Protective Immunity To Pre-Erythrocytic Stage Malaria, Robert J. Schwenk, Thomas L. Richie Jan 2011

Protective Immunity To Pre-Erythrocytic Stage Malaria, Robert J. Schwenk, Thomas L. Richie

United States Army: Publications

The development of a vaccine against malaria is a major research priority given the burden of disease, death and economic loss inflicted upon the tropical world by this parasite. Despite decades of effort, however, a vaccine remains elusive. The best candidate is a subunit vaccine termed RTS,S but this provides only partial protection against clinical disease. This review examines what is known about protective immunity against pre-erythrocytic stage malaria by considering the humoral and T cell-mediated immune responses that are induced by attenuated sporozoites and by the RTS,S vaccine. On the basis of these observations a set of research priorities …

United States Army: Publications

Percutaneous exposure to the chemical warfare nerve agent VX was evaluated in African green monkeys (n = 9). Doses of VX (7.5–100 μg/kg) were applied to the skin for 60 min and residual agent was quantified (before decontamination) to estimate the absorbed dose. Monkeys were evaluated for the presence or absence of clinical signs of toxicity and blood was sampled periodically (30 min–12 weeks) following exposure to measure the degree of circulating acetylcholinesterase (AChE) inhibition. Monkeys were also evaluated for behavioral changes fromVXexposure using a serial probe recognition (SPR) task. The lowest observable adverse effect level (LOAEL) for the production …

Mechanical Response Of Pig Skin Under Dynamic Tensile Loading, Jaeyoung Lim, Jihye Hong, Weinong W. Chen, Tusit Weerasooriya

United States Army: Publications

Uniaxial tensile experiments were performed on pig skin to investigate the tensile stressestrain response at both quasi-static and dynamic rates of deformation. A Kolsky tension bar, also called a split Hopkinson tension bar (SHTB), was modified to conduct the dynamic experiments. Semiconductor strain gages were used to measure the low levels of the transmitted signal from pig skin. A pulse shaper technique was used for generating a suitable incident pulse to ensure stress equilibrium and approximate constant strain rate in the specimen of a thin skin sheet wrapped around the ends of the bars for minimizing radial inertia. In order …

Detection Of Six Serotypes Of Botulinum Neurotoxin Using Fluorogenic Reporters, Daniel R. Ruge, F. Mark Dunning, Timothy M. Piazza, Brian E. Molles, Michael Adler, Füsûn N. Zeytin, Ward C. Tucker

United States Army: Publications

Methods that do not require animal sacrifice to detect botulinum neurotoxins (BoNTs) are critical for BoNT antagonist discovery and the advancement of quantitative assays for biodefense and pharmaceutical applications. Here we describe the development and optimization of fluorogenic reporters that detect the proteolytic activity of BoNT/A, B, D, E, F, and G serotypes in real time with femtomolar to picomolar sensitivity. Notably, the reporters can detect femtomolar concentrations of BoNT/A in 4 h and BoNT/E in 20 h, sensitivity that equals that of animal-based methods. The reporters can be used to determine the specific activity of BoNT preparations with intra- …

Prophylaxis With Human Serum Butyrylcholinesterase Protects Guinea Pigs Exposed To Multiple Lethal Doses Of Soman Or Vx, Ashima Saxena, Wei Sun, James M. Fedorko, Irwin Koplovitz, Bhupendra P. Doctor

United States Army: Publications

Human serum butyrylcholinesterase (Hu BChE) is currently under advanced development as a bioscavenger for the prophylaxis of organophosphorus (OP) nerve agent toxicity in humans. It is estimated that a dose of 200 mg will be required to protect a human against 2 × LD50 of soman. To provide data for initiating an investigational new drug application for the use of this enzyme as a bioscavenger in humans, we purified enzyme from Cohn fraction IV-4 paste and initiated safety and efficacy evaluations in mice, guinea pigs, and non-human primates. In mice, we demonstrated that a single dose of enzyme that …

