Development Of Molecular And Cellular Imaging Tools To Evaluate Gene And Cell Based Therapeutic Strategies In Vivo, Jixiang Xia Jan 2011

Development Of Molecular And Cellular Imaging Tools To Evaluate Gene And Cell Based Therapeutic Strategies In Vivo, Jixiang Xia

Electronic Theses and Dissertations

Molecular imaging modalities are important tools to evaluate the efficacy of gene delivery systems and cell-based therapies. Development and application of these modalities will advance our understanding of the mechanism of transgene expression and cell fate and functions. Physical gene transfer methods hold many advantages over viral vectors among gene therapeutic strategies. Here, we evaluated the efficacy of biolistic (“gene gun”) gene targeting to tissues with non-invasive bioluminescence imaging (BLI) methods. Plasmids carrying the firefly luciferase reporter gene were transfected into mouse skin and liver using biolistics, and BLI was measured at various time points after transfer. With optimized DNA …

Electronic Theses and Dissertations

The objective of this research is to determine if a better understanding of the “molecule of life”, deoxyribonucleic acid or DNA can be obtained through Molecular Dynamics (MD) modeling and simulation (M&S) using contemporary MD M&S. It is difficult to overstate the significance of the DNA molecule. The now-completed Human Genome Project stands out as the most significant testimony yet to the importance of understanding DNA. The Human Genome Project (HGP) enumerated many areas of application of genomic research including molecular medicine, energy sources, environmental applications, agriculture and livestock breeding to name just a few. (Science, 2008) In addition to …

Transitions How Individuals In The Transgender Community Use Impression Management, Candice S. Campanaro Jan 2011

Transitions How Individuals In The Transgender Community Use Impression Management, Candice S. Campanaro

Electronic Theses and Dissertations

There exists an extensive body of literature focusing on impression management, with numerous studies addressing the effects of gender performance on members of the GLBT community. Despite a growing body of literature that centers on the differences between the genders, there exists a lack of research regarding the transgender community‟s ability to use impression management. This exploratory study addresses subjective experiences regarding transgender identity, and concentrates on self-definition and impression management in everyday life. It was found through interviews that “transition” is a temporary state that relies heavily on “passing” through impression management to create a new gender status. Once …

Framing Racial Inequality Reassessing The Effect Of Religion On Racial Attitudes, Jerrold C. Kaufman Ii Jan 2011

Framing Racial Inequality Reassessing The Effect Of Religion On Racial Attitudes, Jerrold C. Kaufman Ii

Electronic Theses and Dissertations

Building on previous work on racial attitudes among the religious, this study reassesses the effects of religion on individuals’ beliefs about racial inequality. This study relies on recent developments in the sociology of culture, which conceives of culture as a frame through which individuals interpret the world in which they inhabit (Benford and Snow 2000; Harding 2007; Small 2002, 2004). Religion is held to be an important social institution that provides substance to the frames that individuals employ for interpreting racial inequality. Two particular developments from this literature inform this study: first, that individuals can employ different, even contradictory, frames …

Mathematical Model To Assess The Relative Effectiveness Of Rift Valley Fever Countermeasures, Holly Gaff, Colleen Burgess, Jacqueline Jackson, Tianchan Niu, Yiannis Papelis, David Hartley

Biological Sciences Faculty Publications

Mathematical modeling of infectious diseases is increasingly used to explicate the mechanics of disease propagation, impact of controls, and sensitivity of countermeasures. The authors demonstrate use of a Rift Valley Fever (RVF) model to study efficacy of countermeasures to disease transmission parameters. RVF is a viral infectious disease that propagates through infected mosquitoes and primarily affects animals but also humans. Vaccines exist to protect against the disease but there is lack of data comparing efficacy of vaccination with alternative countermeasures such as managing mosquito population or destroying infected livestock. This paper presents a compartmentalized multispecies deterministic ordinary differential equation model …

Recovery From The Impact Of Light Reduction On The Seagrass Amphibolis Griffithii, Insights For Dredging Management, Kathryn Mcmahon, Paul Lavery, Michael Mulligan

Research outputs 2011

A large-scale, manipulative experiment was conducted to examine the extent and rate of recovery of meadows of the temperate Australian seagrass, Amphibolis griffithii to different light-reduction scenarios typical of dredging operations, and to identify potential indicators of recovery from light reduction stress. Shade cloth was used to mimic different intensities, durations and start times of light reduction, and then was removed to assess the recovery. The meadow could recover from 3 months of light stress (5–18% ambient) following 10 months re-exposure to ambient light, even when up to 72% of leaf biomass was lost, much faster recovery rates than has …

Conservation Genetics Of Sauger In The Upper Missouri River Drainage, Daniel Michael Bingham Jan 2011

