Synthesis And Characterization Of Amphiphilic Cyclic Diblock Copolypeptoids From N-Heterocyclic Carbene-Mediated Zwitterionic Polymerization Of N-Substituted N-Carboxyanhydride, Chang Uk Lee, Thomas P. Smart, Li Guo, Thomas H. Epps, Donghui Zhang

N-Heterocyclic carbene (NHC)-mediated ring-opening polymerization of N-decyl-N-carboxylanhydride monomer (De-NCA) has been shown to occur in a controlled manner, yielding cyclic poly(N-decylglycine)s (c-PNDGs) with polymer molecular weights (MW) between 4.8 and 31 kg mol -1 and narrow molecular weight distributions (PDI < 1.15). The reaction exhibits pseudo-first-order kinetics with respect to monomer concentration. The polymer MW increases linearly with conversion, consistent with a living polymerization. ESI MS and SEC analyses confirm the cyclic architectures of the forming polymers. DSC and WAXS studies reveal that the c-PNDG homopolymers are highly crystalline with two prominent first-order transitions at 72-79 °C (T m,1) and 166-177 °C (T m,2), which have been attributed to side chain and main chain melting, respectively. A series of amphiphilic cyclic diblock copolypeptoids [i.e., poly(N-methylglycine)-b-poly(N- decylglycine) (c-PNMG-b-PNDG)] with variable molecular weight and composition were synthesized by sequential NHC-mediated polymerization of the corresponding N-methyl-N-carboxyanhydride (Me-NCA) and De-NCA monomers. 1H NMR analysis reveals that adjusting the initial monomer to NHC molar ratio can readily control the block copolymer chain length and composition. Time-lapsed light scattering and cryogenic transmission electron microscopy (cryo-TEM) analyses of c-PNDG-b-PNMG samples showed that the amphiphilic cyclic block copolypeptoids self-assemble into spherical micelles that reorganize into micrometer-long cylindrical micelles with uniform diameter in room temperature methanol over the course of several days. An identical morphological transition has also been noted for the linear analogues, which occurs more rapidly than for the cyclic copolypeptoids. We tentatively attribute this difference to the different crystallization kinetics of the solvophobic block (i.e., PNDG) in the cyclic and linear block copolypeptoids. © 2011 American Chemical Society.

Podocytes are highly differentiated and polarized epithelial cells located on the visceral side of the glomerulus. They form an indispensable component of the glomerular filter, the slit diaphragm, formed by several transmembrane proteins and adaptor molecules. Disruption of the slit diaphragm can lead to massive proteinuria and nephrotic syndrome in mice and humans. CD2AP is an adaptor protein that is important for the maintenance of the slit diaphragm. Together with its paralogue, CIN85, CD2AP belongs to a family of adaptor proteins that are primarily described as being involved in endocytosis and downregulation of receptor tyrosine kinase activity. High levels of prenatal cortisol have been previously reported to retard fetal growth. Although cortisol plays a pivotal role in prenatal maturation, heightened exposure to cortisol can result in lower body weights at birth, which have been shown to be associated with adult diseases like hypertension and cardiovascular disease. This study examines the relationship between natural variation in gestational cortisol and fetal and postnatal growth in marmoset monkeys. Urinary samples obtained during the mother’s gestation were analyzed for cortisol. Marmoset body mass index (BMI) was measured from birth through 540 days in 30- or 60-day intervals. This research focuses on the design and control of a fleet of robotic kayaks, and presents experimental data regarding the functionality and performance of the system. One of the key technical challenges in fielding multi-robot systems for real-world applications is the coordination and relative motion control of the individual units. Coordinated formation control of the fleet is implemented through the use of the cluster space control architecture, which is a full-order controller that treats the fleet as a virtual, articulating, kinematic mechanism. It is one of the marvels of our time that the nuclear industry managed to resurrect itself from its ruins at the end of the last century, when it crumbled under its costs, inefficiencies, and mega-accidents. Chernobyl released hundreds of times the radioactivity of the Hiroshima and Nagasaki bombs combined, contaminating more than 40% of Europe and the entire Northern Hemisphere. Demensia Alzheimer menempati urutan kesembilan penyebab kematian di Amerika Serikat. Demensia adalah kondisi yang sering dialami yang berhubungan dengan berbagai faktor dan gaya hidup terutama diet. Penelitian ini bertujuan untuk mengetahui hubungan defisiensi asupan riboflavin (vitamin B12) dengan demensia pada usia lanjut (usila). Penelitian ini menggunakan desain studi cross sectional dan dilaksanakan pada bulan September 2007 sampai dengan Januari 2008. Neuromolecular Imaging (NMI) based on adsorptive electrochemistry, combined with Dual Laser Doppler Flowmetry (LDF) is presented herein to investigate the brain neurochemistry affected by enoxaparin (Lovenox®), an antiplatelet/antithrombotic medication for stroke victims. NMI with miniature biosensors enables neurotransmitter and neuropeptide (NT) imaging; each NT is imaged with a response time in milliseconds. A semiderivative electronic reduction circuit images several NT’s selectively and separately within a response time of minutes. Spatial resolution of NMI biosensors is in the range of nanomicrons and electrochemically-induced current ranges are in pico- and nano-amperes. Background: Dynamic Bayesian Network (DBN) is an approach widely used for reconstruction of gene regulatory networks from time-series microarray data. Its performance in network reconstruction depends on a structure learning algorithm. REVEAL (REVerse Engineering ALgorithm) is one of the algorithms implemented for learning DBN structure and used to reconstruct gene regulatory networks (GRN). However, the two-stage temporal Bayes network (2TBN) structure of DBN that specifies correlation between time slices cannot be obtained by score metrics used in REVEAL.

