A New Emoia Samoensis Group Lizard (Squamata: Scincidae) From The Cook Islands, South-Central Pacific, George R. Zug, Alison M. Hamilton, Christopher C. Austin

Faculty Publications

The first published report of a large treeskink from Rarotonga, Cook Islands, appeared in 1988. The first museum voucher specimen was collected in 1984. Although this skink seems likely to be a recent arrival to the island of Rarotonga, it represents a unique member of the Emoia samoensis species group. We compare this population with other members of the E. samoensis group and describe the population as Emoia tuitarere n. sp., distinguished by a suite of external characters including SVL, number of dorsal scale rows, and number of subdigital lamellae of the fourth toe. We provide preliminary definitions for the …

Rho Activation Of Mdia Formins Is Modulated By An Interaction With Inverted Formin 2 (Inf2), Hua Sun, Johannes S. Schlondorff, Elizabeth J. Brown, Henry N. Higgs, Martin R. Pollak

Dartmouth Scholarship

Inverted formin 2 (INF2) encodes a member of the diaphanous subfamily of formin proteins. Mutations in INF2 cause human kidney disease characterized by focal and segmental glomerulosclerosis. Disease-causing mutations occur only in the diaphanous inhibitory domain (DID), suggesting specific roles for this domain in the pathogenesis of disease. In a yeast two-hybrid screen, we identified the diaphanous autoregulatory domains (DADs) of the mammalian diaphanous-related formins (mDias) mDia1, mDia2, and mDia 3 as INF2_DID-interacting partners. The mDias are Rho family effectors that regulate actin dynamics. We confirmed in vitro INF2_DID/mDia_DAD binding by biochemical assays, confirmed the in vivo interaction of these …

Linkage Disequilibrium Based Genotype Calling From Low-Coverage Shotgun Sequencing Reads, Jorge Duitama, Justin Kennedy, Sanjiv Dinakar, Yözen Hernández, Yufeng Wu, Ion I. Măndoiu

Publications and Research

Background Recent technology advances have enabled sequencing of individual genomes, promising to revolutionize biomedical research. However, deep sequencing remains more expensive than microarrays for performing whole-genome SNP genotyping. Results In this paper we introduce a new multi-locus statistical model and computationally efficient genotype calling algorithms that integrate shotgun sequencing data with linkage disequilibrium (LD) information extracted from reference population panels such as Hapmap or the 1000 genomes project. Experiments on publicly available 454, Illumina, and ABI SOLiD sequencing datasets suggest that integration of LD information results in genotype calling accuracy comparable to that of microarray platforms from sequencing data of …

Real-Time Dynamic Imaging Of Virus Distribution In Vivo, Sean E. Hofherr, Kristen E. Adams, Christopher Y. Chen, Shannon May, Eric A. Weaver, Michael A. Barry Feb 2011

Real-Time Dynamic Imaging Of Virus Distribution In Vivo, Sean E. Hofherr, Kristen E. Adams, Christopher Y. Chen, Shannon May, Eric A. Weaver, Michael A. Barry

School of Biological Sciences: Faculty Publications

The distribution of viruses and gene therapy vectors is difficult to assess in a living organism. For instance, trafficking in murine models can usually only be assessed after sacrificing the animal for tissue sectioning or extraction. These assays are laborious requiring whole animal sectioning to ascertain tissue localization. They also obviate the ability to perform longitudinal or kinetic studies in one animal. To track viruses after systemic infection, we have labeled adenoviruses with a near-infrared (NIR) fluorophore and imaged these after intravenous injection in mice. Imaging was able to track and quantitate virus particles entering the jugular vein simultaneous with …

Computing Multiscale Entropy With Orthogonal Range Search, Yu-Hsiang Pan, Wei-Yen Lin, Yung-Hung Wang, Kuo-Tien Lee

Journal of Marine Science and Technology–Taiwan

Multi-scale entropy (MSE) is a measurement of a system's complexity. It has received a great deal of attention in recent years, and its effectiveness has been verified, and applied in a number of different fields. However, the algorithms proposed in past studies required O(N2 ), which represented a degree of execution time insufficient for on-line applications, or for applications with long-term correlations. In this study, we showed that the probability function in the entropy term could be transformed into an orthogonal range search in the field of computational geometry. We then developed an efficient new algorithm for computing multi-scale entropy. …

Engaging Chicago Residents In Climate Change Action: Results From Rapid Ethnographic Inquiry, Lynne M. Westphal, Jennifer L. Hirsch Feb 2011