Conservation Genetics Of Sauger In The Upper Missouri River Drainage, Daniel Michael Bingham

Graduate Student Theses, Dissertations, & Professional Papers

Hybridization with non-native walleye may play a substantial role in sauger declines throughout the upper Missouri River drainage of Montana and Wyoming. I identified 11 microsatellite loci to detect interspecific hybridization and describe the genetic population structure of sauger. Two major population groupings of sauger were revealed by principal component analysis. The first consisted of samples from the Missouri and lower Yellowstone River drainages, which showed no evidence for genetic divergence among each other. The second major grouping contained samples from the Bighorn River drainage and the upper Yellowstone River. Samples from the upper Bighorn River drainage were genetically distinct …

Effects Of Different Canopy Tree Species On Belowground Biogeochemistry In A Wet Lowland Tropical Forest, Adrienne Blair Keller Jan 2011

Effects Of Different Canopy Tree Species On Belowground Biogeochemistry In A Wet Lowland Tropical Forest, Adrienne Blair Keller

Graduate Student Theses, Dissertations, & Professional Papers

Tropical rain forests are known for their tremendous biological diversity, but the effects of plant diversity on important ecosystem processes remain unclear. Interspecies differences in both the demand for nutrients and in foliar nutrient concentrations could drive differences in litter chemistry that affect both pools and fluxes of belowground resources. Yet, our understanding of the effects of aboveground biogeochemical heterogeneity on belowground ecosystems is poor, especially in the species-rich forests of the wet tropics. To investigate the effects of tree species diversity on belowground biogeochemical processes, I examined how carbon (C), nitrogen (N), and phosphorus (P) cycles vary under canopy …

Essential Elements Of Ecological Literacy And The Pathways To Achieve It: Perspectives Of Ecologists, Brooke Baldauf Mcbride

Graduate Student Theses, Dissertations, & Professional Papers

National assessments have led many to conclude that the level of ecological literacy among the general population in the United States is too low to enable effective social responses to current environmental challenges. However, the actual meaning of ecological literacy varies considerably between academic fields and has been a topic of intensive deliberation for several decades. Within the field of ecology in particular, a driving purpose behind this ongoing discussion has been to advance a complete, pedagogy-guiding, and broadly applicable framework for ecological literacy, allowing for the establishment of guidelines and tools for assessing educational achievement; yet, a widely accepted …

Heterogeneity Of Stand Structure, Fuels, And Wildfire Hazard In Dry And Moist Old-Growth Ponderosa Pine Forests: Case Studies From Western Montana, Kyle Crosby Stover Jan 2011

Heterogeneity Of Stand Structure, Fuels, And Wildfire Hazard In Dry And Moist Old-Growth Ponderosa Pine Forests: Case Studies From Western Montana, Kyle Crosby Stover

Graduate Student Theses, Dissertations, & Professional Papers

Silvicultural restoration treatments are increasingly proposed to restore ecological function and remediate catastrophic wildfire hazards in old-growth ponderosa pine stands throughout the western United States. However, stand heterogeneity has been inadequately addressed in restoration and fuel treatment planning. In response, we analyzed two old-growth ponderosa pine forest restoration projects in western Montana with different moisture regimes. We utilized available fixed-area plot data to describe heterogeneity by analyzing the distributions of current and anticipated post-treatment plot conditions, and contrasted these with stand-average depictions. The study’s findings illustrate that distributions of overstory structure, fuels, and modeled fire hazards are typically non-normal, skewed, …

Interactive Effects Of Wildfire And Disturbance History On Amphibians And Their Parasites, Blake Roger Hossack Jan 2011

Interactive Effects Of Wildfire And Disturbance History On Amphibians And Their Parasites, Blake Roger Hossack

Graduate Student Theses, Dissertations, & Professional Papers

Climate-driven changes in wildfire and other disturbance regimes are expected to affect populations and communities worldwide. Understanding how these changes will affect native species is critical for future conservation efforts, especially on managed forests. Using data from several wildfires that burned between 1988 and 2003 in and next to Glacier National Park, Montana, I examined how fire affected the distribution, abundance, and infection status of 3 native amphibians. In Chapter 1, I used long-term data on wetland occupancy to show the long-toed salamander (Ambystoma macrodactylum) and Columbia spotted frog (Rana luteiventris) were resistant to change during the first 6 years …

Modeling Risk Of Hunting Pressure On Large Mammals In Northwestern Argentina, Loretta Kathleen Baker Jan 2011

Modeling Risk Of Hunting Pressure On Large Mammals In Northwestern Argentina, Loretta Kathleen Baker

