The Role Of Ezh2 In The Regulation Of The Activity Of Matrix Metalloproteinases In Prostate Cancer Cells., Yong Jae Shin, Jeong-Ho Kim Jan 2012

The Role Of Ezh2 In The Regulation Of The Activity Of Matrix Metalloproteinases In Prostate Cancer Cells., Yong Jae Shin, Jeong-Ho Kim

Biochemistry and Molecular Medicine Faculty Publications

Degradation of the extracellular matrix (ECM), a critical step in cancer metastasis, is determined by the balance between MMPs (matrix metalloproteinases) and their inhibitors TIMPs (tissue inhibitors of metalloproteinases). In cancer cells, this balance is shifted towards MMPs, promoting ECM degradation. Here, we show that EZH2 plays an active role in this process by repressing the expression of TIMP2 and TIMP3 in prostate cancer cells. The TIMPgenes are derepressed by knockdown of EZH2 expression in human prostate cancer cells but repressed by overexpression of EZH2 in benign human prostate epithelial cells. EZH2 catalyzes H3K27 trimethylation and subsequent DNA methylation …

Kno.e.sis Publications

Background: Research on the biology of parasites requires a sophisticated and integrated computational platform to query and analyze large volumes of data, representing both unpublished (internal) and public (external) data sources. Effective analysis of an integrated data resource using knowledge discovery tools would significantly aid biologists in conducting their research, for example, through identifying various intervention targets in parasites, and in deciding the future direction of ongoing as well as planned projects. A key challenge in achieving this objective is the heterogeneity between the internal lab data, usually stored as flat files, Excel spreadsheets or custom-built databases, and the external …

Energy-Based Yield Criterion For Pmma From Large-Scale Molecular Dynamics Simulations, Eugenio Jaramillo, Nathaniel Wilson, Stephen Christensen, Jonathan Gosse, Alejandro Strachan Jan 2012

Energy-Based Yield Criterion For Pmma From Large-Scale Molecular Dynamics Simulations, Eugenio Jaramillo, Nathaniel Wilson, Stephen Christensen, Jonathan Gosse, Alejandro Strachan

Birck and NCN Publications

We use molecular dynamics (MD) with the DREIDING force field to characterize the ultimate mechanical response of amorphous poly(methyl methacrylate). We characterize how volumetric and deviatoric strains contribute to yield for a wide range of loading conditions from pure deviatoric, volume-conserving cases to isotropic volume expansion. We propose and apply an energy-based yield criterion to define yield consistently for all cases. Our results show that permanent deformation occurs when either the deviatoric or volumetric strains reach critical values, except in a narrow region around the transformation between deviatoric- and volumetric-dominated yield where the two strain invariants interact. In contrast, the …

Concept Inventory Design For Determining Students’ Conceptual Understanding Of Oceanography, Leilani Arthurs Jan 2012

Concept Inventory Design For Determining Students’ Conceptual Understanding Of Oceanography, Leilani Arthurs

Discipline-Based Education Research Group Speaker Series

Concept inventories are relatively new types of diagnostic instruments intended to measure student learning. Concept inventories exist for astronomy, biology, chemistry, engineering, fluid mechanics, geology, and physics. None is yet available for oceanography, and development of the Introductory Oceanography Concept Inventory Survey (IO‐CIS) serves to fill this gap. A context‐driven development strategy was designed to create this instrument. Qualitative methods utilizing grounded theory and classical test theory were used to construct it. Quantitative methods, including statistical methods associated with classical test theory and item response theory, were used to evaluate and further refine the IO‐CIS. The instrument is valid and …

Successful mass drug administration (MDA) campaigns have brought several countries near the point of Lymphatic Filariasis (LF) elimination. A diagnostic tool is needed to determine when the prevalence levels have decreased to a point that MDA campaigns can be discontinued without the threat of recrudescence. A six-country study was conducted assessing the performance of seven diagnostic tests, including tests for microfilariae (blood smear, PCR), parasite antigen (ICT, Og4C3) and antifilarial antibody (Bm14, PanLF, Urine SXP). One community survey and one school survey were performed in each country. A total of 8,513 people from the six countries participated in the study, …

Small-Scale Variation In Fuel Loads Differentially Affects Two Co-Dominant Bunchgrasses In A Species-Rich Pine Savanna, Paul R. Gagnon, Kyle E. Harms, William J. Platt, Heather A. Passmore, Jonathan A. Myers Jan 2012

Small-Scale Variation In Fuel Loads Differentially Affects Two Co-Dominant Bunchgrasses In A Species-Rich Pine Savanna, Paul R. Gagnon, Kyle E. Harms, William J. Platt, Heather A. Passmore, Jonathan A. Myers

Faculty Publications

Ecological disturbances frequently control the occurrence and patterning of dominant plants in high-diversity communities like C 4 grasslands and savannas. In such ecosystems disturbance-related processes can have important implications for species, and for whole communities when those species are dominant, yet mechanistic understanding of such processes remains fragmentary. Multiple bunchgrass species commonly co-dominate disturbance-dependent and species-rich pine savannas, where small-scale fuel heterogeneity may influence bunchgrass survival and growth following fires. We quantified how fire in locally varying fuel loads influenced dynamics of dominant C 4 bunchgrasses in a species-rich pine savanna in southeastern Louisiana, USA. We focused on two congeneric, …

Cardiac-Specific Expression Of The Tetracycline Transactivator Confers Increased Heart Function And Survival Following Ischemia Reperfusion Injury, Laila Elsherif, Xuerong Wang, Milana Grachoff, Beata Wolska, David Geenen, John O'Bryan Jan 2012

