Certain fungi thrive in highly radioactive environments including the defunct Chernobyl nuclear reactor. Cryptococcus neoformans (C. neoformans), which uses L-3,4-dihydroxyphenylalanine (L-DOPA) to produce melanin, was used here to investigate how gamma radiation under aqueous aerobic conditions affects the properties of melanin, with the aim of gaining insight into its radioprotective role. Exposure of melanized fungal cell in aqueous suspensions to doses of γ-radiation capable of killing 50 to 80% of the cells did not lead to a detectable loss of melanin integrity according to EPR spectra of melanin radicals. Cryopreservation Of Spin-Dried Mammalian Cells, Nilay Chakraborty, Michael A. Menze, Jason Malsam, Alptekin Aksan, Steven C. Hand, Mehmet Toner

This study reports an alternative approach to achieve vitrification where cells are pre-desiccated prior to cooling to cryogenic temperatures for storage. Chinese Hamster Ovary (CHO) cells suspended in a trehalose solution were rapidly and uniformly desiccated to a low moisture content (< 0.12 g of water per g of dry weight) using a spin-drying technique. Trehalose was also introduced into the cells using a high-capacity trehalose transporter (TRET1). Fourier Transform Infrared Spectroscopy (FTIR) was used to examine the uniformity of water concentration distribution in the spin-dried samples. 62% of the cells were shown to survive spin-drying in the presence of trehalose following immediate rehydration. The spin-dried samples were stored in liquid nitrogen (LN 2) at a vitrified state. It was shown that following re-warming to room temperature and re-hydration with a fully complemented cell culture medium, 51% of the spin-dried and vitrified cells survived and demonstrated normal growth characteristics. Spin-drying is a novel strategy that can be used to improve cryopreservation outcome by promoting rapid vitrification. © 2011 Chakraborty et al.

Recent evidence suggests that yawning is a thermoregulatory behavior. To explore this possibility further, the frequency of contagious yawning in humans was measured while outdoors in a desert climate in the United States during two distinct temperature ranges and seasons (winter: 22°C; early summer: 37°C). As predicted, the proportion of pedestrians who yawned in response to seeing pictures of people yawning differed significantly between the two conditions (winter: 45%; summer: 24%). Across conditions yawning occurred at lower ambient temperatures, and the tendency to yawn during each season was associated with the length of time spent outside prior to being tested. …

Nanotechnology has recently emerged as a strong contributor toward research efforts to develop targeted systems of drug delivery in cancer therapy. Our work investigates the therapeutic potential of Targeted Charge-Reversal Nanoparticles (TCRNs), a novel nanoparticle with in vitro evidence of nuclear drug delivery. Using M12 prostate cancer cells, MDA-MB-231 breast cancer cells, and modified derivatives of these cell lines, we investigated the ability of Folic Acid-tagged TCRNs to deliver Nile Red and Dimethyl Indole Redfluorescent (DiR) fluorescent dyes to the nucleus of cells using confocal microscopy and in vivo biphontonic imaging using Xenogen® Technology. Long-term changes in the state of the Bottom Shelf Water (BSW) on the Western shelf of the Black Sea are assessed using analysis of intra-seasonal and inter-annual temperature variations. For the purpose of this study the BSW is defined as such shelf water mass between the seabed and the upper mixed layer (bounded by the σθ = 14.2 isopycnal) which has limited ability to mix vertically with oxygen-rich surface waters during the warm season due to formation of a seasonal pycnocline. RNA contains over 100 post-transcriptional chemical modifications. Of these, the most abundant is pseudouridine (Ψ), the 5’ ribosyl isomer of uridine. The formation of Ψ occurs at the polynucleotide level, and is catalyzed by Ψ synthase enzymes. To date, five families of Ψ synthases have been identified. Our work deals with the fifth and last family to be discovered, the Ψ synthase TruD family. TruD homologs are present in organisms across the three kingdoms of life. A sequence alignment of TruD homologs reveals distinct sequence insertions at specific and conserved locations in homologs from higher organisms. We have carried out …

Understanding selection of breeding habitat is critical to conserving and restoring habitats for the Greater Sage-Grouse (Centrocercus urophasianus), particularly in xeric landscapes (≤25 cm annual precipitation). We monitored radio-marked female sage-grouse in south-central Wyoming in 2008 and 2009 to assess microhabitat use during nesting and brood rearing. For each model we grouped variables into three hypothesis sets on the basis of the weight of support from previous research (a priori information). We used binary logistic regression to compare habitat used by grouse to that at random locations and used an information-theoretic approach to identify the best-supported models. This study was done to examine the effect of choice on off task behaviors in high school students attending an alternative school. The participants were from various backgrounds, ability levels, and presence of disabilities. The students were observed performing three target behaviors; specifically talking off topic, being out of a seat needlessly, and being out of the classroom. This was initiated during a baseline observation period, and again over a five day period after being introduced to the choice process. The overall success of a brain computer interface (BCI) is largely dependent on the features used to make decisions. Noise in the electroencephalography (EEG) increases the difficulty of acquiring meaningful features. Previous literature suggests teaching subjects meditation and relaxation techniques may improve features relevant to BCI operation. The purpose of this study was to investigate performance on several cognitive protocols for both individuals who use meditation techniques and those who do not use these techniques. Both groups were given a motor imagery based BCI protocol, a P300 speller BCI, a verbal learning task, and an N-back test. No significant difference …
