Haiti Earthquake January 2010: What Actions And Policies Can The Government Of Haiti Implement To Improve Emergency Management Response, Steve Cohen

Master's Projects

In 2010, Haiti experienced a devastating earthquake that destroyed much of its capital city and the governmental offices that should have guided the response to the disaster. This research focuses on how Haiti can benefit from the Caribbean Disaster Management Agency’s standards for disaster resilience as it works to recover from the earthquake. Unfortunately, Haiti has long been dependent on assistance from non-governmental organizations due to its extreme poverty; its recovery is complicated by the need to integrate disaster assistance and on-going economic and social assistance into its development of a more resilient society.

Role Of Matrix Metalloproteinase 3-Mediated Alpha-Synuclein Cleavage In Dopaminergic Cell Death, Dong-Hee Choi, Youn-Jung Kim, Young-Gun Kim, Tong H. Joh, M. Flint Beal, Yoon-Seong Kim

Faculty Bibliography 2010s

Evidence suggests that the C-terminal truncation of alpha-synuclein is equally important as aggregation of alpha-synuclein in Parkinson disease (PD). Our previous results showed that an endopeptidase, matrix metalloproteinase-3 (MMP3), was induced and activated in dopaminergic (DA) cells upon stress conditions. Here, we report that MMP3 cleaved alpha-synuclein in vitro and in vivo and that alpha-synuclein and MMP3 were co-localized in Lewy bodies (LB) in the postmortem brains of PD patients. Incubation of alpha-synuclein with the catalytic domain of MMP3 (cMMP3) resulted in generation of several peptides, and the peptide profiles of WT alpha-synuclein (WTsyn) and A53T mutant (A53Tsyn) were different. …

A Functional Nuclear Epidermal Growth Factor Receptor, Src And Stat3 Heteromeric Complex In Pancreatic Cancer Cells, Soumya Jaganathan, Peibin Yue, David C. Paladino, Jelena Bogdanovic, Qun Huo, James Turkson

Faculty Bibliography 2010s

Evidence is presented for the nuclear presence of a functional heteromeric complex of epidermal growth factor (EGFR), Src and the Signal Transducer and Activator of Transcription (Stat) 3 proteins in pancreatic cancer cells. Stat3 remains nuclear and associated with Src or EGFR, respectively, upon the siRNA knockdown of EGFR or Src, demonstrating the resistance of the complex to the modulation of EGFR or Src alone. Significantly, chromatin immunoprecipitation (ChIP) analyses reveal the nuclear EGFR, Src and Stat3 complex is bound to the c-Myc promoter. The siRNA knockdown of EGFR or Src, or the pharmacological inhibition of Stat3 activity only marginally …

Faculty Bibliography 2010s

Invasive melanoma is the most lethal form of skin cancer. The treatment of melanoma-derived cell lines with 5-aza-2'-deoxycytidine (5-Aza-dC) markedly increases the expression of several miRNAs, suggesting that the miRNA-encoding genes might be epigenetically regulated, either directly or indirectly, by DNA methylation. We have identified a group of epigenetically regulated miRNA genes in melanoma cells, and have confirmed that the upstream CpG island sequences of several such miRNA genes are hypermethylated in cell lines derived from different stages of melanoma, but not in melanocytes and keratinocytes. We used direct DNA bisulfite and immunoprecipitated DNA (Methyl-DIP) to identify changes in CpG …

Faculty Bibliography 2010s

Adrenaline and noradrenaline are produced within the heart from neuronal and non-neuronal sources. These adrenergic hormones have profound effects on cardiovascular development and function, yet relatively little information is available about the specific tissue distribution of adrenergic cells within the adult heart. The purpose of the present study was to define the anatomical localization of cells derived from an adrenergic lineage within the adult heart. To accomplish this, we performed genetic fate-mapping experiments where mice with the cre-recombinase (Cre) gene inserted into the phenylethanolamine-n-methyltransferase (Pnmt) locus were cross-mated with homozygous Rosa26 reporter (R26R) mice. Because Pnmt serves as a marker …

Modulation Of Toxin Stability By 4-Phenylbutyric Acid And Negatively Charged Phospholipids, Supriyo Ray, Michael Taylor, Mansfield Burlingame, Suren A. Tatulian, Ken Teter

Faculty Bibliography 2010s

AB toxins such as ricin and cholera toxin (CT) consist of an enzymatic A domain and a receptor-binding B domain. After endocytosis of the surface-bound toxin, both ricin and CT are transported by vesicle carriers to the endoplasmic reticulum (ER). The A subunit then dissociates from its holotoxin, unfolds, and crosses the ER membrane to reach its cytosolic target. Since protein unfolding at physiological temperature and neutral pH allows the dissociated A chain to attain a translocation-competent state for export to the cytosol, the underlying regulatory mechanisms of toxin unfolding are of paramount biological interest. Here we report a biophysical …