Cardiac-Specific Expression Of The Tetracycline Transactivator Confers Increased Heart Function And Survival Following Ischemia Reperfusion Injury, Laila Elsherif, Xuerong Wang, Milana Grachoff, Beata Wolska, David Geenen, John O'Bryan

Peer Reviewed Articles

Mice expressing the tetracycline transactivator (tTA) transcription factor driven by the rat a-myosin heavy chain promoter (a-MHC-tTA) are widely used to dissect the molecular mechanisms involved in cardiac development and disease. However, these a-MHC-tTA mice exhibit a gain-of-function phenotype consisting of robust protection against ischemia/reperfusion injury in both in vitro and in vivo models in the absence of associated cardiac hypertrophy or remodeling. Cardiac function, as assessed by echocardiography, did not differ between a-MHC-tTA and control animals, and there were no noticeable differences observed between the two groups in HW/TL ratio or LV end-diastolic and end-systolic dimensions. Protection against ischemia/reperfusion …

Assessing Ecological Risk Of Proposed Mines: Can Valid Assessments Be Done Pre-Design?, Bob Loeffler Jan 2012

Assessing Ecological Risk Of Proposed Mines: Can Valid Assessments Be Done Pre-Design?, Bob Loeffler

ISER Reports

Large resource development projects take years to plan. During that planning time, the public frequently debates the potential benefits and risks of a project, but with incomplete information. In these debates, some people might assert that a project would have great benefits, while others might assert that it would certainly harm the environment. At the same time, the developer will be assessing different designs, before finally submitting one to the government permitting agencies for evaluation and public scrutiny. For large mines in Alaska, the government permitting process takes years, and often includes an ecological risk assessment. This assessment is a …

Functional Intimate Association Analysis: A Doctrinal Shift To Save The Roberts Framework, Joshua P. Roling Jan 2012

Functional Intimate Association Analysis: A Doctrinal Shift To Save The Roberts Framework, Joshua P. Roling

Duke Law Journal

In Roberts v. U.S. Jaycees, the Supreme Court recognized intimate association as one of the two distinct senses of the freedom of association. In doing so, the Court identified two essential functions that justify constitutional protection for the relationships that provide them: intimate relationships cultivate and transmit shared ideals and beliefs, and they provide opportunities for emotional enrichment and self-identification by facilitating the creation of close bonds among members. Then, recognizing that familial relationships often exemplify these functions, the Court identified four aspects of family relationships that would help distinguish intimate from nonintimate associations: size, purpose, selectivity, and seclusion from …

Seasonal Affective Disorder And Seasonality: A Review, Leo Sher M.D.

Jefferson Journal of Psychiatry

Seasonal affective disorder is a condition where depressions in fall and winter alternate with nondepressed periods in the spring and summer. The Degree to which seasonal changes affect mood, energy, sleep, appetite, food preference, or the wish to socialize with other people has been called "seasonality." The author reviews historical aspects, clinical features, epidemiology, genetics, pathophysiology, and treatment of seasonal affective disorder and seasonality. Better understanding of the contemporary concept of seasonal affective disorder, seasonality, and light treatment will improve patient care and promote scientific advances in behavioral sciences.

The Influence Of Caspase-3 On The Calpain Enzyme System During Meat Aging, D.A. Mohrhouse, S.A. Kern, K.R. Underwood, A.D. Weaver

South Dakota Beef Report, 2012

Tenderness is a key component of palatability, which influences consumers’ perception of meat quality. There are a variety of factors that contribute to variations in tenderness, including postmortem proteolysis. A more complete understanding of this biological mechanism regulating tenderness is needed to ensure consistently tender beef. Numerous reports indicate μ-calpain is primarily responsible for the degradation of proteins postmortem. Additionally, it has been shown that caspase-3 can cleave calpastatin, the inhibitor of μ-calpain. Therefore, the objective of this study was to determine if in vitro degradation of calpastatin by caspase-3 can enhance the postmortem breakdown of myofibrillar proteins by μ-calpain. …

Optimally Enhanced Optical Emission In Laser-Induced Breakdown Spectroscopy By Combining Spatial Confinement And Dual-Pulse Irradiation, L. B. Guo, B. Y. Zhang, X. N. He, C. M. Li, Y. S. Zhou, T. Wu, J. B. Park, X. Y. Zeng, Yongfeng Lu

Department of Electrical and Computer Engineering: Faculty Publications

In laser-induced breakdown spectroscopy (LIBS), a pair of aluminum-plate walls were used to spatially confine the plasmas produced in air by a first laser pulse (KrF excimer laser) from chromium (Cr) targets with a second laser pulse (Nd:YAG laser at 532 nm, 360 mJ/pulse) introduced parallel to the sample surface to re-excite the plasmas. Optical emission enhancement was achieved by combing the spatial confinement and dual-pulse LIBS (DP-LIBS), and then optimized by adjusting the distance between the two walls and the interpulse delay time between both laser pulses. A significant enhancement factor of 168.6 for the emission intensity of the …

Sheathless Size-Based Acoustic Particle Separation, Rasim Guldiken, Myeong Chan Jo, Nathan D. Gallant, Utkan Demirci, Jiang Zhe

Mechanical Engineering Faculty Research

Particle separation is of great interest in many biological and biomedical applications. Flow-based methods have been used to sort particles and cells. However, the main challenge with flow based particle separation systems is the need for a sheath flow for successful operation. Existence of the sheath liquid dilutes the analyte, necessitates precise flow control between sample and sheath flow, requires a complicated design to create sheath flow and separation efficiency depends on the sheath liquid composition. In this paper, we present a microfluidic platform for sheathless particle separation using standing surface acoustic waves. In this platform, particles are first lined …