Engaging Chicago Residents In Climate Change Action: Results From Rapid Ethnographic Inquiry, Lynne M. Westphal, Jennifer L. Hirsch

Cities and the Environment (CATE)

Addressing climate change requires action at all levels of society, from neighborhood to international levels. Using Rapid Ethnography rooted in Asset Based Community Development theory, we investigated climate-friendly attitudes and behaviors in two Chicago neighborhoods in order to assist the City with implementation of its Climate Action Plan. Our research suggests a means to effectively engage urban residents at the household and neighborhood level: understand the issues of importance in each neighborhood, assess the ways these are related to climate change mitigation or adaptation actions, and engage residents from this perspective, meeting mutually compatible goals. This builds upon the concept …

Animal Welfare In Multipurpose Cattle Production Systems And Its Implications On Beef Quality, Saymore P. Ndou, Voster Muchenje, Michael Chimonyo Feb 2011

Animal Welfare In Multipurpose Cattle Production Systems And Its Implications On Beef Quality, Saymore P. Ndou, Voster Muchenje, Michael Chimonyo

Farm Animal Welfare Collection

Animal welfare and its influence on beef production are major considerations in many developed countries. In the developing world, where food insecurity and poverty are prevalent, the welfare of animals receives low priority due to factors such as traditional customs and beliefs, lack of knowledge in animal handling and sub standard handling facilities. This is worsened by the fact that cattle are used for several purposes, such as meat production, milk production, draught power and traditional ceremonies. Research on animal welfare and how it relates to animal production have been done in the developed world. The objective of this study …

Increasing Liana Abundance And Biomass In Tropical Forests: Emerging Patterns And Putative Mechanisms, Stefan A. Schnitzer, Frans Bongers Feb 2011

Increasing Liana Abundance And Biomass In Tropical Forests: Emerging Patterns And Putative Mechanisms, Stefan A. Schnitzer, Frans Bongers

Biological Sciences Faculty Research and Publications

Tropical forests are experiencing large‐scale structural changes, the most apparent of which may be the increase in liana (woody vine) abundance and biomass. Lianas permeate most lowland tropical forests, where they can have a huge effect on tree diversity, recruitment, growth and survival, which, in turn, can alter tree community composition, carbon storage and carbon, nutrient and water fluxes. Consequently, increasing liana abundance and biomass have potentially profound ramifications for tropical forest composition and functioning. Currently, eight studies support the pattern of increasing liana abundance and biomass in American tropical and subtropical forests, whereas two studies, both from Africa, do …

Fabrication Of Covalently Crosslinked And Amine-Reactive Microcapsules By Reactive Layer-By-Layer Assembly Of Azlactone-Containing Polymer Multilayers On Sacrificial Microparticle Templates, Eric M. Saurer, Ryan M. Flessner, Maren E. Buck, David M. Lynn

Chemistry: Faculty Publications

We report on the fabrication of covalently crosslinked and amine-reactive hollow microcapsules using 'reactive' layer-by-layer assembly to deposit thin polymer films on sacrificial microparticle templates. Our approach is based on the alternating deposition of layers of a synthetic polyamine and a polymer containing reactive azlactone functionality. Multilayered films composed of branched poly(ethylene imine) (BPEI) and poly(2-vinyl-4,4-dimethylazlactone) (PVDMA) were fabricated layer-by-layer on the surfaces of calcium carbonate and glass microparticle templates. After fabrication, these films contained residual azlactone functionality that was accessible for reaction with amine-containing molecules. Dissolution of the calcium carbonate or glass cores using aqueous ethylenediaminetetraacetic acid (EDTA) or …

Genetically Engineered Alginate Lyase-Peg Conjugates Exhibit Enhanced Catalytic Function And Reduced Immunoreactivity, John W. Lamppa, Margaret E. Ackerman, Jennifer I. Lai, Thomas C. Scanlon, Karl E. Griswold

Dartmouth Scholarship

Alginate lyase enzymes represent prospective biotherapeutic agents for treating bacterial infections, particularly in the cystic fibrosis airway. To effectively deimmunize one therapeutic candidate while maintaining high level catalytic proficiency, a combined genetic engineering-PEGylation strategy was implemented. Rationally designed, site-specific PEGylation variants were constructed by orthogonal maleimide-thiol coupling chemistry. In contrast to random PEGylation of the enzyme by NHS-ester mediated chemistry, controlled mono-PEGylation of A1-III alginate lyase produced a conjugate that maintained wild type levels of activity towards a model substrate. Significantly, the PEGylated variant exhibited enhanced solution phase kinetics with bacterial alginate, the ultimate therapeutic target. The immunoreactivity of the …
