Saccharomyces Cerevisiae As A Model Organism To Delineate Initial Lesion Detection Events In Chromatin Repair: A Focus On Ddb2-Mediated Gg-Ner, Kristi Lyn Jones

Open Access Dissertations

DNA damage repair is an essential and complex cellular process. Although the basic mechanisms of nucleotide excision repair (NER) have been studied for decades, some mechanistic details remain elusive. The lesion detection step remains one of the most elusive in the process of NER in the contest of chromatin. The work described herein addresses the initial events in the lesion detection step of chromatin repair, also referred to as global genome repair (GG-NER). Both the role of post-translational modifications of lesion identification proteins, and the initial sequence of events in recruitment of repair and remodeling factors are investigated. First, the …

Bobcats Do Not Exhibit Rub Response Despite Presence At Hair Collection Stations, Chirstopher E. Comer, Symmank M. E., James C. Kroll

Faculty Publications

We evaluated the detection rate for hair snare sampling for bobcats (Lynx rufus) using colocated hair snares and infrared-triggered cameras at 20 locations on private property in eastern Texas. Hair snares and cameras were placed together at survey stations that included both visual and olfactory attractants. In 1,680 trap-nights we photographically documented 15 visits by bobcats but collected only one bobcat hair sample. Our observations suggested limited rubbing behavior by bobcats at hair snares despite presence at hair collection stations. The explanation for this behavior remains unclear but is consistent with observations of low and variable hair trap …

Intramolecular C-H Amination With Sulfonyl Azides, X. Peter Zhang, Joshua V. Ruppel

USF Patents

Cobalt (II) complexes of porphyrins are effective catalysts for intramolecular nitrene insertion of C—H bonds with arylsulfonyl azides. The cobalt-catalyzed process can proceed efficiently under mild and neutral conditions in low catalyst loading without the need of other reagents or additives, generating nitrogen gas as the only byproduct. Using the simple tetraphenylporphyrin (TPP) as the ligand, the cobalt-catalyzed intramolecular amidation can be applied to primary, secondary, and tertiary C—H bonds and suitable for a broad range of arylsulfonyl azides, leading to the syntheses of various benzosultam derivatives in excellent yields

It is well accepted that lateral redistribution of the phytohormone auxin underlies the bending of plant organs towards light. In monocots, photoreception occurs at the shoot tip above the region of differential growth. Despite more than a century of research, it is still unresolved how light regulates auxin distribution and where this occurs in dicots. Here, we establish a system in Arabidopsis thaliana to study hypocotyl phototropism in the absence of developmental events associated with seedling photomorphogenesis. We show that auxin redistribution to the epidermal sites of action occurs at and above the hypocotyl apex, not at the elongation zone. …

Aintegumenta And Aintegumenta-Like6 Regulate Auxin-Mediated Flower Development In Arabidopsis, Beth A. Krizek Jun 2011

Aintegumenta And Aintegumenta-Like6 Regulate Auxin-Mediated Flower Development In Arabidopsis, Beth A. Krizek

Faculty Publications

Background: Two related genes encoding AP2/ERF-type transcription factors, AINTEGUMENTA (ANT) and AINTEGUMENTA-LIKE6 (AIL6), are important regulators of floral growth and patterning in Arabidopsis. Evidence suggests that these genes promote several aspects of flower development in response to auxin. To investigate the interplay of ANT, AIL6 and auxin during floral development, I have examined the phenotypic consequences of disrupting polar auxin transport in ant, ail6 and ant ail6 mutants by either genetic or chemical means.

Results: Plants containing mutations in ANT or AIL6 alone or in both genes together exhibit increased sensitivity to disruptions in polar auxin transport. …

A Randomized Clinical Trial Of A Brief Motivational Intervention (Bmi) For Obesity In College Students, Joanna Buscemi

Electronic Theses and Dissertations Archive

Young adults are at an increased risk for weight gain as they begin college and this has implications for the onset of future health consequences such as Type II Diabetes, heart disease, hypertension, and some cancers. Brief motivational interventions (BMIs) have been found to be effective with college students for reducing risky health behaviors such as alcohol consumption, but have not been developed and tested with a primary goal of reducing obesity. BMIs have been developed and tested for the treatment of obesity and weight-related health behaviors (WRHB) in other populations, such as adults and adolescents, with promising results. The …

Efficient Unfolding Pattern Recognition In Single Molecule Force Spectroscopy Data, Bill Andreopoulos, Dirk Labudde Jun 2011

Efficient Unfolding Pattern Recognition In Single Molecule Force Spectroscopy Data, Bill Andreopoulos, Dirk Labudde

Faculty Publications, Computer Science

BackgroundSingle-molecule force spectroscopy (SMFS) is a technique that measures the force necessary to unfold a protein. SMFS experiments generate Force-Distance (F-D) curves. A statistical analysis of a set of F-D curves reveals different unfolding pathways. Information on protein structure, conformation, functional states, and inter- and intra-molecular interactions can be derived.ResultsIn the present work, we propose a pattern recognition algorithm and apply our algorithm to datasets from SMFS experiments on the membrane protein bacterioRhodopsin (bR). We discuss the unfolding pathways found in bR, which are characterised by main peaks and side peaks. A main peak is the result of the pairwise …

