Interactions Of Burkholderia Pseudomallei And Acanthamoeba Castellanii And Their Effects On Virulence In Human Monocytes, Emily Ann Moore Jul 2010

Interactions Of Burkholderia Pseudomallei And Acanthamoeba Castellanii And Their Effects On Virulence In Human Monocytes, Emily Ann Moore

Theses and Dissertations

Burkholderia pseudomallei, the etiological agent of melioidosis, is a saprophytic bacterium existing endemically in the water and soil of SE Asia and Northern Australia. This organism has shown the ability to remain dormant in its host for decades. B. thailandensis is a closely related non-pathogenic near neighbor that is also found in these soils. It has been suggested that free-living amoeba could be natural reservoirs for these organisms. The interactions of Burkholderia species and Acanthamoeba castellanii, a species of free-living amoeba, were studied to better understand the natural ecology of these organisms and to determine the effects amoeba …

Ribosomal Synthesis Of N-Methylated Peptides, Sara Ahadi Jul 2010

Ribosomal Synthesis Of N-Methylated Peptides, Sara Ahadi

Theses and Dissertations

Natural peptide products isolated from various organisms often contain N-methylated backbones. Such a modification of backbone of the peptide changes its conformational rigidity. This modification improves the biological properties of the peptide, such as improved target affinity, proteolytic stability or membrane permeability. Therefore synthesis of N-methylated peptide libraries is valuable in screening for drug-like peptides suitable for therapeutic uses. Protein synthesis using recombinant elements (PURE) and Flexizyme were used in order to reassign specific codons to N-methyl amino acids. mRNA-dependent translation system enable us to make our desired peptides with N-methyl amino acids. This technology is a convenient tool for …

Reproductive Senescence In The Florida Scrub-Jay (Aphelocoma Coerulescens), Travis Eli Wilcoxen Jul 2010

Reproductive Senescence In The Florida Scrub-Jay (Aphelocoma Coerulescens), Travis Eli Wilcoxen

Electronic Theses and Dissertations Archive

Analysis of twenty years of historical reproductive data from our study population of Florida Scrub-Jays reveals a quadratic relationship between breeder age and number of fledglings produced, with the youngest and oldest birds having the lowest reproductive success. Such a decrease in reproductive performance in older birds, despite the likely benefits of increased experience, is often attributed to senescence. My dissertation research considered reproductive endocrinology, stress physiology, immunology, egg hatchability, life history trade-offs, and parental behavior. The oldest and youngest females have greater rates of hatching failure than middle-aged females. Results also indicate that circulating levels of reproductive hormones decrease …

Evolutionary Relationships In Afro-Malagasy Schefflera (Araliaceae) Based On Nuclear And Plastid Markers, Morgan Gostel

Theses and Dissertations

The genus Schefflera is the largest in Araliaceae, with approximately 900 species. Recent studies have shown that Schefflera is polyphyletic and represents no fewer than five distinct clades, each corresponding to a specific geographic region including Asia, continental Africa and Madagascar, Melanesia, the Neotropics, and a small clade distributed throughout several islands in the insular Pacific Ocean. The Afro-Malagasy clade contains nearly 50 species distributed throughout tropical, sub-Saharan Africa, Madagascar, the Comoros, and the Seychelles islands. Previous studies have suggested that this group is monophyletic, identifying two smaller subclades within Afro-Malagasy Schefflera corresponding roughly to informal groups identified as “Meiopanax” …

Human Induced Rotation And Reorganization Of The Brain Of Domestic Dogs, Taryn Roberts, Paul Mcgreevy, Michael Valenzuela Jul 2010

Human Induced Rotation And Reorganization Of The Brain Of Domestic Dogs, Taryn Roberts, Paul Mcgreevy, Michael Valenzuela

Anatomy Collection

Domestic dogs exhibit an extraordinary degree of morphological diversity. Such breed-to-breed variability applies equally to the canine skull, however little is known about whether this translates to systematic differences in cerebral organization. By looking at the paramedian sagittal magnetic resonance image slice of canine brains across a range of animals with different skull shapes (N = 13), we found that the relative reduction in skull length compared to width (measured by Cephalic Index) was significantly correlated to a progressive ventral pitching of the primary longitudinal brain axis (r = 0.83), as well as with a ventral shift in the position …

Moral Identification: An Alternative Approach To Framing Second-Generation Immigrants' Ethnic Identity Ambivalence, Benjamin R. Brady Jul 2010

