Arsenic and cadmium (Cd2+) are environmental carcinogens that have been implicated in the development of bladder cancer. However, there are no good in vitro model systems to study the development of bladder cancer caused by these carcinogens. Previous studies from this laboratory have shown that arsenite (As3+) and Cd2+ exposure can cause direct malignant transformation of an immortalized human urothelial cell line, the UROtsa. In the present study the repeatability of the transformation process was examined by isolating and characterizing 6 additional Cd2+- transformed lines and 5 additional As3+- transformed cell lines. Post-Translational Regulation Of The Dopamine Transporter, Amy E. Moritz

The dopamine transporter (DAT) is an integral membrane phosphoprotein found on dopaminergic neurons that mediates the presynaptic reuptake of dopamine and is a site of action for psychostimulants. Numerous DAT functions including efflux, uptake activity, and surface expression are regulated by protein kinase C (PKC) through unknown mechanisms. Basal and PKC stimulated phosphorylation of DAT occurs on multiple serines in the distal N-terminus, but the exact sites modified and the kinases involved are unknown. To address these issues, we examined in vitro phosphorylation of a recombinant DAT N-terminal tail peptide (NDAT). We found that NDAT is phosphorylated at multiple sites …

We used X-ray fluorescence (XRF) to compare the heavy metal chemistry of sediments in caves in rural and urban areas to the chemistry of sediments from a control cave in a relatively undisturbed watershed in the Springfield (MO) Plateau. Sediment from Smallin Cave near Ozark, MO, the control cave, has the smallest peak sizes for Zn and Mn and a moderately-sized Pb peak. Sediment from the rural cave exhibited larger peaks of Zn and Mn and a smaller Pb peak. Sediment from the urban cave had the largest Zn, Mn and Pb peaks. Using Macroinvertebrate Community Composition To Distinguish Between Natural And Anthropogenic Sedimentation, Amanda E. Schutt

Excess fine sediment from human activity is a major pollutant to streams across the U.S.; however, distinguishing human-induced sedimentation from natural fine sediment is complex. The U.S. Environmental Protection Agency recently implemented a protocol for the quantitative field assessment of human-induced sedimentation using measurements of stream geomorphology. Macroinvertebrate community composition, streambed sediment stability, and sediment composition were studied at 49 sites in the James River watershed in central Virginia. Sediment composition was found to be a stronger driver of community composition than sediment stability. Although I was not able to show that macroinvertebrate metrics were related to sediment stability independently …

Many mammals travel on both tree branches and flat ground in their natural habitat. Forelimbs were found to have the dominant role in braking on both substrates, while hindlimbs had the dominant role in propulsion on both substrates. However, the forelimb aided in propulsion more so than the hindlimbs contributed to braking on both substrates.

Cancer is the leading cause of death worldwide. The current treatment options available for lung cancer are limited and have drawbacks such as poor bioavailability, numerous side effects, poor efficacy and drug resistance. 3D model serve as a novel approach for drug screening purposes and the evaluation of compounds in the platform can help identify potent compounds for further in vivo xenograft studies.

Functional Morphology Of Rat Hands And Feet: Correlation With The Ability To Grip Tree Branches During Locomotion, Jessica Elaine Fonce

Anatomy and function are usually closely related. Since locomotion on tree branches is common among mammals, we expect to find that the anatomy is well adapted for gripping narrow cylindrical branch-like substrates. Based on the anatomy alone, rats are built for locomotion across cylindrical branches as well as terrestrial substrates.

BACKGROUND: Recombination activation gene 1 deficient (rag1(-/-)) mutant zebrafish have a reduced lymphocyte-like cell population that lacks functional B and T lymphocytes of the acquired immune system, but includes Natural Killer (NK)-like cells and Non-specific cytotoxic cells (NCC) of the innate immune system. The innate immune system is thought to lack the adaptive characteristics of an acquired immune system that provide enhanced protection to a second exposure of the same pathogen. It has been shown that NK cells have the ability to mediate adaptive immunity to chemical haptens and cytomegalovirus in murine models. In this study we evaluated the ability …

Oncostatin M (OSM) is an interleukin-6 (IL-6) family cytokine that has been implicated in a number of biological processes including inflammation, hematopoiesis, immune responses, development, and bone homeostasis. Recent evidence suggests that OSM may promote breast tumor invasion and metastasis. We investigated the role of OSM in the formation of bone metastases in vivo using the 4T1.2 mouse mammary tumor model in which OSM expression was knocked down using shRNA (4T1.2-OSM). 4T1.2-OSM cells were injected orthotopically into Balb/c mice, resulting in a greater than 97% decrease in spontaneous metastasis to bone compared to control cells. Surface recognition and penetration are critical steps in the infection cycle of many plant pathogenic fungi. In Magnaporthe oryzae, cAMP signaling is involved in surface recognition and pathogenesis. Deletion of the MAC1 adenylate cyclase gene affected appressorium formation and plant infection. In this study, we used the affinity purification approach to identify proteins that are associated with Mac1 in vivo. One of the Mac1-interacting proteins is the adenylate cyclase-associated protein named Cap1. CAP genes are well-conserved in phytopathogenic fungi but none of them have been functionally characterized. Deletion of CAP1 blocked the effects of a dominant RAS2 allele …

Self-Love And Self-Respect In The Meaningful Life, Erica Stonestreet

Most people have the sense that there's something wrong with living a meaningless life. Since most meaningless lives seem morally blameless, however, it's not obvious exactly what is wrong with it. Starting with a plausible conception of a meaningful life as a life engaged with values beyond oneself, I suggest that the problem is that someone living outside of this conception is not according herself a kind of recognition she deserves as a human being. Comparing self-respect and self-love as candidates for this recognition, I argue that lacking self-love is actually the more fitting explanation for what goes wrong in …
