Retroviral infections cause a variety of cancers in animals and a number of diverse diseases in humans such as leukemia and acquired immune deficiency syndrome. Productive and efficient proviral integration is critical for retroviral function and is the key step in establishing a stable and productive infection, as well as the mechanism by which host genes are activated in leukemogenesis. Host factors are widely anticipated to be involved in all stages of the retroviral life cycle, and the identification of integrase interacting factors has the potential to increase our understanding of mechanisms by which the incoming virus might appropriate cellular …

On September 24, 2009, Theatre VCU opened its production of Psycho Beach Party, written by Charles Busch and directed by Steve Perigard. In our production, two male actors, Tommy Callan and Kyle Cornell, were cross-sex cast to play female characters, Chicklet and Marvel Ann respectively. In addition to serving as vocal coach for the production, I was to help Callan and Cornell vocally transform into female characters. I have documented our exploration of gender, specifically the vocal transformation from male to female, in Theatre VCU’s production of Psycho Beach Party. Neither actor had played a female character onstage before, and …

Extracellular signal-regulated kinases (ERKs) modulate cellular activities in response to extracellular stimuli and play important biological roles. Thus, perturbed kinase pathways induce pathological conditions, such as tumor development. Rit, a novel member of the Ras family GTPases, activase ERK6, and its over-expression confers tumorigenicity. We hypothesized the presence of scaffolding molecules specific to ERK6, similar to other known MAP kinases. We performed yeast two-hybrid assays using ERK6 as bait, and Scribble was identified as a binding partner. Scribble contains 16 LRR domains and four PDZ domains. We performed immunoprecipitation (IP) assays and discovered ERK2 as another binding partner. Surprisingly, no …

The mechanism by which the type la regulatory subunit (RIa) of cAMP-dependent protein kinase (PKAI) is localized to cell membranes is unknown, however, RIa may be localized through hydrophobic interaction with membrane lipids. To determine if post-translational lipid modification of RIa is important for membrane association, both beef skeletal muscle cytosolic RI and beef heart membrane-associated RI were characterized by electrospray ionization mass spectrometry. Total sequence coverage was 98% for both the membrane and cytosolic forms of RI after digestion with trypsin or AspN protease. Hyperthermophilic archaea are of great interest in evolutionary microbiology, owing to their ability to withstand high temperatures, and often extremes of pressure, pH and salinity. Enzymes from these organisms1 may offer particular opportunities for asymmetric synthesis, complementary to approaches with mesophilic enzymes,2 or those involving enzyme3 and pathway4 reengineering. However, perhaps due to a bias that hyperthermophilic enzymes have “narrow substrate specificities,”5 archaeal extremophiles remain a largely untapped resource in asymmetric synthesis.6

Red grouper (Epinephelus morio) are long-lived, commercially important, soniferous fish belonging to the family Epinephelidae. Found throughout the western North Atlantic and Gulf of Mexico, they are protogynous hermaphrodites, and peak spawning occurs from March through May. Unlike many grouper species, red grouper do not form large spawning aggregations; rather, they form small polygynous groups, and remain in relatively close proximity to rocky depressions excavated in the sandy bottom by males. This excavation activity creates structure and habitat for a wide variety of species, and as a result, red grouper are a keystone species on the West Florida Shelf. While …

Often only one parent in a same-sex relationship is permitted to have legal parenting rights to the children, even when they have planned the children together. Prohibiting parents from being legally recognized as parents to their children is an issue that is unique to same-sex couples. Based on sexual minority stress theory, I hypothesized that same-sex parents without legal rights would report more sexual minority stress (i.e., less disclosure of sexual orientation and experience significantly more stigma consciousness, internalized homophobia, and discrimination) and more parental stress (including family worry and the need for parental justification) than same-sex parents with legal …

The study and utilization of bionanomotors represents a rapid and progressing field of nanobiotechnology. Here, we demonstrate that poly(amidoamine) (PAMAM) dendrimers are capable of supporting heavy meromyosin dependent actin motility of similar quality to that observed using nitrocellulose, and that microcontact printing of PAMAM dendrimers can be exploited to produce tracks of active myosin motors leading to the restricted motion of actin filaments across a patterned surface. These data suggest that the use of dendrimer surfaces will increase the applicability of using protein biomolecular motors for nanotechnological applications.

