Stoichiometry is the ratio of elements in a substance or microbial biomass. This paper investigates microbial stoichiometry and the responses of microbes to dissolved nutrient concentrations and ratios. The current work assesses both autrotrophic and heterotrophic responses to nutrient enrichment: autotrophic and heterotrophic states are defined mainly by ecosystem primary production and respiration. In ecosystems dominated by autotrophic microbes, nutrient enrichment can lead to increased biomass and biomass-specific rates of primary production. Heterotrophic ecosystems rely on subsides of organic carbon from outside the system. Their enrichment with N and/or P can accelerate microbial respiration rates and result in carbon losses …

Iron-Binding Activity Of Human Iron-Sulfur Cluster Assembly Protein Hisca1, Jianxin Lu, Jacob P. Bitoun, Guoqiang Tan, Wu Wang, Wenguang Min, Huangen Ding

A human homologue of the iron-sulfur cluster assembly protein IscA (hIscA1) has been cloned and expressed in Escherichia coli cells. The UV-visible absorption and EPR (electron paramagnetic resonance) measurements reveal that hIscA1 purified from E. coli cells contains a mononuclear iron centre and that the iron binding in hIscA1 expressed in E. coli cells can be furthermodulated by the iron content in the cell growth medium. Additional studies show that purified hIscA1 binds iron with an iron association constant of approx. 2×1019 M-1, and that the iron-bound hIscA1 is able to provide the iron for the iron-sulfur cluster assembly in …

In this work, a zwitterionic molecular micelle, poly-ε-sodium- undecanoyl lysinate (poly-ε-SUK), was synthesized and employed as a coating in open tubular capillary electrochromatography (OT-CEC) for protein separation. The zwitterionic poly-ε-SUK containing both carboxylic acid and amine groups can be either protonated or deprotonated depending on the pH of the background electrolyte; therefore, either an overall positively or negatively charged coating can be achieved. This zwitterionic coating allows protein separations in either normal or reverse polarity mode depending on the pH of the background electrolyte. Catalytic Decomposition Of Peroxynitrite And Superoxide By Nafion Films Modified With Iron And Manganese Porphyrin, Amanda Stull, Victor Sarpong

Nafion films alone and modified with inert electrolytes and metalloporphyrins were tested in their effectiveness of decomposition of aqueous peroxynitrite (PN) and superoxide. Films that were modified with a cationic manganese porphyrin, which has been shown to be responsible for catalytic decomposition of superoxide ion, were found to be most effective. Peroxynitrite was generated in solution by the decomposition of 3-morpholinosydnonimime (SIN-1) that generates nitric oxide and superoxide ion, which rapidly combine to form peroxynitrite. The tyrosine analogue 4-hydroxyphenylacetic (4-HPA) was used to trap the PN in pH 7.00 PBS buffer and was its nitrated product was detected by observing …

