Direct Imaging Of Two-State Dynamics On The Amorphous Silicon Surface, S. Ashtekar, Gregory E. Scott, J. Lyding, M. Gruebele

Chemistry and Biochemistry

Amorphous silicon is an important material, amidst a debate whether or not it is a glass. We produce amorphous Si surfaces by ion bombardment and vapor growth, and image discrete Si clusters which hop by two-state dynamics at 295 K. Independent of surface preparation, these clusters have an average diameter of ~5 atoms. Given prior results for metallic glasses, we suggest that this cluster size is a universal feature. The hopping activation free energy of 0.93 ± 0.15 eV is rather small, in agreement with a previously untested surface glass model. Hydrogenation quenches the two-state dynamics, apparently by increasing surface …

Cis 3,4', 5-Trimethoxy-3'-Aminostilbene (Stilbene 5c) Induces Apoptosis And Protective Autophagy In B16f10 Melanoma Cells, Betelehem Asnake

Theses and Dissertations

The weak selectivity of chemotherapeutic drugs against tumors has sustained efforts to develop better chemotherapeutic agents that are more potent and selective at destroying tumor cell populations versus normal tissues. This project focuses on evaluating the cell killing effects of the microtubule inhibitor, stilbene 5c, against melanoma cancer. We utilized an in vitro murine melanoma model to study the effects of stilbene 5c on tumor proliferation and survival, as well as growth arrest and cell death. Our findings indicate that stilbene 5c promotes dose dependent cell death in melanomas with the induction of apoptosis and autophagy. The role of autophagy …

Antioxidants In Cancer Research And Prevention: Assay Comparison, Structure-Function Analysis, And Food Product Analysis, Andrew Robert Garrett Jun 2011

Antioxidants In Cancer Research And Prevention: Assay Comparison, Structure-Function Analysis, And Food Product Analysis, Andrew Robert Garrett

Theses and Dissertations

Recent epidemiological studies have suggested that the development and progression of several chronic diseases may be initiated or augmented by oxidative stress. Reactive oxygen species and reactive nitrogen species react readily with and can damage nucleic acids, proteins, and lipids. While biological systems are equipped antioxidant defenses to cope with oxidative stress, oxidative damage may still occur when oxidative stress overwhelms antioxidant defenses. This damage, if left unchecked, may lead to a variety of degenerative diseases, including heart disease, Alzheimer's Disease, Parkinson's Disease and cancer. Several assays have been designed to describe the antioxidant activity of various phytochemicals, vitamins, and …

Journal Articles: Biochemistry & Molecular Biology

Sialyl Lewis x (sLe(x)) plays an important role in cancer metastasis. But, the mechanism for its production in metastatic cancers remains unclear. The objective of current study was to examine the effects of a proinflammatory cytokine on the expression of glycosyltransferase and sulfotransferase genes involved in the synthesis of selectin ligands in a prostate cancer cell line. Androgen-independent human lymph node-derived metastatic prostate cancer cells (C-81 LNCaP), which express functional androgen receptor and mimic the castration-resistant advanced prostate cancer, were used. TNFα treatment of these cells increased their binding to P-, E- and L-selectins, anti-sLe(x) antibody, and anti-6-sulfo-sialyl Lewis x …

The Fight In My Classroom: A Story Of Intersectionality In Practitioner Research, Zachary A. Casey Jun 2011

The Fight In My Classroom: A Story Of Intersectionality In Practitioner Research, Zachary A. Casey

i.e.: inquiry in education

Using the story of a fight between an African American male student and a white male student in a high school classroom, I trace not only the raced and classed actors involved, but also the story of my sense making as a practicing teacher and researcher. Drawing on the concept of intersectionality, I show how a story that seems as simple as a racist white student and a reactionary African American student is far more nuanced and complex. Finally, I discuss what this complexity means for us as researchers of our own practice, and find that intersectionality as a theoretical …

Cooperative And Antagonistic Contributions Of Two Heterochromatin Proteins To Transcriptional Regulation Of The Drosophila Sex Determination Decision, Hui Li, Janel Rodriguez, Youngdong Yoo, Momin Mohammed Shareef, Ramakrishna Badugu, Jamila I. Horabin, Rebecca Kellum Jun 2011

Horabin, Rebecca Kellum

Biology Faculty Publications

Eukaryotic nuclei contain regions of differentially staining chromatin (heterochromatin), which remain condensed throughout the cell cycle and are largely transcriptionally silent. RNAi knockdown of the highly conserved heterochromatin protein HP1 in Drosophila was previously shown to preferentially reduce male viability. Here we report a similar phenotype for the telomeric partner of HP1, HOAP, and roles for both proteins in regulating the Drosophila sex determination pathway. Specifically, these proteins regulate the critical decision in this pathway, firing of the establishment promoter of the masterswitch gene, Sex-lethal (Sxl). Female-specific activation of this promoter, Sxl(Pe), is essential to females, as it provides SXL …

