A Stress Inducible Sumo Conjugating Enzyme Gene (Sasce9) From A Grass Halophyte Spartina Alterniflora Enhances Salinity And Drought Stress Tolerance In Arabidopsis, Ratna Karan, Prasanta K. Subudhi

Faculty Publications

Background: SUMO (Small Ubiquitin related Modifier) conjugation is a post translational regulatory process found in all eukaryotes, mediated by SUMO activating enzyme, SUMO conjugating enzyme, and SUMO ligase for the attachment of SUMO to its target protein. Although the mechanism for regulation of SUMO conjugation pathway genes under abiotic stress has been studied to certain extent, the role of SUMO conjugating enzyme in improving abiotic stress tolerance to plant is largely unexplored. Here, we have characterized a SUMO conjugating enzyme gene 'SaSce9' from a halophytic grass Spartina alterniflora and investigated its role in imparting abiotic stress tolerance. Results: SaSce9 gene encodes …

Tributyltin And Dibutyltin Alter Secretion Of Tumor Necrosis Factor Alpha From Human Natural Killer Cells And A Mixture Of T Cells And Natural Killer Cells, Kelsi Hurt, Tasia Hurd-Brown, Margaret Whalen

Chemistry Faculty Research

Butyltins (BTs) have been in widespread use. Tributyltin (TBT) has been used as a biocide in a variety of applications and is found in human blood samples. Dibutyltin (DBT) has been used as a stabilizer in polyvinyl chloride plastics and as a de-worming agent in poultry. DBT, like TBT, is found in human blood. Human natural killer (NK) cells are the earliest defense against tumors and viral infections and secrete the cytokine tumor necrosis factor-alpha (TNF-α). TNF-α is an important regulator of adaptive and innate immune responses. TNF-α promotes inflammation and an association between malignant transformation and inflammation has been …

Force Optimization And Flow Field Characterization From A Flapping Wing Mechanism, Nathaniel Stephen Naegle Oct 2012

Force Optimization And Flow Field Characterization From A Flapping Wing Mechanism, Nathaniel Stephen Naegle

Theses and Dissertations

Flapping flight shows promise for micro air vehicle design because flapping wings provide superior aerodynamic performance than that of fixed wings and rotors at low Reynolds numbers. In these flight regimes, unsteady effects become increasingly important. This thesis explores some of the unsteady effects that provide additional lift to flapping wings through an experiment-based optimization of the kinematics of a flapping wing mechanism in a water tunnel. The mechanism wings and flow environment were scaled to simulate the flight of the hawkmoth (Manduca sexta) at hovering or near-hovering speeds. The optimization was repeated using rigid and flexible wings …

Towards Defining Nutrient Conditions Encountered By The Rice Blast Fungus During Host Infection, Richard A. Wilson, Jessie Fernandez, Cristian Fernando Quispe, Julien Gradnigo, Anya Seng, Etsuko N. Moriyama, Janet D. Wright

Department of Plant Pathology: Faculty Publications

Fungal diseases cause enormous crop losses, but defining the nutrient conditions encountered by the pathogen remains elusive. Here, we generated a mutant strain of the devastating rice pathogen Magnaporthe oryzae impaired for de novo methionine biosynthesis. The resulting methionine-requiring strain grew strongly on synthetic minimal media supplemented with methionine, aspartate or complex mixtures of partially digested proteins, but could not establish disease in rice leaves. Live-cell-imaging showed the mutant could produce normal appressoria and enter host cells but failed to develop, indicating the availability or accessibility of aspartate and methionine is limited in the plant. This is the first report …

Spatio-Temporal Pattern Recognizers Using Spiking Neurons And Spike-Timing-Dependent Plasticity., James Humble, Susan Denham, Thomas Wennekers Oct 2012

Spatio-Temporal Pattern Recognizers Using Spiking Neurons And Spike-Timing-Dependent Plasticity., James Humble, Susan Denham, Thomas Wennekers

School of Engineering, Computing and Mathematics

It has previously been shown that by using spike-timing-dependent plasticity (STDP), neurons can adapt to the beginning of a repeating spatio-temporal firing pattern in their input. In the present work, we demonstrate that this mechanism can be extended to train recognizers for longer spatio-temporal input signals. Using a number of neurons that are mutually connected by plastic synapses and subject to a global winner-takes-all mechanism, chains of neurons can form where each neuron is selective to a different segment of a repeating input pattern, and the neurons are feed-forwardly connected in such a way that both the correct input segment …

Human Nad(P)H:Quinone Oxidoreductase Type I (Hnqo1) Activation Of Quinone Propionic Acid Trigger Groups, Maria F. Mendoza, Nicole M. Hollabaugh, Suraj U. Hettiarachchi, Robin L. Mccarley

