The Effect Of E-Cadherin Loss On Melanoma Formation And Metastasis, Lara Rosenbaum
The Effect Of E-Cadherin Loss On Melanoma Formation And Metastasis, Lara Rosenbaum
Yale Medicine Thesis Digital Library
Melanoma is the most lethal form of skin cancer and accounts for the majority of skin cancer related mortality. Early metastases are characteristic of melanoma and are an ominous sign, as current therapeutic interventions have little effect on overall survival. The lack of accurate prognostic indicators and effective therapies emphasize the need for a better understanding of the genetic and phenotypic changes in melanoma formation and progression. One such change is the loss of E-cadherin, which normally plays a role in cell-cell adhesion and is thought to be a central feature of epithelial-mesenchymal transition (EMT). Maintenance of E-cadherin in melanoma …

The Structural Requirements Of Histone Deacetylase (Hdac) Inhibitors: Suberoylanilide Hydroxamic Acid (Saha) Analogues Modified At C3, C6, And C7 Positions Enhance Selectivity, Sun Ea Choi
Wayne State University Dissertations
Histone deacetylase (HDAC) proteins are targets for drug design towards the treatment of cancers since overexpression of HDAC is linked to cancer. Several HDAC inhibitors, including the FDA approved drug suberoylanilide hydroxamic acid (SAHA, Vorinostat), have cleared clinical trials and emerged as anti-cancer drugs. However, SAHA inhibits all of the 11 metal ion-dependent HDAC proteins. Therefore, we synthesized several libraries of small molecule HDAC inhibitors based on SAHA to help understand the structural requirements of inhibitory potency and isoform selectivity.
In previous work, SAHA analogues functionalized at the C2 position (C2-SAHA analogues) near the metal binding hydroxamic acid displayed decreased …

Research Publications (2011 to 2015)
Leea indica is a medicinal plant used traditionally to cure cancer. In this study, the cytotoxic compounds of L. indica were isolated using bioassay-guided approach. Two cycloartane triterpenoid glycosides, mollic acid arabinoside (MAA) and mollic acid xyloside (MAX), were firstly isolated from L. indica. They inhibited the growth of Ca Ski cervical cancer cells with IC(50) of 19.21 μM (MAA) and 33.33 μM (MAX). MRC5 normal cell line was used to calculate selectivity index. MAA and MAX were about 8 and 4 times more cytotoxic to Ca Ski cells compared to MRC5. The cytotoxicity of MAA was characterized by both …
Chemical Reporters For Investigating Lipidated Proteins At The Host-Pathogen Interface, Guillaume Charron
Chemical Reporters For Investigating Lipidated Proteins At The Host-Pathogen Interface, Guillaume Charron
Student Theses and Dissertations
Lipidation of proteins regulates many cellular processes such as signaling transduction and membrane sorting by modulating protein localization and proteinprotein interactions. As such, defects in protein lipidation can render host cells more susceptible to microbial infection and are also associated with a variety of human diseases ranging from cancer to neurological disorders. Moreover, viral and bacterial pathogens can exploit and modulate the host lipidation machinery to enhance infection. Robust biochemical methods for characterizing lipidated proteins are therefore important for understanding fundamental physiology and disease mechanisms. In this thesis, I report the development of alkyne-lipid chemical reporters that afford more sensitive …

Role Of Hoga And Sskb Protein Kinases In Mycotoxin Production In The Filamentous Fungus Aspergillus Nidulans, Lauren Jurkowski
Honors Capstones
Aspergillus nidulans is a filamentous fungus that is often used as a model organism in genetics and molecular biology studies. The species has a well- characterized life-cycle and its genome has been sequenced. Aspergillus species are commonly found in the environment. Some species can cause health problems in humans. Some Aspergillus species produce aflatoxin, a toxic compound that causes acute necrosis, cirrhosis, and carcinoma of the liver in some animal species and can potentially cause similar symptoms in humans (Center for Integrated Fungal Research, 2005). A. nidulans produces the carcinogenic mycotoxin sterigmatocystin (ST), a precursor in the aflatoxin biosynthetic pathway …

A series of 3-[(4-substitutedpiperazin-1-yl)methyl]-1H-indole derivatives were synthesized, and their structures were confirmed by spectral analysis. All the compounds were tested for their cytotoxic activity in vitro against 3 human tumor cell lines: human liver (HUH7), breast (MCF7), and colon (HCT116). Among the designed derivatives, most of the compounds showed significant cytotoxicity against liver and colon cancer cell lines with lower IC_{50} concentrations than the standard drug 5-fluorouracil. Compound 3s, with 3,4-dichlorophenyl substituent on the piperazine ring, was the most active in suppressing the growth of all screened cancer cells.

