Characterization Of Aquaporin 4 Protein Expression And Localization In Tissues Of The Dogfish (Squalus Acanthias), Christopher P. Cutler, Sheena Harmon, Jonathan Walsh, Kia Burch

Biology: Faculty Publications

The role of aquaporin water channels such as aquaporin 4 (Aqp4) in elasmobranchs such as the dogfish Squalus acanthias is completely unknown. This investigation set out to determine the expression and cellular and sub-cellular localization of Aqp4 protein in dogfish tissues. Two polyclonal antibodies were generated (AQP4/1 and AQP4/2) and these showed somewhat different characteristics in Western blotting and immunohistochemistry. Western blots using the AQP4/1 antibody showed two bands (35.5 and 49.5 kDa) in most tissues in a similar fashion to mammals. Liver had an additional band of 57 kDa and rectal gland two further faint bands of 37.5 and …

Desorption electrospray ionization (DESI) coupled to high-resolution Orbitrap mass spectrometry (MS) was used to study the reactivity of a (β-amino alcohol)(arene)RuCl transfer hydrogenation catalytic precursor in methanol (CH3OH). By placing [(p-cymene)RuCl2]2 on a surface and spraying a solution of β-amino alcohol in methanol, two unique transient intermediates having lifetimes in the submillisecond to millisecond range were detected. These intermediates were identified as Ru (II) and Ru (IV) complexes incorporating methyl formate (HCOOCH3). The Ru (IV) intermediate is not observed when the DESI spray solution is sparged with Ar gas, indicating that …

Justin's Story is part of Pediatric End-of-Life Case Studies, one of seven teaching modules drawn from the Emmy Award-winning documentary, A Lion in the House. A Lion in the House portrays an unprecedented, intimate look at the realities of childhood cancer through the journeys of five families and their professional caregivers over the course of 6 years. Justin's Story provides a forum for open-ended discussion and consideration of compassionate approaches for navigating pediatric end-of-life. It offers content that is unpredictable and not always best practice. Learners reflect on the complex impact of childhood cancer on Justin, his family, and …

Dynamics Of Substrate Interactions In Trna (M1g37) Methyltransferase: Implications For Drug Discovery, Maria Kiouppis Palesis Feb 2012

Dynamics Of Substrate Interactions In Trna (M1g37) Methyltransferase: Implications For Drug Discovery, Maria Kiouppis Palesis

Theses and Dissertations

The bacterial enzyme t-RNA (m1G37) methyltransferase (TrmD) is an ideal anti-microbial drug target since it is found in all eubacteria, serves an essential role during protein synthesis, and shares very little sequence or structural homology with its eukaryotic counterpart, Trm5. TrmD, a homodimeric protein, methylates the G37 nucleotide of tRNA using S-adenosyl-L-methionine (SAM) as the methyl donor and thus enables proper codon-anticodon alignment during translation. The two deeply buried binding sites for SAM seen in X-ray crystallography suggest that significant conformational changes must occur for substrate binding and catalytic turnover. Results from molecular dynamics simulations implicate a flexible loop region …

Humanized Mice As A Model To Study Human Viral Pathogenesis And Novel Antiviral Drugs, Freddy Mauricio Sanchez Tumbaco Feb 2012

Humanized Mice As A Model To Study Human Viral Pathogenesis And Novel Antiviral Drugs, Freddy Mauricio Sanchez Tumbaco

Theses and Dissertations

Animal models have greatly contributed to the understanding of different aspects of human biology, as well as a variety of human-related pathogens and diseases. In order to study them, humanized mice susceptible to pathogens that replicate in human immune cells have been developed (e.g., humanized Rag2-/-γc-/- mice). These animals are engrafted with human hematopoietic stem cells (HSCs), resulting in the de novo development and maturation of the major functional components of the human adaptive immune system and the production of a variety of human cell types. Primary and secondary lymphoid organs in the mouse are populated with human cells, and …

The Ecology Of Pulse Events: Insights From An Extreme Climatic Event In A Polar Desert Ecosystem, U. N. Nielsen, D. H. Wall, B. J. Adams, R. A. Virginia

Dartmouth Scholarship

Climate change is occurring globally, with wide ranging impacts on organisms and ecosystems alike. While most studies focus on increases in mean temperatures and changes in precipitation, there is growing evidence that an increase in extreme events may be particularly important to altering ecosystem structure and function. During extreme events organisms encounter environmental conditions well beyond the range normally experienced. Such conditions may cause rapid changes in community composition and ecosystem states. We present the impact of an extreme pulse event (a flood) on soil communities in an Antarctic polar desert. Taylor Valley, McMurdo Dry Valleys, is dominated by large …

Sharpin Is Essential For Cytokine Production, Nf-Κb Signaling, And Induction Of Th1 Differentiation By Dendritic Cells, Zhe Wang, Anna Sokolovska, Rosemarie Seymour, John P. Sundberg, Harm Hogenesch Feb 2012

Sharpin Is Essential For Cytokine Production, Nf-Κb Signaling, And Induction Of Th1 Differentiation By Dendritic Cells, Zhe Wang, Anna Sokolovska, Rosemarie Seymour, John P. Sundberg, Harm Hogenesch

Department of Comparative Pathobiology Faculty Publications

Spontaneous mutations of the Sharpin (SHANK-associated RH domain-interacting protein, other aliases: Rbckl1, Sipl1) gene in mice result in systemic inflammation that is characterized by chronic proliferative dermatitis and dysregulated secretion of T helper1 (Th1) and Th2 cytokines. The cellular and molecular mechanisms underlying this inflammatory phenotype remain elusive. Dendritic cells may contribute to the initiation and progression of the phenotype of SHARPIN-deficient mice because of their pivotal role in innate and adaptive immunity. Here we show by flow cytometry that SHARPIN- deficiency did not alter the distribution of different DC subtypes in the spleen. In response …

Sparse Integrative Clustering Of Multiple Omics Data Sets, Ronglai Shen, Sijian Wang, Qianxing Mo Feb 2012

Sparse Integrative Clustering Of Multiple Omics Data Sets, Ronglai Shen, Sijian Wang, Qianxing Mo

Memorial Sloan-Kettering Cancer Center, Dept. of Epidemiology & Biostatistics Working Paper Series

High resolution microarrays and second-generation sequencing platforms are powerful tools to investigate genome-wide alterations in DNA copy number, methylation, and gene expression associated with a disease. An integrated genomic profiling approach measuring multiple omics data types simultaneously in the same set of biological samples would render an integrated data resolution that would not be available with any single data type. In a previous publication (Shen et al., 2009), we proposed a latent variable regression with a lasso constraint (Tibshirani, 1996) for joint modeling of multiple omics data types to identify common latent variables that can be used to cluster patient …