Background: Along with obesity, physical inactivity, and family history of metabolic disorders, African American ethnicity is a risk factor for type 2 diabetes (T2D) in the United States. However, little is known about the differences in gene expression and transcriptomic profiles of blood in T2D between African Americans (AA) and Caucasians (CAU), and microarray analysis of peripheral white blood cells (WBCs) from these two ethnic groups will facilitate our understanding of the underlying molecular mechanism in T2D and identify genetic biomarkers responsible for the disparities.

Mutations in the insulin gene can impair proinsulin folding and cause diabetes mellitus. Although crystal structures of insulin dimers and hexamers are well established, proinsulin is refractory to crystallization. Although an NMR structure of an engineered proinsulin monomer has been reported, structures of the wild-type monomer and hexamer remain undetermined. We have utilized hydroxyl radical footprinting and molecular modeling to characterize these structures. Differences between the footprints of insulin and proinsulin, defining a "shadow" of the connecting (C) domain, were employed to refine the model. This study compared academically talented students (n = 23) who were not at risk for school failure and resilient at-risk students (n = 27) on risk factors and protective/promotive factors. Participants’ risk status was determined by student assignment. The academically talented students were attending a summer program at a major research university and the at-risk students were graduates of a continuation high school for students who had had numerous infractions at regular high schools. As expected, the two groups differed on risk factors and on factors related to academic status. However, the groups did not differ on psychosocial variables related …

Pentachlorophenol (PCP) is an organochlorine pesticide that decreases the tumor-cell killing (lytic) function of human natural killer (NK) cells. NK cells defend against tumor cells and virally infected cells. They bind to these targets, utilizing a variety of cell-surface proteins. This study examined concentrations of PCP that decrease lytic function for alteration of NK binding to tumor targets. Levels of PCP that caused loss of binding function were then examined for effects on expression of cell-surface proteins needed for binding. 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR) inhibitors, commonly known as statins, may possess cancer preventive and therapeutic properties. Statins are effective suppressors of cholesterol synthesis with a well-established risk-benefit ratio in cardiovascular disease prevention. Mechanistically, targeting HMGCR activity primarily influences cholesterol biosynthesis and prenylation of signaling proteins. Pravastatin is a hydrophilic statin that is selectively taken up by a sodium-independent organic anion transporter protein-1B1 (OATP1B1) exclusively expressed in liver. Simvastatin is a hydrophobic statin that enters cells by other mechanisms. There is currently no accepted method for quantitative measurement of acquisition program interdependency. While some methods exist for measuring dependency or interdependency at the component or system level, these methods do not translate well to program interdependency measurement. The objective of this thesis is to provide a model for measuring acquisition program interdependency accurately and quantitatively. The model presented in this thesis uses four Interdependency Factors to identify dependency relationships between programs. Specific Interdependency Levels are then used to measure the strengths of those dependencies. The model also accounts for measurement of dependencies upon programs that are not directly connected, …

