Drosophila Activated Cdc42 Kinase Has An Anti-Apoptotic Function., Jessica A. Schoenherr, J Michelle Drennan, Juan S. Martinez, Madhusudana Rao Chikka, Mark C. Hall, Henry C. Chang, James C. Clemens

Department of Biochemistry Faculty Publications

Activated Cdc42 kinases (Acks) are evolutionarily conserved non-receptor tyrosine kinases. Activating somatic mutations and increased ACK1 protein levels have been found in many types of human cancers and correlate with a poor prognosis. ACK1 is activated by epidermal growth factor (EGF) receptor signaling and functions to regulate EGF receptor turnover. ACK1 has additionally been found to propagate downstream signals through the phosphorylation of cancer relevant substrates. Using Drosophila as a model organism, we have determined thatDrosophila Ack possesses potent anti-apoptotic activity that is dependent on Ack kinase activity and is further activated by EGF receptor/Ras signaling. Ack anti-apoptotic signaling …

Community Involvement As A Tool For International Policy: Realizing The Great Lakes Water Quality Agreement, Michael D. Eriksen Jr. May 2012

Community Involvement As A Tool For International Policy: Realizing The Great Lakes Water Quality Agreement, Michael D. Eriksen Jr.

Honors Theses

Whether present in abundance or known for its scarcity, water defines the regional and geographical identities of people. It defines political and ecological boundaries globally. Water paucity and quality in burgeoning populations has been a catalyst for creative resource management policies. Conservation of water resources, in practice, has still not improved however. As a result, the health and stability of the Great Lakes as a natural resource is threatened. The Great Lakes region includes two countries, eight states, two provinces, and over 200 tribal and native influences. International agreements, such as the Boundary Waters Treaty of 1909 (BWT) and the …

An Experimental Study On The Interaction Of Coaxial And Co-Rotating Vortex Rings, Jagannadha Reddy Satti May 2012

An Experimental Study On The Interaction Of Coaxial And Co-Rotating Vortex Rings, Jagannadha Reddy Satti

Engineering

The study investigated the role of formation time, Reynolds Number, and non-dimensional frequency number, the three most significant parameters in the dynamics of vortex rings, in the interaction between co-axial and co-rotating vortex rings and in the ring behaviors of merging and leapfrogging. To generate and investigate vortex rings with the required characteristics, two laminar vortex rings were generated consecutively from a piston-cylinder apparatus such that the rings propagated in the same direction and that the spatial separation between them decreased until they began merging. Using digital particle image velocimetry to measure the flow fields as well as the trajectory …

Reflections Of Self, Melissa Athey May 2012

Reflections Of Self, Melissa Athey

Theses and Dissertations

I vacillate between all extremes, beauty vs. ugly, internal vs. external, micro vs. macro. It is these disparate notions that inspire what I make. We cannot ever see ourselves objectively, but does that mean we shouldn’t try? This thesis is my attempt to dissect what I created in my 2 years at Virginia Commonwealth University, my exploration of the illness within and the psychological nature of how we go about hiding our insecurities.

Shoreline Distribution And Landscape Genetics Of Bears In A Recently Deglaciated Fjord: Glacier Bay, Alaska, Tania M. Lewis May 2012

Shoreline Distribution And Landscape Genetics Of Bears In A Recently Deglaciated Fjord: Glacier Bay, Alaska, Tania M. Lewis

Biological Sciences

To further knowledge of mammalian colonization patterns following deglaciation, I used occupancy modeling to estimate black and brown bear shoreline distribution of Glacier Bay and how these distributions relates to the number years of land exposure and post glacial plant and stream succession. I also conducted microsatellite genetic analysis of brown bear hair and tissue to determine contemporary population structure throughout the park and how it relates to landscape features and surrounding populations. Closed forest cover within 1 km of the study site was a strong positive predictor of black bear occurrence. Brown bears were detected at 100% of sites …

Central Co2 Chemosensitivity In Tadpoles: Impairment And The Role Of Serotonin, Spencer D. Audie May 2012

Central Co2 Chemosensitivity In Tadpoles: Impairment And The Role Of Serotonin, Spencer D. Audie

Biological Sciences

Nicotine and ethanol are known neuroteratogens and prenatal exposure correlates with Sudden Infant Death Syndrome (SIDS), which is thought to result from failure to maintain pH homeostasis through respiratory adjustments. This failed homeostatic control is believed to be serotonergic in origin. In previous studies nicotine or ethanol exposure ablated the robust hypercapnic response of early-stage tadpoles. These findings lead us to question if the ablation occurred through a serotonindependent mechanism. This study investigated the role of serotonin (5- HT) in the nicotine- or ethanol-induced abolishment of the hypercapnic response. We found that toxin-exposed animals were insensitive to hypercapnia and also …

Biochemical And Microbiological Assessments Of Dried Alaska Pink Salmon, Red Salmon And Pacific Cod Heads, Huseyin Biceroglu May 2012

Biochemical And Microbiological Assessments Of Dried Alaska Pink Salmon, Red Salmon And Pacific Cod Heads, Huseyin Biceroglu

Fisheries

Fish heads are generally considered as unsuitable byproducts for human consumption in the United States. The initial objective was to compare the moisture content and water activity levels on dried pink salmon (Oncorhynchus gorbuscha) and dried red salmon (O. nerka) using different temperature and time integration. The secondary objective was to compare shelf life characteristics, rancidity and mold growth, between dried pink dried salmon and dried Pacific cod (Gadus macrocephalus) heads stored for up to 180 days at the ambient temperature (21°C) for East African seafood markets. The third objective was to assess the antioxidant effects for frozen and dried …

