many psychiatric disorders. Decreased transcriptional activity of the S allele (“risk allele”) …

The highly conserved phosphatase calcineurin (CaN) plays vital roles in numerous processes including T-cell activation, development and function of the central nervous system, and cardiac growth. It is activated by the calcium sensor calmodulin (CaM). CaM binds to a regulatory domain (RD) within CaN, causing a conformational change that displaces an autoinhibitory domain (AID) from the active site, resulting in activation of the phosphatase. This is the same general mechanism by which CaM activates CaM-dependent protein kinases. Previously published data have hinted that the RD of CaN is intrinsically disordered. In this work, we demonstrate that the RD is unstructured …

God Has No Grandchildren, Steve Case Jan 2012

God Has No Grandchildren, Steve Case

180 Symposium Publications

Biological births create second and third generations. A spiritual birth creates a child of God regardless of that person’s age or relationship to other biological generations. That’s why God has no grandchildren; only children. The second birth may lack obvious, tangible markers. Those who grow up with religious parents might not be able to name a date or place for their own spiritual re-birth. New environments seem more conducive for new-birth experiences. We need additional markers. Complications, misunderstanding, and power plays may arise when the first, second and third generations flatten into one generation called “Children of God.”

Uterine Dysfunction In Biglycan And Decorin Deficient Mice Leads To Dystocia During Parturition., Zhiping Wu, Abraham W Aron, Elyse E Macksoud, Renato V Iozzo, Chi-Ming Hai, Beatrice E Lechner

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

Cesarean birth rates are rising. Uterine dysfunction, the exact mechanism of which is unknown, is a common indication for Cesarean delivery. Biglycan and decorin are two small leucine-rich proteoglycans expressed in the extracellular matrix of reproductive tissues and muscle. Mice deficient in biglycan display a mild muscular dystrophy, and, along with mice deficient in decorin, are models of Ehlers-Danlos Syndrome, a connective tissue anomaly associated with uterine rupture. As a variant of Ehlers-Danlos Syndrome is caused by a genetic mutation resulting in abnormal biglycan and decorin secretion, we hypothesized that biglycan and decorin play a role in uterine function. Thus, …

Altering Iron Oxide Nanoparticle Surface Properties Induce Cortical Neuron Cytotoxicity, Christopher J. Rivet, Yuan Yuan, Diana Andra Borca-Tasciuc, Ryan J. Gilbert

Chemical and Biochemical Engineering Faculty Research & Creative Works

Superparamagnetic iron oxide nanoparticles, with diameters in the range of a few tens of nanometers, display the ability to cross the blood-brain barrier and are envisioned as diagnostic and therapeutic tools in neuro-medicine. However, despite the numerous applications being explored, insufficient information is available on their potential toxic effect on neurons. While iron oxide has been shown to pose a decreased risk of toxicity, surface functionalization, often employed for targeted delivery, can significantly alter the biological response. This aspect is addressed in the present study, which investigates the response of primary cortical neurons to iron oxide nanoparticles with coatings frequently …

Case Report Of Dramatic Resolution Of Psychotic Symptoms During Cross-Over To Clozapine, Leo Sher M.D., Alan Mendelowitz M.D.

Jefferson Journal of Psychiatry

Clozapine is the first antipsychotic drug with proven superiority over conventional antipsychotics in the management of treatment-resistant patients. We describe a case of tratment-resistant schizoaffective disorder in a young woman who improved rapidly on clozapine. The patient was started on a low dose of clozapine while fluphenazine was decreased. She improved significantly during the first several days of treatment. This improvement took place well before the expected effect of clozapine. Possible explanations for this unusual response include: 1) the placebo effect; 2) fluphenazine dose-response curve; 3) acute clozapine neurochemical mechanisms; 4) inaccurate original diagnosis. Better understanding of the mechanisms of …

Case Report: Failure Of Group Therapy With Elderly Russian-Jewish Immigrants (An Exploratory Analysis), Gennady Berezkin M.D.

Jefferson Journal of Psychiatry

This paper is a description of an unsuccessful attempt to carry out group therapy with elderly Russian-Jewish immigrants.

An attempt was made to conduct group therapy following general principals of traditional psychotherapy with geriatric populations.

In the final analysis, we conclude that culturally based preconceptions toward psychotherapy, culturally based stereotypes of interpersonal relationships and culturally-based transference to the therapist were the primary reasons for the development of negative transference and resistance which culminated in the ultimate failure of group psychotherapy with this group of elderly immigrants.

An equine SNP genotyping array was developed and evaluated on a panel of samples representing 14 domestic horse breeds and 18 evolutionarily related species. More than 54,000 polymorphic SNPs provided an average inter-SNP spacing of ∼43 kb. The mean minor allele frequency across domestic horse breeds was 0.23, and the number of polymorphic SNPs within breeds ranged from 43,287 to 52,085. Genome-wide linkage disequilibrium (LD) in most breeds declined rapidly over the first 50-100 kb and reached background levels within 1-2 Mb. The extent of LD and the level of inbreeding were highest in the Thoroughbred and lowest in the …