A comprehensive study of the electroluminescence of four GaAs/AlGaAs microcavity devices with InAs/GaInAs quantum dot active regions emitting near 1.3 µm was conducted. The four molecular beam epitaxial grown samples with AlAs oxide aperture confinement layers were fabricated, characterized, and optically modeled. Optical power transmission of the samples was modeled using Matlab and compared with measured transmission data. Resonant cavity light emitting diodes (RCLEDs) and three vertical cavity surface emitting laser (VCSEL) samples were fabricated and electro-optically characterized over a range of injection currents and temperatures. The buildings represent the culmination of the University’s “Higher Ground Campaign,” and they manifest the generous and continued support of the University’s most faithful friends and donors. The link between EBV infection and Burkitt lymphoma (BL) is strong, but the mechanism underlying that link has been elusive. We have developed a mouse model for EBV-associated BL in which LMP2A, an EBV latency protein, and MYC are expressed in B cells. Our model has demonstrated the ability of LMP2A to accelerate tumor onset, increase spleen size, and bypass p53 inactivation. Here we describe the results of total gene expression analysis of tumor and pretumor B cells from our transgenic mouse model. It is important for industries to find ways to find green chemistries to protect the environment and the paper industry is no exception. Using renewable resources such as plant material will be an excellent substitute for the chemicals that are currently used as paper binders. In this project, carbohydrates, proteins and phenolic compounds were applied to air laid, pressed paper in order to obtain a binder that is as strong as the currently used factory binders while being organic and renewable. In addition to its catalytic activity, AChE appears to play a significant non-catalytic role of neurotropic activity on dorsal root ganglion (DRG) neurons of rat. However the mechanism responsible for AChE's neurotrophic activity is not fully understood. Earlier experiments in Dr. Srivatsan's laboratory demonstrated the binding of fluorophore conjugated AChE to cell surface of DRG neurons. Mercury is found throughout the environment with a distribution on a global scale in nearly all terrestrial to marine environments. Biotransformation of divalent mercury and subsequent bioaccumulation of organo-mercury species is catalyzed by sulfate reducing bacteria. Trophic transport of the resultant methyl mercury, a highly toxic species, leads to upward accumulation in the trophic structure of aquatic environments and ultimately to human exposure through consumption of methyl mercury laden fish. Severe droughts were evaluated using a tree-ring network of 12 chronologies (1700−2005) from the Qilian Mountains on the northeastern Tibetan Plateau. A total of 98 drought years, including 78 moderate, 15 severe, and 5 extreme events, were identified for the past 306 yr. Following the starting year of a drought phase, drought commonly continued for 1 to 2 yr. The spatial patterns of moisture conditions in central and eastern China corresponding to 19 major drought years during the period 1700−2000 were grouped into 3 clusters, with one particular pattern showing droughts of nation-wide impact (i.e. 1721, 1928 and 1966). 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR) inhibitors, commonly known as statins, may possess cancer preventive and therapeutic properties. Statins are effective suppressors of cholesterol synthesis with a well-established risk-benefit ratio in cardiovascular disease prevention. Mechanistically, targeting HMGCR activity primarily influences cholesterol biosynthesis and prenylation of signaling proteins. Pravastatin is a hydrophilic statin that is selectively taken up by a sodium-independent organic anion transporter protein-1B1 (OATP1B1) exclusively expressed in liver. Simvastatin is a hydrophobic statin that enters cells by other mechanisms. As in other eukaryotes, protein kinases play major regulatory roles in filamentous fungi. Although the genomes of many plant pathogenic fungi have been sequenced, systematic characterization of their kinomes has not been reported. The wheat scab fungus Fusarium graminearum has 116 protein kinases (PK) genes. Although twenty of them appeared to be essential, we generated deletion mutants for the other 96 PK genes, including 12 orthologs of essential genes in yeast. All of the PK mutants were assayed for changes in 17 phenotypes, including growth, conidiation, pathogenesis, stress responses, and sexual reproduction. Overall, deletion of 64 PK genes resulted in …

The Atlantic killifish (Fundulus heteroclitus) is an environmental sentinel organism used extensively for studies on environmental toxicants and salt (NaCl) homeostasis. Previous research in our laboratory has shown that rapid acclimation of killifish to seawater is mediated by trafficking of CFTR chloride channels from intracellular vesicles to the plasma membrane in the opercular membrane within the first hour in seawater, which enhances chloride secretion into seawater, thereby contributing to salt homeostasis. Acute transition to seawater is also marked by an increase in both mRNA and protein levels of serum glucocorticoid kinase 1 (SGK1) within 15 minutes of transfer. …

In the tropical Indo-Pacific, most phylogeographic studies have focused on the shallow-water taxa that inhabit reefs to approximately 30 m depth. Little is known about the large predatory fishes, primarily snappers (subfamily Etelinae) and groupers (subfamily Epinephelinae) that occur at 100–400 m. These long-lived, slow-growing species support fisheries across the Indo-Pacific, yet no comprehensive genetic surveys within this group have been conducted. Here we contribute the first range-wide survey of a deepwater Indo-Pacific snapper, Pristipomoides filamentosus, with special focus on Hawai'i. Although viruses have been implicated in central nervous system (CNS) diseases of unknown etiology, including multiple sclerosis and amyotrophic lateral sclerosis, the reproducible identification of viral triggers in such diseases has been largely unsuccessful. Here, we explore the hypothesis that viruses need not replicate in the tissue in which they cause disease; specifically, that a peripheral infection might trigger CNS pathology. To test this idea, we utilized a transgenic mouse model in which we found that immune cells responding to a peripheral infection are recruited to the CNS, where they trigger neurological damage. The feasibility and outcomes of a 12-week extracurricular family-based intervention led by physical therapists that was designed to increase physical activity (PA) in three Hispanic male middle-school children was examined. This intervention has limited feasibility and may increase physical activity levels for overweight Hispanic middle school children. In a second study, differences in motor proficiency, strength, endurance, and PA among healthy weight, overweight and obese children were examined, and correlations between BMI and physical impairments were explored. Obese children demonstrated impairments in motor proficiency, strength, and endurance when compared to healthy weight children. Among overweight children, higher BMI was associated …