Fishes demonstrate the greatest change throughout ontogeny in body size of all vertebrates, some becoming twelve times their original length. Panther groupers, Cromileptes altivelis, are no exception. This vast size change influences other aspects of their lives including their feeding behavior, the prey they consume, and the capabilities of the mechanisms they use to capture prey. The change in the feeding performance of the panther grouper, Cromileptes altivelis, was quantified through buccal pressure recordings and high-speed videography. From this data, we can deduce that the larger juvenile was able to generate greater negative pressure within the mouth. It appears that …

Kinematic Evidence For Superfast Locomotory Muscle In Two Species Of Teneriffiid Mites, Grace C. Wu, Jonathan C. Wright, Dwight L. Whitaker, Anna N. Ahn

Locomotory muscles typically operate over a narrow range of contraction frequencies, characterized by the predominant fiber types and functional roles. The highest documented frequencies in the synchronous sound-producing muscles of insects (550 Hz) and toadfish (200 Hz) far exceed the contraction frequencies observed in weight-bearing locomotory muscles, which have maximum documented frequencies below 15-30 Hz. Laws of scaling, however, predict that smaller arthropods may employ stride frequencies exceeding this range. In this study we measured running speed and stride frequency in two undescribed species of teneriffiid mites from the coastal sage scrub of southern California. Relative speeds of both species …

This report details the structure and content of targeted regions of the strawberry genome. The data indicate that the strawberry genome is gene-dense, with an average of one protein-encoding gene or pseudogene per 5.9 kb. Current overall EST coverage is sparse. The unexpected gene duplications and their differential patterns of EST support suggest possible subfunctionalization or pseudogenization of these sequences. This report provides a high-resolution depiction of targeted gene neighborhoods that will aid whole-genome sequence assembly, provide valuable tools for plant breeders and advance the understanding of strawberry genome evolution.

A Rock Strikes Back: Women's Struggles For Equality In The Development Of The South African Constitution, Thuto Seabe Thipe

In 1991, South African women’s organisations formed the Women's National Coalition (WNC) to identify and advocate for women's primary needs in the post-apartheid Constitution. The outcome of this advocacy was South Africa’s adoption, in 1996, of one of the most comprehensive protections of gender and sexuality rights of any national constitution. I argue that the WNC became a key actor in the development of the Constitution by drawing from a tradition of women’s organising in South Africa that emphasised women’s legitimacy in and value to public politics. The WNC rejected masculinist framings of politics and instead demanded that political structures …

Strawberry (Fragaria spp.) is the familiar name of a group of economically important crop plants and wild relatives that also represent an emerging system for the study of gene and genome evolution. Its small stature, rapid seed-to-seed cycle, transformability and miniscule basic genome make strawberry an attractive system to study processes related to plant physiology, development and crop production; yet it lacks substantial genomics-level resources. Methylating agents are involved in carcinogenesis, and the DNA repair protein O(6)-methylguanine-DNA methyltransferase (MGMT) removes methyl group from O(6)-methylguanine. Genetic variation in DNA repair genes has been shown to contribute to susceptibility to squamous cell carcinoma of the head and neck (SCCHN). We hypothesize that MGMT polymorphisms are associated with risk of SCCHN. In a hospital-based case-control study of 721 patients with SCCHN and 1234 cancer-free controls frequency-matched by age, sex and ethnicity, we genotyped four MGMT polymorphisms, two in exon 3, 16195C>T and 16286C>T and two in the promoter region, 45996G>T and 46346C>A. The research was conducted in southern Nigeria federal university libraries to identify the extent of ICT use for serials functions. A questionnaire was distributed to staff from the 11 federal university libraries in southern Nigeria. The results reveal that serials functions are still predominately manual, with ICT being more prominent in the serials public services and preservation functions; public services activities involving the use of ICT are mainly access to and retrieval of serials and article titles. It is recommended that university libraries should make a decision on whether to equip serials units with ICT facilities or have a systems …
