Extended Interferon-Alpha Therapy Accelerates Telomere Length Loss In Human Peripheral Blood T Lymphocytes, Joel M. O'Bryan, James A. Potts, Herbert L. Bonkovsky, Anuja Mathew, Alan L. Rothman

Institute for Immunology and Informatics Faculty Publications

Background:

Type I interferons have pleiotropic effects on host cells, including inhibiting telomerase in lymphocytes and antiviral activity. We tested the hypothesis that long-term interferon treatment would result in significant reduction in average telomere length in peripheral blood T lymphocytes.

Methods/Principal Findings:

Using a flow cytometry-based telomere length assay on peripheral blood mononuclear cell samples from the Hepatitis-C Antiviral Long-term Treatment against Cirrhosis (HALT-C) study, we measured T cell telomere lengths at screening and at months 21 and 45 in 29 Hepatitis-C virus infected subjects. These subjects had failed to achieve a sustained virologic response following 24 weeks of pegylated-interferon-alpha …

The Characterization Of Avian Polyomavirus, Satellite Tobacco Mosaic Virus, And Bacteriophage Cw02 By Means Of Cryogenic Electron Microscopy, Peter S. Shen

Theses and Dissertations

Viruses are the most abundant biological entity in the biosphere and are known to infect hosts from all domains of life. The aim of my work is to identify conserved and non-conserved features among the capsid structures of related and divergent icosahedral viruses via cryogenic electron microscopy, sequence analysis, molecular modeling, and other techniques. Bird polyomaviruses often cause severe disease in their hosts whereas mammalian polyomaviruses generally do not. Avian polyomavirus is a type of bird polyomavirus with an unusually broad host range compared to the restricted tropism of other polyomaviruses. Although most polyomaviruses have a conserved, rigid capsid protein …

Spite And The Scale Of Competition In Pseudomonas Aeruginosa, R. Inglis, Patrick Roberts, Andy Gardner, Angus Buckling Aug 2011

Spite And The Scale Of Competition In Pseudomonas Aeruginosa, R. Inglis, Patrick Roberts, Andy Gardner, Angus Buckling

Biology Department Faculty Works

Scale of competition has been shown to be an important factor in shaping the evolution of social interactions. Although many theoretical and experimental studies have examined its effect on altruistic cooperation, relatively little research effort has been focused on spiteful behaviors—actions that harm both the actor and the recipient. In this study, we expand on existing theory by investigating the importance of the global frequency of spiteful alleles, and we determine experimentally how the scale of competition affects selection for spite in the bacterial pathogen Pseudomonas aeruginosa under high and intermediate spatial relatedness. Consistent with our theoretical results, we found …

Inhibition And Mechanism Of The 1,4-Dihydroxy-2-Naphthoyl-Coa Synthase (Menb) From Mycobacterium Tuberculosis, Xiaokai Li Aug 2011

Inhibition And Mechanism Of The 1,4-Dihydroxy-2-Naphthoyl-Coa Synthase (Menb) From Mycobacterium Tuberculosis, Xiaokai Li

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)

Tuberculosis is a common, deadly, and contagious disease that is caused by the Gram positive bacterium Mycobacterium tuberculosis. In order to better treat TB, novel antibiotics with shorter treatment periods and improved activity against drug-resistant and latent TB infections are urgently needed. The redox cofactor menaquinone is an essential component of the electron transport chain in M. tuberculosis and enzymes in the biosynthetic pathway for this cofactor are promising targets for anti-TB drug discovery. 1,4-Dihydroxy-2-naphthoyl-CoA synthase (MenB), is the sixth enzyme in the menaquinone biosynthesis pathway, and catalyzes the formation of DHNA-CoA from o-succinylbenzoate-CoA via Claisen condensation. In order to …

To Get The Shot Or Not: Narratives, Rhetoric, And The Childhood Vaccination Crisis, Katherine M. Hurley Aug 2011

To Get The Shot Or Not: Narratives, Rhetoric, And The Childhood Vaccination Crisis, Katherine M. Hurley

Electronic Theses and Dissertations

Parents in developed countries like the United States are questioning the need for and safety of childhood vaccinations. Incidences of disease have risen as fewer parents have vaccinated their children. Perhaps the most significant public figure to reinforce the choices of parents not-to-vaccinate is Jenny McCarthy, whose best-selling book details her theory about the cause of, and cure for, her son‘s autism. As I demonstrate, the study of the narratives is vital for understanding the vaccination crisis, not the least because of the extent to which McCarthy‘s (2007) story has echoed through parenting communities. I examine whether chosen anti- and …

Exploring Protein-Protein And Protein-Ligand Interactions In The Bacterial Fatty Acid Biosynthesis Pathway, Janine Borgaro Aug 2011

Exploring Protein-Protein And Protein-Ligand Interactions In The Bacterial Fatty Acid Biosynthesis Pathway, Janine Borgaro

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)