A Diagnostic Approach To Pruritus, Brian V. Reamy, Christopher W. Bunt, Stacy Fletcher Jan 2011

A Diagnostic Approach To Pruritus, Brian V. Reamy, Christopher W. Bunt, Stacy Fletcher

United States Air Force: Publications

Pruritus can be a symptom of a distinct dermatologic condition or of an occult underlying systemic disease. Of the patients referred to a dermatologist for generalized pruritus with no apparent primary cutaneous cause, 14 to 24 percent have a systemic etiology. In the absence of a primary skin lesion, the review of systems should include evaluation for thyroid disorders, lymphoma, kidney and liver diseases, and diabetes mellitus. Findings suggestive of less serious etiologies include younger age, localized symptoms, acute onset, involvement limited to exposed areas, and a clear association with a sick contact or recent travel. Chronic or generalized pruritus, …

Talking And Not Talking: Sexual Education And Ethics For Young Women Within The Evangelical Movement In America, Kate Sargent Jan 2011

Talking And Not Talking: Sexual Education And Ethics For Young Women Within The Evangelical Movement In America, Kate Sargent

CGU Theses & Dissertations

In this paper I intend to discuss the ways in which sex, sexuality, and the female body are treated in the discourse within the Evangelical movement, particularly with regard to teenaged and young girls. I will focus on the way young women are talked to about sex, and how they are socialized and educated to regard sexuality and gender. I will spend most of my time with the popular literature aimed at young women, analyzing the underlying theology and theory at work. I will extrapolate, through the use of the (admittedly limited) data regarding sexual activity among young Evangelicals, including …

Children’S Ability To Recall Unique Aspects Of One Occurrence Of A Repeated Event, Sonja P. Brubacher, Una Glisic, Kim P. Roberts, Martine B. Powell

Psychology Faculty Publications

Preschool and school-age children’s memory and source monitoring were investigated by questioning them about one occurrence of a repeated lab event (n = 39). Each of the four occurrences had the same structure, but with varying alternatives for the specific activities and items presented. Variable details had a different alternative each time; hi/lo details presented the identical alternative three times and changed once. New details were present in one occurrence only and thus had no alternatives. Children more often confused variable, lo, and new details across occurrences than hi details. The 4- to 5-year-oldchildren were less …

Money For Nothing: Molière’S Miser And The Risky World Of Early Modern France, Michael Call Jan 2011

Money For Nothing: Molière’S Miser And The Risky World Of Early Modern France, Michael Call

Quidditas

Molière’s 1668 comedy L’Avare, or The Miser, takes place during a significant shift in the way that early modern Europeans thought about chance and risk. Staged at the same historical moment that Pascal, Huygens, and Leibnitz were developing the first foundations of probability mathematics, L’Avare conjures up an atmosphere of uncertainty, setting in opposition risk-takers and the risk-averse. As the characters encounter and debate the concepts of usury, life expectancy, gambling, and the risks of maritime travel, they grapple palpably with the consequences of an uncertain world in which Divine Providence can no longer be assumed—a new world …

Love And Marriage On The Medieval English Stage: Using The English Cycle Plays As Sources For Social History, James H. Forse Jan 2011

Love And Marriage On The Medieval English Stage: Using The English Cycle Plays As Sources For Social History, James H. Forse

Quidditas

Much scholarship concerning the concept of “companionate” marriage traces its origins to the early modern period as clergymen, especially Protestant ones, began to publish “guides” to the relationships and respective duties of husbands and wives in the 1500s and 1600s. Studies of marriage in the Middle ages concentrate on marriage among the nobility, since there is more documentary evidence about the medieval elites. Examinations of sermons reveal that the Church, especially after the twelfth century, stressed the sanctity of marriage as an institution created by God and blessed by Christ at the marriage at Cana, but sermons say little about …