Delayed Effects Of Oil Exposure On Fish, Corinne E. Hicken May 2012

Delayed Effects Of Oil Exposure On Fish, Corinne E. Hicken

Fisheries

Polycyclic aromatic hydrocarbons (PAHs) are continuously added to aqueous environments through point source and non-point source pollution and can cause deleterious effects on exposed fish populations. Historically, studies have shown that acute PAH exposure causes only short-term effects in adult fish which were resolved when the exposure ended. Chronic exposure to PAHs, however -- even at the less susceptible juvenile and adult stages -- can cause a host of effects including lesions, lower body length and weight, and reduced swimming ability. More recently studies of embryonic fish have demonstrated that much lower PAH concentrations can cause lethal and sub-lethal effects …

Optical Properties& Energy Transfer Dynamics Of Atmospheric Species, Daniel K. Havey

Science Enabled by Photon Sources: Local Research Interests in Virginia

Our research group is interested in how light interacts with small molecules and particulate matter that are important to atmospheric chemistry and climate change. An active project currently being performed in the James Madison University Undergraduate Laser Laboratory involves a detailed mapping of energy transfer rates from excited or metastable states of atomic or molecular species. This talk describes a specific example study with potential relevance to the JLAMP VUV/Soft X-ray User Facility that would investigate relaxation dynamics of metastable Krypton atoms using two-photon photoacoustic spectroscopy at 819 nm and 124 nm. A study like this would provide useful reference …

The Federal Democratic Republic Of Ethiopia: Liberal Democratic Or Authoritarian Regime?, Solomon Terfa May 2012

The Federal Democratic Republic Of Ethiopia: Liberal Democratic Or Authoritarian Regime?, Solomon Terfa

African Social Science Review

The leaders of the Federal Democratic Republic of Ethiopia adopted their constitution on December 8, 1994. This analysis argues that the ethno-linguistic federal political system adopted by the founding members is problematic because the framers superimposed the constitution on the citizens dogmatically without thoroughly examining the country’s objective reality. Hence, the author contends that the ethnic federalist paradigm adopted in Ethiopia is diametrically opposed to the wishes and aspirations of the people as validated by a survey he conducted between 1992 and 1993. He draws from this, perhaps the first and only study on this pertinent topic, to argue his …

Integrin Activation By A Cold Atmospheric Plasma Jet, Olga Volotskova, Mary Ann Stepp, Michael Keidar May 2012

Integrin Activation By A Cold Atmospheric Plasma Jet, Olga Volotskova, Mary Ann Stepp, Michael Keidar

Anatomy and Regenerative Biology Faculty Publications

Current breakthrough research on cold atmospheric plasma (CAP) demonstrates that CAP has great potential in various areas, including medicine and biology, thus providing a new tool for living tissue treatment. In this paper, we explore potential mechanisms by which CAP alters cell migration and influences cell adhesion. We focus on the study of CAP interaction with fibroblasts and corneal epithelial cells. The data show that fibroblasts and corneal epithelial cells have different thresholds (treatment times) required to achieve maximum inhibition of cell migration. Both cell types reduced their migration rates by ~30–40% after CAP compared to control cells. Also, the …

Farber Institute for Neuroscience Faculty Papers

DARPP-32 (dopamine and adenosine 3', 5'-cyclic monophosphate cAMP-regulated phosphoprotein, 32 kDa) is a striatal-enriched protein that mediates signaling by dopamine and other first messengers in the medium spiny neurons. The transcriptional mechanisms that regulate striatal DARPP-32 expression remain enigmatic and are a subject of much interest in the efforts to induce a striatal phenotype in stem cells. We report the identification and characterization of a conserved region, also known as H10, in intron IV of the gene that codes for DARPP-32 (Ppp1r1b). This DNA sequence forms multiunit complexes with nuclear proteins from adult and embryonic striata of mice and rats. …

Faculty Publications

Background: The pathway-focused association approach offers a hypothesis driven alternative to the agnostic genome-wide association study. Here we apply the pathway-focused approach to an association study of hypertension, systolic blood pressure (SBP), and diastolic blood pressure (DBP) in 1614 Nigerians with genome-wide data. Methods and Results: Testing of 28 pathways with biological relevance to hypertension, selected a priori, containing a total of 101 unique genes and 4,349 unique single-nucleotide polymorphisms (SNPs) showed an association for the adrenergic alpha 1 (ADRA1) receptor pathway with hypertension (pp>10-5) SNPs offers a novel method for detecting the "missing heritability" of hypertension. These findings …

Publications and Research

The use of Global Positioning Systems (GPS) and Geographical Information Systems (GIS) in disease surveys and reporting is becoming increasingly routine, enabling a better understanding of spatial epidemiology and the improvement of surveillance and control strategies. In turn, the greater availability of spatially referenced epidemiological data is driving the rapid expansion of disease mapping and spatial modeling methods, which are becoming increasingly detailed and sophisticated, with rigorous handling of uncertainties. This expansion has, however, not been matched by advancements in the development of spatial datasets of human population distribution that accompany disease maps or spatial models. Where risks are heterogeneous …

Testing Crayfish Evolutionary Hypotheses With Phylogenetic Methods, Jesse W. Breinholt May 2012

Testing Crayfish Evolutionary Hypotheses With Phylogenetic Methods, Jesse W. Breinholt

Theses and Dissertations

This dissertation focuses on increasing the understanding of the evolution processes that have contributed to the diversification of freshwater crayfish. Chapter one estimates the divergence time of the three crayfish families and tests the hypothesis that diversification is tied to the break-up of Pangaea, Gondwanna, and Laurasia. I find that the families of crayfish diverged prior to or in association with the break-up of the three super continents. Chapter two addresses the evolutionary history of the genus Cambarus, using molecular data to test hypotheses of relationships based on chela and carapace morphology. The results provide evidence that the morphology used …