The bacterial fatty acid biosynthesis (FASII) pathway is a promising target for antibacterial drug discovery and current research is focused on elucidating the substrate specificity of the b-ketoacyl-ACP synthase (KAS) enzymes in this pathway, identify the interactions between the components of the FASII pathway and discovering the unknown dehydratase, and studying the interaction of current enzyme inhibitors with FASII components in whole cells. Substrates are shuttled between the FASII enzymes by acyl carrier protein, a phosphopantetheinylated protein with a MW of 13 kDa in Mycobacterium tuberculosis and 7 kDa in Escherichia coli. While the KAS enzyme(s) involved in fatty acid …

Mechanism And Inhibition Studies Of Enoyl-Acp Reductases And Dihydroxynaphthoyl-Coa Synthase In Pathogenic Bacteria, Nina Liu Aug 2011

Mechanism And Inhibition Studies Of Enoyl-Acp Reductases And Dihydroxynaphthoyl-Coa Synthase In Pathogenic Bacteria, Nina Liu

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)

Resistance to existing antimicrobial agents is a global threat to human health care, and new drugs with novel mechanisms of action are required in order to keep pace with the emergence of drug resistant pathogens. The bacterial fatty acid biosynthesis (FAS-II) and menaquinone pathways represent attractive yet relatively unexploited targets for new antibiotic development and consequently there is significant interest in developing potent inhibitors of enzymes in these two pathways. My research is mainly focused on the mechanism and inhibition of the enoyl-ACP reductases from the FAS-II pathway and dihydroxynaphthoyl-CoA synthase from the menaquinone pathway. The mechanism and inhibition of …

Department of Emergency Medicine

NO is involved in normal kidney function and perturbed in acute kidney injury (AKI). We hypothesized that urinary concentration of NO metabolites, nitrite, and nitrate would be lower in children with early AKI presenting to the emergency department (ED), when serum creatinine (SCr) was uninformative. Patients up to 19 y were recruited if they had a urinalysis and SCr obtained for routine care. Primary outcome, AKI, was defined by pediatric Risk, Injury, Failure, Loss of function, End-stage renal disease (pRIFLE) criteria. Urinary nitrite and nitrate were determined by HPLC. A total of 252 patients were enrolled, the majority (93%) of …

Altered Toll-Like Receptor 2-Mediated Endotoxin Tolerance Is Related To Diminished Interferon Β Production, Ss Zaric, Wa Coulter, Ce Shelburne, Cr Fulton, Et Al Aug 2011

Altered Toll-Like Receptor 2-Mediated Endotoxin Tolerance Is Related To Diminished Interferon Β Production, Ss Zaric, Wa Coulter, Ce Shelburne, Cr Fulton, Et Al

Peninsula Dental School

Induction of endotoxin tolerance leads to a reduced inflammatory response after repeated challenge by LPS and is important for resolution of inflammation and prevention of tissue damage. Enterobacterial LPS is recognized by the TLR4 signaling complex, whereas LPS of some non-enterobacterial organisms is capable of signaling independently of TLR4 utilizing TLR2-mediated signal transduction instead. In this study we report that Porphyromonas gingivalis LPS, a TLR2 agonist, fails to induce a fully endotoxin tolerant state in a human monocytic cell line (THP-1) and mouse bone marrow-derived macrophages. In contrast to significantly decreased production of human IL-8 and TNF-α and, in mice, …

This case study, utilizing ethnographic tools of observation, interviews, and investigation of artifacts, examines educators' perceptions of the role of the school in the prevention of this epidemic, how perceptions affect practices, and the barriers to prevention efforts. The lens of caring theory, social justice, and critical theory frame the analysis of how educators in a medium-sized elementary school contend with the problem of childhood obesity. Educators in this setting perceived their role in the prevention …

Investigating Nitrate-Dependent Humic Substance Oxidation And In-Service K-12 Teachers' Understanding Of Microbiology, Nastassia N. Jones Aug 2011

Investigating Nitrate-Dependent Humic Substance Oxidation And In-Service K-12 Teachers' Understanding Of Microbiology, Nastassia N. Jones

Dissertations

Humic substances (HS) are the humified portions of totally decomposed soil organic matter that are ubiquitous in nature. Although these substances have been studied for more than 200 years, neither their metabolic capabilities nor a specific chemical structure has yet to be determined. HS have been studied as a carbon source in many environments where they are degraded; however, previous studies have shown that some microorganisms are capable of utilizing humic substances as electron acceptors and electron donors in anaerobic respiration. Even though there have been humic-reducing and humic-oxidizing microorganisms isolated and studied in recent years, the mechanism of humics …

School of Natural Resources: Dissertations, Theses, and Student Research

Highly-pathogenic avian influenza virus (HPAIV H5N1) poses risks to wild birds, poultry, and humans. Personnel with the United States Department of Agriculture-Animal Plant Health Inspection Service-Wildlife Services, state, and tribal wildlife agencies collected 168,940 samples from migratory birds from 2007 to 2009 to test for presence of HPAIV H5N1. No HPAIV was found, but other subtypes were discovered, including H5 and H7. I estimated prevalence of avian influenza virus by flyway and found prevalence was lowest each year in the Atlantic Flyway (6.7%–8.3%), highest in the Pacific Flyway in 2007 (13.3%) and 2008 (13.4%), and highest in the Mississippi Flyway …

