How is it that Americans are so obsessed with nutrition and dieting and yet remain unhealthy? This project attempts to give a theoretically driven answer to this great paradox within the Western diet. Constance Calice analyzes the practice and rhetoric of dieting as a crystallization of a problematic relationship to food using a Foucauldian understanding of discipline. Using examples from the media, she illustrates the way in which outside forces effect our food choices and the power relationships formed in this exchange. Population Dynamics Of Stored-Product Insects At A Rice Mill In Northeast Arkansas, Amanda Lea White

The flight activity, seasonal and spatial distributions, and effects of fumigations on the lesser grain borer [Rhyzopertha dominica (F.)], warehouse beetle [Trogoderma variabile (Ballion)], cigarette beetle [Lasioderma serricorne (F.)], and Indianmeal moth [Plodia interpunctella (Hûbner)] were examined at a rice mill in Northeast Arkansas from June 2008 to October 2010. Stored-product insect activity was monitored using pheromone-baited glue traps (N = 99) on the exterior walls or fences around the perimeter and each major building of interest on the property. The warehouse beetle was the most abundant species which comprised 72.3% of all stored-product insects captured. The warehouse beetle showed …

Chlamydomonas reinhardtii is a unicellular green alga. In C. reinhardtii, three types of photoreceptors are known to be present: rhodopsins, phototropins, and cryptochromes. The single cryptochrome is the most likely photoreceptor for adjusting the circadian clock to the daily light/dark cycles, because cryptochromes are involved in clock entrainment in higher plants and insects. In this segment of the research, C. reinhardtii strains, which were genetically modified through transformation with a RNA interference construct, were screened for reduction in cryptochrome compared to the control strain. The World Health Organization (WHO) implemented the Millennium Declaration in 2000 to establish global health goals to be achieved by 2015. Two of these Millennium Development Goals (MDG’s) are directly related to maternal, neonatal and child health. Healthcare during the prenatal period can help achieve the WHO MDG’s for maternal health and child mortality. Evidence has shown that women in Kenya have limited access to prenatal care. This study focused on the prenatal care and postnatal outcome trends of the women in Kasigau, Kenya. Immunoglobulin E (IgE)-conjugated gold nanoparticles were produced via in situ conjugation of gold nanoparticles with immunoglobulin E by laser ablation of Au in a liquid solution. The colloidal stability and the size distribution of the resulting bio-nanoconjugates were examined with UV-Visible spectroscopy (UV-Vis), dynamic light scattering (DLS), and transmission electron microscopy (TEM). These techniques showed that the Au nanoparticles in aqueous solutions were highly monodispersed spherical particles with a very narrow size distribution. The particles net diameter using TEM, was found to be D5 =3.8±0.9nm and D10 =4.7±1.3nm while the hydrodynamic diameter obtained with DLS was found to be h5 …

Bioanalytical Screening Of Riboflavin Antagonists For Targeted Drug Delivery - A Thermodynamic And Kinetic Study, Anna Plantinga, Amanda Witte, Ming Hsin Li, Andrew Harmon

The present study screened riboflavin mimicking small molecules to determine their binding activity for the riboflavin binding protein. We performed thermodynamic and kinetic binding studies of these molecules using a combination of two analytical approaches: isothermal titration calorimetry and surface plasmon resonance spectroscopy. Screening of a biased set of nonriboflavin-based small molecules by microcalorimetry led to the discovery of two known drug molecules, quinacrine and chloroquine, as favorable ligands for the riboflavin receptor with KD values of 264 and 2100 nM, respectively. We further demonstrated that quinacrine is a competitive ligand for the receptor as measured by surface plasmon resonance. …

Opioids have been widely applied in clinics as one of the most potent pain relievers for centuries, but their abuse has deleterious physiological effects beyond addiction. However, the underlying mechanism by which microglia in response to opioids remains largely unknown. Here we show that morphine induces the expression of Toll-like receptor 9 (TLR9), a key mediator of innate immunity and inflammation. Interestingly, TLR9 deficiency significantly inhibited morphine-induced apoptosis in microglia. Similar results were obtained when endogenous TLR9 expression was suppressed by the TLR9 inhibitor CpGODN. Possible Molecular Mechanism To Account For Wavelength Dependence Of Equilibration Rates Of Patman And Laurdan In Phosphatidylcholine Bilayers, Hannabeth A. Franchino