Faculty Publications

NAD(P)H:quinone oxidoreductase type I (NQO1) is a target enzyme for triggered delivery of drugs at inflamed tissue and tumor sites, particularly those that challenge traditional therapies. Prodrugs, macromolecules, and molecular assemblies possessing trigger groups that can be cleaved by environmental stimuli are vehicles with the potential to yield active drug only at prescribed sites. Furthermore, quinone propionic acids (QPAs) covalently attached to prodrugs or liposome surfaces can be removed by application of a reductive trigger stimulus, such as that from NQO1; their rates of reductive activation should be tunable via QPA structure. We explored in detail the recombinant human NAD(P)H:quinone …

We report on the synthesis and characterization of a PEGylated IR786 GUMBOS (Group of Uniform Materials Based on Organic Salts). The synthesis of this material was accomplished using a three step protocol: (1) substitution of chloride on the cyclohexenyl ring in the heptamethine chain of IR786 by 6-aminohexanoic acid, (2) grafting of methoxy polyethylene glycol (MeOPEG) onto the 6-aminohexanoic acid via an esterification reaction, and (3) anion exchange between [PEG786][I] and lithium bis(trifluoromethylsulfonyl)imide (LiNTf 2) or sodium bis(2-ethylhexyl)sulfosuccinate (AOT) in order to obtain PEG786 GUMBOS. Examination of spectroscopic data for this PEG786 GUMBOS indicates a large stokes shift (122 nm). …

Dartmouth Scholarship

The antiapoptotic Bcl-2 family member Mcl-1 is a PEST protein (containing sequences enriched in proline, glutamic acid, serine, and threonine) and is subject to rapid degradation via multiple pathways. Impaired degradation leading to the maintenance of Mcl-1 expression is an important determinant of drug resistance in cancer. Phosphorylation at Thr 163 in the PEST region, stimulated by 12-O-tetradecanoylphorbol acetic acid (TPA)-induced activation of extracellular signal-regulated kinase (ERK), is associated with Mcl-1 stabilization in BL41-3 Burkitt lymphoma cells. This contrasts with the observation that Thr 163 phosphorylation in normal fibroblasts primes glycogen synthase kinase (GSK3)-induced phosphorylation at Ser 159, producing a …

The Role Of Micrornas In Breast Cancer Migration, Invasion And Metastasis, Joy Tang, Fazlul H. Sarkar, Aamir Ahmad

Honors College Theses

MicroRNAs (miRNAs) are a major class of small, noncoding RNA molecules that regulate gene expression by targeting mRNAs to trigger either translational repression or mRNA degradation. They have recently been more widely investigated due to their potential role as targets for cancer therapy. Many miRNAs have been implicated in several human cancers, including breast cancer. miRNAs are known to regulate cell cycle and development, and thus may serve as useful targets for exploration in anticancer therapeutics. The link between altered miRNA signatures and breast cancer development and metastasis can be observed either through the loss of tumor suppressor miRNAs, such …

School Food Environment The Frontline For Childhood Obesity Prevention: A Mixed-Method Study Of Nutritional Competencies And Skills Of School Nutrition Professionals In Nebraska, Zainab Rida

Department of Nutrition and Health Sciences: Dissertations, Theses, and Student Research

The purpose of this statewide study was to address the nutrition knowledge, attitudes, and perceptions of school foodservice personnel in Nebraska regarding offering/serving healthy school meals. Moreover, this study identified some potential barriers and avenues of action for offering/serving healthy school meals. Data obtained from this study indicates that there is a strong correlation (r= .103, p r .237, p< .01) between the foodservice staff’s self-efficacy and their practices of offering/serving healthy school meals. Fortunately, the relationship between foodservice staff practices of offering/serving healthy school meals and their self-efficacy was positive and significantly predicted practices scores, β =.237, P< 0.01. The finding of the present study also identifies many barriers including lack of time and support that face the foodservice personnel in offering/serving healthy school meals. The findings suggested that there is an urgent need of a full school approach to promote and encourage healthy eating habits among students. Future research is needed to evaluate school wellness policies regarding healthy eating practices in schools. Moreover, establish partnerships with communities and universities for intervention that target students and their parents.

The Unclothed Body In Francisco Goya's The Disasters Of War, Elizabeth Sanderford Oct 2012

The Unclothed Body In Francisco Goya's The Disasters Of War, Elizabeth Sanderford

Theses and Dissertations

Francisco Goya (1746-1828) created a series of prints entitled The Disasters of War (1810-1820) (Los Desastres de la Guerra) depicting the Napoleonic Wars in Spain, the famine that subsequently occurred in Madrid, and symbolic representations similar to his print series Los Caprichos (1797-1799). This thesis will consider Goya’s use of the unclothed human body in his series of prints The Disasters of War. This thesis will place these representations within the contexts of other works produced by Goya that portray the unclothed figures, as well as within the Spanish artistic tradition of Goya’s time. Through this investigation, I will situate …
