Reducing The Effects Of Intracellular Accumulation Of Thermolabile Collagen Ii Mutants By Increasing Their Thermostability In Cell Culture Conditions., Katarzyna Gawron, Deborah A. Jensen, Andrzej Steplewski, Andrzej Fertala
Department of Dermatology and Cutaneous Biology Faculty Papers
Mutations in collagen II are associated with spondyloepiphyseal dysplasia, a group of heritable diseases whose common features include aberrations of skeletal growth. The mechanisms through which mutations in collagen II affect the cartilaginous tissues are complex and include both intracellular and extracellular processes. One of those mechanisms involves cellular stress caused by excessive accumulation of misfolded collagen II mutants. We investigated whether stabilizing the structure of thermolabile R789C and R992C collagen II mutants would improve their secretion from cells, thereby reducing cellular stress and apoptosis. Employing glycerol and trimethylamine N-oxide (TMAO), chemicals that increase the thermostability of collagen triple helices, …

The northern Gulf of Mexico (GOM) is geologically diverse basin, described as the most complex continental slope region in the world. Regional topography of the slope consists of basins, knolls, ridges, and mounds derived from the dynamic adjustments of salt and the introduction of large volumes of sediment over long time scales. More than 99% of the sea floor in the GOM consists of soft sediment made up of various mixtures of primarily silt and clay. These wide-spread soft bottom communities are described in reports from major MMS studies by Gallaway et al. (1998) and Rowe and Kennicutt (2009). Relative …

An Empirical Approach To Evaluating Sufficient Similarity: Utilization Of Euclidean Distance As A Similarity Measure, Scott Marshall
Theses and Dissertations
Individuals are exposed to chemical mixtures while carrying out everyday tasks, with unknown risk associated with exposure. Given the number of resulting mixtures it is not economically feasible to identify or characterize all possible mixtures. When complete dose-response data are not available on a (candidate) mixture of concern, EPA guidelines define a similar mixture based on chemical composition, component proportions and expert biological judgment (EPA, 1986, 2000). Current work in this literature is by Feder et al. (2009), evaluating sufficient similarity in exposure to disinfection by-products of water purification using multivariate statistical techniques and traditional hypothesis testing. The work of …

Role Of Capsule In The Interaction Of Porphyromonas Gingivalis With Host, Amrita Singh
Theses and Dissertations
Periodontal disease is a chronic oral disease that is triggered by bacteria. One of these bacteria is Porphyromonas gingivalis. Some strains of P. gingivalis produce capsule, however, so far the role of capsule in the interactions with host cells in P. gingivalis is not well understood. Here, we investigated the contribution of capsule to triggering host response as well as its protective role on bacterial internalization by host cells with subsequent killing. qRT-PCR analysis showed more upregulation of expression of various groups of genes in macrophages challenged with the non-capsulated strain than in those challenged with the capsulated one with …

State Versus Ethnic Identification In Tibati, Cameroon, Brian Hillery
Capstone Projects – Politics and Government
The purpose of this paper is to analyze state versus ethnic identification among residents of Tibati, Cameroon, and to question whether identification suggests the existence of tribalism. This study explores how residents of Tibati who more strongly identify with their ethnic group than the state are more likely to express dissatisfaction with the Cameroonian government, using information collected through informal conversations about the level of satisfaction of Tibatians with regards to both their ethnic group and the Cameroonian government.
This paper provides a synopsis of the major ethnic groups in Tibati and a brief historical summary of Cameroonian state evolution …

The Economic And Environmental Effects Of An Eu Ban On Illegal Logging Imports. Insights From A Cge Assessment, Francesco Bosello, Ramiro Parrado, Renato Rosa
Fondazione Eni Enrico Mattei Working Papers
Illegal logging is widely recognized as a major economic problem and one of the causes of environmental degradation. Increasing awareness of its negative effects has fostered a wide range of proposals to combat it by major international conservation groups and political organizations. Following the 2008 US legislation which prohibits the import of illegally harvested wood and wood products, the European Union (EU) is now discussing a legislation proposal which would ban illegal timber from the EU market. In this study we use the ICES computable general equilibrium model to estimate the reallocation of global demand and timber imports following the …

Synthesis Of Polyethylene/Starch Hybrids Using Aqueous Mini Emulsion Polymerization, Brinda Shah
Theses
Sustainability regarding the most produced plastic, polyethylene, was the main motivation factor behind research of this thesis. The aim was to synthesize hybrids of polyethylene and starch in an aqueous mini emulsion polymerization process using single-site catalysts. Two types of water soluble starches were applied: potato and hydroxyethyl starch. A trifluoromethane bearing nickel enolate catalyst and a 5-halo 3-methoxy salicyl aldiminate nickel pyridyl catalyst (with Cl and Br as halogen atoms) were used as polymerization catalysts, the latter two known to results in higher molecular weight polymer. The first catalyst turned to be sufficiently active with increased loads of starch …

