Flow cytometry (FCM) and computer-assisted sperm motion analysis (CASA) methods were developed and validated for use with endangered razorback suckers Xyrauchen texanus collected (nZ64) during the 2006 spawning season. Sperm motility could be activated within osmolality ranges noted during milt collections (here 167-343 mOsm/kg). We hypothesized that sperm quality of milt collected into isoosmotic (302 mOsm/kg) or hyperosmotic (500 mOsm/kg) Hanks' balanced salt solution would not differ. Pre-freeze viabilities were similar between osmolalities (79%G6 (S.E.M.) and 76%G7); however, post-thaw values were greater in hyperosmotic buffer (27%G3 and 12%G2; PZ0.0065), as was mitochondrial membrane potential (33%G4 and 13%G2; PZ0.0048). Visual estimates …

BACKGROUND: Plasmids containing hylEfm (pHylEfm) were previously shown to increase gastrointestinal colonization and lethality of Enterococcus faecium in experimental peritonitis. The hylEfm gene, predicting a glycosyl hydrolase, has been considered as a virulence determinant of hospital-associated E. faecium, although its direct contribution to virulence has not been investigated. Here, we constructed mutants of the hylEfm-region and we evaluated their effect on virulence using a murine peritonitis model.

RESULTS: Five mutants of the hylEfm-region of pHylEfmTX16 from the sequenced endocarditis strain (TX16 [DO]) were obtained using an adaptation of the PheS* system and were evaluated in a commensal strain TX1330RF to …

Modeling Infectious Diseases : Two Strain Diseases In Metapopulations, Daniel Rossi, Sydney Philipps, Rachel Vonarb Jan 2011

Modeling Infectious Diseases : Two Strain Diseases In Metapopulations, Daniel Rossi, Sydney Philipps, Rachel Vonarb

Symposium on Research and Creative Expression (SORCE)

Infectious diseases often mutate and are carried between regions. We consider a mathematical model which begins to account for these factors. We assume two disjoint populations that only occasionally comingle, and two strains of a disease present in these populations. Of interest are the equations describing the dynamics of this system, the conditions under which epidemics will occur, and the long term behavior of the system under various initial conditions. We find that in many ways this system is similar to a simpler one-population model. However, we find evidence that there may be conditions under which both disease strains can …

Epidemiology Of Blood-Borne Infections Among Georgian Health Care Workers, Maia Butsashvili Jan 2011

Epidemiology Of Blood-Borne Infections Among Georgian Health Care Workers, Maia Butsashvili

Legacy Theses & Dissertations (2009 - 2024)

INTRO: The prevalence of hepatitis in central Asia is considered high (>8% endemnicity) by the World Health Organization. Universal precautions among healthcare workers (HCWs) and newborn vaccination programs are important public health interventions to reduce the spread of hepatitis and other blood borne pathogens. This comprehensive evaluation of the risk of blood borne pathogen exposure among HCWs in the Republic of Georgia measures the following: 1) nosocomial risk events; 2) prevalence of HIV, HBV and HCV among Georgian HCWs; 3) predictive factors for HCWs recommending HBV vaccination for children; 4) prevalence of and willingness to report behavioral risk factors …

Gene And Environment Interactions Modulate Immune System Influences On Social Behavior, Yubin Zhang Jan 2011

Gene And Environment Interactions Modulate Immune System Influences On Social Behavior, Yubin Zhang

Legacy Theses & Dissertations (2009 - 2024)

Autism is a neurodevelopmental disorder with unknown etiology, affecting about 1 in 110 children in the US. Here, we show that developmental exposure to anti-brain Abs impaired mouse social behaviors. BTBR mice, a mouse strain with high serum anti-brain Abs and behaviors that resemble autism, have more activated splenic B cells, CD4+ T cells (Th1, Th2 and Th17 subsets), and CD4+ T cells with expression of Vb6 chains than B6 mice (the control H-2 identical strain). Elevated numbers of activated (CD25+) CD4+Vb6+ cells also were detected in thymic preparations. All of the immunological and behavioral parameters of BTBR mice were …