Rhizocephalan Parasites Of Mud Crabs In South Carolina Estuaries, Erin Burnette

Rhizocephalan parasites often infect commercially important crustacean species such as the blue crab (Callinectes sapidus). In this experiment, the prevalence of rhizocephalan parasites was determined by sampling mud crabs (Panopeus herbstii) in three different locations; Huntington Beach State Park, Waites Island, and Murrells Inlet. Crabs were determined to be parasitized by the presence of an externae extruding from their apron. Unparasitized crabs were also collected to serve as a control group. The externa were removed and DNA extractions were performed. Polymerase chain reactions (PCR) were done to prove whether the crabs were parasitized by Loxothylacus texanus or Loxothylacus panopaei. We suggest a possible realization of a solid-state memory capacitive (memcapacitive) system. Our approach relies on the slow polarization rate of a medium between plates of a regular capacitor. To achieve this goal, we consider a multilayer structure embedded in a capacitor. The multilayer structure is formed by metallic layers separated by an insulator so that nonlinear electronic transport (tunneling) between the layers can occur. The suggested memcapacitor shows hysteretic charge-voltage and capacitance-voltage curves, and both negative and diverging capacitance within certain ranges of the field. Studying genetic variation presents a dilemma. While the genetic variation of greatest interest is that causing variation in traits and disease risk in natural populations, natural populations have characteristics that make them challenging to study. In this work, I have assessed the use of cell culture methods as a solution to some of these challenges. In particular, I studied genetic variation in the budding yeast Saccharomyces cerevisiae that was generated by selection in the lab as a model for natural genetic variation. Measurement of growth of the euryhaline teleost Mugil cephalus is quite common in open ocean settings, but growth rates of M. cephalus in isolated areas is non-existent. Two isolated ponds in Huntington Beach State, Horry County, South Carolina were selected to study the growth rate of M. cephalus; Sandpiper Pond with virtual no hydrologic connection to the ocean and Jetty Pond with adequate hydrologic connection to the ocean were the sites of sample collection. Bacteriophage is of great interest because of its potential role in controlling bacterial populations in our environment. UV exposure has a damaging effect on the virus decreasing lytic ability. This study set out to test the effects of UV radiation, in amounts comparable to local environmental conditions, on bacteriophage T2. The virus was placed in a Petri dish in a PBS medium and exposed to UV radiation at 365nm. The irradiated virus was allowed to infect E. coli and plated. The main goal of this study was to evaluate the effectiveness of two biological control agents in reducing population growth and spread of the invasive tree Melaleuca quinquenervia, a subtropical tree native to Australia, and invasive in Florida, Puerto Rico, and the Bahamas. While in Florida two insects Oxyops vitiosa (weevil), and Boreioglycaspis melaleucae (psyllid) have been established as biocontrol agents, in Puerto Rico only psyllids are present, and in the Bahamas no biocontrol agents are present. Context: Several recovery strategies have been used, with limited effectiveness, to reduce the muscle discomfort or pain and the diminished muscle performance following a bout of unaccustomed physical activity, a condition known as delayed onset of muscle soreness (DOMS). Muscle damage in this condition is associated with mechanical disruption of the muscle and connective tissue and inflammation and increased oxidative stress. Low frequency, low intensity, whole body periodic acceleration (WBPA) that increases nitric oxide (NO) release from vascular endothelium into the circulation through increased pulsatile shear stress offers a potential solution. One motivation of systems biology research is to understand gene functions and interactions from functional genomics data such as that derived from microarrays. Up-to-date structural and functional annotations of genes are an essential foundation of systems biology modeling. We propose that the first essential step in any systems biology modeling of functional genomics data, especially for species with recently sequenced genomes, is gene structural and functional re-annotation. Individual differences in behavior are significant because they serve as the substrate for natural selection. Within the Behavioral Syndromes framework, researchers study individual differences in behavior of animals. Behavioral Syndromes are defined as correlations between behaviors in different environmental contexts or testing situations. In this study, I examined the effects of litter size and sex ratio, familial relationships, and age and sex on exploratory behavior of prairie voles, Microtus ochrogaster. Exploratory behavior, defined as spontaneous behavioral responses to unfamiliar stimuli, was examined in three novel situations: an open-field with novel objects, a two-way novel choice apparatus, and a complex maze. …

Driven in part by the imminent threats of population growth and climate destabilization, recent studies suggest that urban areas face severe water scarcity, with some areas in Australia and the United States already instituting moratoria on water use. While water managers traditionally avoid such crises by developing demand forecasts based on population estimates, technological developments, and weather predictions, their analysis are often at a regional scale with aggregate measures of water consumption. Disability in chronic low back pain patients has been established as a function of variables across three areas: pain, cognitive-behavioral variables, and social variables. New technology has improved the ability to accurately measure physical activity, a significant component of disability, through the use of actigraphy for real-time ambulatory monitoring. The current study assessed between and within patient changes in physical activity as a function of current pain, anticipated pain, pain sensitivity, depression, pain anxiety, catastrophizing, and significant others' responses.