Inhibition Of Proliferation And Induction Of Apoptosis In Multiple Myeloma Cell Lines By Cd137 Ligand Signaling, Charles A. Gullo Phd, Liang Kai Koh, Wan Lu Pang, Kian Tong Ho, Shi Hao Tan, Herbert Schwarz
Biochemistry and Microbiology
Background
Multiple myeloma (MM) is a malignancy of terminally-differentiated plasma cells, and the second most prevalent blood cancer. At present there is no cure for MM, and the average prognosis is only three to five years. Current treatments such as chemotherapy are able to prolong a patient's life but rarely prevent relapse of the disease. Even hematopoietic stem cell transplants and novel drug combinations are often not curative, underscoring the need for a continued search for novel therapeutics. CD137 and its ligand are members of the Tumor Necrosis Factor (TNF) receptor and TNF superfamilies, respectively. Since CD137 ligand cross-linking enhances …

Functionalization Of Fibers Using Azlactone-Containing Polymers: Layer-By-Layer Fabrication Of Reactive Thin Films On The Surfaces Of Hair And Cellulose-Based Materials, Maren E. Buck, David M. Lynn
Chemistry: Faculty Publications
We report an approach to the functionalization of fibers and fiber-based materials that is based on the deposition of reactive azlactone-functionalized polymers and the "reactive" layer-by-layer assembly of azlactone-containing thin films. We demonstrate (i) that the azlactone-functionalized polymer poly(2-vinyl-4,4-dimethylazlactone) (PVDMA) can be used to modify the surfaces of a model protein-based fiber (horsehair) and cellulose-based materials (e.g., cotton and paper), and (ii) that fibers functionalized in this manner can be used to support the fabrication of covalently cross-linked and reactive polymer multilayers assembled using PVDMA and poly(ethyleneimine) (PEI). The growth, chemical reactivity, and uniformity of films deposited on these substrates …

Ocean Engineering and Marine Sciences Faculty Publications
The threespot damselfish, Stegastes planifrons (Cuvier), is important in mediating interactions among corals, algae, and herbivores on Caribbean coral reefs. The preferred microhabitat of S. planifrons is thickets of the branching staghorn coral Acropora cervicornis. Within the past few decades, mass mortality of A. cervicornis from white-band disease and other factors has rendered this coral a minor ecological component throughout most of its range. Methodology/Principal Findings:Survey data from Jamaica (heavily fished), Florida and the Bahamas (moderately fished), the Cayman Islands (lightly to moderately fished), and Belize (lightly fished) indicate that distributional patterns of S. planifrons are positively correlated with live …

A Comparison Of Probe-Level And Probeset Models For Small-Sample Gene Expression Data, John R. Stevens, Jason L. Bell, Kenneth I. Aston, Kenneth L. White
Mathematics and Statistics Faculty Publications
Background: Statistical methods to tentatively identify differentially expressed genes in microarray studies typically assume larger sample sizes than are practical or even possible in some settings.
Results: The performance of several probe-level and probeset models was assessed graphically and numerically using three spike-in datasets. Based on the Affymetrix GeneChip, a novel nested factorial model was developed and found to perform competitively on small-sample spike-in experiments.
Conclusions: Statistical methods with test statistics related to the estimated log fold change tend to be more consistent in their performance on small-sample gene expression data. For such small-sample experiments, the nested factorial model can …

This paper analyzes the economic and investment implications of a series of climate mitigation scenarios, characterized by different levels of ambition in terms of long term stabilization goals and the transition to attain them. In particular, the implications of fairly ambitious scenarios are investigated for the first time by means of the model WITCH. Although milder climate objectives can be achieved at moderate costs, our results show that stringent stabilization paths, compatible with the target of the European Union and the G8, might have important economic repercussions. The timing of mitigation action influences the cost of meeting a target as …

This paper presents results from recent work using structured light laser profile imaging to create high resolution bathymetric maps of underwater archaeological sites. Documenting the texture and structure of submerged sites is a difficult task and many applicable acoustic and photographic mapping techniques have recently emerged. This effort was completed to evaluate laser profile imaging in comparison to stereo imaging and high frequency multibeam mapping. A ROV mounted camera and inclined 532 nm sheet laser were used to create profiles of the bottom that were then merged into maps using platform navigation data. These initial results show very promising resolution …

A novel organic cation transporter (OCT) gene, OCT 6, and use thereof is described. The OCT6 gene is preferentially expressed in human hematopoietic tissues, including CD34+ cells and leukemia cells. Its narrow tissue distribution, substrate specificity, and close homology to other cell membrane transporters make OCT6 an attractive target for the treatment of myeloid diseases.