Patman is a fluorescent membrane probe related to Laurdan. The structural distinctions between the two probes are the lengths of the aliphatic tails (eleven carbons in Laurdan and fifteen in Patman) and the presence of a trimethylammonium group on Patman that produces a positively-charged head. Preliminary studies exploring Patman as a probe to detect membrane properties during apoptosis revealed that the fluorescence intensity of two edges of the emission spectrum (435 and 500 nm) stabilizes at different rates as the probe binds to the cell membrane. The purpose of the current studies was to identify messages that Latino parents communicate to their offspring about the use of legal and illegal drugs and to determine associations between parental messages and substance use outcomes. Previous research has identified parent-child communication as protective against tobacco, alcohol, and other drug use. However, most of these studies have failed to examine the specific messages communicated and those that have focused almost exclusively on non-Hispanic Caucasians. Hydrologic science has largely built its understanding of the hydrologic cycle using contemporary data sources (i.e., last 100 years). However, as we try to meet water demand over the next 100 years at scales from local to global, we need to expand our scope and embrace other data that address human activities and the alteration of hydrologic systems. For example, the accumulation of human impacts on water systems requires exploration of incompletely documented eras. The ocean and atmosphere are characterized mainly by stable stratification which sustains propagation of particular ocurrances called internal gravity waves. These internal waves are generated as long as a perturbation to the stratification occurs at a frequency lower than the buoyancy (natural) frequency of the medium. These waves, once generated, propagate with wavelengths which can vary from a few meters to hundreds of meters in the vertical and thousands of meters in the horizontal. These waves propagate through the ocean and atmosphere interacting with other flow phenomena and eventually overturn and break, dissipating their energy. We present a linear type ion trap consisting of metal lines that are lithographically patterned onto two ceramic substrates. Trapping fields are realized by applying specific voltage potentials to each line. This arrangement allows miniaturization approaches that are immune to surface roughness issues and increased complexities of precision machining of electrode surfaces. We also present how these traps allow a thorough study of higher order multipoles and their effect on mass analyzer performance.

Design Of A Flapping Wing Mechanism For Force Analysis And Optimization, Ryan B. George, Scott L. Thomson

The design of a robotic flapping wing mechanism is discussed. The design allows for dynamic adjustment of flapping trajectory in fluid with three rotational degrees of freedom, while keeping all motors and encoders out of the fluid (i.e., water or oil) to protect critical equipment from potential failure and increase reliability. Mechanism control is discussed. Preliminary optimization using a Box-Behnkin design approach is used and shows successful parameter optimization. Mechanism limitations are addressed.

Characterization Of Changes In Megalagrion Opsin Genes To Detect Signatures Of Selection, Bhagya G. Janananda

Megalagrion damselflies have radiated into new breeding habitats independently at least six times in the Hawaiian archipelago, and have evolved bright body coloration numerous times. We hypothesize that these radiations are correlated with specific changes in the opsin proteins. We isolated and characterized two opsin genes from nine different Megalagrion species. The opsin phylogeny is consistent with the phylogeny based on breeding habitat preference of Megalagrion species supporting the correlation between the evolutionary changes of vision and habitat shifts. dN/dS ratios of opsin sequences show that these genes are evolving under purifying selection, though some sites of the opsin genes …

The island continent of Madagascar spans nearly 13.5o of latitude in the SW Indian Ocean. Its coastline includes a number of well developed coral reefs, ranging from tropical Nosy Bé (NW Madagascar, 12oS) and Vohemar (Volhmarina, NE Madagascar, 13oS) to subtropical Tuléar (Toliara , SW Madagascar, 23.5oS), as well as temperate coral communities at Fort Dauphin (Tolagnaro, SE Madagascar, 25oS). Given the range of environmental conditions experienced by reef corals at these different sites, Madagascar represents an ideal location to study the distribution of algal symbionts (Symbiodinium spp.) in these coral hosts.