Department of Botany and Plant Pathology Faculty Publications

The plant-pathogenic fungus Mycosphaerella graminicola (asexual stage: Septoria tritici) causes septoria tritici blotch, a disease that greatly reduces the yield and quality of wheat. This disease is economically important in most wheat-growing areas worldwide and threatens global food production. Control of the disease has been hampered by a limited understanding of the genetic and biochemical bases of pathogenicity, including mechanisms of infection and of resistance in the host. Unlike most other plant pathogens, M. graminicola has a long latent period during which it evades host defenses. Although this type of stealth pathogenicity occurs commonly in Mycosphaerella and other Dothideomycetes, …

Principles Of Learning: A Conceptual Framework For Domain-Specific Theories Of Learning, Christian J. Weibell Jun 2011

Principles Of Learning: A Conceptual Framework For Domain-Specific Theories Of Learning, Christian J. Weibell

Theses and Dissertations

This study is predicated on the belief that there does not now exist, nor will there ever exist, any single theory of learning that is broad enough to account for all types of learning yet specific enough to be maximally useful in practical application. Perhaps this dichotomy is the reason for the apparent gap between existing theories of learning and the practice of instructional design. As an alternative to any supposed grand theory of learning—and following the lead of prominent thinkers in the fields of clinical psychology and language teaching—this study proposes a shift toward principles. It presents a principle-based …

Imaging Mass Spectrometry Detection Of Gangliosides Species Within The Mouse Brain Following Transient Focal Cerebral Ischemia, Shawn N. Whitehead, Kenneth H. N. Chan, Sandhya Gangaraju, Jacqueline Slinn, Jianjun Li, Sheng T. Hou

Anatomy and Cell Biology Publications

Gangliosides, a member of the glycosphingolipid family, are heterogeneously expressed in biological membranes and are particularly enriched within the central nervous system. Gangliosides consist of mono- or poly-sialylated oligosaccharide chains of variable lengths attached to a ceramide unit and are found to be intimately involved in brain disease development. The purpose of this study is to examine the spatial profile of ganglioside species using matrix-assisted laser desorption/ionization (MALDI) imaging (IMS) following middle cerebral artery occlusion (MCAO) reperfusion injury in the mouse. IMS is a powerful method to not only discriminate gangliosides by their oligosaccharide components, but also by their carbon …

Polar Patterns In Active Fluids, Luca Giomi, M. Cristina Marchetti Jun 2011

Polar Patterns In Active Fluids, Luca Giomi, M. Cristina Marchetti

Physics - All Scholarship

We study the spatio-temporal dynamics of a model of polar active fluid in two dimensions. The system exhibits a transition from an isotropic to a polarized state as a function of density. The uniform polarized state is, however, unstable above a critical value of activity. Upon increasing activity, the active fluids displays increasingly complex patterns, including traveling bands, traveling vortices and chaotic behavior. The advection arising from the particles self-propulsion and unique to polar fluids yields qualitatively new behavior as compared to that obtain in active nematic, yielding traveling-wave structures. We show that the nonlinear hydrodynamic equations can be mapped …

Smoking Cessation And Relapse Among Pregnant African-American Smokers In Washington, Dc, Ayman El-Mohandes, M. Nabil El-Khorazaty, Michele Kieley, Marie G. Gantz

Publications and Research

Smoking is the single most preventable cause of perinatal morbidity. This study examines smoking behaviors during pregnancy in a high risk population of African Americans. The study also examines risk factors associated with smoking behaviors and cessation in response to a cognitive behavioral therapy (CBT) intervention. This study is a secondary analysis of data from a randomized controlled trial addressing multiple risks during pregnancy. Five hundred African-American Washington, DC residents who reported smoking in the 6 months preceding pregnancy were randomized to a CBT intervention. Psycho-social and behavioral data were collected. Self-reported smoking and salivary cotinine levels were measured prenatally …

The Regulation Of Liv-1 Mrna In Mda-Mb-231 Human Breast Cancer Cells And Its Association With E-Cadherin (Cdh1), Leelyn Chong Jun 2011

The Regulation Of Liv-1 Mrna In Mda-Mb-231 Human Breast Cancer Cells And Its Association With E-Cadherin (Cdh1), Leelyn Chong

Master's Theses

LIV-1, the estrogen-regulated gene encoding a member of the ZIP family of zinc transporters (SLC39A6), has been implicated in both growth and metastasis of estrogen-receptor (ER) positive (+) and negative (-) breast cancer cells. In ER+ breast cancer cells, LIV-1 expression is induced by both estrogen and insulin. In embryonic cells, LIV-1 has also been negatively associated with the expression of the cell adhesion protein, E-cadherin (CDH1), thought to play a role in metastasis. In the present study, we investigated the regulation of LIV-1 and its relationship to CDH1 in MDA-MB-231 ER- breast cancer cells. Cells were …