Growth Phase-Dependent Gene Regulation In Vivo In Sulfolobus Solfataricus, Melissa Deyoung, Michelle Thayer, John Van Der Oost, Kenneth M. Stedman Jun 2011

Growth Phase-Dependent Gene Regulation In Vivo In Sulfolobus Solfataricus, Melissa Deyoung, Michelle Thayer, John Van Der Oost, Kenneth M. Stedman

Biology Faculty Publications and Presentations

Ribosomal genes are strongly regulated dependent on growth phase in all organisms, but this regulation is poorly understood in Archaea. Moreover, very little is known about growth phase-dependent gene regulation in Archaea. SSV1- based lacS reporter gene constructs containing the Sulfolobus 16S/23S rRNA gene core promoter, the TF55a core promoter, or the native lacS promoter were tested in Sulfolobus solfataricus cells lacking the lacS gene. The 42-bp 16S/23S rRNA gene and 39-bp TF55a core promoters are sufficient for gene expression in S. solfataricus. However, only gene expression driven by the 16S/23S rRNA gene core promoter is dependent on …

Thermal adaptation of gross primary production and ecosystem respiration has been well documented over broad thermal gradients. However, no study has examined their interaction as a function of temperature, i.e. the thermal responses of net ecosystem exchange of carbon (NEE). In this study, we constructed temperature response curves of NEE against temperature using 380 site-years of eddy covariance data at 72 forest, grassland and shrubland ecosystems located at latitudes ranging from ~29° N to 64° N. The response curves were used to define two critical temperatures: transition temperature (Tb) at which ecosystem transfer from carbon source to sink and optimal …

Store-Operated Ca(2+) Entry (Soce) Contributes To Normal Skeletal Muscle Contractility In Young But Not In Aged Skeletal Muscle, Angela M. Thornton, Xiaoli Zhao, Noah Weisleder, Leticia S. Brotto, Sylvain Bougoin, Thomas M. Nosek, Michael B. Reid, Brian Hardin, Zui Pan, Jianjie Ma, Jerome Parness, Marco Brotto

Physiology Faculty Publications

Muscle atrophy alone is insufficient to explain the significant decline in contractile force of skeletal muscle during normal aging. One contributing factor to decreased contractile force in aging skeletal muscle could be compromised excitation-contraction (E-C) coupling, without sufficient available Ca(2+) to allow for repetitive muscle contractility, skeletal muscles naturally become weaker. Using biophysical approaches, we previously showed that store-operated Ca(2+) entry (SOCE) is compromised in aged skeletal muscle but not in young ones. While important, a missing component from previous studies is whether or not SOCE function correlates with contractile function during aging. Here we test the contribution of extracellular …

The Impact Of Depression And Health On Sexual Satisfaction For Older Couples, Victoria Celeste Scott Jun 2011

The Impact Of Depression And Health On Sexual Satisfaction For Older Couples, Victoria Celeste Scott

Theses and Dissertations

This study of 535 older married couples examined the relationship between depression and health and sexual satisfaction directly and when mediated by communication. The sample included 535 older couples who completed a survey questionnaire known as Project Couple Retire. Among the items in the questionnaire were measures of depression, health, perception of sexual intimacy, communication and other demographic information. Results from Structural Equation Modeling indicated that for women, health was a significant predictor of sexual satisfaction. For both genders, the results suggest that depression, when mediated by communication, is a predictor of sexual satisfaction among older couples.

The Influence Of Executive Functioning On The Intention-Behavior Link For Sexual Health Behavior Among Young Men, Robert John Sawyer Ii Jun 2011

The Influence Of Executive Functioning On The Intention-Behavior Link For Sexual Health Behavior Among Young Men, Robert John Sawyer Ii

Electronic Theses and Dissertations Archive

Typically, research and interventions have relied upon social-cognitive theories like the Theory of Planned Behavior (TPB) to understand sexual health behavior. Of particular import in the TPB model is the intention-behavior link where strength of intentions to enact a particular behavior are significantly related to actual behavior performance. Recently, investigations have sought to understand possible moderators of the intention-behavior relationship. One moderating variable examined is the common neuropsychological construct executive functioning. Executive functioning consists of skills for self-regulation, inhibition of impulsivity, organization, planning, and so forth that impact attainment of previously established goals. This study examined the possible influence of …

Progress Toward Theonellamide F, Jennifer Dawn Bettale Jun 2011

Progress Toward Theonellamide F, Jennifer Dawn Bettale

Theses and Dissertations

Theonellamide is a bicyclic peptide isolated from a marine sponge, which shows interesting biological activity. It contains several unnatural amino acids, among which are (2S,3R)-3-hydroxyasparagine (L-erythro-β-hydroxyasparagine) (β-OHAsn) and τ-L-histindino-D-alanine (τ-HAL). Although there were previous synthetic efforts toward each of these unnatural amino acids, the efforts were not ideal due to expensive starting material, time-consuming steps, and poor regioselectivity. The presented work demonstrates an inexpensive, enantioselective synthesis of β-OHAsn, which can be completed in a matter of weeks, as well as several attempts at a novel regiospecific approach toward τ-HAL, including work on a model study.