Primary Provider Beliefs And Practice Patterns Contribute To The Lack Of Hiv/Hcv Coinfected Patients Undergoing Liver Transplantation In The United States, Jonathan M. Fenkel, Md, Victor J. Navarro, Md Aug 2011

Primary Provider Beliefs And Practice Patterns Contribute To The Lack Of Hiv/Hcv Coinfected Patients Undergoing Liver Transplantation In The United States, Jonathan M. Fenkel, Md, Victor J. Navarro, Md

Division of Gastroenterology and Hepatology Faculty Papers

Conclusions:

1. The majority of primary providers were ambivalent toward or against LT for HIV/HCV coninfected patients.

2. Half of all respondents were unlikely to refer cirrhotic coinfected patients for LT evaluation.

3. HIV specialists were significantly more likely to believe transplant should be offered, but reported no difference in likelihood of LT referral.

4. These findings suggest that primary provider beliefs and self-reported practice patterns may partially explain the paucity of coinfected US liver transplant recipients.

Assessment Of Adherence To Guidelines For Hepatocellular Carcinoma Screening In Hiv/Hcv Coinfected Patients, Jonathan M. Fenkel, Md, Victor J. Navarro, Md Aug 2011

Assessment Of Adherence To Guidelines For Hepatocellular Carcinoma Screening In Hiv/Hcv Coinfected Patients, Jonathan M. Fenkel, Md, Victor J. Navarro, Md

Division of Gastroenterology and Hepatology Faculty Papers

Conclusions:

1. Self-reported adherence with published guidelines for HCC screening is poor among primary providers for HIV/HCV coinfected patients, including HIV specialists and University-based providers.

2. Unnecessary imaging is also frequently ordered on non-cirrhotics, particularly by University-based providers.

3. Improved adherence to guidelines is needed among primary providers as over 50% of HCC's may be missed, and many patients many not be referred for subspecialty GI or Liver care, where screening practices may differ.

Synthesis Of A Chemical Probe For Identification Of Ger Receptor Proteins Of Bacillus Anthracis, Hyelee Lee Aug 2011

Synthesis Of A Chemical Probe For Identification Of Ger Receptor Proteins Of Bacillus Anthracis, Hyelee Lee

UNLV Theses, Dissertations, Professional Papers, and Capstones

Bacillus anthracis,a gram positive, endospore-forming, rod-shaped, aerobic bacterium, is the causative agent of anthrax. A key step in the pathogenic cycle is the spore germination, where the dormant spore transforms into a metabolically active cell. This allows B. anthracis to proliferate and secrete toxins. It is believed that spore germination is triggered when small molecules, known as germinants, are recognized by germination (Ger) receptor proteins located in the inner membrane of spores. Most Ger proteins are encoded by tricistronic operons, resulting in three distinct products, the A-, B-, C-subunits. Genomic sequencing showed that B. anthracis has seven ger receptor …

Gambling With Lives: A History Of Occupational Health In Southern Nevada, 1905--2010, Michelle Ann Turk

UNLV Theses, Dissertations, Professional Papers, and Capstones

In April 2009, the Pulitzer committee awarded its public service prize to the Las Vegas Sun for its coverage of the high fatalities on Las Vegas Strip construction sites. The newspaper attributed failures in safety policy to "the exponential growth in the Las Vegas market." In fact, since Las Vegas' founding in 1905, rapid development in the region has always strained occupational health standards. From transporting hazardous railroad cargoes to building Hoover Dam, chemical processing at Basic Magnesium, nuclear testing, and dense megaresort construction on the Strip, workers, residents, and tourists alike have been exposed to the threat of living …

Determining The Roles Of Dendritic Cells And Icam-1 In The Transpresentation Of Il-15 To Cd8 T Cells, Spencer W. Stonier Aug 2011

Determining The Roles Of Dendritic Cells And Icam-1 In The Transpresentation Of Il-15 To Cd8 T Cells, Spencer W. Stonier

Dissertations and Theses (Open Access)

The maintenance and generation of memory CD8 T cells is dependent on the cytokine IL-15. IL-15 is delivered by a novel mechanism termed transpresentation: IL-15 is presented by a cell expressing IL-15Ralpha to the CD8 T cell which responds via IL-2Rbeta/gammac. The identity of what cells transpresent IL-15 to support the survival and homeostatic proliferation of memory CD8 T cells is unknown. Using a transgenic mouse model that limits IL-15 transpresentation to DCs, I have demonstrated that DCs transpresent IL-15 to CD8 T cells. DCs transpresent IL-15 to CD8 T cells during the contraction of an immune response …