Moral Identification: An Alternative Approach To Framing Second-Generation Immigrants' Ethnic Identity Ambivalence, Benjamin R. Brady

Theses and Dissertations

Contemporary sociological research on second-generation immigrants living in the United States is lined with questions of ethnic inclusion and transnational participation. Many scholars are interested in how the children of immigrants relate to their parents' ethnic identity while being raised in a new land. Noting that the majority of scholars in this field approach ethnic identity within a social constructionist perspective, in this study I explore the ways that identity ambivalence and ethnic belonging are framed. Specifically, I critically question the ways that an ethnic identity is assumed to be valued and asserted in a constructionist model. After presenting a …

The Effects Of Hydrology And Vegetation On Microbial Community Structure And Soil Function In The Sediments Of Freshwater Wetlands, Christine Prasse Jul 2010

The Effects Of Hydrology And Vegetation On Microbial Community Structure And Soil Function In The Sediments Of Freshwater Wetlands, Christine Prasse

Theses and Dissertations

In wetland soils, hydrology is considered to be one of the primary factors shaping wetland function and microbial community structure, but plant-soil interactions are also important mechanisms affecting microbial nutrient transformations. The research presented here considered the interactive effect to describe how hydrology and the presence of plants alter the soil profile, the development of the bacterial community, and their associated functions. To achieve this goal, plots were established in three hydrologically-distinct regimes (Wet, Intermediate, and Dry) within a non-tidal freshwater wetland along the James River (Charles City County, Virginia). Inside each main plot, ten subplots were cleared of all …

The Relationship Among Maternal Infant Bonding, Spirituality, And Maternal Perception Of Childbirth Experience, Linda Bennington Jul 2010

The Relationship Among Maternal Infant Bonding, Spirituality, And Maternal Perception Of Childbirth Experience, Linda Bennington

Theses and Dissertations

The beginning of life is an intense experience for both mother and baby and sets the foundation for future interactions. Researchers have theorized that maternal infant bonding begins prenatally and continues on through the postnatal period. Mṻller (1996) examined that process to determine if prenatal bonding was related to postnatal bonding and discovered that there was only a modest correlation between the two. This led to speculation as to what variables, besides prenatal bonding, could influence postnatal bonding. Klaus & Kennell (1976) noted the detrimental effects of a lack of bonding in terms of abuse and attachment disorders and emphasized …

In Vitro Enabling Technologies For Use In The Aquatic Environment, Colm J. O'Dowd Jul 2010

In Vitro Enabling Technologies For Use In The Aquatic Environment, Colm J. O'Dowd

Doctoral

O’Dowd, C., Mothersill, C.E., Cairns, M.T., Austin, B., Lyng, F.M., McClean, B. and Murphy, J.E.J. Assessing the mitochondrion as a biomarker of fish tissue damage using g radiation as a stress model in vitro.

There is an ever-increasing need for biomarkers to identify toxic stress in the aquatic environment. Such techniques need to be accurate, expeditious, ethical and economical. Typically, in vitro based platforms fit these criteria however many of these systems often undergo ‘assay drift’ and consequently do not fully represent the real-life situation.

In recent years, there has been growing interest in the mitochondrion and its (dys)function …

Chemistry and Biochemistry Faculty Publications

A negative-ion proton-transfer chemical ionization mass spectrometric technique (NI-PT-CIMS), using acetate as the reagent ion, was applied to the measurement of volatile inorganic acids of atmospheric interest: hydrochloric (HCl), nitrous (HONO), nitric (HNO(3)), and isocyanic (HNCO) acids. Gas phase calibrations through the sampling inlet showed the method to be intrinsically sensitive (6-16 cts/pptv), but prone to inlet effects for HNO(3) and HCl. The ion chemistry was found to be insensitive to water vapor concentrations, in agreement with previous studies of carboxylic acids. The inlet equilibration times for HNCO and HONO were 2 to 4s, allowing for measurement in biomass burning …

Phylotree – A Toolkit For Computing Experiments With Distance-Based Methods For Genome Coevolution, Elissaveta Arnaoudova, Jerzy W. Jaromczyk, Neil Moore, Christopher L. Schardl, Ruriko Yoshida

Computer Science Presentations

We have developed software called Phylotree as a toolkit for running experiments to study gene cophylogenies for genome evolution using distance-based methods. In particular, the toolkit has been instrumental in conducting processing-heavy experiments with the new “difference of means” statistical method. Phylotree was used to run experiments using simulated data as well as biological sequences of well known host and parasite species, and is distributed with data and configuration files allowing these experiments to be reproduced.