Graduate Student Theses, Dissertations, & Professional Papers

The subtropical Yungas and Chaco forests of northwestern Argentina are two of the most biodiverse and threatened biomes in South America. It is unclear how development pressure and increased human presence may be affecting wildlife in this increasingly fragmented and degraded landscape. We initiated a broad-scale analysis of the spatial distribution and magnitude of anthropogenic factors that may influence large mammal mortality due to potential human hunting pressure in a landscape linkage connecting these threatened forests. We conducted a literature review of Neotropical study sites that reported hunting of large mammals by indigenous people or colonists, and used this information …

Opposition And Barriers To Conservation Easements: Insights From Montana Landowners, Alayna S. Dupont Jan 2011

Opposition And Barriers To Conservation Easements: Insights From Montana Landowners, Alayna S. Dupont

Graduate Student Theses, Dissertations, & Professional Papers

A conservation easement (CE) is a voluntary agreement that perpetually limits development on private land in order to protect open space, agricultural land, and scenic amenities, as well as ecological or historic resources. Due to the fact that CEs are voluntary, an understanding of the landowner’s decision about whether or not to participate in conservation easement programs is imperative if CE use is to continue and expand. Moreover, understanding how non-participating landowners view CEs, how they regard the incentives, and how CEs fit or do not fit with their ideas regarding private property and conservation is of critical importance for …

Predicting Amphibian Occurrence Based On Wetland And Landscape Level Factors In Montana, Catherine Anne Mcintyre Jan 2011

Predicting Amphibian Occurrence Based On Wetland And Landscape Level Factors In Montana, Catherine Anne Mcintyre

Graduate Student Theses, Dissertations, & Professional Papers

Amphibians have a complex life history that requires a mosaic of habitats, including breeding, foraging, and over-wintering areas. Historically, regulators have focused on wetland breeding habitat quality to explain amphibian presence. Recently, other habitat requirements including landscape level factors have been examined. Data collected from amphibian surveys in Montana were used to determine if wetland quality factors or landscape level factors were better determinants of amphibian occurrence at breeding sites. Twenty-six habitat models were constructed a priori for eight species of amphibians in Montana. This included five models containing parameters associated with wetland quality, ten landscape level models, and ten …

Running Economy: Improvements In Physiological Efficiency Attained Through Changes In Muscle Structural Morphology, Joseph Kenneth Pellegrino Jan 2011

Running Economy: Improvements In Physiological Efficiency Attained Through Changes In Muscle Structural Morphology, Joseph Kenneth Pellegrino

Graduate Student Theses, Dissertations, & Professional Papers

Running Economy (RE) is a crucial determinant for running performance. While strategies for improving RE have been determined, the mechanisms governing this phenomenon have eluded the scientific community. My objective was to determine what adaptations, physiological, morphological, or otherwise, occur to bring about the altered RE associated with plyometric training. Specifically this project was designed to examine whether measureable transformations in muscle protein isoform makeup brought on through specific training will result in better RE in moderately trained runners. Participants (n=25) were placed into either a plyometrics-training or control group. All participants underwent similar testing before and after the 6-week …

Salud, Dignidad, Justicia: Articulating "Choice" And "Reproductive Justice" For Latinas In The United States, Kathleen Maire De Onis Jan 2011

Salud, Dignidad, Justicia: Articulating "Choice" And "Reproductive Justice" For Latinas In The United States, Kathleen Maire De Onis

Graduate Student Theses, Dissertations, & Professional Papers

This investigation explores the rhetorical evolution of the National Latina Institute for Reproductive Health. The author argues that the rhetors of the reviewed texts reveal the organization’s dual goals of both altering and resonating with the values and attitudes of Latinas regarding their concerns, as well as the necessity of combating systemic barriers and oppressive dominant ideology hindering Latinas’ ability to exercise a full range of choices. Although this two-fold purpose is persistent throughout the artifacts, as the Latina Institute embraces the language of reproductive justice, the agency gradually shifts its rhetorical efforts from addressing internal issues directed toward its …

Specifying The Links Between Adolescent Substance Use And Traditional Delinquency Theories: A Negative Bionomial Approach, Anthony James Feuchter

Graduate Student Theses, Dissertations, & Professional Papers

The consumption of alcohol and illicit drugs among adolescents has been a growing concern in sociology and related disciplines (Hayes-Smith and Whaley 2009; Embry, Hankins, Biglan, and Boles 2009; Herman-Stahl, Krebs, Kroutil, Heller 2007). The societal and financial effects of substance use in Montana have been detrimental. For example, methamphetamine use accounted for 33 percent of children in foster care and over 50 percent of children were in foster care for drug-related reasons in 2006 (Methamphetamine in Montana 2008). Also, Bryant, Schulenberg, O’Malley, Bachman, and Johnston (2003) found consistent patterns in the prior literature that high truancy rates and low …