Background: Despite decades of new discoveries in biomedical research, the overwhelming complexity of cells has been a significant barrier to a fundamental understanding of how cells work as a whole. As such, the holistic study of biochemical pathways requires computer modeling. Due to the complexity of cells, it is not feasible for one person or group to model the cell in its entirety.
Results: The Cell Collective is a platform that allows the world-wide scientific community to create these models collectively. Its interface enables users to build and use models without specifying any mathematical equations or computer code - addressing …

Gaining Insight Into The Physics Of Dynamic Atomic Force Microscopy In Complex Environments Using The Veda Simulator, Daniel Kiracofe, John Melcher, Arvind Raman
Birck and NCN Publications
Dynamic atomic force microscopy (dAFM) continues to grow in popularity among scientists in many different fields, and research on new methods and operating modes continues to expand the resolution, capabilities, and types of samples that can be studied. But many promising increases in capability are accompanied by increases in complexity. Indeed, interpreting modern dAFM data can be challenging, especially on complicated material systems, or in liquid environments where the behavior is often contrary to what is known in air or vacuum environments. Mathematical simulations have proven to be an effective tool in providing physical insight into these non-intuitive systems. In …
Examining Chronic Non-Cancer Pain Among A Sample Of Individuals In Opioid Treatment Programs, Erin Stevenson
Examining Chronic Non-Cancer Pain Among A Sample Of Individuals In Opioid Treatment Programs, Erin Stevenson
Theses and Dissertations--Social Work
National rates of chronic non-cancer pain (CNCP) are rising alongside increasing reports of prescription opioid abuse and mortality. Associations between the rise in CNCP and in opioid abuse seem logical, yet research on CNCP among individuals with opioid dependence is currently limited due to the complicated nature of comorbid conditions in research and treatment. This study aims to expand the CNCP knowledge base by responding to the question: Do individuals with CNCP participating in an opiate treatment program have better or worse treatment outcomes than individuals without CNCP?
This study used a secondary dataset including 483 adults from Kentucky’s Opiate …

Turkish Journal of Botany
Six populations of Isoetes panchganiensis G.K.Srivast., D.D.Pant & P.K.Shukla growing in different environmental conditions of the Western Ghats have been studied to determine the range of variation within the species. The present investigation does not show any variation in habitat (aquatic), rhizomorph (trilobed), velum (fenestra), and presence or absence of peripheral strands; however, there is a subtle variation in the length of plants, number of leaves, size of megasporangia, and size and ornamentation of the megaspores. The basic organisation of the megaspores of all the populations is almost same, but the finer details of the ornamentation of the megaspores exhibit …
Design, Synthesis, And Biological Evaluation Of Novel Polyamine Transport System Probes And Their Application To Human Cancers, Aaron Muth
Electronic Theses and Dissertations
The mammalian polyamine transport system (PTS) has been of interest due to its roles in cancer and maintaining cellular homeostasis. Polyamines are essential growth factors which are tightly controlled via a balance of biosynthesis, metabolism, import, and export. This work focused on the development and biological testing of polyamine transport probes to help understand the molecular requirements of the PTS. This was mediated through the use of a CHO (PTS active) and CHO-MG* (PTS deficient) screen, where compounds demonstrating high toxicity in CHO and low toxicity in CHO-MG* were considered PTS selective. The first chapter focused on the development of …

Mechanism Of Inhibition Of The Glua2 Receptors By N-3 Derivatives Of 2,3-Benzodiazepines With C-4 Methyl Group, Congzhou Wang
Legacy Theses & Dissertations (2009 - 2024)
α-Amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors are one of the three subtypes of ionotropic glutamate receptors. AMPA receptors mediate fast synaptic neurotransmission in the central nervous system (CNS). Over-activation of calcium permeable AMPA receptors causes intracellular calcium overload, which leads to neurodegeneration and cell death. As such, AMPA receptors have been implicated in a number of neurological disorders and diseases, such as epilepsy, amyotrophic lateral sclerosis (ALS), and Parkinson's disease. 2,3-Benzodiazepine derivatives (or GYKI compounds) are a group of structurally similar compounds synthesized as inhibitors of AMPA receptors, and they have been used as potential drug candidates for the treatment of various …

Isolation, Structure Elucidation And Biological Evaluation Of Natural Products From Plants And Fungi, Xiaoning Wang
Electronic Theses and Dissertations
Natural products have provided considerable value for drug discovery over the past half-century. Nearly four fifth of drugs were natural products or inspired by natural compounds. Bioactive secondary metabolites isolated from natural source, either fungal or terrestrial, are relatively safer for human health and the environment and can serve as drug or as drug leads for the development of synthetic or semi-synthetic analogs. Natural products have also been successfully used in agriculture as plant growth regulators, signal transductions, fungicides, herbicides and tranquilizers. In our continued efforts to identify bioactive compounds, seventy-seven plant and fungal extracts were evaluated using a direct-bioautography …