The Importance Of Disease And Behavior In Mammalian Ecology And Behavior, Christopher R. Collins Jan 2011

The Importance Of Disease And Behavior In Mammalian Ecology And Behavior, Christopher R. Collins

Legacy Theses & Dissertations (2009 - 2024)

Causes of mortality reflect the evolutionary forces shaping the physiology and behavior of species. To evaluate the relative importance of these in North American Mammals I compared published data from 69 populations across 27 species. Of the known causes, humans are the main cause of mortality (51.8%), followed by natural causes (48.5%), with predation being the largest natural cause. Populations in remote areas experienced higher hunting mortality, and those closer to human populations suffered increased vehicle collision. Predation mortality was negatively correlated with body mass.

Turkish Journal of Biology

Sonchus erzincanicus (Asteraceae) is a plant species that is endemic to Turkey and grows in Erzincan. Lipid peroxidation inhibition, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and total phenolic content of the methanol and water extracts of the plant were determined. Antimicrobial activity of the extracts was also tested against 107 clinical isolates of human pathogenic microorganism strains belonging to Enterobacter aerogenes, Escherichia coli, Proteus mirabilis, Pseudomonas aeruginosa, Staphylococcus aureus, and Candida albicans using the disc-diffusion method, and the minimal inhibition concentration (MIC) value of each active extract was determined. The extracts were observed to have antioxidant potential, although no better than …

Biochemical Identification And Numerical Taxonomy Of Aeromonas Spp. Isolated From Food Samples In Turkey, Belgi̇n Erdem, Ergi̇n Kari̇ptaş, Eli̇f Çi̇l, Kami̇l Işik

Turkish Journal of Biology

Numerical taxonomy was used to analyze phenotypic data obtained from 126 new isolates of Aeromonas strains taken from red meat, raw chicken, minced meat, and fish samples. Each strain was tested for 86 characters but only the final data including 63 characters were analysed using the S_{SM} coefficients and the UPGMA clustering algorithm. At S_{SM} values of => 83%, the strains clustered into 10 aggregate groups consisting of 7 major (5 and up strains) and 3 minor (2-4 strains), and 5 single member clusters, each of which was identified as A. hydrophila, A. caviae, and A. sobria, respectively. It was …

Turkish Journal of Biology

In recent years, the incidence of fungal infections has been rising all over the world. Although the amount of research in the field of pathogenic fungi has also increased, there is still a need for studies on fungal virulence. In this investigation, we focused on testing bloodstream-identified Candida albicans in experimental in vitro virulence assays. The proteinase enzyme activities of 30 C. albicans strains isolated from the bloodstream and sterile body fluids were investigated (15 colonizations, 15 infections). The spectrophotometric and plate methods were used to determine proteinase activity. The correlations between the 2 methods were compared. This study indicates …

Clonal Propagation And Cryogenic Storage Of Virus-Free Grapevine (Vitis Vinifera L.) Via Meristem Culture, Mohamad Shatnawi, Ghandi Anfoka, Rida Shibli, Mohammed Al-Mazra'awi, Wesam Shahrour, Areej Arebiat

Turkish Journal of Agriculture and Forestry

A protocol for production of virus-free Vitis vinifera using meristem culture was developed. Meristems (0.1-0.2 mm) of V. vinifera infected with grapevine fanleaf virus and grape leafroll associated viruses were excised from 1-year-old growing vines. Shoot tips were cultured on half-strength Murashige and Skoog (MS) medium, supplemented with 0.02 mg L^{-1} of benzylaminopurine (BAP) and 0.01 mg L^{-1} of naphthalene acetic acid (NAA). BAP and kinetin resulted in differences in the number of new shoots per explant, shoot height, and number of new leaves per explant. BAP at 0.8 mg L^{-1} gave the highest in vitro multiplication rate, with 5.25 …
