Characterisation Of The Metacaspase Gene Family In Arabidopsis Thaliana, Paige N. Cox

Caspases are known to be involved in animal programmed cell death (PCD). The objective of this thesis was to use gene expression analysis and reverse genetics to determine if Arabidopsis metacaspase (AtMC) genes play a role in plant PCD. The majority of AtMC genes were found to be expressed nearly constitutively in various tissues, developmental stages, and under various inductive treatments. Transgenic Arabidopsis plants generated with AtMCpromoter::AtMCgene::GUS fusions showed expression of the reporter gene in leaves, vasculature, trichomes, siliques, anthers, and during embryo development. Preliminary phenotypic characterization of single and double Arabidopsis AtMC loss-of-function mutants suggested that the …

Skeletal muscle force evaluation is difficult to implement in a clinical setting. Muscle force is typically assessed through either manual muscle testing, isokinetic/isometric dynamometry, or electromyography (EMG). Manual muscle testing is a subjective evaluation of a patient’s ability to move voluntarily against gravity and to resist force applied by an examiner. Muscle testing using dynamometers adds accuracy by quantifying functional mechanical output of a limb. However, like manual muscle testing, dynamometry only provides estimates of the joint moment. EMG quantifies neuromuscular activation signals of individual muscles, and is used to infer muscle function. Despite the abundance of work performed to …

The invasive fruit fly Bactrocera invadens (Drew, Tsurata & White 2005) is causing extensive socio-economic losses in West Africa as a pest in mango production. In addition to reducing crop quality and decreasing fruit production, the presence of the fruit fly has detrimental impact on the export market due to international trade restrictions. To manage the damage being caused by B.invadens to the mango industry in Africa, an Integrated Pest Management (IPM) strategy with biological control as one of its components was initiated by the African Fruit Fly Programme (AFFP) based at ICIPE. The horticultural sub-sector in Kenya is a key source of foreign exchange earnings, employment creation and also plays a major role in ensuring food security to many Kenyan households. Longitudinal entomological sampling was executed to study physiological and ecological adaptations of malaria vectors at two semi-arid villages (Kamarimar and Tirion) in Kenya. Adult mosquitoes were sampled using CDC light traps and pyrethrum spray catch collections. Mosquito larvae were sampled using standard dipping methods. Parity rates, length of gonotophic cycle duration and insemination rates of female mosquitoes were assessed. Host choice and sporozoite rates were determined by ELISAs. Sub-species of the Anopheles gambiae complex were identified by PCR. Tsetse flies use olfaction in search for food, mates and larviposition sites. Olfactory proteins [(odorant binding proteins (OBPs), pheromone binding proteins (PBPs), chemosensory protein (CSPs), odorant degrading enzymes (ODEs) and odorant receptors (Ors)], located within the antennae, play key role in this process. In this work, presence of olfactory proteins was investigated by constructing and sequencing cDNA libraries from Glossina pallidipes Austen, antennae; Glossina palpalis gambiensis Vanderplank, head and Glossina tachinoides Westwood, head. This study focused on the development of practical agro-ecosystem measures aimed at enhancing agricultural productivity while simultaneously reducing malaria vector populations for improved household incomes in irrigated rice farming. The main strategy entailed testing alternation of rice with legumes. Soya bean (Glycine max L.JMarr) and Sesbania sesban were tested as the preceding crops against the conventional rice-fallow and the rice-rice cultivation systems. Among the preceding crops, rice produced the highest amount of biomass followed by Soya bean varieties EAI 3600 and Duiker. However, Soya bean residue had a higher nutrient content than rice straw and fallow treatment (weeds). The fallow-rice …