Molecular Characterization Of Novel Sulfotransferases From The Tick, Ixodes Scapularis, Sivakamasundari Pichu, Emine Bihter Yalcin, José Mc Ribeiro, Roberta S. King, Thomas N. Mather

Biomedical and Pharmaceutical Sciences Faculty Publications

Background: Ixodes scapularis, commonly known as the blacklegged or deer tick, is the main vector of Lyme disease in the United States. Recent progress in transcriptome research has uncovered hundreds of different proteins expressed in the salivary glands of hard ticks, the majority of which have no known function, and include many novel protein families. We recently identified transcripts coding for two putative cytosolic sulfotransferases in these ticks which recognized phenolic monoamines as their substrates. In this current study, we characterize the genetic expression of these two cytosolic sulfotransferases throughout the tick life cycle as well as the …

THE ROLE OF THE SPARC ACIDIC DOMAIN AND EGF-LIKE MODULE IN GLIOMA MIGRATION, INVASION, AND SIGNALING

HEATHER M. MCCLUNG

Advisor: Sandra A. Rempel, Ph.D.

Major: Pharmacology

Degree: Doctor of Philosophy

We have previously shown that Secreted Protein Acidic and Rich in Cysteine (SPARC) is upregulated in all astrocytoma grades and increases tumor cell migration and invasion. It is thought that different domains within the protein may regulate SPARC functions, suggesting domain-specific targeting to inhibit invasion. To enhance our understanding of SPARC-mediated invasion, we first confirm, at the protein level, our previous cDNA array results, that SPARC increases expression of the …

Lung cancer is the leading cause of cancer deaths in the US, in part due to poor diagnosis. Prognosis can be improved, if detected at an early stage. The objective of this study was to use the metabolomic approach to investigate early potential biomarkers for lung cancer. Golden Syrian hamsters were randomized into two groups (treatment and control). Lung cancer was induced in treatment group by a chemical carcinogen, N-nitrosobis-2-(oxopropyl) amine (BOP) while the control animals were injected with saline. Urine was collected weekly throughout the study and frozen at -800 C. Proton nuclear magnetic resonance (1H NMR) spectra, acquired …

Reinforced Chitosan-Based Heart Valve Scaffold And Utility Of Bone Marrow-Derived Mesenchymal Stem Cells For Cardiovascular Tissue Engineering, Mohammad Z. Albanna Jan 2011

Reinforced Chitosan-Based Heart Valve Scaffold And Utility Of Bone Marrow-Derived Mesenchymal Stem Cells For Cardiovascular Tissue Engineering, Mohammad Z. Albanna

Wayne State University Dissertations

Recent research has demonstrated a strong correlation between the differentiation profile of mesenchymal stem cells (MSCs) and scaffold stiffness. Chitosan is being widely studied for tissue engineering applications due to its biocompatibility and biodegradability. However, its use in load-bearing applications is limited due to moderate to low mechanical properties. In this study, we investigated the effectiveness of a fiber reinforcement method for enhancing the mechanical properties of chitosan scaffolds. Chitosan fibers were fabricated using a solution extrusion and neutralization method and incorporated into porous chitosan scaffolds. The effects of different fiber/scaffold mass ratios, fiber mechanical properties and fiber lengths on …

Identification Of The Role Of The Sal Locus In Streptococcus Pyogenes Virulence During Host-Pathogen Interactions, Phanramphoei Namprachan-Frantz Jan 2011

Identification Of The Role Of The Sal Locus In Streptococcus Pyogenes Virulence During Host-Pathogen Interactions, Phanramphoei Namprachan-Frantz

Wayne State University Dissertations

The pathogenesis of Streptococcus pyogenes is due to its ability to overcome and adapt to the harsh environment created by the host immune response. The focus of this project was the SalKR two-component regulatory system, which facilitates bacterial adaptation by responding to environmental signals during host pathogen-interactions. The first goal of this project was to determine a role in virulence for the SalKR regulatory system. The complete deletion of the salKR genes in the wild type S. pyogenes strain HSC5 produced a highly attenuated mutant in a Zebrafish infection model. The ΔsalKR mutant appeared to lose the ability to survive …