Faculty Bibliography 2010s

Background: RC-101, a cationic peptide retrocyclin analog, has in vitro activity against HIV-1. Peptide drugs are commonly prone to conformational changes, oxidation and hydrolysis when exposed to excipients in a formulation or biological fluids in the body, this can affect product efficacy. We aimed to investigate RC-101 stability under several conditions including the presence of human vaginal fluids (HVF), enabling the efficient design of a safe and effective microbicide product. Stability studies (temperature, pH, and oxidation) were performed by HPLC, Circular Dichroism, and Mass Spectrometry (LC-MS/MS). Additionally, the effect of HVF on formulated RC-101 was evaluated with fluids collected from …

Faculty Bibliography 2010s

Reduced brain metabolism is an invariant feature of Alzheimer Disease (AD) that is highly correlated to the decline in brain functions. Decreased activities of key tricarboxylic acid cycle (TCA) cycle enzymes may underlie this abnormality and are highly correlated to the clinical state of the patient. The activity of the alpha-ketoglutarate dehydrogenase complex (KGDHC), an arguably rate-limiting enzyme of the TCA cycle, declines with AD, but the mechanism of inactivation and whether it can be reversed remains unknown. KGDHC consists of multiple copies of three subunits. KGDHC is sensitive to oxidative stress, which is pervasive in AD brain. The present …

A Therapeutic Chemical Chaperone Inhibits Cholera Intoxication And Unfolding/Translocation Of The Cholera Toxin A1 Subunit, Michael Taylor, Tuhina Banerjee, Fernando Navarro-Garcia, Jazmin Huerta, Shane Massey, Mansfield Burlingame, Abhay H. Pande, Suren A. Tatulian, Ken Teter

Faculty Bibliography 2010s

Cholera toxin (CT) travels as an intact AB(5) protein toxin from the cell surface to the endoplasmic reticulum (ER) of an intoxicated cell. In the ER, the catalytic A1 subunit dissociates from the rest of the toxin. Translocation of CTA1 from the ER to the cytosol is then facilitated by the quality control mechanism of ER-associated degradation (ERAD). Thermal instability in the isolated CTA1 subunit generates an unfolded toxin conformation that acts as the trigger for ERAD-mediated translocation to the cytosol. In this work, we show by circular dichroism and fluorescence spectroscopy that exposure to 4-phenylbutyric acid (PBA) inhibited the …

Walking Abnormalities Are Associated With Copd: An Investigation Of The Nhanes Iii Dataset, Jenna M. Yentes, Harlan Sayles, Jane Meza, David M. Mannino, Stephen I. Rennard, Nikolaos Stergiou

Journal Articles

Research on the peripheral effects of COPD has focused on physiological and structural changes. However, different from muscular weakness or decreased physical activity, mechanical abnormalities of the muscular system, e.g. walking, have yet to be investigated. Our purpose was to utilize the National Health and Nutritional Examination Survey (NHANES) dataset to determine whether walking abnormalities are associated with COPD severity. To determine if walking abnormalities were independently associated with COPD severity, our analysis aimed to investigate the association of physical activity levels with COPD severity and with walking abnormalities. The NHANES III dataset that contains data for 31,000 persons that …

Weathering The Storm: Adolescent Girls And Climate Change, Anita Swarup, Irene Dankelman, Kanwal Ahluwalia, Kelly Hawrylyshyn

East Africa

The impacts of climate change, whether they are gradual changes in agriculture and living conditions or the more cataclysmic effects of a cyclone or flood, are different for different populations. But inevitably children everywhere are badly affected, and girls in particular bear the greater burden. Their lives, prospects and human rights must be better protected. We will never achieve climate justice without addressing the gender dimensions of climate change, and girls themselves, their skills, knowledge and energy, must be part of the search for solutions. During UNFCCC’s COP-16 in Cancún, Mexico, in December 2010 the girls who attended demonstrated their …

Second National Communication Of Seychelles, Government Of Seychelles

East Africa

The Initial National Communication of Seychelles which was submitted to UNFCCC in October 2000 was important in bringing attention to climate change issues in Seychelles; it had nonetheless some limitations. It focused primarily on reporting under the Convention and that strategic opportunities for feeding into national development were not fully exploited and there was limited participation of stakeholders from public and private sectors. The Investigations were essentially academic in nature; hence of limited relevance to policy decision making, for instance the climate risk management issue.

Anterior-Posterior And Medial-Lateral Control Of Sway In Infants During Sitting Acquisition Does Not Become Adult-Like, Fabien Cignetti, Anastasia Kyvelidou, Regina T. Harbourne, Nikolaos Stergiou

Journal Articles

We examined (1) how sitting postural control in infants develops in the anterior–posterior (A/P) and medial–lateral (M/L) directions of sway, and (2) whether this control is already adult-like during the late phase of infant's sitting acquisition. COP data were acquired from 14 healthy infants (from the onset of sitting until independent sitting) and 21 healthy adults while sitting on a force platform. Attractor dimensionality (CoD: correlation dimension), attractor predictability (LyE: largest Lyapunov exponent), and sway variability (RMS: root-mean square) were calculated from the COP data to evaluate postural control. In the A/P direction, sitting was mastered by the infants by …