A Qualitative Study On African American Males' Perceptions Of Reading, Phyllis Marie Stallings Jan 2011

A Qualitative Study On African American Males' Perceptions Of Reading, Phyllis Marie Stallings

Wayne State University Dissertations

African American male students begin experiencing failure when they enter fourth grade. At this grade, the curriculum becomes more focused, students are expected to complete high-stakes standardized tests, teachers become more distant, and students are expected to become independent learners. Many African American male students are not ready for this transformation. They may not have developed the reading skills needed to understand text books for social studies and science, and are deficient in regard to the math skills needed for problem solving. Understanding which factors are contributing to African American male student failure is important, especially in determining if these …

The Impact Of Folate Deficiency On The Base Excision Repair Pathway: Analysis Of Enzyme Coordination In Response To Dna Damage And Imbalanced Repair, Amanda Pilling Jan 2011

The Impact Of Folate Deficiency On The Base Excision Repair Pathway: Analysis Of Enzyme Coordination In Response To Dna Damage And Imbalanced Repair, Amanda Pilling

Wayne State University Dissertations

The condition of folate deficiency has been implicated in carcinogenesis, with the strongest evidence formulated in colon cancer. The role of folate in DNA repair, DNA synthesis and methylation reactions renders this nutrient an valuable target for studying the onset and progression of cancer. Using molecular techniques to determine gene and protein expression, enzyme activity and methylation status elucidates the mechanism of DNA repair and damage in folic acid deficient animals in response to carcinogen. The findings presented in this study indicate failure to remove and repair damage in the condition of folate deficiency and suggest that the accumulation of …

An Investigation Of The Biomechanical Response From Shock Wave Loading To The Head, Alessandra Dal Cengio Leonardi

Wayne State University Dissertations

There is a pressing need for a comprehensive explanation of the mechanism of brain injury after exposure to blast and several hypotheses have been suggested. The focus of this research was to investigate one of the hypotheses for primary brain injury due to blast: multimodal skull flexure. The significance of this research is twofold. First, resolution of the mode of energy transfer and of the induced stresses within the skull-brain system will allow for creations of mitigation/protective techniques/equipment, as well as design of experiments investigating live-cell response using more reliable physical models. Second, the data obtained experimentally will be available …

Hedgehog Signaling: A Potential Therapeutic Target For Non-Small Cell Lung Cancer, Ma'in Yehya Maitah

Wayne State University Dissertations

The American Cancer Society estimated that 222,520 Americans were diagnosed with lung cancer and 157,300 died of lung cancer in 2010 (Jemal et al. 2009, 225-249;Jemal et al. 2011, 69-90). The clinical outcome of patients diagnosed with non-small cell lung cancer (NSCLC), the major lung cancer sub-types, is very poor, which calls for innovative research for finding novel therapeutic targets and agents for better treatment outcome.

Emerging evidences have suggested that a phenomenon called Epithelial-to-Mesenchymal Transition (EMT), which shares similar molecular characteristics with cancer stem-like cells, contributes to lung cancer treatment failure. In view of the fact that EMT process …

Wayne State University Dissertations

Folate, an essential water soluble vitamin has been implicated in the etiology of many types of cancer especially colorectal cancer. Folate deficiency has been reported to incapacitate DNA repair pathways and thereby affect the genomic stability. Our lab has reported previously and here again through this research that folate deficiency affects the DNA damage inducibility of base excision repair pathway. Our study shows the differential effect folate deficiency has on the expression of the genes involved in this pathway. Further, our study shows the increase in preneoplastic lesions in the colon of mice exposed to folate deficiency in response to …
